Serum proteins differentially expressed in gestational diabetes mellitus assessed using isobaric tag for relative and absolute quantitation proteomics

BACKGROUND As a well-known fact to the public,gestational diabetes mellitus(GDM)could bring serious risks for both pregnant women and *** this important investigation into the linkage between GDM patients and their altered expression in the serum,proteomics techniques were deployed to detect the differentially expressed proteins(DEPs)of in the serum of GDM patients to further explore its pathogenesis,and find out possible biomarkers to forecast GDM *** Subjects were divided into GDM and normal control groups according to the IADPSG diagnostic *** samples were randomly selected from four cases in each group at 24-28 wk of gestation,and the blood samples were identified by applying iTRAQ technology combined with liquid chromatography-tandem mass *** proteins and signaling pathways associated with GDM were identified by bioinformatics analysis,and the expression of key proteins in serum from 12 wk to 16 wk of gestation was further verified using enzyme-linked immunosorbent assay (EliSA).RESULTS Forty-seven proteins were significantly differentially expressed by analyzing the serum samples between the GDMgravidas as well as the healthy ones. Among them, 31 proteins were found to be upregulated notably and the rest16 proteins were downregulated remarkably. Bioinformatic data report revealed abnormal expression of proteinsassociated with lipid metabolism, coagulation cascade activation, complement system and inflammatory responsein the GDM group. EliSA results showed that the contents of RBP4, as well as ANGPTL8, increased in the serumof GDM gravidas compared with the healthy ones, and this change was found to initiate from 12 wk to 16 wk *** GDM symptoms may involve abnormalities in lipid metabolism, coagulation cascade activation, complementsystem and inflammatory response. RBP4 and ANGPTL8 are expected to be early predictors of GDM.

Aging is an inevitable physiological process,often accompanied by age-related bone loss and subsequent bone-related diseases that pose serious health *** on skeletal diseases caused by aging in humans is challenging due to lengthy study durations,difficulties in sampling,regional variability,and substantial ***,mice are preferred for such studies due to their similar motor system structure and function to humans,ease of handling and care,low cost,and short generation *** this review,we present a comprehensive overview of the characteristics,limitations,applicability,bone phenotypes,and treatment methods in naturally aging mice and prematurely aging mouse models(including SAMP6,POLG mutant,LMNA,SIRT6,ZMPSTE24,TFAM,ERCC1,WERNER,and KL/KL-deficient mice).We also summarize the molecular mechanisms of these aging mouse models,including cellular DNA damage response,senescence-related secretory phenotype,telomere shortening,oxidative stress,bone marrow mesenchymal stem cell(BMSC)abnormalities,and mitochondrial ***,this review aims to enhance our understanding of the pathogenesis of aging-related bone diseases.

Dynamic recrystallization mechanism, texture evolution development and mechanical characteristics of a Mg–8.7Gd–4.18Y–0.42Zr magnesium alloy by ECAP

Equal channel angle pressing is recognized for its ability to refine alloy grains and alter grain orientation, thereby achieve better mechanical performance of the magnesium alloy. This study investigates the microstructures,dynamic recrystallization mechanism, texture development, and mechanical performance of GW94K(Mg–8.7Gd–4.18Y–0.42Zr wt. %) Mg alloy following ECAP-4 passes at 400°C and 3 mm/min. Results show that when high-temperature deformation is undertaken, twin formation is suppressed while dislocation slip is facilitated, increasing dislocation density during deformation. Following ECAP deformation, the sample displayed higher fracture elongation, TYS, and UTS than the as-solutioned GW94K alloy. In particular, the GW94K alloy performed well mechanically after ECAP-4 passes, with an ultimate TYS of 231 MPa, an UTS of 290 MPa, and an elongation of 14.8 %. DDRX and shear bands induce CDRX, both of which are important in plastic deformation. as well as in modifying microstructure and grain orientation during ECAP deformation.

Tel1 and Rif2 oppositely regulate telomere protection at uncapped telomeres in Saccharomyces cerevisiae

It has been well documented that Tel1 positively regulates telomere-end resection by promoting Mre11-Rad50-Xrs2（MRX） activity, while Rif2 negatively regulates telomere-end resection by inhibiting MRX activity. At uncapped telomeres, whether Tel1 or Rif2 plays any role remains largely unknown. In this work, we examined the roles of Tel1 and Rif2 at uncapped telomeres in yku70△ and/or cdc13-1 mutant cells cultured at non-permissive temperature. We found that deletion of TEL1 exacerbates the temperature sensitivity of both yku70△ and cdc13-1 cells. Further epistasis analysis indicated that MRX and Tel1 function in the same pathway in telomere protection. Consistently, TEL1 deletion increases accumulation of Exo1-dependent telomeric single-stranded DNA（ssDNA） at uncapped telomeres, which stimulates checkpoint-dependent cell cycle arrest. Moreover, TEL1 deletion in yku70△ cells facilitates Rad51-dependent Y0 recombination. In contrast, RIF2 deletion in yku70△ cells decreases the accumulation of telomeric ssDNA after 8 h of incubation at the non-permissive temperature of 37℃ and suppresses the temperature sensitivity of yku70△ cells, likely due to the increase of Mre11 association at ***, our findings indicate that Tel1 and Rif2 regulate telomere protection at uncapped telomeres via their roles in balancing MRX activity in telomere resection.

Blimp-1 protein and Hans classification on prognosis of diffuse large B-cell lymphoma and their interrelation

Image Field Deformation of LAMOST due to Differential Atmospheric Refraction

A vectorial expression of the image field deformation of LAMOST due to the differential atmospheric refraction is presented. The calculated results are compared with those from previous analyses based on the traditional spherical trigonometric formulas. It is demonstrated that different tangential displacements of star images during the observation tracking given by various authors are simply due to different reference points adopted. It is pointed out that the observational celestial pole is the center of the apparent diurnal motion, that, by referring to the observational celestial pole, the effect of the differential refraction on the image field of LAMOST during the 1.5-hour tracking period is approximately equivalent to a constant rotation of - 13.65″ for all declination belts. It is therefore unnecessary to design a particular tracking velocity for each observation, and this will be obviously advantageous to the observation implementation. If the maximum tracking error of the fibers is 0.2″, then the fibers are required to be able to re-position during observational tracking for sky regions south of declination ＋20° and north of declination ＋60°.

Paraplegia is a rare but terrible complication following major spinal operation. Intraoperative monitoring should be used to detect the neurological compucauons early so that they can be surgically revised.1 The common procedures for spinal cord monitoring are intraoperative wake-up test and neurophysiological methods. The latter includes somatosensory evoked potentials （SSEP） and motor evoked potentials （MEP）. The wake-up test has been routinely used to monitor voluntary motor function of the lower limbs during corrective spinal surgery.
