This experiment aimed to discuss and reveal the effect and mechanism of mannanase on intestinal inflammation in broilers triggered by a soybean meal diet. In this experiment, 384 Arbor Acres broilers at1 d old were randomly divided into 3 treatment groups. The broilers were fed a corn-soybean meal basal diet, a low-energy diet(metabolizable energy reduced by 50 kcal/kg), and a low-energy diet supplemented with 100 mg/kg mannanase for 42 d. The low-energy diet increased feed conversion ratio from0 to 42 d, reduced ileal villus height and villus height-to-crypt depth ratio and upregulated the expression of nuclear factor kappa B(NF-κB) in the ileum(P lium of broilers(P < 0.05). In addition, mannanase upregulated the expression of claudin-1(CLDN1) and zonula occludens-1(ZO-1) in the jejunum at 21 d,downregulated the expression of ileal NF-κB, and increased the content of isobutyric acid in the cecum of broilers(P < 0.05). The results for the ileal microbiota showed that a low-energy diet led to a decrease in the relative abundance of Lactobacillus reuteri in the ileum of broilers. The addition of mannanase increased the relative abundance of Lactobacillus-KC45b and Lactobacillus johnsonii in broilers. Furthermore, a low-energy diet reduced the relative abundance of Butyricicoccus in the intestine of broilers and inhibited oxidative phosphorylation and phosphoinositol metabolism. Mannanase increased the relative abundance of Odoribacter, promoted energy metabolism and N-glycan biosynthesis, and increased the activities of GH3 and GH18. Neuropeptides are crucial in regulation of a rich variety of developmental,physiological,and behavioral functions throughout the life cycle of *** an integrated approach of multiomics,we identified neuropeptide precursors in the greater wax moth Galleria mellonella,which is a harmful pest of honeybee hives with a worldwide ***,a total of 63 and 67 neuropeptide precursors were predicted and annotated in the *** genome and transcriptome,in which 40 neuropeptide precursors were confirmed in the *** ***,we identified 12 neuropeptide precursor genes present in *** but absent in honeybees,which may be potential novel pesticide target *** hives were contaminated with heavy metals such as lead,enabling its bioaccumulation in *** bodies through the food chain,we performed transcriptome sequencing to analyze the effects of Pb stress on the mRNA expression level of *** neuropeptide *** treatment by Pb,the expression of neuropeptide F1 was found to be significantly downregulated,implying that this neuropeptide might be associated with responding to the heavy metal stress in *** study comprehensively identified neuropeptide precursors in ***,and discussed the effects of heavy metals on insect neuropeptides,with the example of *** results are valuable for future elucidation of how neuropeptides regulate physiological functions in *** and contribute to our understanding of the insect's environmental plasticity and identify potential new biomarkers to assess heavy metal toxicity in insects.

Covalent organic frameworks(COFs)have garnered increasing attention as promising electrode materials for sodium-ion batteries(SIBs)due to their ordered backbones,uniform pore sizes,and high surface ***,they also face challenges,including low capacity,unsatisfactory rate performance,and limited cycling stability,which pose the primary obstacles to their practical ***,a pyrenoimidazole-based COF,denoted as PyIm-COF,has been synthesized as a novel electrode material for highperformance SIBs using an efficient synthetic *** extended fully conjugated backbones and readily accessible redox-active sites,PyIm-COF demonstrates remarkable high-rate performance,delivering around 250 m Ah g^(−1)and excellent cycling stability,retaining 97.2%of its capacity even after 2500 cycles at 5 A g^(−1),which is higher than that of most previously reported COF-based *** work provides valuable insights into the development of nitrogen-rich conjugated COF electrode materials for rechargeable SIBs.

Previous studies through targeted mutagenesis of K-D-K-E motif have demonstrated that 2′-O-MTase activity is essential for efficient viral replication and immune ***,the K-D-K-E catalytic motif of 2′-O-MTase is highly conserved across numerous viruses,including flaviviruses,vaccinia viruses,coronaviruses,and extends even to ***,we observed a stronger 2′-O-MTase activity in SARS-CoV-2 compared to SARS-CoV,despite the presence of a consistently active catalytic *** further identified critical residues(Leu-36,Asn-138 and Ile-153)which served as determinants of discrepancy in 2′-O-MTase activity between SARS-CoV-2 and *** residues significantly enhanced the RNA binding affinity of 2′-O-MTase and boosted its versatility toward RNA *** interest,a triple substitution(Leu^(36)→Ile^(36),Asn^(138)→His^(138),Ile^(153)→Leu^(153),from SARS-CoV-2 to SARS-CoV)within nsp16 resulted in a proportional reduction in viral 2′-O-methylation and impaired viral ***,it led to a significant upregulation of type I interferon(IFN-I)and proinflammatory cytokines both in vitro and vivo,relying on the cooperative sensing of melanoma differentiation-associated protein 5(MDA5)and laboratory of genetics and physiology 2(LGP2).In conclusion,our findings demonstrated that alterations in residues other than K-D-K-E of 2′-O-MTase may affect viral replication and subsequently influence *** changes in nsp16 residues is crucial as it may aid in identifying and assessing future alteration in viral pathogenicity resulting from natural mutations occurring in nsp16.

The aqueous zinc ion batteries(AZIBs)are thought as promising competitors for electrochemical energy storage,though their wide application is curbed by the uncontrollable dendrite growth and gas evolution side ***,to stabilize both zinc anodes and water molecules,we developed a modified electrolyte by adding a trace amount of N,N-diethylformanmide(DEF)into the ZnSO_(4)electrolyte for the first time in zinc ion *** effectiveness of DEF is predicted by the comparison of donor number and its preferential adsorption behavior on the zinc anode is further demonstrated by several spectroscopy characterizations,electrochemical methods,and molecular dynamics *** modified electrolyte with 5%*** content can ensure a stable cycling life longer than 3400 h of Zn‖Zn symmetric cells and an ultra-reversible Zn stripping/plating process with a high coulombic efficiency of 99.7%.The Zn‖VO_(2)full cell maintains a capacity retention of 83.5%and a 104 mA h g^(-1)mass capacity after *** work provides insights into the role of interfacial adsorption behavior and the donor number of additive molecules in designing low-content and effective aqueous electrolytes.
