AIM: To show a new paradigm of simultaneously testing whether breast cancer therapies impact other causes of death. MeTHODS: MA.14 allocated 667 postmenopausal women to 5 years of tamoxifen 20 mg/daily ± 2 years of octreotide 90 mg, given by depot intramuscular injections monthly. event-free survival was the primary endpoint of MA.14; at median 7.9 years, the tamoxifen+octreotide and tamoxifen arms had similar event-free survival(P = 0.62). Overall survival was a secondary endpoint, and the two trial arms also had similar overall survival(P = 0.86). We used the median 9.8 years follow-up to examine by intention-to-treat, the multivariate time-to-breast cancer-specific(Br Ca) and other cause(OC) mortality with log-normal survival analysis adjusted by treatment and stratification factors. We tested whether baseline factors including Insulin-like growth factor 1(IGF1), IGF binding protein-3, C-peptide, body mass index, and 25-OH vitamin D were associated with(1) all cause mortality, and if so; and(2) cause-specific mortality. We also fit step-wise forward cause-specific adjusted ***: The analyses were performed on 329 patients allocated tamoxifen and 329 allocated tamoxifen+octreotide. The median age of MA.14 patients was 60.1 years: 447(82%) ears and 120(18%) ≥ 70 years. There were 170 deaths: 106(62.3%) BrC a; 55(32.4%) OC, of which 24 were other malignancies, 31 other causes of death; 9(5.3%) patients with unknown cause of death were excluded from competing risk assessments. BrC a and OC deaths were not significantly different by treatment arm(P = 0.40): tamoxifen patients experienced 50 BrC a and 32 OC deaths, while tamoxifen + octreotide patients experienced 56 Br Ca and 23 OC deaths. Proportionately more deaths(P = 0.004) were from BrC a for patientsears, where 70% of deaths were due to Br Ca, compared to 54% for those ≥ 70 years of age. Advances in cellular and molecular biology underpin most current therapeutic advances in *** advances for neurological and neurodegenerative diseases are hindered by the lack of similar specimens. It is becoming increasingly evident that greater access to human brain tissue is necessary to understand both the cellular biology of these diseases and their variation. Research in these areas is vital to the development of viable therapeutic options for these currently untreatable diseases. The development and coordination of human brain specimen collection through brain banks is evolving. This perspective article from the Sydney Brain Bank reviews data concerning the best ways to collect and store material for different research purposes.

Paleoecological Response of Corals to the end-Triassic Mass extinction： An Integrational Analysis

The end-Triassic （also Triassic-Jurassic） mass extinction severely affected life on planet earth 200 million years ago. Paleoclimate change triggered by the volcanic eruptions of the Central Atlantic Magmatic Province （CAMP） caused a great loss of marine biodiversity, among which 96% coral genera were get lost. However, there is precious little detail on the paleoecology and growth forms lost between the latest Triassic extinction and the early Jurassic recovery. Here a new pilot study was conducted by analyzing corallite integration levels among corals from the latest Triassic and early Jurassic times. Integration levels in corals from the Late Triassic and early Jurassic were determined through both the Paleobiology Database as well as from a comprehensive museum collection of fossil corals. Results suggest that in addition to a major loss of diversity following the end-Triassic mass extinction, there also was a significant loss of highly integrated corals as clearly evidenced by the coral data from the early Jurassic. This confirms our hypothesis of paleoecological selectivity for corals following the end-Triassic mass extinction. This study highlights the importance of assigning sim- ple to advanced paleoecological characters with integration levels, which opens a useful approach to understanding of mass extinction and the dynamics of the recovery.

