Body color plasticity of Diaphorina citri reflects a response to environmental stress

Body color polyphenism is common in Diaphorina citri.Previous studies compared physiological characteristics in D.citri,but the ecological and biological significance of its body color polyphenism remains poorly understood.We studied the ecological and molecular effects of stressors related to body color in D.citri.Crowding or low temperature induced a high proportion of gray morphs,which had smaller bodies,lower body weight,and greater susceptibility to the insecticide dinotefuran.We performed transcriptomic and metabolomics analysis of 2 color morphs in D.citri.Gene expression dynamics revealed that the differentially expressed genes were predominantly involved in energy metabolism,including fatty acid metabolism,amino acid metabolism,and carbohydrate metabolism.Among these genes,plexin,glycosidase,phospholipase,take out,trypsin,and triacylglycerol lipase were differentially expressed in 2 color morphs,and 6 hsps(3 hsp70,hsp83,hsp90,hsp68)were upregulated in gray morphs.The metabolome data showed that blue morphs exhibited a higher abundance of fatty acid and amino acid,whereas the content of carbohydrates was elevated in gray morphs.This study partly explains the body color polyphenism of D.citri and provides insights into the molecular changes of stress response of D.citri.

Electrochemical Analysis of Single Glucose Oxidase with a Nanopipette

Nanopore-based electrochemical technique is a promising tool for detecting single proteins.However,detecting single proteins using a nanopipette in their native state without labeling is challenging due to the rapid translocation,which results in an inefficient signal identification.In our study,we finely tuned the driving force equilibrium between electrophoretic force(EPF)and electroosmotic flow(EOF)inside the nanopipette for efficient sensing of single glucose oxidase(GOD)molecules.The duration time of GOD within the nanopipette is extended to about 4 ms.This strategy provided clear ionic current signals with a signal-to-noise ratio of 3.3.As EPF increased in the direction opposite to the motion of GOD,we observed a nonlinear growth in GOD’s duration time.This extended the duration to about 4.4 times longer at−1000 mV compared to at−800 mV.Hence,nanopore-based electrochemical sensing could be used for single GOD molecule analysis as an ultrasensitive method.

The Mass Assembly History for Galaxies with MaNGA

How galaxies assemble masses through their own star formation or interaction with the external environment is still an important topic in the field of galaxy formation and evolution.We use Value Added Catalogs with galaxy features that are spatially and temporally resolved from Sloan Digital Sky Survey Data Release 17 to investigate the mass growth histories of early-type galaxies(ETGs)and late-type galaxies(LTGs).We find that the mass growth of ETGs is earlier than that of LTGs for massive galaxies(M_(*)>10^(10)M_⊙),while low-mass(M_(*)≤10^(10)M_⊙)ETGs have statistically similar mass assembly histories as low-mass LTGs.The stellar metallicity of all massive galaxies shows a negative gradient and basically does not change with time.However,in low-mass galaxies,the stellar metallicity gradient of elliptical galaxies is negative,and the stellar metallicity gradient of lenticular and spiral galaxies evolves from positive to negative.ETGs are not all in a high-density environment,but exhibit mass dependence.As the tidal strength increases,the star formation rate of low-mass ETGs rapidly decreases.These results support a picture where massive galaxies exhibit inside-out quenching mode,while low-mass galaxies show outside-in quenching mode.Environmental effects play an important role in regulating the mass assembly histories of low-mass ETGs.

Ultrathin surface coating of Cu enabling long-life Zn metal anodes

Aqueous zinc-ion batteries （AZIBs） are regarded as promising energy storage devices due to their low cost,high capacity,and ecological safety.Nevertheless,the severe dendritic growth and side reactions hinder their practical applicability significantly.Herein,an ultrathin Cu coating layer （～200 nm） is decorated on zinc foils via filtered cathodic vacuum arc deposition technology,aiming to achieve high-performance AZIBs.The Cu layer effectively suppresses chemical corrosion and hydrogen evolution reactions and enables preferential （002） Zn deposition during the stripping/plating cycles.Consequently,the Cu@Zn anode represents an elongated cycle life over 4,000 h at 2 mA·cm-2/2 mAh·cm-2.Even in conditions of high current density of 5 and 10 mA·cm-2,the Cu@Zn anode shows prolonged cycling stability exceeding 4000 and 2000 h,respectively.Such advantages also bring high Coulombic efficiency of 99.87% under 5 mAh·cm-2in Cu@Ti||Zn half-cell over 1500 cycles.Moreover,the Cu@Zn||MnO2full cell demonstrates a superior cyclability with a specific capacity of 203 mAh·g-1after 500 cycles a1 A·g-1.

