Lacticaseibacillus rhamnosus Fmb14 ameliorates hyperuricemia-induced hepatocyte pyroptosis via NLRP3 inflammasome cascade inhibition

Hyperuricemia is a high-risk factor for the development of gout and renal fibrosis,but the adverse effects of hyperuricemia on the liver have been seriously *** research investigated the ameliorating effect of Lacticaseibacillus rhamnosus Fmb14 on hyperuricemia induced liver dysfunction both in vitro and in *** free extracts of high dose *** Fmb14 treatment reduced the death rate of HepG2 cell lines from 24.1%to 14.9%by inhibiting NLRP3 recruitment,which was mainly activated by reactive oxygen species release and mitochondrial membrane potential *** purine dietary induced hyperuricemia(PDIH)mice model,liver oedema and pyroptosis were ameliorated after *** Fmb14 administration through downregulating the expression levels of NLRP3,caspase-1 and gasdermin-D from 1.61 to 0.86,3.15 to 1.01 and 5.63 to 2.02,*** Fmb14 administration restored mitochondrial inner membrane protein(MPV17)and connexin 43 from 2.83 and 0.73 to 0.80 and 0.98 respectively in PDIH mice,indicating that dysbiosis of mitochondrial membrane potential was restored in ***,PDIH pyroptosis stimulates the process of apoptosis,which leads to severe leakage of hepatocytes,and both of pyroptosis and apoptosis were decreased after *** Fmb14 ***,*** Fmb14 is a promising biological resource to maintain homeostasis of the liver in hyperuricemia and the prevention of subsequent complications.

Experimental study of the damage characteristics of rocks containing non-penetrating cracks under cyclic loading

The damage evolution process of non-penetrating cracks often causes some unexpected engineering *** specimens containing non-penetrating crack(s)are used to study the damage evolution and characteristics under cyclic *** results show that under cyclic loading,the relationship between the number of non-penetrating crack(s)and the characteristic parameters(cyclic number,peak stress,peak strain,failure stress,and failure strain)of the pre-cracked specimens can be represented by a decreasing linear *** damage evolution equation is fitted by calibrating the accumulative plastic strain for each cycle,and the damage constitutive equation is proposed by the concept of effective ***,non-penetrating cracks are more likely to cause uneven stress distribution,damage accumulation,and local failure of *** local failure can change the stress distribution and relieve the inhibition of non-penetrating crack extension and eventually cause a dramatic destruction of the ***,the evolution process caused by non-penetrating cracks can be regarded as one of the important reasons for inducing *** results are expected to improve the understanding of the process of spalling formation and rockburst and can be used to analyze the stability of rocks or rock structures.

A New Method for the Aging Evaluation of Oil-Paper Insulation Using η-Butanol and Methanol

To improve the accuracy and efficiency of the aging life prediction and assessment of transformer oil-paper insulation,and to make up for the deficiencies of traditional characterizers,such as 2-furfural,carbon monoxide,and carbon dioxide,a method for the simultaneous determination of methanol,ethanol,n−propanol,and η-butanol in oil with a single injection is established by headspace-gas chromatography-mass *** measured results show that the determination limits of the four alcohol characterizers can be controlled to 10μg/kg *** on this method,the change patterns of the above four alcohols with thermal aging time and degree of polymerization are obtained through thermal aging experimental ***,η-propanol,and η-butanol in oil indicate nearly linear correlations with thermal aging time and degree of polymerization,similar to that of *** analyzing 52 sets of measured data of 500 kV EHV transformers in operation,η-butanol is found to have excellent performance,and a new method to evaluate the aging state of oil-paper insulation employing η-butanol and methanol is proposed along with the aging attention value *** measured data of 500 kV EHV transformers in operation indicate that the combination of η-butanol and methanol as the preferred characterizers can effectively compensate for the shortcoming of traditional characterizers in the early stages of aging,and the feasibility of the method is *** possible pathways for the generation of η-butanol by cellulose cleavage during the aging of oil-paper are proposed from the chemical structure of cellulose.

