Methodology of the Tetrazolium Test for Evaluating Physiological Quality in Pitaya (<i>hylocereus undatus</i>) Seeds

The germination test is routinely used for evaluating physiological quality of seeds, but it has not been satisfactory, since it requires relatively long periods to obtain results. In this sense, there is the possibility to resort to vigor tests, although the absence of standardized methodologies has hindered your applicability and reproducibility. The goal of the present study was to establish a methodology for the tetrazolium test that is effective for evaluating physiological quality in pitaya seeds. So, we used five seed batches obtained from mature fruits of pitaya (hylocereus undatus), harvested in the years of 2008, 2009, 2010, 2011, and 2012. The experimental design was the completely randomized design, with four replicates of 50 seeds. The tetrazolium test was conducted in a 5 × 4 × 3 factorial plot, corresponding to five batches of pitaya seeds (2008, 2009, 2010, 2011, and 2012), four concentrations of the tetrazolium solution (0.075%, 0.1%, 0.5%, and 1.0%) and three immersion periods (2, 3, and 4 h). In addition, the germination test for the seed batches was carried out and the analyzed variables were: percentage of germination (PG), germination speed index (GSI) and mean germination time (MGT). Data were submitted to ANOVA and means were compared by the Scott-Knott test (p ≤ 0.05). Tetrazolium test conducted at a solution concentration of 0.5% and an immersion period of 3 h proved to be efficient for evaluating physiological quality of pitaya seeds, in order to stratify the seed batches into more viability levels compared to the germination test.

＜正＞ A calibration bench was developed and constructed to calibrate domestic water meters, especially for the needs of the water distribution companies. The bench could calibrate up to 6 water meters at the same time in the present version. It was designed to work with water and in normal operation with two reservoirs and a battery of working standards, electromagnetic flowmeters of special grade. The mag meters assure an uncertainty of less than 0,3%, because they are calibrated on the spot: there is an electronic scale in the circuit of the bench that can be used at any time to calibrate the secondary standards or the meters under test. With this, this bench is almost a "primary" laboratory. A centrifugal pump feeds the constant head level tank and the water in excess turns back to the lower reservoir, assuring that there is no influence of the level of water, or of its oscillation due to the pump movement, in the calibration of the meter. For the highest flow rates, the pump is used in direct connection to the meters under test, with no passage through the upper reservoir. The range of operation is from 2 L/h up to 7500 L/h. The control is totally automated using a PLC and data is stored in an electronic data sheet.

Background Spontaneous supratentorial intracerebral haemorrhage accounts for 20% of all stroke- related sudden neurological deficits, has the highest morbidity and mortality of all stroke, and the role of surgery remains controversial. We undertook a prospective randomised trial to compare early surgery with initial conservative treatment for patients with intracerebral haemorrhage. Methods A parallel- group trial design was used. Early surgery combined haematoma evacuation (within 24 h of randomisation) with medical treatment. Initial conservative treatment used medical treatment, although later evacuation was allowed if necessary. We used the eight- point Glasgow outcome scale obtained by postal questionnaires sent directly to patients at 6 months follow- up as the primary outcome measure. We divided the patients into good and poor prognosis groups on the basis of their clinical status at randomisation. For the good prognosis group, a favourable outcome was defined as good recovery or moderate disability on the Glasgow outcome scale. For the poor prognosis group, a favourable outcome also included the upper level of severe disability. Analysis was by intention to treat. Findings 1033 patients from 83 centres in 27 countries were randomised to early surgery (503) or initial conservative treatment (530). At 6 months, 51 patients were lost to follow- up, and 17 were alive with unknown status. Of 468 patients randomised to early surgery, 122 (26% ) had a favourable outcome compared with 118 (24% ) of 496 randomised to initial conservative treatment (odds ratio 0.89 [95% CI 0.66- 1.19], p=0.414); absolute benefit 2.3% (- 3.2to 7.7), relative benefit 10% (- 13 to 33). Interpretation Patients with spontaneous supratentorial intracerebral haemorrhage in neurosurgical units show no overall benefit from early surgery when compared with initial conservative treatment.

Evaluation of Thermal Comfort Level in Social Interest housing with Earth Tubes for the Brazilian Bioclimatic Zones 1, 2 and 7

The expansion of social housing in Brazil is solving the great demand among low-income populations, ltowever, these projects are not considering climatic factors and thermal comfort strategies. There is a standardization of architectural projects, which leads to the user discomfort. Bioclimatic solutions that improve the thermal performance of buildings should be valued in the projects of buildings. This paper presents a comparative study noticing the thermal comfort provided by a earth tube ventilation system with a natural ventilation system based in the opening of the windows. In this study, three Brazilian bioclimatic zones defined by NBR （Brazilian Standard） 15220 （2005）, Zone 1 （Curitiba）, 2 （Camaqu~,） and 7 （Cuiabfi）, were considered. To make this comparison, computer simulation was used to observe the thermal confort conditions in RTQ-R （Quality Technical Standard for Energy Efficiency Level in Residential Buildings）（2010）, with the help of Energy Plus software and Schektch Up with the plug in Open Studio for modeling the residence. After the energy modeling, the results were analyzed with the Analisys Bio software in order to obtain the percentage of hours of comfort for the year （8,760 h）. The conclusion is drawn that the natural ventilation strategy with the opening and closing of the windows is more efficient in Bioclimatic Zone 1. For the Bioclimatic Zone 2, the earth tubes should be used only in the summer and, in Bioclimatic Zone 7, earth tubes are efficient throughout the year because of the high temperatures in the exterior environment.