The rodent running-wheel recording apparatus is a reliable approach for studying cir-cadian rhythm.This study demonstrated how to construct a simple and intelligent running-wheel recording system.The running wheel was attached to the cage's base,whereas the Hall sensor was attached to the cage's cover.Then,the RJ25 adaptor relayed the running signal to the main control board.Finally,the main control board was connected to the USB port of the computer with the USB connection.Data were collected using the online-accessible,self-created software Magturning.Through Magturning,generated data were saved and exported in real time.Afterward,the device was validated by collecting data on the locomotor activities of mice under dif-ferent light conditions.In conclusion,this new device can record circadian activity of rodents.Our device is appropriate for interdisciplinary investigations related to biological clock research.

Proteomic profiling of cerebrospinal fluid in pediatric myelin oligodendrocyte glycoprotein antibody-associated disease

Background Myelin oligodendrocyte glycoprotein(MOG)antibody-associated disease(MOGAD)is an autoimmune demy-elinating disorder of the central nervous system.Methods Extracted proteins from 34 cerebrospinal fluid(CSF)samples[patients with MOGAD(MOG group,n=12);healthy controls(HC group,n=12);patients with MOG seronegative and metagenomics next-generation sequencing-negative inflammatory neurological diseases(IND group,n=10)]were processed and subjected to label-free quantitative proteomics.Supervised partial least squares-discriminant analysis(PLS-DA)and orthogonal PLS-DA(O-PLS-DA)models were also performed based on proteomics data.Functional analysis of differentially expressed proteins(DEPs)was performed using Gene Ontology,InterPro,and Kyoto Encyclopedia Genes and Genomes.An enzyme-linked immunosorbent assay was used to determine the complement levels in serum from patients with MOGAD.Results Four hundred and twenty-nine DEPs(149 upregulated and 280 downregulated proteins)were identified in the MOG group compared to the HC group according to the P value and fold change(FC).Using the O-PLS-DA model,872 differentially abundant proteins were identified with variable importance projection(VIP)scores>1.Five proteins(gamma-glutamyl hydrolase,cathepsin F,interalpha-trypsin inhibitor heavy chain 5,latent transforming growth factor beta-binding protein 4 and leukocyte-associated immunoglobulin-like receptor 1)overlapping between the top 30 DEPs with top-ranked P value and FC and top 30 proteins in PLS-DA VIP lists were acquired.Functional analysis revealed that the dysregulated proteins in the MOG group were primarily involved in complement and coagulation cascades,cell adhesion,axon guidance,and glycosphingolipid biosynthesis compared to the HC group.Conclusion The proteomic alterations in CSF samples from children with MOGAD identified in the current study might provide opportunities for developing novel biomarker candidates.

Spatial-temporal changes and driving factors of eco-environmental quality in the Three-North region of China

Eco-environmental quality is a measure of the suitability of the ecological environment for human survival and socioeconomic development.Understanding the spatial-temporal distribution and variation trend of eco-environmental quality is essential for environmental protection and ecological balance.The remote sensing ecological index(RSEI)can quickly and objectively quantify eco-environmental quality and has been extensively utilized in regional ecological environment assessment.In this paper,Moderate Resolution Imaging Spectroradiometer(MODIS)images during the growing period(July-September)from 2000 to 2020 were obtained from the Google Earth Engine(GEE)platform to calculate the RSEI in the three northern regions of China(the Three-North region).The Theil-Sen median trend method combined with the Mann-Kendall test was used to analyze the spatial-temporal variation trend of eco-environmental quality,and the Hurst exponent and the Theil-Sen median trend were superimposed to predict the future evolution trend of eco-environmental quality.In addition,ten variables from two categories of natural and anthropogenic factors were analyzed to determine the drivers of the spatial differentiation of eco-environmental quality by the geographical detector.The results showed that from 2000 to 2020,the RSEI in the Three-North region exhibited obvious regional characteristics:the RSEI values in Northwest China were generally between 0.2 and 0.4;the RSEI values in North China gradually increased from north to south,ranging from 0.2 to 0.8;and the RSEI values in Northeast China were mostly above 0.6.The average RSEI value in the Three-North region increased at an average growth rate of 0.0016/a,showing the spatial distribution characteristics of overall improvement and local degradation in eco-environmental quality,of which the areas with improved,basically stable and degraded eco-environmental quality accounted for 65.39%,26.82%and 7.79%of the total study area,respectively.The Hurst exponent of the RSEI ranged from 0.20 to 0.76 an

