Enhanced therapeutic effects of combined chemotherapeutic drugs and midkine antisense oligonucleotides for hepatocellular carcinoma

AIM: To evaluate the effect of combined antisense oligonucleotides targeting midkine (MK-AS) and chemotherapeutic drugs [cisplatin(DDP), 5-fluorouracil (5-FU) and adriamycin (ADM)] on inhibition of HepG2 cell proliferation, and to analyze the efficacy of MK-AS used in combined ADM in in situ human hepatocellular carcinoma (HCC) model. METHODS: HepG2 cells were treated with MK-AS and/or chemotherapeutic drugs mediated by Lipofectin, and cell growth activity was determined by MTS assay. An in situ HCC model was used in this experiment. MK- AS, ADM and MK-AS + ADM were given intravenously for 20 d, respectively. The animal body weight and their tumor weight were measured to assess the effect of the combined therapy in vivo. RESULTS: Combined treatment with MK-AS reduced the IC50 of DDP, 5-FU and ADM in HepG2 cells. MK-AS significantly increased the inhibition rate of DDP, 5-FU and ADM. Additionally, synergism (Q 1.15) occurred at a lower concentration of ADM, 5-FU and DDP with combined MK-AS. Combined treatment with MK-AS and ADM resulted in the more growth inhibition on in situ human HCC model compared with treatment with chemotherapeutic drugs alone. CONCluSION: MK-AS increases the chemosensitivity in HepG2 cells and in situ human HCC model, and thecombination of MK-AS and ADM has a much better in vitro and in vivo synergism.

AIM: To evaluate the effect of antisense oligonucleotide targeting midkine (MK-AS) on angiogenesis in chick chorioallantoic membrane (CAM) and in situ human hepatocellular carcinoma (HCC). METHODS: An in situ human hepatocellular carcinoma (HCC) model and CAM assay were used in this experiment. The effect of MK-AS on angiogenesis was evaluated by cell proliferation assay and hematoxylin- eosin (HE) staining. RESULTS: MK-AS significantly inhibited human umbilical vein endothelial cells (HUVEC) and in situ human HCC growth. At the same time, MK-AS suppressed the angiogenesis both in human hepatocellular carcinoma cell line (HEPG2)-induced CAM and in situ human HCC tissues. CONCluSION: MK-AS is an effective antiangiogenesis agent in vivo.

Applications of TP-AGB synthesis to Mira variables

Employing recent TP-AGB synthesis, we have carried out a detailed study of Mira variables by means of a population synthesis code. We estimate that the total number of Mira variables in the Galaxy is from about 100 000 to 600 000 and their average lifetimes are between about 150 000 and 830 000 yr. The ratio of the number of O-Mira variables to that of C-Mira variables ranges from about 0.68 to 6.2. The effects of input physical parameters （mass loss rate, the minimum core mass for the third dredge-up, the third dredge-up efficiency and theoretical criterion for selecting Mira variables） on the Mira population are discussed.

Contributions of source population and incubation temperature to phenotypic variation of hatchling Chinese skinks

Phenotypic plasticity and local adaptation are viewed as the main factors that result in between-population variation in phenotypic traits,but contributions of these factors to phenotypic variation vary between traits and between species and have only been explored in a few species of ***,we incubated eggs of the Chinese skink(Plestiodon chinensis)from 7 geographically separated populations in Southeast China at 3 constant temperatures(24,28,and 32℃)to evaluate the combined effects of dutch origin,source population,and incubation temperature on hatchling *** relative importance of these factors varied between *** all examined hatchling traits,including body mass,snout-vent length(SVL),tail length,head size,limb length,tympanum diameter,and locomotor speed,varied among populations and were affected by incubation *** for hatchling size(body mass and SVL)varied considerably among *** population explained much of the variation in hatchling body mass,whereas incubation temperature explained much of the variation in other examined *** results indicate that between-population variation in hatchling traits of *** likely reflects the difference in natural incubation conditions and genetic divergence.

Effect of Panax notoginseng saponins on the expression of beta-amyloid protein in the cortex of the parietal lobe and hippocampus, and spatial learning and memory in a mouse model of senile dementia

BACKGROUND： The pharmacological actions of Panax notoginseng saponins （PNS） lie in removing free radicals, anti-inflammation and anti-oxygenation. It can also improve memory and behavior in rat models of Alzheimer＇s disease. OBJECTIVE： Using the Morris water maze, immunohistochemistry, real-time PCR and RT-PCR, this study aimed to measure improvement in spatial learning, memory, expression of amyloid precursor protein （App） and β -amyloid （A β ）, to investigate the mechanism of action of PNS in the treatment of AD in the senescence accelerated mouse-prone 8 （SAMP8） and compare the effects with huperzine A. DESIGN, TIME AND SETTING： A completely randomized grouping design, controlled animal experiment was performed in the Center for Research ＆ Development of New Drugs, Guangxi Traditional Chinese Medical University from July 2005 to April 2007. MATERIALS： Sixty male SAMP8 mice, aged 3 months, purchased from Tianjin Chinese Traditional Medical University of China, were divided into four groups： PNS high-dosage group, PNS low-dosage group, huperzine A group and control group. PNS was provided by Weihe Pharmaceutical Co., Ltd. （batch No.： Z53021485, Yuxi, Yunan Province, China）. Huperzine A was provided by Zhenyuan Pharmaceutical Co., Ltd. （batch No.： 20040801, Zhejiang, China）. METHODS： The high-dosage group and low-dosage group were treated with 93.50 and 23.38 mg/kg PNS respectively per day and the huperzine A group was treated with 0.038 6 mg/kg huperzine A per day, all by intragastric administration, for 8 consecutive weeks. The same volume of double distilled water was given to the control group. A （δ, g）-cage is a δ-regular graph with girth g and with the least possible number of vertices. In this paper, we show that all （δ, g）-cages with odd girth g ≥ 9 are r-connected, where （r - 1）2 ≤δ ＋ x/δ - 2 〈 r2 and all （δ, g）-cages with even girth g 〉 10 are r-connected, where r is the largest integer satisfying r（r-1）2/4 ＋ 1 ＋ 2r（r - 1） ≤δ. These results support a conjecture of Fkl, Huang and Rodger that all （δ, g）-cages are 6-connected.

A Note on the Strong Edge-coloring of Outerplanar Graphs with Maximum Degree 3

A strong k-edge-coloring of a graph G is an assignment of k colors to the edges of G in such a way that any two edges meeting at a common vertex, or being adjacent to the same edge of G, axe assigned different colors. The strong chromatic index of G is the smallest integer k for which G has a strong k-edge-coloring. In this paper, we have shown that the strong chromatic index is no larger than 6 for outerplanax graphs with maximum degree 3.
