Analyses of fin ray types to detect strontium markers in juvenile blunt-snout bream Megalobrama amblycephala

Background Restocking by introducing hatchery-reared fish into wild habitats aids in the restoration of fishery aquatic ecosystems and reefs to increase the abundance of fish resources, restore the ecological balance of water bodies, and enhance ecosystem functioning. Accurately, rapidly, and effectively evaluating the success of restocking using chemical markers（e.g., strontium [Sr]） remains challenging for fisheries management. consequently, for nonlethal fish sampling, hard tissues, such as fin rays, have received increasing attention as a target for marking method. However, data on the differences in marking different types of fin rays remain limited. Therefore, we exposed juvenile blunt snout bream individuals（Megalobrama amblycephala） to 0（control group） or 800 mg/L of Srcl2·6H2O（marked group） for 5 days and transferred them into normal aerated water for post-immersion culture. We sampled their pectoral, dorsal, ventral, anal, and caudal fin rays. The Sr marks among the fin types were sampled at 0 and 20 days post-immersion and evaluated using an electron probe micro-analyzer（EPMA） for the five-day Sr/ca ratios, along with line transect and Sr mapping *** Sr marking signatures were observed in all fin types in the marked group, with a success rate of up to 100%. Although marking efficiency varied among the different fin ray types, the highest Sr/ca ratios were most often detected in the dorsal fin. cross-sectional Sr concentration maps of all fin rays sampled showed high-Sr domains in the marked group; in contrast, the entire cross-sections of the control group displayed low Sr contents, indicating successful marking *** Fin ray Sr marking is a successful method for juvenile M. amblycephala, with the advantages of nonlethality and negligible sampling injuries, facilitating the rapid and effective evaluation of Sr marking in restocking M. amblycephala.

Asian carp are expanding their range throughout the Mississippi River; however, abundance is thought to be highest in reaches within close proximity to the Illinois River. In the Mississippi River, lock and Dam 19(L&D 19) at Keokuk, Iowa is the primary barrier to slow the expansion upstream. As Asian carp abundance increases below L&D 19, it is important to investigate potential means of control(i.e., reduction through harvest and barriers) that will prevent complete invasion of the Mississippi River above L&D 19. Silver carp and Bighead carp were collected below L&D 19, a subsample were implanted with ultrasonic transmitters to evaluate passage through the lock chamber and the remainder were used to determine population dynamics at the leading edge of invasion. Although the dam portion of the structure poses a complete barrier to upstream expansion, we documented lock chamber passage demonstrating the lock chamber has the capability to provide passage upstream. Based on the results of the population assessment, in order to induce recruitment overfishing at this leading edge of invasion, Asian carp will need to be intensively harvested at 300 mm and larger. The combination of commercial fishing efforts and research investigating ways to prevent passage upstream must be employed.

The establishment of a highly efficient planktivore, Silver carp(Hypophthalmichthys molitrix), in the Mississippi River Basin has been documented as having negative impacts on native planktivore populations. Silver carp have the potential, to alter the trophic food web through negative interactions with native planktivores in large river systems. Due to increasing Silver carp ranges, interactions between Silver carp and other native planktivores is likely to increase. One abundant native planktivore that is often found in similar habitats as Silver carp is Gizzard shad(Dorosoma cepedianum). Potential competition for resources exists because of overlapping habitat use and forage. Given the similarities between these two planktivores, we sought to evaluate daily growth rates and determine hatch dates of age-0 Silver carp and Gizzard shad in the Middle Mississippi River. Silver carp and Gizzard shad were collected from the Middle Mississippi River using an electrified surface trawl from June through September with sizes ranging from 21—65 mm total length for Gizzard shad and 24—85 mm total length for Silver carp. A sample of age-0 fish(n=697) was collected at the end of the growing season including 292 Gizzard shad and 405 Silver carp. Size structure(total length in mm) of Silver carp(49.05±0.41; ± SE) was significantly greater(T= –16.43, df=695, Pch dates were similar between Silver carp(July 26 th to August 18 th) and Gizzard shad(July 28 th to August 19 th). Growth of Silver carp(2.24 mm per day) was significantly greater(Pcarp(28.1%)and Gizzard shad(25.1%) at this early life stage. The faster growth rates in Silver carp result in Silver carp entering the winter season larger in size with likely an increased capacity to consume plankton and decreased predation ultimately leading to higher recruitment. Silver carp Hypophthalmichthys molitrix were introduced into the U.S. to control water quality in aquaculture ponds. From this point of origin, silver carp escaped into nearby rivers through multiple flood events. Because of their documented negative effects on native biota, silver carp have been labeled as problematic. Therefore, evaluating the biology and ecology of these non-indigenous species is critical. Multiple parameters are needed to evaluate silver carp populations(length, weight, age, and sex). Furthermore, developing methods for rapidly acquiring these data are needed. In relation to sex determination, sexual dimorphism was observed where males exhibit distinct pectoral fin ray features. Specifically, males have pronounced ridges or a "rough patch" on the dorsal surface of pectoral fins. Therefore, to test if this was an applicable way of determining silver carp gender; silver carp were collected from Midwestern U.S. rivers(N=2015) in the fall of 2011(N=870), spring of 2012(N=645), winter of 2013—2014(N=202) and summer2015(N=323) via electrofishing. For each silver carp collected, presence(e.g., rough patch) or absence(e.g.,smooth) of pronounced ridges on the top side of the pectoral fins was recorded, and an incision was made in the body cavity to identify gender. Based on the results of our evaluation, gender was correctly identified over99% of the time(2006 out of 2015) based on the pectoral fin dimorphism. In the samples taken in the winter of 2013—2014 and summer of 2015, accuracy for silver carp shorter than 300 mm and longer than 800 mm was 53.7%(19 out of 41) while accuracy for silver carp between 300 mm and 800 mm total length was 98.9%(289 out of 292). This study provides a rapid assessment approach for evaluating silver carp gender.

