Inherited retinal dystrophies (IRDs) are major causes of visual impairment and irreversible blindness worldwide, while the precise molecular and genetic mechanisms are still elusive. n6-methyladenosine (m^(6)A) modification is the most prevalent internal modification in eukaryotic mRnA. YTH domain containing 2 (YTHDC2), an m^(6)A reader protein, has recently been identified as a key player in germline development and human cancer. However, its contribution to retinal function remains unknown. Here, we explore the role of YTHDC2 in the visual function of retinal rod photoreceptors by generating rod-specific Ythdc2 knockout mice. Results show that Ythdc2 deficiency in rods causes diminished scotopic ERG responses and progressive retinal degeneration. Multi-omics analysis further identifies Ppef2 and Pde6b as the potential targets of YTHDC2 in the retina. Specifically, via its YTH domain, YTHDC2 recognizes and binds m^(6)A-modified Ppef2 mRnA at the coding sequence and Pde6b mRnA at the 5′-UTR, resulting in enhanced translation efficiency without affecting mRnA levels. Compromised translation efficiency of Ppef2 and Pde6b after YTHDC2 depletion ultimately leads to decreased protein levels in the retina, impaired retinal function, and progressive rod death. Collectively, our finding highlights the importance of YTHDC2 in visual function and photoreceptor survival, which provides an unreported elucidation of IRD pathogenesis via epitranscriptomics.

Covalent organic frameworks/carbon nanotubes composite with cobalt(II)pyrimidine sites for bifunctional oxygen electrocatalysis

With characteristics and advantages of functional composite materials, they are commendably adopted in numerous fields especially in oxygen electrocatalysis, which is due to the significant synergies between various components. Herein, a novel bifunctional oxygen electrocatalyst（Co-CnT@COF-Pyr） has been synthesized through in-situ growth of covalent organic frameworks（COFs） layers on the outer surface of highly conductive carbon nanotubes（CnTs） followed by coordination with Co（II）. For electrocatalytic OER, Co-CnT@COF-Pyr reveals a low overpotential（438 mV） in alkaline electrolyte（1.0 M aqueous solution of KOH） with a current density of 10 mA cm-2, which is comparable to most discovered COF-based catalysts. For electrocatalytic ORR, CoCnT@COF-Pyr exhibits a low H2O2yield range（9.0 %–10.1 %） and a reaction pathway close to 4e-（n =3.82–3.80） in alkaline electrolyte（0.1 M aqueous solution of KOH） within the test potential range of 0.1–0.6 V ***, which is superior to most reported COF-based catalysts. Hence, this research could not only offer an innovative insight into the construction of composites, but also facilitate the practical application of renewable fuel cells, closed water cycle, and rechargeable metal-air batteries.

Lithium metal shows a great advantage as the most promising anode for its unparalleled theoretical specific capacity and extremely low electrochemical ***,uncontrolled lithium dendrite growth and severe side reactions of the reactive intermediates and organic electrolytes still limit the broad application of lithium metal ***,we propose 4-nitrobenzenesulfonyl fluoride(nBSF)as an electrolyte additive for forming a stable organic-inorganic hybrid solid electrolyte interphase(SEI)layer on the lithium *** abundance of lithium fluoride and lithium nitride can guarantee the SEI layer's toughness and high ionic conductivity,achieving dendrite-free lithium ***,the phenyl group of nBSF significantly contributes to both the chemical stability of the SEI layer and the good adaptation to volume changes of the lithium *** lithium-oxygen batteries with nBSF exhibit prolonged cycle lives and excellent cycling *** simple approach is hoped to improve the development of the organic-inorganic SEI layer to stabilize the lithium anodes for lithium-oxygen batteries.

The disparity in the transfer of carriers(electrons/mass)during the reaction in zinc-air batteries(ZABs)results in sluggish kinetics of the oxygen reduction reaction(ORR)and oxygen evolution reaction(OER),along with elevated overpotentials,thereby imposing additional constraints on its ***,the pre-design and target-development of inexpensive,high-performance,and long-term stable bifunctional catalysts are urgently *** this work,an apically guiding dual-functional electrocatalyst(Ag-Fen_(x)-n-C)was prepared,in which a hierarchical porous nitrogen-doped carbon with three-dimensional(3D)hollow star-shaped structure is used as a substrate and high-conductivity Ag nanoparticles are coupled with iron nitride(Fen_(x))*** calculations indicate that the Mott-Schottky heterojunction as an inherent electric field comes from the two-phase bound of Ag and Fen_(x),of which electron accumulation in the Fen_(x)phase region and electron depletion in the Ag phase region promote orientated-guiding charge *** effective modulation of local electronic structures felicitously reforms the d-band electron-group distribution,and intellectually tunes the masstransfer reaction energy barriers for both ORR/***,the hollow star-s haped hierarchical porous structure provides an apical region for fast mass *** results show that the halfwave potential for ORR is 0.914 V,and the overpotential for OER is only 327 mV at 10 mA cm^(-2).A rechargeable ZAB with Ag-Fen_(x)-n-C as the air cathode demonstrates long-term cycling performance exceeding 1500 cycles(500 h),with a power density of 180 mW cm^(-2).Moreover,when employing AgFen_(x)-n-C as the air cathode,flexible ZABs demonstrate a notable open-circuit voltage of 1.42 V and achieve a maximum power density of 65.6 mW cm^(-2).Ag-Fen_(x)-n-C shows guiding electron/mass transfer route and apical reaction microenvironment for the electrocatalyst architecture in the explo

Immunotherapy has attracted wide attention because it can not only kill tumor cells but also effectively prevent tumor recurrence. However, due to the poor efficacy of the monotherapy in tumor immunotherapy, combination therapies are needed to enhance their efficacy,such as chemodynamic therapy/immunotherapy, photodynamic therapy/immunotherapy, sonodynamic therapy/immunotherapy and so on. nanoscale metal–organic frameworks(nMOFs) are widely used for drug delivery because of their rich and variable composition, high drug loading capacity and good biodegradability. Compared with other conventional drug delivery systems(DDSs),nMOFs play more advantageous role in inducing immune cell death of tumor cells and combination therapies. This article focuses on the application of n MOFs in tumor immunotherapy in terms of their use as gatekeepers, supplementary agents and drug carriers, respectively, with the aim of providing some ideas and references for the design of smart nanoscale DDS for tumor immunotherapy.

With abundant potassium resources and high capacity,potassium metal batteries(PMBs)present a compelling option for the next generation of energy storage ***,PMBs suffer from an unstable anode interface caused by uncontrolled dendrite growth,which results in unsatisfactory cyclability and safety *** investigations suggest that significant progress has been made in enhancing the interfacial stability of *** various effective strategies for stabilizing interfaces can ultimately be attributed to the regulation of the sluggish ion transfer kinetics and irregular deposition,i.e.,the arrangement of ion transport behaviors at the *** modulation of ions transport rate and ions deposition directions makes it possible to obtain a dendrite-free and smooth deposition ***,the influencing factors and action mechanism of K^(+)interface transport behaviors are discussed to understand the nature of material design for constructing stable anode interfaces,including regulating the solvation and desolvation structures,accelerating K^(+)transport kinetics and controlling K^(+)deposition *** addition,the deficiencies and prospects of the research on electrolyte,separators and designed electrode involved in the manufacturing and testing and ion transport process of PMBs are *** review is expected to provide some possible directions for constructing dendrite-free interfaces in advanced PMBs-related research and offer significant insights for prospective experimental research and commercial applications.