Objective: IMpower210(NCT02813785) explored the efficacy and safety of single-agent atezolizumab vs.docetaxel as second-line treatment for advanced non-small cell lung cancer(NSCLC) in East Asian patients.Methods: Key eligibility criteria for this phase Ⅲ, open-label, randomized study included age ≥18 years;histologically documented advanced NSCLC per the Union for International Cancer Control/American Joint Committee on Cancer staging system(7th edition);Eastern Cooperative Oncology Group performance status of 0 or 1;and disease progression following platinum-based chemotherapy for advanced or metastatic NSCLC. Patients were randomized 2:1 to receive either atezolizumab(1,200 mg) or docetaxel(75 mg/m^(2)). The primary study endpoint was overall survival(OS) in the intention-to-treat(ITT) population with wild-type epidermal growth factor receptor expression(ITT EGFR-WT) and in the overall ITT population.Results: Median OS in the ITT EGFR-WT population(n=467) was 12.3 [95% confidence interval(95% CI),10.3-13.8] months in the atezolizumab arm(n=312) and 9.9(95% CI, 7.8-13.9) months in the docetaxel arm[n=155;stratified hazard ratio(HR), 0.82;95% CI, 0.66-1.03]. Median OS in the overall ITT population was 12.5(95% CI, 10.8-13.8) months with atezolizumab treatment and 11.1(95% CI, 8.4-14.2) months(n=377) with docetaxel treatment(n=188;stratified HR, 0.87;95% CI, 0.71-1.08). Grade 3/4 treatment-related adverse events(TRAEs) occurred in 18.4% of patients in the atezolizumab arm and 50.0% of patients in the docetaxel arm.Conclusions: IMpower210 did not meet its primary efficacy endpoint of OS in the ITT EGFR-WT or overall ITT populations. Atezolizumab was comparatively more tolerable than docetaxel, with a lower incidence of grade3/4 TRAEs.

Dual labeling of an RNA can provide Förster resonance energy transfer(FRET)sensors for studying RNA folding,miRNA maturation,and RNA-protein interactions.Here,we report the development of a highly efficient strategy for direct dual-terminal labeling of any RNA of interest.We explored new Michael cycloaddition for facile labeling of 5′-terminal RNA with improved efficiency.Direct chemical tetrazinylation of RNA at the 3′-terminus was achieved with the highly efficient and catalysis-free tetrazine-cycloalkyne ligation.Both single-terminal labeling methods were combined for dual-terminal labeling of an RNA including short hairpin RNA,pre-miRNA,riboswitch,and noncoding RNA.Notably,these dual-labeled RNA-based FRET sensors were used to monitor RNA-ligand interactions in vitro and in live cells.It is anticipated that these universal RNA labeling strategies will be useful to study RNA structures and functions.

无监督域自适应在解决训练集(源域)和测试集(目标域)分布不一致的问题上已经取得了一定的成功.在面向低能耗场景和开放动态任务环境时,在资源约束和开放类别出现的情况下,现有的无监督域自适应方法面临着严峻的挑战.源域无关开集域自适应(SF-ODA)旨在将源域模型中的知识迁移到开放类出现的无标签目标域,从而在无源域数据资源的限制下辨别公共类和检测开放类.现有的源域无关开集域自适应的方法聚焦于设计准确检测开放类别的源域模型或增改模型的结构.但是,这些方法不仅需要额外的存储空间和训练开销,而且在严格的隐私保护场景下难以实现.提出了一个更加实际的场景:主动学习的源域无关开集域自适应(ASF-ODA),目标是基于一个普通训练的源域模型和少量专家标注的有价值的目标域样本来实现鲁棒的迁移.为了达成此目标,提出了局部一致性主动学习(LCAL)算法.首先,利用目标域中局部特征标签一致的特点,LCAL设计了一种新的主动选择方法:局部多样性选择,来挑选更有价值的阈值模糊样本来促进开放类和公共类分离.接着,LCAL基于信息熵初步筛选出潜在的公共类集合和开放类集合,并利用第一步得到的主动标注样本对这两个集合进行匹配纠正,得到两个对应的可信集合.最后,LCAL引入开集损失和信息最大化损失来进一步促使公共类和开放类分离,引入交叉熵损失来实现公共类的辨别.在Office-31、Office-Home和VisDA-C这3个公开的基准数据集上的大量实验表明:在少量有价值的目标域样本的帮助下,LCAL不仅显著优于现有的源域无关开集域自适应方法,还大幅度超过了现有的主动学习方法的表现,在某些迁移任务上可以提升20%.