X-ray micro-computed tomography(μ-CT)can be used to provide both qualitative and quantitative information on the structure of three-dimensional(3D)bioactive *** performed in a dry state,μ-CT accurately reflects the structure of collagen-based scaffolds,but imaging in a wet state offers challenges with *** we have used phosphotungstic acid(PTA)as a contrast agent to visualise fully hydrated collagen scaffolds in a physiologically relevant environment.A systematic investigation was performed to understand the effects of PTA on the results of μ-CT imaging by varying sample processing variables such as crosslinking density,hydration medium and staining *** samples in 0.3% PTA solution overnight completely stained the samples and the treatment provided a successful route forμ-CT analysis of crosslinked ***,significant structural artefacts were observed for samples which were either non-crosslinked or had low levels of crosslinking,which had a heterogeneous interior architecture with collapsed pores at the scaffold *** work highlights the importance of optimising the choice of processing and staining conditions to ensure accurate visualisation for hydrated 3D collagen scaffolds in an aqueous medium.

Airborne laser scanning of natural forests in New Zealand reveals the influences of wind on forest carbon

Background： Forests are a key component of the global carbon cycle, and research is needed into the effects of human-driven and natural processes on their carbon pools. Airborne laser scanning （ALS） produces detailed 3D maps of forest canopy structure from which aboveground carbon density can be estimated. Working with a ALS dataset collected over the 8049-km2 Wellington Region of New Zealand we create maps of indigenous forest carbon and evaluate the influence of wind by examining how carbon storage varies with aspect. Storms flowing from the west are a common cause of disturbance in this region, and we hypothesised that west-facing forests exposed to these winds would be shorter than those in sheltered east-facing sites. Methods： The aboveground carbon density of 31 forest inventory plots located within the ALS survey region were used to develop estimation models relating carbon density to ALS information. Power-law models using rasters of top-of-the-canopy height were compared with models using tree-level information extracted from the ALS dataset. A forest carbon map with spatial resolution of 25 m was generated from ALS maps of forest height and the estimation models. The map was used to evaluate the influences of wind on forests. Results： Power-law models were slightly less accurate than tree-centric models （RMSe 35% vs 32%） but were selected for map generation for computational efficiency. The carbon map comprised 4.5 million natural forest pixels within which canopy height had been measured by ALS, providing an unprecedented dataset with which to examine drivers of carbon density. Forests facing in the direction of westerly storms stored less carbon, as hypothesised. They had much greater above-ground carbon density for a given height than any of 14 tropical forests previously analysed by the same approach, and had exceptionally high basal areas for their height. Hepatitis C in the pediatric population: Transmission, natural history, treatment and liver transplantation

The number of children affected by the hepatitis C virus (HCV) in the United States is estimated to be between 23000 to 46000. The projected medical cost for children with HCV in the United States is upwards of 200 million over the next decade. The implementation of routine screening of blood supply has virtually eliminated transmission via transfusion and vertical transmission is now the most common mode of infection in children. Infections acquired during infancy are more likely to spontaneously resolve and fibrosis of the liver tends to increase with age suggesting slow progressive histologic injury. Anti-viral treatment may be warranted in children with persistently elevated liver enzymes or with significant fibrosis on liver biopsy. Current standard of care includes weekly pegylated interferon and ribavirin twice daily. Predictors of high sustained viral response include genotype 2 and 3 and low viral load in children with genotype 1 (e therapy is associated with a significantly higher rate of sustained virologic response (> 90%). Only 34 pediatric patients were transplanted with hepatitis C between January 2008 and April 2013. The majority of pediatric patients were born prior to universal screening of blood products and, as of June 2013, there are only two pediatric patients awaiting liver transplantation for end-stage liver disease secondary to hepatitis C. Pediatric survival rates post-transplant are excellent but graft survival is noticeably reduced compared to adults (73.73% for pediatric patients at one year compared to 87.69% in adult patients). New safe potent, and all-oral effective antiviral therapies for recurrent HCV should help increase graft survival.

