Benefits of the Combined Use of NSAID and Early Exercise on Tendon Healing in a Murine Model

Achilles tendon rupture is one of the most frequent tendon injuries. The aim of this experimental study was to evaluate the effect of a non-steroidal anti-inflammatory drug (NSAID) and of aerobic exercise, in an isolated fashion and combined, on cell proliferation and on the biomechanical aspects of the Achilles tendon during healing process after a complete tenotomy in 156 male Wistar rats. Half of the animals received an intra-muscular injection of tenoxicam for 7 days and exercise was initiated on the 8th day for half the animals of each group. Animals were sacrificed at 1, 2, 4 and 8 weeks after the tenotomy and cell proliferation was evaluated by immunohistochemistry for PCNA, biomechanical evaluation was performed with ultimate load and gait cycle analysis was also carried out. Animals were divided into four treatment groups: A) no NSAID and no exercise (control group);B) NSAID plus exercise;C) no NSAID, with exercise;D) NSAID and no exercise. We used the test of two-way analysis of variance with equal cells number, Kruskal-Wallis test and also, Bonferroni method, in the R Project program 3.5.0. The highest intensity of PCNA immunostaining was found at 2 weeks in all groups except for group A (control) that had the highest intensity at 1 week. Animals submitted to exercise had significantly higher (P = 0.02) ultimate loads when compared to animals that were not submitted to exercise at 8 weeks after injury. The animals that received NSAID presented with a more stable gait cycle than the animals that did not receive NSAID. Aerobic exercise, initiated early after a complete Achilles tendon tenotomy, was beneficial to the biomechanical aspects of the tendon that occur during tendon regeneration and the combined use of NSAID improved the gait’s characteristics which could be protective against re-ruptures.

Introduction: Among the extrinsic factors, homocysteine (Hy) stands out, which is an intermediate amino acid of the intracellular metabolism of methionine involved in the process of cellular oxidation, which promotes the installation of atheromatous plaques and, therefore, is considered as an emerging cardiovascular risk factor. Objective: To evaluate the plasma homocysteine levels (Hy) in overweight or obese children and adolescents and their relation with cardiovascular risk factors. Methods: A cross-sectional study was conducted from July 2011 to May 2012 with overweight or obesity children and adolescents aged 2 to 18 years followed at the Center for Childhood Obesity (IOC), Campina Grande-PB. A structured form was used to record demographic, socioeconomic and clinics variables and the patients underwent laboratory tests to define their lipid and glucose profiles and measurement of plasma Hy levels. Results: The study evaluated a total of 165 children and adolescents with mean age of 12.5 (±2.5) years;the majority were female (57.0%). Regarding the lipid profile, there was more individuals with low HDL cholesterol (88.5%). Plasma Hy levels were high in 24.2% of the sample. The mean Hy levels ranged from 4.3 to 18.9 μmol/L, being higher in males, obese adolescents and also in patients with high insulin levels and resistance. Conclusions: The results shown in this study emphasize the importance of detecting and controlling the plasma Hy levels as an independent cardiovascular risk factor, and the need for further studies to evaluate the clinical and biological factors related to alterations in its metabolism.

Freshwater environments are the main ecosystems subjected to various anthropogenic impacts, which have caused the waterborne transmission of diseases caused mainly by bacteria. The main objective of this work was to carry out a systematic analysis through the literature on the main microorganisms found in impacted aquatic environments and to relate the data with the risks to public health. This review was planned, conducted and reported following the PRISMA protocol, which was used the PECO strategy. There performed searches for studies using the electronic databases PubMed, Medline and LILACS in September 2021, using the controlled search strategy: “Bacteria” and “Ecosystem” and “Environment” and “Aquatic” and “Impacted. The studies that met the eligibility criteria were considered eligible, with the participation of two independent reviewers. There found 1058 studies, of which were chosen only 31 to compose the results of this systematic review. Because of the results, there observed that the predominant bacterial genera in freshwater are: Pseudomonas, Aeromonas, and Escherichia, with the Asian continent presenting more varieties of bacterial species detected. These microorganisms are responsible for causing diseases that can lead to death, which shows the importance of constantly monitoring these ecosystems.

Electrochemical Evaluation of Aminoguanidine Hydrazone derivative with Potential Anticancer Activity:Studies of Glassy Carbon/CNT and Gold Electrodes Both Modified with PAMAM

Aminoguanidine hydrazones (AGHs) are a class of compounds that have interesting pharmacological activities. They are derived from the same chemical group as aminoguanidine, so it has mixed properties (receptor and donor) in the formation of hydrogen bonds. Its anticancer agent properties were recently highlighted, but the molecules of this class have solubility in aqueous solutions that can be considered low. The identification of this class, by a simple, sensitive and low-cost technique, such as electrochemistry, which also allows the evaluation of its solubilization process through agents such as PAMAM dendrimer is the main objective of the work described here. The electrochemical response of the LQM10 (AGH derivative) was evaluated, as well as its behavior in different electrochemical sensors. Electrochemical experiments were performed in buffered (phosphate at pH 7.02 and acetate at 4.5). LQM10 has a reversible oxidation peak with a potential of +0.22 V. It was efficiently detected in different electrodes tested (glass carbon/CNT, glass carbon/CNT/PAMAM), which proves the viability of the electrodes for various analyses and has the determination of the apparent constant association, indicating its interaction with the analysis that is higher in the presence of the PAMAM encapsulating agent. This was corroborated by the results for the modified gold electrode with MUA and PAMAM. The sum of the results shows the possibility of electrochemically evaluating the Aminoguanidine hydrazone derivative, the viability of electrodes employed and the greater solubilization of LQM10 in the presence of the PAMAM dendrimer.

