Thioredoxin pathway regulated live-cell synthesis of CdSe quantum dots in Saccharomyces cerevisiae

Currently, the application of synthetic biology to artificially manipulate and utilize organisms for the synthesis of desired products such as nanomaterials with excellent fluorescence properties is attracting considerable attention. However, it is still difficult to obtain designed products efficiently due to insufficient knowledge of the biosynthetic mechanisms. The thioredoxin(TRX) and glutathione(GSH) pathways are generally conserved thiol-reductase systems that protect organisms from oxidative stress and are involved in selenium(Se) metabolism. In this study, we revealed the pivotal role of cytoplasmic TRX pathway in regulating the metabolism of Na_(2)SeO_(3) during the live-cell synthesis of cadmium-selenium quantum dots(Cd Se QDs) in Saccharomyces cerevisiae by regulating the expression level of genes related to TRX pathway and measuring the intracellular content of selenocysteine(Se Cys). The determination of Se Cys metabolism in yeast with GSH pathway-related genes deleted demonstrated that the TRX pathway played a more significant role in Se Cys metabolism than GSH pathway. A 6.4-fold enhancement in the synthetic yield of Cd Se QDs was achieved through the overexpression of TRX pathway-related genes,improvement of synthetic procedure, and supplementation of GSH based on the understanding of biological *** the mechanism of CdSe QDs live-cell synthesis facilitates the precise manipulation of biological processes for the synthesis of inorganic nanomaterials.

A novel Ni-capped complex H4{[Ni（H2O）3]2[CeMo12O42] }· 13.5H2O has been synthesized, which is the first Silverton-type anion capped with two Ni units via three terminal oxygen atoms. Crystallographic data for the title compound： Trigonal, space group R-3, a = 13.0950（19）, b = 13.0950（19）, c = 33.029（7） A, α = β= γ = 58.727（17）A °, V = 4905.1（14） A °^3, Z = 3, Mr= 2437.17, Dc = 2.475 g/cm^3, F（000） = 3462, T = 293（2） K, μ（MoKa） = 3.564 mm^-1, Rt = 0.0369, wR2 = 0.0905 and GOF= 1.079 for 151 parameters and 1963 reflections with I 〉 2σ（I） in the range of 1.85≤θ≤25.50°.

Catalytic Oscillatory Oxidation of CO on Pt(110): on-off Control to Chaos

An improved mean-field model,which specially takes into account pair correlation,was proposed to describe catalytic oscillatory oxidation of CO on Pt(110).By applying on-off control strategies under different frequency,the chemical oscillator shows an abundance of periodic doubling processes,even chaos response,and the route to the chaos and out of the chaos were both identified as period-doubling through analysis of its Lyapunov exponent *** response results reveal that structure delay effect plays an important role to the complexity of this system.

First-principles Investigation of the Structural, Electronic and Elastic Properties of Al_2Ca and Al_4Sr Phases in Mg-Al-Ca(Sr) Alloy

First-principles calculations have been carried out to investigate the structural stabilities, electronic structures and elastic properties of Mg17Al12, Al2Ca and Al4Sr phases. The optimized structural parameters are in good agreement with the experimental and other theoretical values. The calculated formation enthalpies and cohesive energies show that Al2Ca has the strongest alloying ability, and Al4Sr has the highest structural stability. The densities of states （DOS）, Mulliken electronic populations, and electronic charge density difference are obtained to reveal the underlying mechanism of structural stability. The bulk modulus, shear modulus, Young＇s modulus, and Poisson＇s ratio are estimated from the calculated elastic constants. The mechanical properties of these phases are further analyzed and discussed. The Gibbs free energy and Debye temperature are also calculated and discussed.

Reactive oxygen species, nitric oxide and plant cell death associated with caspase-like protease activity during somatic embryogenesis in Fraxinus mandshurica

Programmed cell death occurs in browning explants of Fraxinus mandshurica during somatic embryogenesis, but the underlying mechanism is unclear. In this study, single cotyledons of zygotic embryos of F. mandshurica were used as explants. Mitochondrial structure and function, caspase-3-like protease activity, hydrogen peroxide metabolism, and nitric oxide accumulation induced by high concentrations of sucrose and plant growth regulators were studied. The results show that plant growth regulators induced somatic embryogenesis and also promoted explant browning. High sucrose concentrations had similar effects. High concentrations of sucrose and plant growth regulators led to the accumulation of hydrogen peroxide and nitric oxide which induced changes in mitochondrial structure and function such as modifications in mitochondrial morphology, increased membrane permeability, decreased membrane potential, and the release of cytochrome c into the cytoplasm. An increase in caspase-3-like protease activity triggered programmed cell death in some browning explant cells. During somatic embryogenesis there were increased activities of superoxide dismutase, peroxidase, and catalase, which are associated with hydrogen peroxide metabolism and jointly maintain reactive oxygen species levels. Intracellular nitric oxide synthase and nitrate reductase activities were not significantly correlated with nitric oxide content. Instead, intracellular nitric oxide may be derived from non-enzymatic reactions. Our results indicate that hydrogen peroxide and nitric oxide may function as signals, playing key roles in somatic embryogenesis and programmed cell death of explant cells of F. mandshurica. The interaction between nitric oxide and reactive oxygen species determines the occurrence of programmed cell death in explant cells;somatic embryogenesis and programmed cell death are positively regulated by hydrogen peroxide. However, the regulation of nitric oxide is complex.