胰腺囊性纤维化(cystic fibrosis,cF)是一种单基因缺陷导致的致死性遗传疾病,在高加索人种中广泛分布。这种疾病在其它人种的发生率非常低,但据报道大部分人种中发现有该基因的突变。本文对cF发生的分子和病理生理学基本概念进行阐述。首先,阐述了cF的病理学和遗传特征,其基因产物囊性纤维化跨膜电导调节体(cystic fibrosis transmembrane con- ductance regulator,cFTR)的分子结构、特征、功能和调控。其次,由于突变的主要表现是电解质转运失调,其病理学效应和机制在两个典型受累器官中得到了很好的阐明,一个是汗腺,其病理发生是由于分泌过多Nacl,另一个是胰腺,其病理发生是由于分泌太少HcO_3^-。然而,cF的发病率和死亡率主要来自难治性呼吸道感染,其发生机制存存争议,我们推断可能的机制为阴离子转运失调导致cF肺部慢性感染。

BAcKGROUND： The placenta is recognized as an endocrine organ, largely due to its secretions of steroid hormones, including progesterone, androgens, and estrogens. Steroid hormones play an essential role in the progression of pregnancy, fetal development, and growth. Furthermore, steroids are necessary for establishment and maintenance of a normal pregnancy, preparing the endometrium for implantation, stimulating endometrial secretions, and regulating uterine blood flow, however the exact mechanism of sex steroid signaling through their receptors in placental function is unknown. OBJEcTIVE： In this review, we will provide an overview of the current knowledge on sex steroid receptors in normal placental development, as well as evidence of abnormal signaling associated with placental dysfunction. METHODS： A systematic literature search was performed using the NcBI PubMed search engine, including the following key works： estrogen receptor, androgen receptor, placenta, placental development, cytotrophoblast, and differentiation. RESULTS： Of the over 700 articles that were returned, 125 studies focused on estrogen and androgen receptors in human placenta development and function during normal and abnormal pregnancy, as well as in rodents and ruminants placentae. cONcLUSION： Receptors for both estrogens and androgens have been localized within the mammalian placenta, but surprisingly little is known about their signaling in trophoblast cell differentiation and function. An emerging picture is developing in which estrogen receptors possibly play role in cytotrophoblast proliferation and extravillous trophoblast invasion, whereas androgen receptors are involved in syncytiotrophoblast differentiation and function.

Accurately quantifying rates of soil erosion requires capturing both the volumetric nature of the visible,convergent fluvial pathways(also known as rills)and the subtle nature of the less-visible,diffuse pathways(interrill areas).The aim of this study was to use Rare Earth Oxide(REO)tracers and Structure-from-Motion(SfM)photogrammetry to elucidate retrospective information about soil erosion rates and sediment sources during different soil erosion conditions,within a controlled laboratory *** experimental conditions created erosion events consistent with diffuse and convergent erosion *** tracers allowed the sediment transport distances of over 2 m to be described,and helped resolved the relative contribution of diffuse and convergent soil erosion;interrill areas were also iden-tified as a significant sediment sources soil loss under convergent erosion *** the potential for SfM photogrammetry to resolve sub-millimetre elevations changes was demonstrated,under some conditions non-erosional changes in surface elevation,such as compaction,exceeded volumes of soil loss via diffuse *** discrepancies between SfM Photogrammetry calculations and REO tagged sediment export were beneficial,identifying that during soil erosion events sediment in both aggregate and particle form is deposited within the convergent features,even when the rill extended the full length of the soil *** combination of SfM photogrammetry and REO tracers has provided a novel platform for building a spatial understanding of patterns of soil loss and source apportionment between rill and interrill erosion.

Ovine hippocampal mRNA expression in offspring from dams supplemented with fishmeal and stress challenged in late pregnancy with endotoxin

Previous research has shown that adulthood disease can be attributed to stress events that occur during gestation. The objective of the present study was to determine whether maternal stress during late pregnancy, using a bacterial endotoxemia model, causes changes in hippocampal mRNA expression of candidate genes related to hypothalamic-pituitary-adrenal axis(HPAA) regulation in sheep. This study also sought to investigate whether maternal diets supplemented with fishmeal(FM) rich in omega-3 polyunsaturated fatty acids(PUFAs) offer protection to the fetus when subjected to maternal endotoxin stress. Using RT-qPcR, relative mRNA expression was assessed in both fetal lambs and 6-month-old lambs from dams supplemented with soybean meal(SM) or FM and challenged with either endotoxin or saline. From this it was found that fetal mineralocorticoid receptor(MR) gene expression had a tendency to be altered during endotoxin challenge, however, in the 6-month-old offspring, MR and glucocorticoid receptor(GR) gene expression were differently altered across treatment groups. These results suggest that gene regulation within the hippocampus is altered into adulthood by maternal endotoxin stress and that dietary supplementation affects outcome.
