Application of multidisciplinary in situ simulation training in the treatment of acute ischemic stroke: a quality improvement project

BACKGROUND:Ischemic stroke refers to a disorder in the blood supply to a local area of brain tissue for various reasons and is characterized by high morbidity,mortality,and *** reperfusion of brain tissue at risk of injury is crucial for the treatment of acute ischemic *** purpose of this study was to evaluate comfort levels in managing acute stroke patients with hypoxemia who required endotracheal intubation after multidisciplinary in situ simulation training and to shorten the door-to-image ***:This quality improvement project utilized a comprehensive multidisciplinary in situ simulation exercise.A total of 53 participants completed the two-day in situ simulation *** main outcome was the self-reported comfort levels of participants in managing acute stroke patients with hypoxemia requiring endotracheal intubation before and after simulation training.A 5-point Likert scale was used to measure participant comfort.A paired-sample t-test was used to compare the mean self-reported comfort scores of participants,as well as the endotracheal intubation time and door-to-image time on the fi rst and second days of in situ simulation *** door-to-image time before and after the training was also ***:The findings indicated that in situ simulation training could enhance participant comfort when managing acute stroke patients with hypoxemia who required endotracheal intubation and shorten door-to-image *** the emergency management of hypoxemia or tracheal intubation,the mean post-training self-reported comfort score was signifi cantly higher than the mean pre-training comfort score(hypoxemia:4.53±0.64 vs.3.62±0.69,t=-11.046,P<0.001;tracheal intubation:3.98±0.72 vs.3.43±0.72,t=-6.940,P<0.001).We also observed a decrease in the tracheal intubation and door-to-image time and a decreasing trend in the door-to-image time,which continued after the ***:Our study demonstrates that the implementation of in s

Amultifunctional liposomal polydopamine nanoparticle(MPM@Lipo)was designed in this study,to combine chemotherapy,photothermal therapy(PTT)and oxygen enrichment to clear hyperproliferating inflammatory cells and improve the hypoxic microenvironment for rheumatoid arthritis(RA)***@Lipo significantly scavenged intracellular reactive oxygen species and relieved joint hypoxia,thus contributing to the repolarization of M1 macrophages into M2 ***,MPM@Lipo could accumulate at inflammatory joints,inhibit the production of inflammatory factors,and protect cartilage in vivo,effectively alleviating RA progression in a rat adjuvant-induced arthritis ***,upon laser irradiation,MPM@Lipo can elevate the temperature to not only significantly obliterate excessively proliferating inflammatory cells but also accelerate the production of methotrexate and oxygen,resulting in excellent RA treatment ***,the use of synergistic chemotherapy/PTT/oxygen enrichment therapy to treat RA is a powerful potential strategy.

PD-L1-driven efficient enrichment and elimination of circulating cancer cells by magnetic MoSe_(2) nanosheet

Objective: Preparing folate(FA)-modified chitosan nanoparticles carrying mouse interferon-induced protein-10(mIP-10) gene(FA-chitosan/m IP-10) and explored the effect of combination of FA-chitosan/m IP-10 and DC/tumor cell fusion vaccine against hepatocellular carcinoma(HCC) and investigated its underlying mechanisms. Methods: Mouse IP-10 plasmid was encapsulated into FA-chitosan to obtain FA-chitosan/m IP10 by complex *** transform infrared spectroscopy(FTIR) were to evaluate the production of mannoselipid. Dynamic light scattering instrument(DLS) and scanning electron microscope(SEM) were to characterize the size, morphology, zeta potential of FA chitosan/m IP10. The gel retardation assay was to ensure that IP-10 plasmid was successfully encapsulated into the conjugated compounds of FA-chitosan. The phagocytosis experiment confirmed the specifically targeting to Hepa1-6 of FA-chitosan. We employed a mouse model of Hepa1-6 in C57BL/6 mice, and subcutaneous injection of FA-chitosan/m IP10 and DC2.4/Hepa1-6 fusion cells was applied to treat HCC in mice and measured tumor growth and mouse survival. Then using flow cytometry to measure the frequency of CXCR3+CD8T cells and myeloid-derived suppressor cells(MDSCs). Enzyme-linked immunospot assay(ELISPOT) were used to measure the levels of interferongamma(IFN-γ). And the IP-10 protein expression of local tumor vascular, density, proliferation, and apoptosis of tumor cells were evaluated using immunological histological chemistry staining. Results: Our results showed that combination of FA-chitosan/IP-10 and DC/tumor fusion vaccine effectively inhibited the growth of implanted HCC tumors and prolonged the survival of mice. Furthermore, the combination therapy significantly reduced Myeloid-derived suppressor cell(MDSC) in mouse spleen, local tumor, and bone marrow, but increased tumor-specific IFN-γ responses in mice. Circulating tumor cells(CTCs)are important biomarkers in the development and progression of lung cancer because they can reach other organs through the blood circulation and form distant metastases,exacerbating lung cancer *** presence of CTCs is also the main reason for the failure of nanomedicine-based lung cancer ***,magnetic MoSe_(2) nanosheets loaded with programmed death-ligand 1(PD-L1),named PD-L1-MFP NS,were employed here to precisely capture lung cancer CTCs in the blood circulation through the tumor-targeting effect of PD-L1 killing CTCs with highly effective photothermal therapy(PTT).In addition,by increasing the expression of cytomegalovirus UL16-binding protein(ULBP)ligands on tumor cells,the PD-L1-MFP NS further activated natural killer(NK)cells and triggered NK cell-induced cancer immunotherapy,thereby enhancing the overall tumor-killing *** summary,this material designed to capture CTCs provides a substantial advancement for personalized PTT-triggered immunotherapy and has great clinical translational potential.

