Does granulocyte-colony stimulating factor administration induce damage or repair response in schistosomiasis?

AIM:To introduce Granulocyte-colony stimulating factor (G-CSF) as a new therapeutic modality for schistosomiasis through stem cell mobilization,immunomodulation or fibrosis remodeling. METHODS:In this study,a 5 d course of human recombinant G-CSF (100 μg/kg sc) was applied to Schis-tosoma mansoni-infected mice at different stages of disease (5 d before infection as well as 3,5 and 7 wk post-infection). The animals were sacrificed at 10 d as well as 4,6 and 8 wk post infection. Mice were examined for:(1) Total leukocyte count which is an accepted surrogate marker for the stem cell mobilization into the circulation; (2) Egg count in intestine and liver tissue to assess the parasitic load; and (3) Histopathological changes in Hx/E and Masson trichrome stained sections as well as collagen content in Sirius redstained liver sections to determine the severity of liver fibrosis. RESULTS:Mice developed leukocytosis. The egg load and the number of granulomas were not affected by the G-CSF treatment but there was an obvious change in the composition of granulomas towards an increased cellularity. Moreover,fibrosis was significantly decreased in treated groups compared to untreated animals (collagen content either preinfection or at 3 and 5 wk post infection:5.8 ± 0.5,4.7 ± 0.5,4.0 ± 0.7 vs 8.2 ± 0.9; P ≤ 0.01). CONCLUSION:Although G-CSF did not cause direct elimination of the parasite,it enhanced granulomatous reaction and reduced the fibrosis. Further investigation of the underlying mechanisms of these two actions is warranted.

This study aimed to evaluate the anti-hyperglycemic and antioxidant role of Actinidia deliciosa(kiwifruits)aqueous extract in streptozotocin-treated *** were distributed into;control,*** aqueous extract(ADAE;1 g/kg orally),streptozotocin(STZ;50 mg/kg,i.p,single dose),and STZ plus ADAE,*** showed that ADAE had high antioxidant and radical scavenging *** in blood sugar level,lipid peroxidation(LPO),kidney function biomarkers,and perturbations in hematological parameters were observed in diabetic ***,enzymatic and non-enzymatic antioxidants,protein content,and alkaline phosphatase(ALP)activity ***,histological,immunohistochemical alpha-smooth muscle actin immunoreactivity(α-SMA-ir)and histochemical(collagen,total protein,DNA,and RNA)alterations were observed in rat ***,STZ produced upregulation of inflammatory associated genes(tumor necrosis factor-alpha;TNF-αand transforming growth factorβ1;TGF-β1)and triggered apoptosis by upregulating apoptotic related gene[Bcl2-associated X protein(Bax)]and downregulating anti-apoptotic related gene B-cell lymphoma-2(Bcl-2)based on real-time PCR ***,diabetic rats administered with ADAE showed significant restoration in LPO,antioxidant status,and biochemical indices besides tissue architecture,and genes improvement regarding STZ ***,*** has a valuable ameliorative infl uence and can restore glucose levels and improve kidney dysfunction in diabetic rats.

The rapid population growth results in a crucial problem in the early detection of diseases inmedical *** all the cancers unveiled,breast cancer is considered the second most severe ***,an exponential rising in death cases incurred by breast cancer is expected due to the rapid population growth and the lack of resources required for performing medical *** recent advances in machine learning could help medical staff in diagnosing diseases as they offer effective,reliable,and rapid responses,which could help in decreasing the death *** this paper,we propose a new algorithm for feature selection based on a hybrid between powerful and recently emerged optimizers,namely,guided whale and dipper throated *** proposed algorithm is evaluated using four publicly available breast cancer *** evaluation results show the effectiveness of the proposed approach from the accuracy and speed *** prove the superiority of the proposed algorithm,a set of competing feature selection algorithms were incorporated into the conducted *** addition,a group of statistical analysis experiments was conducted to emphasize the superiority and stability of the proposed *** best-achieved breast cancer prediction average accuracy based on the proposed algorithm is 99.453%.This result is achieved in an average time of 3.6725 s,the best result among all the competing approaches utilized in the experiments.

Bariatric surgeries are often complicated by de-novo gastroesophageal reflux disease(GERD)or worsening of pre-existing *** growing rates of obesity and bariatric surgeries worldwide are paralleled by an increase in the number of patients requiring post-surgical GERD ***,there is currently no standardized approach for the assessment of GERD in these *** this review,we delineate the relationship between GERD and the most common bariatric surgeries:sleeve gastrectomy(SG)and Roux-en-y gastric bypass(RyGB),with a focus on pathophysiology,objective assessment,and underlying anatomical and motility *** suggest a stepwise algorithm to help diagnose GERD after SG and RyGB,determine the underlying cause,and guide the management and treatment.

目的对分化阶段中过表达RBP4的MC3T3⁃E1细胞进行定量蛋白组学分析,为后续研究颅缝早闭发病机制中异常成骨的研究奠定基础。方法使用LENTI⁃RBP4转染小鼠MC3T3⁃E1成骨细胞,使其过表达RBP4;培养收集传代细胞,提取蛋白,进行质谱分析,并通过Western blot实验来验证蛋白组学的结果。结果过表达RBP4小鼠成骨细胞中的差异蛋白主要集中在PI3K⁃AKT信号通路和MAPK信号通路。经Western blot验证,RBP4过表达组的Ikbkg、Akt和Col1a1表达量显著高于空载体组,而Nfkb1、Traf2、Cdk4、RAS、Mapk3和Braf表达量则明显下调。结论过表达RBP4可能通过同时激活PI3K信号通路和拮抗MAPK信号通路来促进成骨细胞分化。

Effectiveness of hepatitis B virus vaccination program in Egypt:Multicenter national project

AIM:To assess the effectiveness of hepatitis B virus(HBV) vaccination program among fully vaccinated ***:A national community based crosssectional study was carried out in 6 governorates representing Egypt. A total of 3600 children aged from 9 mo to 16 years who were fully vaccinated with HBV vaccine during infancy were recruited. Face to face interviews were carried out and sera were evaluated for hepatitis B surface antigen(HBsA g),anti-HBV core antibodies(total) and quantitative detection of hepatitis B surface antibody using enzyme linked immunoassays techniques. Samples positive to HBs Ag/anti-HBV core antibodies were subjected to quantitative HBV-DNA detection by real time polymerase chain reaction with 3.8 IU/L detection limit. RESULTS:Sero-protection was detected among 2059 children(57.2%) with geometric mean titers 75.4 ± 3.6 IU/L compared to 3.1 ± 2.1 IU/L among nonseroprotected children. Multivariate logistic analysis revealed that older age and female gender were the significant predicting variables for having non seroprotective level,with adjusted odds ratio 3.3,9.1and 14.2 among children aged 5 to years respectively compared to those years and 1.1 among girls compared to boys with P y of anti-HBc and DNA detection respectively. The prevalence of HBV infection was significantly higher among children aged ≥ 7 years(0.59%) compared to 0.07% among younger children with odds ratio equal to 8.4(95%CI:1.1-64.2) and P ys(0.29%) with P > 0.05. C ON C LU S I ON :T he E gy pt ian c ompuls or y H B V vaccination program provides adequate protection. Occult HBV infection exists among apparently healthy vaccinated children. Adherence to infection control measures is mandatory.