Recently the fetal membranes are regarded to be abundant, ethically acceptable and easily accessible sources of mesenchymal stem cells, which cause only minor immunogenic troubles. In addition, these membranes have been studied as an alternative to adult and embryonic stem cells. The chorion develops firstly as a layer of simple avascular epithelium (trophoblast). The amnion, as the chorion is also an avascular membrane. It is derived from the ectoderm, and forms a very thin membrane which surrounded the embryo in development. The hypoblast and exocoelomic membrane together originates the yolk sac which late will give rise to the allantois. The study of the origin and development of these tissues is of vital importance for obtaining stem cells. Different cell lineages can be obtained from the fetal placental tissues: the hematopoietic lineage is found in the chorion, allantois and, yolk sac, and the mesenchymal lineage is found in the chorion and amnion. The properties and potential for differentiation of fetal stem cells isolated from placental tissues must be characterized in order for them to be used in the treatment of several diseases. The chorion, yolk sac, allantois, and amnion contain heterogeneous cell populations. However, few studies have focused on characterizing these alternative stem cells sources. The amnion has a significant advantage over the other fetal membranes, mainly due to its easily accessibility for collection in humans and rodents and also because it contains populations of pluripotent and multipotent cells. In some other species (dogs, ruminants, horses, cats, and swine) the allantoic fluid is more easily accessible, but there is little information regarding the characterization of the cell population. This review will address advances in the isolation of stem cells from fetal placental tissues and describe their characterization and possible use in cell therapy, as well as their origin and development.

Objective: Although Angelica archangelica is a medicinal and aromatic plant with a long history of use for both medicinal and food purposes, there are no studies regarding the antineoplastic activity of its *** study aimed to evaluate the cytotoxicity and antitumor effects of the crude extract of A. archangelica root(CEAA) on breast ***: The cytotoxicity of CEAA against breast adenocarcinoma cells(4T1 and MCF-7) was evaluated by a 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide(MTT) assay. Morphological and biochemical changes were detected by Hoechst 33342/propidium iodide(PI) and annexin V/PI staining. Cytosolic calcium mobilization was evaluated in cells staining with FURA-4 NW. Immunoblotting was used to determine the effect of CEAA on anti-and pro-apoptotic proteins(Bcl-2 and Bax, respectively).The 4 T1 cell-challenged mice were used for in vivo ***: Using ultra-high-performance liquid chromatography–mass spectrometry analysis, angelicin, a constituent of the roots and leaves of A. archangelica, was found to be the major constituent of the CEAA evaluated in this study(73 mg/mL). The CEAA was cytotoxic for both breast cancer cell lines studied but not for human fibroblasts. Treatment of 4 T1 cells with the CEAA increased Bax protein levels accompanied by decreased Bcl-2 expression, in the presence of cleaved caspase-3 and cytosolic calcium mobilization, suggesting mitochondrial involvement in breast cancer cell death induced by the CEAA in this cell line. No changes on the Bcl-2/Bax ratio were observed in CEAA-treated MCF7 cells. Gavage administration of the CEAA(500 mg/kg) to 4T1 cell-challenged mice significantly decreased tumor growth when compared with untreated ***: Altogether, our data show the antitumor potential of the CEAA against breast cancer cells in vitro and in vivo. Further research is necessary to better elucidate the pharmacological application of the CEAA in breast cancer therapy.

Urinary bladder cancer(UBC)is a heterogeneous disease with highly variable clinical outcomes and responses to *** some advances in the molecular understanding of UBC,this knowledge still has not been translated to the clinic in terms of improvements in the prognosis and treatment of *** urinary bladder tumor models representative of the human disease in terms of histology and behavior are needed to study factors involved in tumor initiation,progression and ***,accurate model systems would facilitate identification of new therapeutic targets and predictive markers that could lead to optimization of existing therapies and development of new *** established cancer cell lines derived from human urinary bladder tumors representing different grades and stages have been used as experimental models for UBC *** cell lines reflect some of the genetic and morphologic alterations observed in human urothelial carcinoma and serve as simplified models to study the behavior of cancer cells in ***,their translational potential is limited due to the artificial conditions,in which the cells are maintained,grown and *** models offer a more complex and realistic model for the establishment,development,and progression of tumors as well as to evaluate new therapeutic *** the years,the authors'group has worked with several UBC cell lines,established and characterized chemically induced UBC models,and patient-derived xenografts *** this study,the authors will provide a summary of the UBC models developed by their group,analyze their translational potential and weaknesses,and define areas that remain to be explored.

Ex-Patients with COVID-19: Observations after Healing and Its Consequences*

During the SARS-Cov-2 or COVID-19 clinical course pandemic of the coronavirus family, we observed symptoms of the respiratory tract, fever, cough, sore throat, and fatigue. The most common symptoms were loss of smell and taste and, when aggravated, it could develop into pneumonia and even death. We selected 40 former COVID-19 patients, who answered a questionnaire with questions about the symptoms and complications after they were cured and which symptoms remained or appeared after the total isolation quarantine for a specific time. Among the answers, the most cited was about “forgetting”. Another observation noted was the number of cases in children, which surprised us, considering that, at the beginning of the pandemic, the danger was more related to the elderly population and those with comorbidities.
