Copper,permalloy,cobalt,and silicon are the materials that have been widely utilised in magnetic *** the size of interest is down to the nanoscale,the inter-diffusion between certain materials becomes *** paper studies the nanoscale friction characteristics between frictional pairs with and without inter-diffusion properties through the atomic force *** distinct evolution features of nanoscale friction force when inter-diffusion is involved are discovered experimentally,which is also confirmed through theoretical ***,through the thin film deposition method,four pairs of contact materials(Cu–Ni_(81)Fe_(19),Si–Ni_(81)Fe_(19),Cu–Co,Cu–Si)are designed for friction tests,in which diffusion occurs at the interface of Cu–Ni_(81)Fe_(19)***,the effects of sliding velocity and loading force on the nano friction of each pair are *** is found that regardless of the diffusion phenomenon:(1)the adhesion force values exhibit a notable correlation to the values of the friction force;(2)the friction force in all four material pairs consistently increases with the growth of the normal loading force,although the growth rate may *** terms of the sliding velocity effect,the friction forces of immiscible materials(Si–Ni_(81)Fe_(19),Cu–Co,and Cu–Si)are found to increase with the increasing sliding ***,the friction force of Cu–Ni_(81)Fe_(19),decreases with the increasing sliding ***,a compositive friction model considering both the velocity and the normal force effect was proposed,which shows good agreement with the experimental results and explains the nano friction behaviour of both miscible and immiscible metals.

Objective: To explore the effect of cytosolic phospholipase A2α(cPLA2α) on hepatocellular carcinoma(HCC) cell adhesion and the underlying ***: Cell adhesion, detachment, and hanging-drop assays were utilized to examine the effect of cPLA2α on the cell-matrix and cell-cell adhesion. Downstream substrates and effectors of cPLA2α were screened via a phospho-antibody *** signaling pathways were identified by the functional annotation tool DAVID. Candidate proteins were verified using Western blot and colocalization was investigated via immunofluorescence. Western blot and immunohistochemistry were used to detect protein expression in HCC tissues. Prognosis evaluation was conducted using Kaplan-Meier and Cox-proportional hazards regression ***: Our findings showed that cPLA2α knockdown decreases cell-matrix adhesion but increases cell-cell adhesion in HepG2 cells. Microarray analysis revealed that phosphorylation of multiple proteins at specific sites were regulated by cPLA2α. These phosphorylated proteins were involved in various biological processes. In addition, our results indicated that the focal adhesion pathway was highly enriched in the cPLA2α-relevant signaling pathway. Furthermore, cPLA2α was found to elevate phosphorylation levels of FAK and paxillin, two crucial components of focal adhesion. Moreover, localization of p-FAK to focal adhesions in the plasma membrane was significantly reduced with the downregulation of cPLA2α. Clinically, cPLA2α expression was positively correlated with p-FAK levels. Additionally, high expression of both cPLA2α and p-FAK predicted the worst prognoses for HCC ***: Our study indicated that cPLA2α may promote cell-matrix adhesion via the FAK/paxillin pathway, which partly explains the malignant cPLA2α phenotype seen in HCC.

Objective:Epithelial cancers often originate from progenitor cells,while the origin of hepatocellular carcinoma(HCC)is still ***,one of the deadliest cancers,is closely linked with liver injuries and chronic inflammation,which trigger massive infiltration of bone marrow-derived cells(BMDCs)during liver ***:To address the possible roles of BMDCs in HCC origination,we established a diethylnitrosamine(DEN)-induced HCC model in bone marrow transplanted *** and frozen tissue immunofluorescence were used to verify DENinduced HCC in the pathology of the *** cellular origin of DEN-induced HCC was further studied by single cell sequencing,single-cell nested PCR,and immunofluorescence-fluorescence in situ ***:Studies by using single cell sequencing and biochemical analysis revealed that HCC cells in these mice were coming from donor mice BMDCs,and not from recipient ***,the copy numbers of mouse orthologs of several HCC-related genes previously reported in human HCC were also altered in our mouse ***-induced HCCs exhibited a similar histological phenotype and genomic profile as human ***:These results suggested that BMDCs are an important origin of HCC,which provide important clues to HCC prevention,detection,and treatments.

OBJECTIVE To investigate the effect of endostatin and doxycycline on melanoma cellular proliferation and tumor *** The effects of endostatin and doxycycline were studied in mice transplanted with B16 melanoma *** mice were divided into 4 groups that were treated as follows:endostatin treatment(E group),doxycycline treatment(D group),endostatin plus doxycycline trearment(DE group),controls(C group) received no *** 9 days of treatment the tumor tissue was removed to compare the differences in the tumor necrotic rate and micro-vessel density(MVD) among the different *** staining was conducted to detect the expression of proliferating cell nuclear antigen(PCNA)in the di?erent groups. RESULTS The MVD of the 3 experimental groups was significantly less than the control group,(F=10.888,P<0.05),indicating that doxycycline and endostatin can inhibit tumor angiogenesis by decreasing the tumor blood *** effect results in inhibition of tumor cellular proliferation and promotion of tumor cell *** tumor cell necrotic rate of the 3 experimental groups were all significantly higher than the C group(F=7.229,P<0.05) and the difference between the DE and C groups also was statistically *** expression in all 3 experimental groups was statistically less than the C group(F=17.729,P<0.05). CONCLUSION The combined use of endostatin and doxycycline in vivo can influence PCNA expression and angiogenesis in melanoma,and significantly inhibit melanoma cellular proliferation.

