BACKGROUND Acute decompensation(AD)of cirrhosis is associated with high short-term mortality,mainly due to the development of acute-on-chronic liver failure(ACLF).Thus,there is a need for biomarkers for early and accurate identification of AD patients with high risk of development of ACLF and *** triggering receptor expressed on myeloid cells-1(sTREM-1)is released from activated innate immune cells and correlated with various inflammatory *** To explore the prognostic value of sTREM-1 in patients with AD of *** A multicenter prospective cohort of 442 patients with cirrhosis hospitalized for AD was divided into a study cohort(n=309)and validation cohort(n=133).Demographic and clinical data were collected,and serum sTREM-1 was measured at *** enrolled patients were followed-up for at least 1 *** In patients with AD and cirrhosis,serum sTREM-1 was an independent prognosis predictor for 1-year survival and correlated with liver,coagulation,cerebral and kidney failure.A new prognostic model of AD(P-AD)incorporating sTREM-1,blood urea nitrogen(BUN),total bilirubin(TBil),international normalized ratio(INR)and hepatic encephalopathy grades was established and performed better than the model for end-stage liver disease(MELD),MELD-sodium(MELD-Na),chronic liver failure-consortium(CLIF-C)ACLF and CLIF-C AD ***,sTREM-1 was increased in ACLF and predicted the development of ACLF during first 28-d *** ACLF risk score incorporating serum sTREM-1,BUN,INR,TBil and aspartate aminotransferase levels was established and significantly superior to MELD,MELD-Na,CLIF-C ACLF,CLIF-C AD and P-AD in predicting risk of ACLF *** Serum sTREM-1 is a promising prognostic biomarker for ACLF development and mortality in patients with AD of cirrhosis.

Single-cell profiling of the pig cecum at various developmental stages

The gastrointestinal tract is essential for food digestion,nutrient absorption,waste elimination,and microbial ***-cell transcriptome profiling of the intestinal tract has greatly enriched our understanding of cellular diversity,functional heterogeneity,and their importance in intestinal tract development and *** such profiling has been extensively conducted in humans and mice,the single-cell gene expression landscape of the pig cecum remains ***,single-cell RNA sequencing was performed on 45572 cells obtained from seven cecal samples in pigs at four different developmental stages(days(D)30,42,150,and 730).Analysis revealed 12 major cell types and 38 subtypes,as well as their distinctive genes,transcription factors,and regulons,many of which were conserved in *** increase in the relative proportions of CD8^(+)T and Granzyme A(low expression)natural killer T cells(GZMA^(low)NKT)cells and a decrease in the relative proportions of epithelial stem cells,Tregs,RHEX^(+)T cells,and plasmacytoid dendritic cells(pDCs)were noted across the developmental ***,the post-weaning period exhibited an up-regulation in mitochondrial genes,COX2 and ND2,as well as genes involved in immune activation in multiple cell ***-cell crosstalk analysis indicated that IBP6^(+)fibroblasts were the main signal senders at D30,whereas IBP6^(−)fibroblasts assumed this role at the other *** cells established interactions with epithelial cells and IBP6^(+)fibroblasts in the D730 cecum through mediation of GZMA-F2RL1/F2RL2 *** study provides valuable insights into cellular heterogeneity and function in the pig cecum at different development stages.

To elucidate the effect of Nano-Se and GSH(SeG) exogenous bio-enhancement on cucumber's resistance to Botrytis cinerea(***) infection,we conducted an in-depth study to explore its mec^*** analysis encompassed transcriptome and metabolome analysis,detection of antioxidant enzyme activity,staining studies,and illness index *** results indicate that leaf application of SeG significantly enhances cucumber growth,increases photosynthetic pigment levels and capacity,and reduces the incidence rate by 41.9% after *** linolenic acid metabolicpathway triggered by SeG activates genes associated with Jasmonic acid(JA),including LOX(210%),LOX4(430%),AOS1(100%),and AOC2(120%).With strengthened JA signal transduction,the expression of cucurbitacin synthesis-related genes OSC(107.5%),P450(440.8%,31.6%),and ACT(414.0%) is activated,thereby promoting the accumulation of cucurbitacin.L-phenylalanine(67.5%),P-coumaric acid(144.8%),sinapic acid(49.0%),and ferulic acid(8.4%) were among the other lignin precursors *** activities of APX(81.7%),MDHAR(43.4%),and GSH-Px(60.5%),essential antioxidant enzymes in the ASA-GSH cycle,display varying degrees of *** summary,SeG bio-enhancement activates the AsA-GSH cycle,boosts the activity of antioxidant enzymes,enhances JA signal transduction,facilitates the accumulation of cucurbit and lignin precursor compounds,thereby enhancing cucumber's resistance to ***.

