Effects of living and metabolically inactive mesenchymal stromal cells and their derivatives on monocytes and macrophages

Mesenchymal stromal cells (MScs) are multipotent and self-renewing stem cellsthat have great potential as cell therapy for autoimmune and inflammatorydisorders, as well as for other clinical conditions, due to their immunoregulatoryand regenerative properties. MScs modulate the inflammatory milieu by releasingsoluble factors and acting through cell-to-cell mechanisms. MScs switch theclassical inflammatory status of monocytes and macrophages towards a nonclassicaland anti-inflammatory phenotype. This is characterized by an increasedsecretion of anti-inflammatory cytokines, a decreased release of pro-inflammatorycytokines, and changes in the expression of cell membrane molecules and inmetabolic pathways. The MSc modulation of monocyte and macrophage phenotypesseems to be critical for therapy effectiveness in several disease models, sincewhen these cells are depleted, no immunoregulatory effects are observed. Here,we review the effects of living MScs (metabolically active cells) and metabolicallyinactive MScs (dead cells that lost metabolic activity by induced inactivation) andtheir derivatives (extracellular vesicles, soluble factors, extracts, and microparticles)on the profile of macrophages and monocytes and the implications forimmunoregulatory and reparative processes. This review includes mechanisms ofaction exhibited in these different therapeutic appro-aches, which induce the antiinflammatoryproperties of monocytes and macrophages. Finally, we overviewseveral possibilities of therapeutic applications of these cells and their derivatives,with results regarding monocytes and macrophages in animal model studies andsome clinical trials.

The growth and production modeling of individual trees of Eucalyptus urophylla plantations

Individual tree models(ITMs)are classified as growth and production models for projecting current and future forest *** are more complex than other growth and production models,show a higher level of detail and,consequently,produce a better modeling ***,the accuracy and efficiency of ITMs have not been properly assessed to *** this study,we estimated the growth in height,diameter,and individual tree volume of a Eucalyptus urophylla plantation by applying an *** used a continuing forest inventory dataset in which 1554 individual trees within 29 permanent plots were measured in the field over a 6-year period(24 to 72 months).Each individual tree volume was estimated for future tree *** achieve this,we adjusted the model to predict the height and diameter growth,and the probability of mortality as a function of the competition *** ITM accuracy was assessed based on the analysis of variance results and,subsequently,the multiple mean comparison test at the 5%significance *** tree volumes predicted by the ITM for the forest stand aged 72 months,beginning at ages 24,36,48,and 60 months,were compared to the field measured tree volume acquired from the 72-month forest inventory that was used as the reference *** and observed tree volumes were similar when the estimation was based on the 48-month forest *** results might help to reduce financial costs of forest inventory because the ITM produces accurate future predictions of forest stand *** estimated ITM for Eucalyptus plantations using measurement intervals up to 2 years is recommended because it significantly reduced the projected volume discrepancy compared to the field measurements.

Effects of cryofrequence on Localized Adiposity: case Study

cryofrequence is the combination of cryotherapy with radiofrequency, which promotes a “thermal shock” to the tissue, working simultaneously to skin flaccidity and localized adiposity. The efficacy of dermatological and hypodermic cryofrequency treatment was evaluated. This study reports the case of a 45-year-old woman with complaints of flaccidity and fat located in the infra-umbilical region in the preoperative period of abdominoplasty. The evaluation was performed before and after treatment through evaluation protocols, ultrasound examinations and histological findings through optic microscopy, microfocal and immunohistochemical analysis. The treatment was performed in 4 sessions, with a weekly application, using the BHS 156 FULL®—BODY HEALTH BRASIL®apparatus, the parameters were temperature of -10°c, power in 70%, duration of 8 minutes. After the surgical procedure, parts of treated and untreated abdominal tissues were removed for evaluation. In the tests performed, significant destruction of adipocytes was detected, with consequent reduction of fat and greater expression of caspase-3 in the treated tissue, being constated the macrophage increase by cD68 expression. There was improvement in flaccidity, evidenced by the significant increase in the number of fibroblasts and inflammatory cells positive for Ki67, bringing more quantity and better quality to the collagen.

