BACKGROUND Studies have reported that microRNA-30c(miR-30c)has vital functions in the development and progression of multiple *** To investigate the clinical significance and role of miR-30c in pancreatic *** MiR-30c and twinfilin 1(TWF1)expression levels were analyzed in Gene Expression Omnibus datasets and validated in human pancreatic cancer by quantitative real-time polymerase chain reaction(RT-qPCR).The effects of miR-30c on pancreatic cancer cell growth,apoptosis,and cell cycle were evaluated by CCK-8 and flow cytometry ***,the in vivo effects were investigated using a subcutaneous xenograft *** gene prediction software and luciferase reporter assays were used to identify TWF1 as a direct target of *** The expression of miR-30c was significantly decreased in pancreatic cancer tissues and associated with ***-and loss-of-function assays showed that miR-30c suppressed pancreatic cancer cell proliferation in vitro and in ***-qPCR,Western blot,and luciferase reporter assays showed that miR-30c directly targeted *** expression level of miR-30c was negatively correlated with TWF1 expression in pancreatic cancer ***,the effects of ectopic miR-30c were rescued by TWF1 *** Our results identified the role of the miR-30c/TWF1 axis in pancreatic cancer progression and demonstrated that miR-30c might serve as a prognostic biomarker and therapeutic target for pancreatic cancer.

HEF-19-induced relaxation of colonic smooth muscles and the underlying mechanisms

The functional diversity index of a phytoplankton body has gradually become a new mean of measuring and *** explore the response of phytoplankton taxonomy and the functional diversity index to interannual environmental changes,a survey on the structure of the phytoplankton community and water physicochemical characteristics of the water was carried out at 28 sampling points in the Harbin section of the Songhua River for three consecutive years in every May from 2021 to *** taxonomy diversity index and the functional diversity index were ***,The relationship between the structure of the characteristics of the community and environmental factors was explored;secondly,we reveal the responses of taxonomic and functional diversity indices to different habitats between years;finally,we explore the main environmental factors that control the taxonomic and functional diversity indices of *** show that,initially,environmental factors in water changes caused by different water levels between years affected the composition of phytoplankton ***,by comparing the taxonomic diversity index and the functional diversity index on a time scale,we found that the taxonomic diversity index was more responsive to environmental ***,the main environmental factors on the phytoplankton taxonomic diversity index were dissolved oxygen,Taxonomic diversity index,and specific conductivity,and the main environmental factors that affected the functional diversity index were dissolved oxygen,turbidity,and water *** study reveals the important role of the taxonomic diversity index in river water quality evaluation,obtained new information on the relative precision of the taxonomic diversity index and the functional diversity index in the evaluation of ecological health of the water,and provided a reliable tool for monitoring river water quality based on aquatic organisms.

Limited Role of Promoter Methylation of MGMT and C130RF18 in Triage of Low-Grade Squamous Intraepithelial Lesion

Background：Promoter methylation of MGMT and C13ORF18 has been confirmed as a potential biomarker for early diagnosis of cervical cancer. The aim of this study was to evaluate the performance of MGMT and C13ORF18 promoter methylation for triage of cytology screening samples and explore the potential ***：Methylation-sensitive high-resolution melting was used to detect promoter methylation of MGMT and C13ORF18 in 124 cervical samples. High-risk human papillomavirus （HR-HPV） was detected by the Digene Hybrid Capture 2？. Gene methylation frequencies in relation to cervical intraepithelial neoplasia （CIN） were analyzed. Frequencies were compared by Chi-square tests. The expression of gene biomarkers and methylation regulators was analyzed by immunohistochemical staining, real-time fluorescence quantitative polymerase chain reaction, and Western ***：For triage of low-grade squamous intraepithelial lesion （LSIL）, gene methylation increased specificity from 4.0% of HR-HPV detection to 30.8% of MGMT （χ2 = 9.873, P = 0.002） and to 50.0% of C13ORF18 （χ2 = 21.814, P = 0.001）. For triage of atypical squamous cells of undetermined significance, HR-HPV detection had higher positive predictive value of 54.8%. Either MGMT or C13ORF18 methylation combined with HR-HPV increased the negative predictive value to 100.0% （χ2 = 9.757, P = 0.002）. There was no relationship between MGMT and C13ORF18 expression and DNA methylation （χ2 = 0.776, P = 0.379 and χ2 = 1.411, P = 0.235, respectively）. MBD2 protein level in cervical cancer was relatively lower than normal cervical tissue （t = 4.11, P = 0.006）.Conclusions：HR-HPV detection is the cornerstone for triage setting of CIN. Promoter methylation of MGMT and C13ORF18 plays a limited role in triage of LSIL. Promoter methylation of both genes may not be the causes of gene silence.