The selective oxidation of cyclohexane is a promising green route for the production of KA oil(cyclohexanol and cyclohexanone),but remains a huge challenge to design high-performing heterogeneous catalysts under mild ***,a pure silica ZSM-12(MTW topology)zeolite was applied as the support to immobilize highly dispersive tetrahedrally coordinated vanadium(V)species and the constructed catalyst 1%V/SZ-12 exhibited high efficiency in the fast and selective oxidation of cyclohexane into cyclohexanone with H_(2)O_(2).The reaction completed within 30 s,affording the selectivity of 99%towards cyclohexanone and the extremely high turnover frequency(TOF)of 5,891 h^(-1).1%V/SZ-12 was facilely reused with stable activity and feasibly extended to the oxidation of various cyclohexane *** one-dimensional 12-membered ring microchannels contributed to the formation and accessibility of tetrahedrally coordinated high valent V5+species as the robust active centers,which activated H_(2)O_(2) to generate V–O–O·,the active oxygen species for the electrophilic attack on cyclohexane to produce cyclohexanone as the sole product.

Marine fauna provides a plentiful repository of peptides and bioactive proteins. Peptides and proteins isolated from marine animals have been studied and applied in the development of food supplements， drugs， and cosmeceutical products because of their special bioactivities， such as anti-inflammatory and antioxidant effects This study focused on exploring the alleviating effects of five major marine animal-derived peptides (Apostichopus japonicus， Acaudina leucoprocta， Melanogrammus aeglefinus， Phascolosoma esculenta and Rhopilema esculentum) on adjuvant-induced arthritis (AIA). The treatment with five marine animals-derived peptides downregulated the expression levels of pro-inflammatory cytokines of IL-1β， IL-17 and TNF-α in the bones of the mice with AIA and alleviated the rough surface of bone tissues significantly. A. japonicus-treatment ameliorates inflammation by restoring NF-κB pathway in AIA mice. High-throughput sequencing of the gut microbiota based on 16S rRNA sequencing revealed that A. japonicus peptide-treated AIA mice showed alterations and imbalance of intestinal flora and an increased abundance of Lactobacillus and Clostridium. Furthermore， metabolomic analysis showed that the level of SCFAs in the feces was enhanced to different degrees in mice treated with five major marine animal-derived peptides. Taken together， we propose that major marine animal-derived peptides can alleviate arthritis by improving the imbalance in the gut flora and increasing SCFA production to varying degrees.

Dater center interconnection has stimulated the development of short-reach optical communication transmission. To increase the capacity of the single sideband(SSB) system with direct-detection(DD), the twin-SSB system can double the system capacity without an extra optical modulator. Recently, the Stokes vector receiver(SVR) bridges the coherent detection and DD. A carrier can be sent along with the signal so that the SVR can utilize the reference carrier to conduct self-coherent detection and realize a linear complex optical detection, which is similar to the coherent receiver. In this paper, we propose a twin-SSB reception without optical SSB filters based on self-coherent detection. The proposed scheme greatly reduces the implementation complexity and has higher spectral efficiency(SE) compared with the traditional twin-SSB signal detection where two narrow-band optical filters and guard band are used for SSB signal extracting and inter crosstalk suppression. Meanwhile, the proposed twin-SSB signal detection with self-coherent detection also helps to relax the receiver digital signal processing(DSP), since the self-coherent detection reduces the inter crosstalk between the twin-SSB. The twin-SSB reception with self-coherent detection scheme makes it suitable for future high-speed short and medium-reach applications, such as the data center interconnect and metro area network.