Phase-sensitive optical time-domain reflectometry(cDOTDR)is a well-established technique that provides spatiotemporal measurements of an environmental variable in real *** unique capability is being leveraged in an everincreasing number of applications,from energy transportation or civil security to *** date,a wide number of different approaches have been implemented,providing a plethora of options in terms of performance(resolution,acquisition bandwidth,sensitivity or range).However,to achieve high spatial resolutions,detection bandwidths in the GHz range are typically required,substantially increasing the system cost and ***,we present a novel OOTDR approach that allows a customized time expansion of the received optical ***,the presented technique reaches cm-scale spatial resolutions over t km while requiring a remarkably low detection bandwidth in the MHz *** approach relies on the use of dual-comb spectrometry to interrogate the fibre and sample the backscattered *** phase-spectral coding is applied to the employed combs to maximize the signal-to-noise ratio of the sensing scheme.A comparison of the proposed method with alternative approaches aimed at similar operation features is provided,along with a thorough analysis of the new *** results demonstrate a radically novel high-resolution OOTDR scheme,which could promote new applications in metrology,borehole monitoring or aerospace.

目的评价颅底骨折患者预防性使用抗生素预防脑膜炎的效果。方法检索cochrane临床对照试验资料库（cENTRAL）（cochrane图书馆2005年第3期）、MEDLINE（1966～2005．09）、EMBASE（1974～2005．06）和LILAcS（1982～2005．09）。电子检索美国神经外科医师协会会议记录（1997～2005．09），手工检索神经外科学会欧洲联合会会议记录摘要（1995，1999，2003）。收集比较抗生素与安慰剂或空白对照的随机对照试验。同时对检出的非随机对照试验单独进行Meta分析以比较结果。至少由两名研究者独立评价每项试验的质量并提取数据。采用RevMan4．2软件进行Meta分析。结果纳入的5个随机对照试验和17个非随机对照试验比较了颅底骨折患者使用不同类型的预防性抗生素与安慰刹或空白对照的效果。但多数试验对方法学细节描述不充分。所有试验均将脑膜炎纳入其主要结局。共评价了符合Meta分析纳入标准的四个随机对照试验，共包括208例受试者。在降低脑膜炎的发生率、全因病死率、脑膜炎病死率和需要手术干预的脑脊液漏的发生率等方面，预防性抗生素组与对照组无显著差异。对伴或不伴脑脊液漏的患者进行亚组分析以评价其主要结局，并对所有检出的非随机对照试验（共包括2168例受试者）进行Meta分析，得出的结果与随机数据一致。结论现有的随机对照试验证据并不支持对颅底骨折患者预防性使用抗生素，无论其有无脑脊液漏的迹象。在更多的研究完成之前，尚无法确定颅底骨折患者使用抗生素的效果，因为迄今发表的研究均存在偏倚，尚需更多设计完善的大样本随机对照试验。

Antibacterial activity and inhibition against Staphylococcus aureus NorA efflux pump by ferulic acid and its esterified derivatives

Objective:To evaluate the inhibitory activity of ferulic acid and four of its esterified derivatives(methyl,ethyl,propyl,and butyl)against resistance mechanisms in Staphylococcus aureus ***:Ferulic acid derivatives were obtained by esterification with methanol,ethanol,propanol,and butanol,and then characterized by hydrogen and carbon-13 nuclear magnetic resonance *** minimum inhibitory concentrations(MIc)of ferulic acid and its esterified derivatives,ethidium bromide,and norfloxacin were obtained using the microdilution test,while the efflux pump inhibition test was conducted by examining reduction in the MIcs of norfloxacin and ethidium *** docking was also carried out using the Schrodinger Suite 2015 molecular modeling software.A three-dimensional model of NorA efflux pump was generated using *** best scoring model was used as a receptor for ligand-receptor ***:The methyl and butyl ester derivatives did not demonstrate significant antimicrobial ***,a significant synergic effect was evidenced when norfloxacin was combined with the ethyl and propyl esterified *** docking study demonstrated favorable energy of interaction between ferulate derivatives and NorA,and amino acid residues TYR57,TYR58,and LEU255 were present commonly in stabilizing all *** PcA analysis corroborated the docking hypothesis that the lipophilic character and hydrogen bond interactions were the most relevant characteristics involved with NorA *** pharmacokinetic parameters of ferulic acid derivatives showed good ADMET properties,demonstrating that they can be easily absorbed and have no effect or inhibit the cytochrome P450 enzyme complex,revealing their potential as drug ***:This study provides strong evidence that the molecular basis for this activity is potentially due to the NorA efflux pump.