Reconstituted type I collagen fibres have received considerable interest as tendon implant materials due to their chemical and structural similarity to the native *** produced through a semi-continuous extrusion process were cross-linked with different concentrations of the zerolength cross-linker 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide(eDC)in combination with N-hydroxysuccinimide(NHS).Tensile properties of the fibres were considered,along with imaging of both surface structure and fibrillar *** of the fibres to bacterial collagenase was investigated and fibre sections seeded with human tendon cells for biological characterization,including cell adhesion and *** work clearly demonstrated that whilst the concentration of eDC and NHS had no significant effect on the mechanics,a higher concentration was associated with higher collagenase resistance,but also provided a less attractive surface for cell adhesion and proliferation.A lower cross-linking concentration offered a more biocompatible material without reduction in mechanics and with a potentially more optimal degradability.

Diversification of SUMO-Activating enzyme in Arabidopsis： Implications in SUMO Conjugation

Sumoylation is an essential posttranslational modification that participates in many biological processes including stress responses. However, little is known about the mechanisms that control Small Ubiquitin-like MOdifier （SUMO） conjugation in vivo. We have evaluated the regulatory role of the heterodimeric e1 activating enzyme, which catalyzes the first step in SUMO conjugation. We have established that the e1 large SAe2 and small SAe1 subunits are encoded by one and three genes, respectively, in the Arabidopsis genome. The three paralogs genes SAela, SAelbl, and SAelb2 are the result of two independent duplication events. Since SAelbl and SAelb2 correspond to two identical cop- ies, only two e1 small subunit isoforms are present in vivo： SAela and SAelb. The e1 heterodimer nuclear localization is modulated by the C-terminal tail of the SAe2 subunit. In vitro, SUMO conjugation rate is dependent on the SAe1 isoform contained in the e1 holoenzyme and our results suggest that downstream steps to SUMO-e1 thioester bond formation are affected. In vivo, SAela isoform deletion in T-DNA insertion mutant plants conferred sumoylation defects upon abi- otic stress, consistent with a sumoylation defective phenotype. Our results support previous data pointing to a regula- tory role of the e1 activating enzyme during SUMO conjugation and provide a novel mechanism to control sumoylation in vivo by diversification of the e1 small subunit.

The Joumey of the Magi： A Lyric Monologue for First and Second Voices and Three-in-One Character（s）

Although T. S. eliot＇s ＂The Journey of the Magi＂ is a religious poem in the profoundest sense, the title of my paper is intended to give only a sly wink at Trinitarianism. My real object is to explain how eliot contrived to manufacture a poem which, at fu＇st glance, resembles a dramatic monologue （generally understood as a poem for one voice----that of a historical/fictional/mythological character addressing a silent listener, group of listeners or reader）, yet which is slowly revealed as a lyrical monologue （for the poet＇s own voice） which yet--and this quite intentionally----contains considerably more than mere echoes of another two speakers： namely a Magus and the biblical translator and, most famously, sermon writer Archbishop Launcelot Andrewes （1555-1626） court preacher to James 1 and Charles 1 of england. I wish to show how eliot, in writing what is ultimately confessional verse, goes out of his way to hoodwink the reader by allowing the first two of his ＂{The} Three Voices of Poetry＂（1957） to overlap with and then incorporate the third. His own descriptions of these voices are （i） lyric, defined as ＂the poet talking to himself＂, （ii） that of the single speakerwho gives a （dramatic） monologuel ＂addressing an {imaginary} audience in an assumed voice＂ and （iii） that of the verse dramatist ＂who attempts to create a dramatic character speaking in verse when he {i.e. the author} is saying.., only what he can say within the limits of one imaginary character addressing another imaginary character＂ yet adding ＂some bit of himself that the author gives to a character may be the germ from which that character starts＂（eliot, 1957, pp. 38, 40）. The basis of my argument is that such an act of＂giving of the self＇ as the raw material for the creation of a dramatic monologue persona as well as a character designed for the stage had been part and parcel of eliot＇s modus operandi up to and including ＂Prufrock＂ and The Waste Land; further, that in ＂The Journey of the Magi＂ an
