cationic antimicrobial protein of Mr 37 kDa:a multifunctional inflammatory protein

PURPOsE: To investigate the role of cationic antimicrobial protein of Mr 37 kDa (cAP37) a neutrophil-derived inflammatory mediator on endothelial cell function. DATA sOURcEs: Endothelial cells used in this study were obtained from human lung microvessels and rat aorta. The latter was a kind gift of Dr. Paula Grammas. The mono-mac 6 cell line used in this study was the generous gift of Dr. H.W. Loms Ziegler-Heitbrock. sTUDY sELEcTION AND DATA EXTRAcTION: Endothelial cell proteins kinase c activity was determined by measuring calcium- and phospholipid-dependent phosphorylation of histone. Endothelial cell migration was determined using costar Transwell apparatus. cell surface expression of adhesion molecules, IcAM-1 and PEcAM-1 was determined using flow cytometry. RT-PcR was used to amplify the cAP37 from endothelial cells treated with LPs. REsULTs: We demonstrated that cAP37 which was originally identified as having potent antimicrobial activity and chemotactic activity for monocytes was capable of modulating endothelial cell functions. cAP37 activated endothelial cell protein kinase c in a dose- and time-dependent fashion. Importantly cAP37 increased the adhesive properties of the endothelium for monocytes. cAP37 upregulated the well known adhesion molecules, IcAM-1 and PEcAM-1 in a dose- and time-dependent manner. In addition, cAP37 promoted endothelial cell migration. Further investigations indicated that cAP37 was induced in endothelial cells in response to pro-inflammatory cytokines such as tumor necrosis factor-alpha and interleukin-1 alpha as well as inflammatory mediators such as lipopolysaccharide. Unstimulated endothelial cells did not constitutively express cAP37. The cDNA sequence of endothelial cAP37 was determined and found to be highly homologous to the sequence obtained for neutrophil-derived cAP37. cONclusIONs: Our studies strongly suggest that cAP37 plays a pivotal role in monocyte-endothelial interactions and the transmigration of monocytes from th

Analysis of 12 variants in the development of gastric and colorectal cancers

AIM To evaluate the relation between 12 polymorphisms and the development of gastric cancer(Gc) and colorectal cancer(cRc).METHODs In this study,we included 125 individuals with Gc diagnosis,66 individuals with cRc diagnosis and 475 cancer-free individuals. All participants resided in the North region of Brazil and authorized the use of their samples. The 12 polymorphisms(in cAsP8,cYP2 E1,cYP19 A1,IL1 A,IL4,MDM2,NFKB1,PAR1,TP53,TYMs,UGT1 A1 and XRcc1 genes) were genotyped in a single PcR for each individual,followed by fragment analysis. To avoid misinterpretation due to population substructure,we applied a previously developed set of 61 ancestryinformative markers that can also be genotyped by multiplex PcR. The statistical analyses were performed in structure v.2.3.4,R environment and sPss *** After statistical analyses with the control of confounding factors,such as genetic ancestry,three markers(rs79071878 in IL4,rs3730485 in MDM2 and rs28362491 in NFKB1) were positively associated with the development of Gc. One of these markers(rs28362491) and the marker in the UGT1 A1 gene(rs8175347) were positively associated with the development of cRc. Therefore,we investigated whether the joint presence of the deleterious alleles of each marker could affect the development of cancer and we obtained positive results in all analyses. carriers of the combination of alleles RP1 + DEL(rs79071878 and rs28361491,respectively) are at 10-times greater risk of developing Gc than carriers of other combinations. similarly,carriers of the combination of DEL + RARE(rs283628 and rs8175347) are at about 12-times greater risk of developing cRc than carriers of other *** These findings are important for the comprehension of gastric and cRc development,particularly in highly admixed populations,such as the Brazilian population.

The key risk factor for glaucoma is increased intraocular pressure (IOP). Glaucoma drainage devices implanted in theeye can reduce IOP and thus stop disease progression. However, most devices currently used in clinical practice arepassive and do not allow for postsurgical IOP control, which may result in serious complications such as hypotony (i.e.,excessively low IOP). To enable noninvasive IOP control, we demonstrate a novel, miniature glaucoma implant that willenable the repeated adjustment of the hydrodynamic resistance after implantation. This is achieved by integrating amagnetic microvalve containing a micropencil-shaped plug that is moved using an external magnet, thereby openingor closing fluidic channels. The microplug is made from biocompatible poly(styrene-block-isobutylene-block-styrene)(sIBs) containing iron microparticles. The complete implant consists of an sIBs drainage tube and a housing elementcontaining the microvalve and fabricated with hot embossing using femtosecond laser-machined glass molds. Usingin vitro and ex vivo microfluidic experiments, we demonstrate that when the microvalve is closed, it can providesufficient hydrodynamic resistance to overcome hypotony. Valve function is repeatable and stable over time. Due to itssmall size, our implant is a promising, safe, easy-to-implant, minimally invasive glaucoma surgery device.