Relationship between Modulator Recognition Factor 2/AT-rich Interaction Domain 5B Gene Variations and Type 2 Diabetes Mellitus or Lipid Metabolism in a Northern Chinese Population

Background： Four single nucleotide polymorphisms （SNPs） in the modulator recognition factor 2/AT-rich interaction domain 5B （MRF2/ARID5B） gene located at chromosome 10q21,2 have been shown to be associated with both type 2 diabetes mellitus （T2DM） and coronary artery disease in a Japanese cohort. This study aimed to investigate the relationship between these SNPs （rs2893880, rs10740055, rs7087507, rs10761600） and new-onset T2DM and lipid metabolism in a Northern Chinese population. Methods： This was a case-control study. The rs2893880, rs10740055, rs7087507, and rs10761600 genetic variants were genotyped by SNPscan and analyzed in relation to T2DM susceptibility in 2000 individuals （090 with newly diagnosed T2DM and 1001 controls without diabetes mellitus）. Associations between the MRF2/ARID5B genetic models and T2DM were determined by multivariate logistic regression. Results： Regarding the rs 10740055 SNP, AA was associated with a higher risk of T2DM compared with codominant-type CC （adjusted by sex, age, and body mass index [BMI], P = 0.041, odds ratio [OR] = 1.421, 95% confidence interval [C1 ] 1.014-1.991）. Meanwhile, AA individuals were at increased risk of presenting with T2DM compared with individuals with CC or a single C （adjusted by sex, age, and BMI, P = 0.034, OR = 1.366, 95% CI 1.023-1.824）. With respect to rsl0761600, AT contributed to a higher risk of T2DM compared with AA （adjusted by sex, age, and BMI, P = 0.013, OR = 1.585, 95% CI 1.101-2.282）, while TT also increased the risk of presenting with T2DM compared with AA or A （adjusted by sex, age, and BMI, P = 0.004, OR = 1.632, 95% CI 1.166-2.284）. High-density lipoprotein cholesterol （HDL-C） levels were significantly different among the three genotypes of rs7087507 in the controls （P = 0.048）（GG〉GA）. Conclusions： The present results identified MRF2/AR1D5B as a potential susceptibility gene for new-onset T2DM in a Northern Chinese population, while the rs7087507 SNP was associated with HDL-C levels

Affection of Single-Nucleotide Polymorphisms in miR-27a, miR-124a, and miR-146a on Susceptibility to Type 2 Diabetes Mellitus in Chinese Han People

Background：Polymorphisms of microRNA （miRNA）,as a novel mechanism,are closely associated with disease states by interfering with miRNA *** correlations have been identified between single-nucleotide polymorphisms （SNPs） in miRNA,but the effect on type 2 diabetes mellitus （T2DM） onset among Chinese population remains ***,the aim of this study was to identify correlations between common SNPs in miR-27a,miR-146a,and miR-124a with T2DM among a Chinese population,as well as to explore diabetic pathological mechanisms and the impact of environmental ***：SNPscan technology was used to genotype 995 patients newly diagnosed with T2DM and 967 *** regression analysis was performed to compare mutation frequencies between cases and ***：We found no significant correlations between all genotypes of these miRNAs and T2DM in our ***,stratification analysis identified a lower risk of T2DM associated with the rs531564GC genotype among younger subjects （age ＜ 45 years）（adjusted P =0.043; odds ratio [OR] =0.73; 95％ confidence interval [CI] =0.54-0.99）.Furthermore,the rs895819CC genotype in overweight people （24 ＜ body mass index [BMI] ＜ 28） was significantly associated with an increased risk of T2DM （adjusted P =0.042; OR =1.73; 95％ CI =1.02-2.94）,while the rs2910164 genotype in miR-146a was not significantly correlated with *** genetic risk score was calculated based on the number of risk alleles of the three SNPs and was found to be correlated to total cholesterol （adjusted P =0.021）.Conclusions：The rs531564GC genotype acted as a protective factor to decrease the risk of T2DM in younger subjects （age ＜ 45 years）,while the presence of the rs895819CC genotype increased the risk of illness among overweight subjects （24 ＜ BMI ＜ 28 kg/m2）.The presence of SNPs in miRNA might promote disease by affecting miRNA expression and gene ***,miRNA mimics or inhibitors that directly regulate miRNA expression

The increase in frequency and intensity of Extreme High-temperature Events (EHEs) over Central-Eastern China (CEC) in recent decades has severely impacted social development and livelihoods. Using observation and reanalysis datasets, this study explores the effect of the East Asian subtropical westerly jet stream (EAJ) on the CEC EHEs for the summers spanning 1979-2020. Considering its general relative location to the right side of the upper-level jet stream exit region, CEC would theoretically suffer more EHEs with a stronger and northwardly-shifted EAJ in summer due to the likelihood of abnormal subsidence induced by the EAJ. However, such an EAJ-EHE connection has been unstable over the past four decades but has displayed an evident interdecadal change. Before the late 1990s, the interannual variation of the EAJ was manifested mainly by its meridional displacement in the northeastern part of East Asia;thus, the atmospheric responses were essentially located to the east of CEC, exerting less of an influence on the CEC EHEs. However, since the late 1990s, the EAJ variation has featured an intensity change in its center over the northwest portion of the CEC, which has resulted in a westward shift in atmospheric responses to cover the CEC region. Therefore, the EAJ could potentially affect the summer CEC EHEs during 2000-21. Our findings offer support for an in-depth understanding of the formation mechanisms of extreme weather/climate events of this nature and thus provide a scientific reference for seasonal climate predictions.