Raman Spectra of Nd:La3Ga5SiO14 and Nd:La3Ga5.5Ta0.5O14 crystals

Single crystals of Nd：La3Ga5SiO14 （NLGS） and Nd：La3Ga5.5Ta0.5O14 （NLGT） were grown by czochralski method. Both crystals exhibit a langasite structure （space group D32 （P321） with Z = 1）, whose general formula is A3Bc2D3O14, where A, B, c and D represent the dodecahedral, octahedral and tetrahedral positions, respectively. The adding of low content of Nd does not change the crystal structure. Although they are isostructural, the distribution of atoms among the symmetric sites is different. While NLGT exhibits a perfect orderly structure, NLGS exhibits an intrinsically disordered structure due to the random distribution of Ga and Si atoms at the c3（2） symmetry sites. To understand what changes occur in the phonon distribution when Ta replaces Si, we recorded and analyzed room temperature polarized Raman spectra. We observe 8A1＋18E modes for NLGS and 10A1＋13E for NLGT crystals. When Si is substituted by Ta, we observe that the wavenumber of some modes related to vibrations involving Ga and O atoms decreases. This agrees with the fact that the Ga–O bonds are shorter in NLGS than in NLGT.

Left-Handed Students and clinical Practice in Dentistry: Adaptations, Difficulties and Realities Experienced in the Academic Environment

Objectives: To verify the existence of difficulties throughout the course of dentistry clinical practices, identify musculoskeletal pain, and evaluate frequency and intensity of pain/discomfort (if existent) in different regions of the body, experienced by left- handed students in comparison to right-handed students of the odontology courses of the state of Rio Grande do Norte (Northeast Brazil). Methods: A general questionnaire was applied to identify left-handed students in a population of 750 students, enrolled in clinical courses. An adaptation of the Grad-corllet Diagram and a specific questionnaire on clinical practices were applied to the selected students. Descriptive statistics was utilized to analyze data of the specific questionnaire, and Mann-Whitney statistics, chi-square or Fisher’s Exact tests (with a 5% significance level) were utilized for the Grad-corllet Diagram, searching for an association be- tween discomfort/pain in different body regions and manual laterality. Results: All students presented higher percentage of pain in the lumbar and neck/cervical re- gions. However, left-handed students presented higher values, with statistical signifi- cance levels of p = 0.041 for lower back, p = 0.002 for neck and p = 0.003 for cervical region. conclusions: The majority of left-handed students classified “moderate” the difficulty of working with equipment designed for right-handers. There was high frequency of musculoskeletal pain/discomfort, with relevant statistical results for left- handed students regarding intensity. It is suggested that the participating institutions evaluate the adoption of inclusion politics, re-adapt the infrastructure to include equipment designed for left-handers, and finally provide adequate work conditions for all students.

The interaction of ultra-intense high-power lasers with solid-state targets has been largely studied for the past 20 years as a future compact proton and ion ***,the huge potential established on the target surface by the escaping electrons provides accelerating gradients of TV/*** process,called target normal sheath acceleration,involves a large number of phenomena and is very difficult to study because of the picosecond scale *** the SPARc LAB Test Facility,the high-power laser FLAME is employed in experiments with solid targets,aiming to study possible correlations between ballistic fast electrons and accelerated *** detail,we have installed in the interaction chamber two different diagnostics,each one devoted to characterizing one *** first relies on electro-optic sampling,and it has been adopted to completely characterize the ultrafast electron *** the other hand,a time-of-flight detector,based on chemical-vapour-deposited diamond,has allowed us to retrieve the proton energy *** this work,we report preliminary studies about simultaneous temporal resolved measurements of both the first forerunner escaping electrons and the accelerated protons for different laser parameters.

Broadband Optical Active Waveguides Written by Femtosecond Laser Pulses in Lithium Fluoride

Broadband waveguiding through light-emitting strips directly written in a blank lithium fluoride crystal with a femtosecond laser is reported. Light guiding was observed at several optical wavelengths, from blue, 458 nm, to near-infrared, at 155nm. Visible photoluminescence spectra of the optically active F2 and F33 color centers produced by the fs laser writing process were measured. The wavelength-dependent refractive index increase was estimated to be in the order of 10-3-10-4 in the visible and near-infrared spectral intervals, which is consistent with the stable formation of point defects in LiF.