Differences in physical and structural characteristics of seeds may indicate variability within and between plant *** the present study,we performed a close characterization of dimension,shape,and tegument delayed chlorophyll fluorescence in seeds obtained from three species of the genus Annona(Annonaceae),i.e.,Annona coriacea,***,*** showed that studied seeds may be sorted as scalene ellipsoids expressing low values for the seed *** morphological estimates suggested differences in seed shape for all species.A high correlation was observed between surface area and volume(r^2>99%)for all the three species suggesting that in addition to structural *** addition,we also observed very high positive correlations(Rho=1.000,psurface area and arithmetic mean diameterof the seeds for all *** first principal component(PcA1)of elliptical Fourier descriptors explained most of the variations in morphological structure of the seeds in the three ***,a less intense tegument delayed chlorophyll fluorescence was observed for *** while the highest intensity was recorded for ***,revealing the potential use of fluorescence spectroscopy in discrimination at the species level by analyzing the frequency domain by means of Fourier Transform spectra as well as the relationship time-frequency of chlorophyll fluorescence.

Adsorbents Derived from Xylan Hemicellulose with Removal Properties of Pollutant Metals

The adsorption capacity of hydrogels derived from modified xylan hemicellulose has been tested in order to develop new bio-based adsorbent materials useful for removing metal ions pollutants,such as cd(Ⅱ),cu(Ⅱ)and Pb(Ⅱ)from an aqueous *** was extracted from bleached kraft pulp of eucalyptus and subsequently modified with different proportions of functional sulfonic acid groups(HA3-HA7)and sulfonate groups(Hs30-Hs70)to generate *** results showed that all the synthesized hydrogels were capable of adsorbing metal ions,being the hydrogels with 30%and 50%xylan the ones that presented the highest adsorption *** capacity studies at different initial concentrations revealed that at an initial concentration of 300 mg·L-1,the HA3 hydrogel presented an adsorption capacity of 193mg Pb(Ⅱ),182 mg cd(Ⅱ),and 66 mg cu(Ⅱ)per g *** HA5 hydrogel presented a capacity of 185 mg Pb(Ⅱ),113 mg cd(Ⅱ),and 48 mg cu(Ⅱ)per g *** Hs30 hydrogel exhibited an adsorption of 205 mg Pb(Ⅱ),174 mg cd(Ⅱ),and 71 mg cu(Ⅱ)per g hydrogel,and Hs50 hydrogel exhibited an adsorption capacity of 273 mg Pb(Ⅱ),143 mg cd(Ⅱ),and 45 mg cu(Ⅱ)per g *** results show that modified Xylan hemicellulose is a promising adsorbent for removal cd(Ⅱ),cu(Ⅱ),and Pb(Ⅱ)ions from aqueous solutions.

Antidiabetic effects of galactomannans from Adenanthera pavonina *** streptozotocin-induced diabetic mice

Phytochemical investigations of the ethanolic extract of leaves of clutia lanceolata(Family:Euphorbiaceae)resulted in the isolation of four compounds viz.3,4-dihydroxy-2-methylbenzoic acid(1),2,20-dihydroxy-1,10-binaphthyl(2),1,3,8-trihydroxy-6-methylanthracene-9,10-dione(3)and 5-hydroxy-1,7-bis(4-hydroxy-3-methoxyphenyl)hepta-1,4,6-trien-3-one(4).Although all the isolated compounds were known but this was the first report from this plant *** structures were established on the basis of chemical and physical evidences *** analysis,FT-IR,1 H-NMR,13cNMR and mass spectral *** of compound 2 and 4 was further authenticated by single-crystal X-ray analysis and density functional theory *** isolated compounds(1–4)were screened for AchE enzyme inhibition assay in which compound 3 and 4 were found to be more potent AchE *** docking study of potent AchE inhibitor was performed to find the probable binding mode of the compounds into the active site of ***,the isolated compounds were also screened for in vivo cytotoxicity by brine shrimp lethality assay.