Effects of Gabaergic Phenols on Phospholipid Bilayers as Evaluated by <sup>1</sup>h-NMR

The phenols propofol and thymol, and lately carvacrol, eugenol and chlorothymol, have been shown to act as positive allosteric modulators on GABAA receptor, which is the main inhibitory receptor of the central nervous system. GABAA receptor is an intrinsic membrane protein which activity may be affected by surface-active compounds and by physical changes in the membrane. Recently, we demonstrated that these phenols interacted with the lipid membrane phase, suggesting their anesthetic activity could be the combined result of their specific (with receptor proteins) as well as nonspecific (with surrounding lipid molecules) interaction modulating the supramolecular organization of the receptor environment. In the current study, by using 1h-NMR spectroscopy, we have investigated the effects of the insertion and the possible preferential location of the five phenol derivatives with GABAergic activity on EPC membranes. The results indicate that all compounds are able to insert in EPC phospholipid vesicles and to locate in the region between the polar group (choline molecule), the glycerol and the first atoms of the acyl chains, being the more lipophilic compounds (propofol and chlorothymol) that seem to prefer a deeper bilayer insertion. The location of the phenol molecules would reduce the repulsive forces among phospholipids head groups allowing closer molecular packing and finally diminishing the mobility of the hydrocarbon chains, as revealed by 1h spin relaxation times.

AIM：To investigate the cytotoxic effects of spray-dried extracts of Phyllanthus niruri in combination with cis- platin on two cancer cell lines. METhODS： Colorectal carcinoma （hT29） and human hepatocellular carcinoma （hepG2） cells were treated with spray-dried extracts of Phyllanthus niruri （SDEPN） either alone or in combination with cisplatin at differ- ent concentrations （0.5 mg/mL and 1 mg/mL） for 4 h and 24 h. To verify and quantify cancer cells treated with these products as well as identify the cell cycle stage and cell viability, we stained the cells with prop- idium iodide and assessed them by flow cytometry. The percentage of cells in different cell cycle phases was quantified and data were expressed as histo- grams. Significant differences between groups were determined using analysis of variance and Bonferroni＇s test, as indicated. A value of P 〈 0.05 was considered to be statistically significant. RESULTS： SDEPN had significantly different cyto- toxic effects on hT29 （2.81 4- 0.11 vs 3.51 4- 1.13, P 〉 0.05） and hepG2 （5.07± 0.3 vs 15.9 ± 1.04, P 〈 0.001） cells when compared to control cells for 4 h. SDEPN also had significantly different cytotoxic effects on hT29 （1.91 ± 0.57 vs 4.53± 1.22, P 〉 0.05） and hepG2 （14.56 ± 1.6 vs 35.67 ± 3.94, P 〈 0.001） cells when compared to control cells for 24 h. Both cell lines were killed by cisplatin in a dose-dependent manner compared to control cells （hepG2 cells for 4 h： 10.78 ± 1.58 vs 53.89 ± 1.53, P 〈 0.001; 24 h： 8.9 ± 1.43 vs 62.78 ± 1.87, P 〈 0.001 and hT29 cells for 4 h： 9.52 ±0.913 vs 49.86 ± 2.89, P 〈 0.001; 24 h： 11.78 ± 1.05 vs 53.34 ± 2.65, P 〈 0.001）. In hT29 cells, pretreat- ment with SDEPN and subsequent treatment with cis-platin resulted in a greater number of cells being killed （12.78 ± 1.01 vs 93.76 ± 1.6, P 〈 0.001）. hepG2 cells showed significant cell killing with treatment with SDEPN when combined with cisplatin （12.87 ± 2.78 vs 78.8 ± 3.02, P 〈 0.001）. Functional Morphology of the Spermatogenesis of Gir Bulls

The purpose of this study was to evaluate the functional morphology of the spermatogenesis of Gir bulls, with emphasis on the testicular biometry and testicular parenchyma histomorphometry. Testicular fragments from eight Gir bulls were used. The fragments were fixed by tissue perfusion with Karnovsck’s solution, and inclusion was subsequently performed using glycol methacrylate. histological sections of 4 μm were made and stained with a 1% toluidine blue-sodium borate solution. The mean age of the bulls was 8.0 ± 1.3 years and the mean body weight was 467.5 ± 26.7 kg. Testicular weight was 289.2 ± 30.5 g on average, and the average gonadosomatic index was 0.12% ± 0.02%. The seminiferous tubules, Leydig cells, stroma, and intertubular tissue composed 80.9% ± 1.7%, 5.4% ± 1.3%, 13.7% ± 1.1%, and 19.1% ± 1.7% of the total testicular weight, respectively. The mean length of the seminiferous tubule per gram of testicle was 17 ± 0.8 m. Losses during spermatogonia mitosis averaged 75.5% ± 3.22%, while losses were 30.6% ± 8.17% during the meiotic phase, which resulted in an average total loss of 81.49% ± 2.58% of cells during the entire process of spermatogenesis. The average daily sperm production per gram of parenchyma was 28.0 × 106 cells. It was concluded that the histometric measurements of testicular parameters in the Gir breed are within the averages reported for other zebu cattle breeds.