Objective:Hepatocellular carcinoma(HCC)is a lethal global disease that requires an accurate *** assessed the potential of 5 serum biomarkers(AFP,AFU,GGT-II,GPC3,and HGF)in the diagnosis of ***:In this retrospective study,we measured the serum levels of each biomarker using ELISAs in 921 participants,including 298 patients with HCC,154 patients with chronic hepatitis(CH),122 patients with liver cirrhosis(LC),and 347 healthy controls from 3 *** negative for hepatitis B surface antigen and hepatitis C antibody(called"NBNC-HCC")and patients positive for the above indices(called"HBV-HCC and HCV-HCC")were *** selected diagnostic model was constructed using a training cohort(n=468),and a validation cohort(n=453)was used to validate our *** operating characteristic analysis was used to evaluate the diagnostic ***:Theα-L-fucosidase(AFU)/α-fetoprotein(AFP)combination was best able to distinguish NBNC-HCC[area under the curve:0.986(95%confidence interval:0.958–0.997),sensitivity:92.6%,specificity:98.9%]from healthy controls in the test *** screening populations at risk of developing HCC(CH and LC),the AFP/AFU combination improved the diagnostic specificity for early-stage HCC[area under the curve:0.776(0.712–0.831),sensitivity:52.5%,specificity:91.6%in the test group].In all-stage HBV-HCC and HCV-HCC,AFU was also the best candidate biomarker combined with AFP[area under the curve:0.835(0.784–0.877),sensitivity 69.1%,specificity:87.4%in the test group].All results were verified in the validation ***:The AFP/AFU combination could be used to identify NBNC-HCC from healthy controls and hepatitis-related HCC from at-risk patients.

Objective:The immunoscore,which is used to quantify immune infiltrates,has greater relative prognostic value than tumor,node,and metastasis(TNM)stage and might serve as a new system for classification of colorectal ***,a comparable immunoscore for predicting lung adenocarcinoma(LUAD)prognosis is currently ***:We analyzed the expression of 18 immune features by immunohistochemistry in 171 *** relationship of immune marker expression and clinicopathologic factors to the overall survival(OS)was analyzed with the Kaplan-Meier method.A nomogram was developed by using the optimal features selected by least absolute shrinkage and selection operator(LASSO)regression in the training cohort(n=111)and evaluated in the validation cohort(n=60).Results:The indicators integrated in the nomogram were TNM stage,neuron-specific enolase,carcino-embryonic antigen,CD8 center of tumor(CT),CD8 invasive margin(IM),Fox P3 CT,and CD45 *** calibration curve showed prominent agreement between the observed 2-and 5-year OS and that predicted by the *** simplify the nomogram,we developed a new immune-serum scoring system(I-SSS)based on the points awarded for each factor in the *** I-SSS was able to stratify same-stage patients into different risk *** combination of I-SSS and TNM stage had better prognostic value than the TNM stage ***:Our new I-SSS can accurately and individually predict LUAD prognosis and may be used to supplement prognostication based on the TNM stage.

目的:探究钙/钙调蛋白依赖性蛋白激酶Ⅱ(calcium/calmodulin-dependent protein kinaseⅡ,CaMKⅡ)在肺腺癌组织中的表达及其促进肺腺癌的侵袭、转移。方法:通过免疫组织化学染色(immunohistochemistry,IHC)分析肺腺癌患者的石蜡组织标本CaMKⅡ表达与临床病理参数关系,同时对肺腺癌组织与其配对淋巴结转移病灶中的CaMKⅡ表达情况进行比较分析,收集2011年1月至2011年12月于天津医科大学肿瘤医院行手术治疗的113例肺腺癌患者及21例配对的原发病灶及淋巴结转移病灶的石蜡组织标本。将肺腺癌细胞系H1299及Calu3进行慢病毒转染,实验组转染高表达CaMKⅡ病毒,对照组转染阴性对照病毒,通过Transwell实验和划痕实验检测肺腺癌细胞侵袭、转移能力,通过Western blot检测CaMKⅡ表达水平与上皮间充质转化(pithelial-mesenchymal transition,EMT)相关指标和表皮生长因子受体(epidermal growth factor receptor,EGFR)通路激活间的关系。结果:CaMKⅡ高表达与肺腺癌TNM分期和淋巴结转移呈正相关,肺腺癌淋巴结转移病灶中的癌组织CaMKⅡ表达明显高于其原发病灶(P<0.05)。细胞实验表明,CaMKⅡ高表达的肺腺癌细胞,其穿膜细胞数目明显增多,伤口愈合能力增强;EGFR通路激活后p-CaMKⅡ水平增加,且CaMKⅡ促进了EMT相关蛋白及转录因子的表达。结论:CaMKⅡ参与EGFR信号传导,促进EMT过程,增加肺腺癌的侵袭转移能力。

将生物基原料氢化松香醇与三乙胺、对羟基苯甲醚、丙烯酰氯反应制备了可聚合的丙烯酸氢化松香醇酯,然后其与一系列丙烯酸及其衍生物通过共聚,制备了高固含低黏度生物基氟碳树脂。结合分子动力学模拟,分析了丙烯酸氢化松香醇酯降黏机理,探究了生物基氟碳树脂成膜后的涂膜性能。结果表明,随着丙烯酸氢化松香醇酯质量分数的增加,改性氟碳树脂运动黏度降低;当丙烯酸氢化松香醇酯质量分数为18.6%时,改性氟碳树脂固含量为78.56%,其运动黏度与石油树脂叔碳酸乙烯酯改性氟碳树脂相当,远低于不含大体积侧基的改性氟碳树脂;与叔碳酸乙烯酯相比,丙烯酸氢化松香醇酯的空间体积更大,侧基体积效应更明显,分子间作用力小,降黏效果好;丙烯酸氢化松香醇酯改性氟碳涂膜硬度、附着力高于叔碳酸乙烯酯改性氟碳涂膜,耐化学腐蚀性略低于叔碳酸乙烯酯改性氟碳涂膜。