目的探讨老年乳腺癌患者术前衰弱与术后短期并发症的关系。方法采用便利抽样的方法,选取2022年7月—12月在北京市某三级甲等医院行择期手术的231例老年乳腺癌患者作为调查对象。术前1 d收集患者的一般资料并对患者进行衰弱评估,于术后30 d追踪患者的并发症发生情况。结果214例老年乳腺癌患者完成随访,37例术后30 d内发生并发症。Logistic回归分析显示,术前衰弱的老年乳腺癌患者术后30 d内并发症发生风险是非衰弱患者的5.532倍(P<0.05)。存在自由活动下降、阻力增加/耐力减退和体重下降问题的老年乳腺癌患者术后并发症的发生风险较高。结论术前衰弱增加老年乳腺癌患者术后30 d内并发症的发生风险,提示应对术前衰弱的老年乳腺癌患者加强围手术期健康管理,积极预防衰弱的发生和发展。

西双版纳黄瓜是我国特有的果肉橙黄色的黄瓜变种资源,不同种质间的β-胡萝卜素含量差异明显。PSY是胡萝卜素生物合成途径中的第1个限速酶。本文以西双版纳黄瓜为试材,分别克隆西双版纳黄瓜八氢番茄红素合成酶(Cs-PSY1)的DNA和c DNA序列,结果显示,DNA长2797 bp,包含5个内含子和6个外显子,c DNA序列长1385 bp,编码421个氨基酸。Psy1推测的氨基酸序列包含该家族的2个特征序列,保守性很高。该蛋白为不稳定蛋白,无明显疏水区,未预测到跨膜结构;系统进化分析结果显示,西双版纳黄瓜的Cs-PSY1蛋白与甜瓜的同源性较高;与栽培黄瓜深度测序材料"9930"和"GY14"的序列进行比较分析,结合115份黄瓜重测序结果,共发现5个SNP,其中2个位于起始密码子上游27 bp处和971 bp处,3个位于内含子区域。其中SNP4在重测序的19份西双版纳黄瓜中的突变率为100%,在96份栽培黄瓜中的特异性为5.3%。转录因子结合位点预测结果显示,在普通栽培黄瓜该位点处存在一个CTAG motif,在西双版纳黄瓜中该位点突变后则不存在该motif。利用实时荧光定量PCR技术分析Cs-Psy1的表达量变化趋势,结果表明,在黄瓜不同果实发育时期,该基因的表达量均呈现先上升后下降的趋势,在西双版纳黄瓜中表达量变化的差异明显,在授粉后50 d达到最大值,是果实发育初期表达量的8倍多,是同时期普通黄瓜的4倍多,而普通黄瓜表达量的总体变化相对平缓。西双版纳黄瓜果实内果皮的表达水平明显高于中果皮,最高相差约5倍,普通黄瓜差异不明显。从上述研究结果推测Psy1基因可能影响西双版纳黄瓜的β-胡萝卜素积累。

Influence of temperature, stress, and grain size on behavior of nano-polycrystalline niobium

Atomic simulations are executed to investigate the creep responses of nano-polycrystalline(NC) niobium established by using the Voronoi algorithm. The effects of varying temperature, applied stress, and grain size(GS) on creep properties and mechanisms are investigated. Notably, the occurrence of tertiary creep is exclusively observed under conditions where the applied stress exceeds 4.5 GPa and the temperature is higher than 1100 K. This phenomenon can be attributed to the significant acceleration of grain boundary and lattice diffusion, driven by the elevated temperature and stress levels. It is found that the strain rate increases with both temperature and stress increasing. However, an interesting trend is observed in which the strain rate decreases as the grain size increases. The stress and temperature are crucial parameters governing the creep behavior. As these factors intensify, the creep mechanism undergoes a sequential transformation: initially from lattice diffusion under low stress and temperature conditions to a mixed mode combining grain boundaries(GBs) and lattice diffusion at moderate stress and mid temperature levels, and ultimately leading to the failure of power-law controlled creep behavior, inclusive of grain boundary recrystallization under high stress and temperature conditions. This comprehensive analysis provides in more detail an understanding of the intricate creep behavior of nano-polycrystalline niobium and its dependence on various physical parameters.