The similarities and differences in inherent mechanism and characteristic frequency between the onedimensional(1D) poroelastic model and the layered White model were investigated. This investigation was conducted under the assumption that the rock was homogenous and isotropic at the mesoscopic scale. For the inherent mechanism, both models resulted from quasi-static flow in a slow P-wave diffusion mode, and the differences between them originated from saturated fluids and boundary conditions. On the other hand, for the characteristic frequencies of the models, the characteristic frequency of the 1D poroelastic model was first modified because the elastic constant and formula for calculating it were misused and then compared to that of the layered White model. Both of them moved towards higher frequencies with increasing permeability and decreasing viscosity and diffusion length. The differences between them were due to the diffusion length. The diffusion length for the 1D poroelastic model was determined by the sample length, whereas that for the layered White model was determined by the length of the representative elementary volume(REV). Subsequently, a numerical example was presented to demonstrate the similarities and differences between the models. Finally, published experimental data were interpreted using the 1D poroelastic model combined with the Cole-Cole *** prediction of the combined model was in good agreement with the experimental data, thereby validating the effectiveness of the 1D poroelastic model. Furthermore, the modified characteristic frequency in our study was much closer to the experimental data than the previous prediction, validating the effectiveness of our modification of the characteristic frequency of the 1D poroelastic model. Objective:To assess the analgesic and anti-inflammatory effects of standardized extract of Cotinus coggygria(***)in different animal ***:*** extracts(25,50,and 100 mg/kg)were administered to rats and mice(n=6)during hot plate,tail-flick,acetic acid-induced writhing,and formalin tests to determine its analgesic *** anti-inflammatory activity of *** extracts was evaluated by histamine and carrageenan-induced paw edema,cotton pellet-induced granuloma,and acetic acid-induced peritoneal capillary dye leakage ***:*** extracts(50 and 100 mg/kg)significantly alleviated thermal and chemical-induced pain in rodents(P<0.05).It also demonstrated notable anti-inflammatory properties by mitigating histamine and carrageenan-induced paw edema,granuloma deposits,and vascular permeability(P<0.05).Moreover,*** extracts remarkably reduced TNF-α,IL-1β,IL-6,COX-2,and oxidative stress in rat paws(P<0.05).Carrageenan-induced histological aberrations in hind paw tissues were effectively(P<0.05)mitigated by treatment with *** ***:*** *** show analgesic and anti-inflammatory effects via inhibition of COX-2 and inflammatory and oxidative mediators.

Synthesis of granulated Li_(1.33)Mn_(1.67)O_(4) via two antisolvent methods for lithium adsorption from gas-produced water

Gas-produced water is an accompanying wastewater in the natural gas extraction process,and it is a potential liquid lithium resource that contains a considerable amount of *** study investigated the feasibility of using manganese-based ion sieves to adsorb and extract lithium from gas-produced *** we focused on the applicability of two different granulation methods,extrusion and droplet,in gas-produced water *** types of H_(1.33)Mn_(1.67)O_(4) particles were prepared by the extrusion method(EHMO)and the droplet method(DHMO).The porosity of DHMO was much higher than that of EHMO,and the adsorption performance of DHMO increased with the decrease of binder *** prepared with a binder concentration of 0.14 g·ml^(-1)exhibited the best adsorption performance in gas-produced water,and the Li^(+)adsorption capacity could reach 25.14 mg·g^(-1).In gas-produced water,the adsorption equilibrium of DHMO only took 9 h,and the adsorption process conformed to the Langmuir model and pseudo-second-order kinetic *** pore diffusion model(PDM)could well describe its adsorption ***,DHMO showed a great selectivity to Li^(+),and the selectivity order of DHMO in gas-produced water was Li^(+)>Ba^(2+)>>Mg^(2+),Ca^(2+),Sr^(2+)>>Na^(+)>>K^(+).After 20 cycles,the Li^(+)adsorption capacity was still higher than 17.30 mg·g^(-1),and the rate of manganese dissolution was less than 1%.

The RNA m^(6)A demethylase ALKBH5 drives emergency granulopoiesis and neutrophil mobilization by upregulating G-CSFR expression

Emergency granulopoiesis and neutrophil mobilization that can be triggered by granulocyte colony-stimulating factor(G-CSF)through its receptor G-CSFR are essential for antibacterial innate ***,the epigenetic modifiers crucial for intrinsically regulating G-CSFR expression and the antibacterial response of neutrophils remain largely unclear.N6-methyladenosine(m^(6)A)RNA modification and the related demethylase alkB homolog 5(ALKBH5)are key epigenetic regulators of immunity and inflammation,but their roles in neutrophil production and mobilization are still *** used cecal ligation and puncture(CLP)-induced polymicrobial sepsis to model systemic bacterial infection,and we report that ALKBH5 is required for emergency granulopoiesis and neutrophil ***5 depletion significantly impaired the production of immature neutrophils in the bone marrow of septic *** addition,Alkbh5-deficient septic mice exhibited higher retention of mature neutrophils in the bone marrow and defective neutrophil release into the circulation,which led to fewer neutrophils at the infection site than in their wild-type *** bacterial infection,ALKBH5 imprinted production-and mobilization-promoting transcriptome signatures in both mouse and human ***,ALKBH5 erased m^(6)A methylation on the CSF3R mRNA to increase the mRNA stability and protein expression of G-CSFR,consequently upregulating cell surface G-CSFR expression and downstream STAT3 signaling in *** RIP-qPCR results confirmed the direct binding of ALKBH5 to the CSF3R mRNA,and the binding strength declined upon bacterial infection,accounting for the decrease in G-CSFR expression on bacteria-infected *** these results collectively,we define a new role of ALKBH5 in intrinsically driving neutrophil production and mobilization through m^(6)A demethylation-dependent posttranscriptional regulation,indicating that m^(6)A RNA modification in neutrop