Restitution coefficient(RC)of garlic bulb is an important mechanical property that is required to establish the kinematics model of bulb collision and research the damage mechanism of bulb *** this study,kinetic equations of bulb collision were established based on Hertz's contact *** kinematics characteristics,elastoplastic deformation and contact damage during bulb collision were analyzed by using high-speed *** effects of bulb mass,moisture content,collision material,material thickness and release height on the RC were investigated by mixed orthogonal experiments and single-factor *** results showed that the movement of bulb in the compression stage was translation,and the movement in the rebound stage was translation and *** collision,the larger the rotational angular velocity of the bulb was,the smaller the measured RC would *** contact damage of bulb included internal damage of the tissue,epidermis stretch and *** significance of effects of factors on RC decreased with the following sequence:collision material,release height,material thickness,bulb mass,and moisture *** material,release height,material thickness,and bulb mass were significant *** RC between the bulb and Q235,nylon,and rubber decreased *** RC decreased with the increase of release height and bulb *** RC increased with the increase of material thickness of Q235,while it decreased with the increase of material thickness of rubber or *** determination coefficients of the regression equations between the significant factors and the RC were all greater than *** results will be helpful for damage mechanism analysis and design of garlic production equipment.

C5a/C5a R pathway is essential for up-regulating Sph K1 expression through p38-MAPK activation in acute liver failure

AIM To investigate the role of the complement 5a(C5a)/C5 a receptor(C5a R) pathway in the pathogenesis of acute liver failure(ALF) in a mouse *** BALB/c mice were randomly assigned to different groups, and intraperitoneal injections of lipopolysaccharide(LPS)/D-galactosamine(D-Gal N)(600 mg/kg and 10 μg/kg) were used to induce ALF. The KaplanMeier method was used for survival analysis. Serum alanine aminotransferase(ALT) levels, at different time points within a 1-wk period, were detected with a biochemistry analyzer. Pathological examination of liver tissue was performed 36 h after ALF induction. Serum complement 5(C5), C5 a, tumor necrosis factor-α(TNF-α), interleukin(IL)-1β, IL-6, high-mobility group protein B1(HMGB1) and sphingosine-1-phosphatelevels were detected by enzyme-linked immunosorbant assay. Hepatic morphological changes at 36 h after ALF induction were assessed by hematoxylin and eosin staining. Expression of C5 a R, sphingosine kinase 1(Sph K1), p38-MAPK and p-p38-MAPK in liver tissue, peripheral blood mononuclear cells(PBMCs) and peritoneal exudative macrophages(PEMs) of mice or RAW 264.7 cells was analyzed by western blotting. C5 a R m RNA levels were detected by quantitative real-time *** Activation of C5 and up-regulation of C5 a R were observed in liver tissue and PBMCs of mice with ALF. Blockade of C5 a R with a C5 a R antagonist(C5a Ra C5 a Ra) significantly reduced the levels of serum ALT, inflammatory cytokines(TNF-α, IL-1β and IL-6) and HMGB1, as well as the liver tissue damage, but increased the survival rates(P < 0.01 for all). Blockade of C5 a R decreased Sph K1 expression in both liver tissue and PBMCs significantly at 0.5 h after ALF induction. C5 a Ra pretreatment significantly downregulated the phosphorylation of p38-MAPK in liver tissues of ALF mice and C5 a stimulated PEMs or RAW 264.7 cells. A Single Image Derain Method Based on Residue Channel Decomposition in Edge Computing

The numerous photos captured by low-price Internet of Things(IoT)sensors are frequently affected by meteorological factors,especially *** causes varying sizes of white streaks on the image,destroying the image texture and ruining the performance of the outdoor computer vision *** methods utilise training with pairs of images,which is difficult to cover all scenes and leads to domain *** addition,the network structures adopt deep learning to map rain images to rain-free images,failing to use prior knowledge *** solve these problems,we introduce a single image derain model in edge computing that combines prior knowledge of rain patterns with the learning capability of the neural ***,the algorithm first uses Residue Channel Prior to filter out the rainfall textural features then it uses the Feature Fusion Module to fuse the original image with the background feature *** results in a pre-processed image which is fed into Half Instance Net(HINet)to recover a high-quality rain-free image with a clear and accurate structure,and the model does not rely on any rainfall *** results on synthetic and real-world datasets show that the average peak signal-to-noise ratio of the model decreases by 0.37 dB on the synthetic dataset and increases by 0.43 dB on the real-world dataset,demonstrating that a combined model reduces the gap between synthetic data and natural rain scenes,improves the generalization ability of the derain network,and alleviates the overfitting problem.

A thermodynamic model for predicting activity of CaO-SiO_(2)-Al_(2)O_(3)-MgO-TiO_(2)-V_(2)O_(3)-FeO slags during electric furnace smelting process

To optimize the comprehensive utilization of vanadium titanomagnetite by direct reduction-smelting processes,it is essential to acquire titanium slag with a higher TiO_(2)content of 45–60 wt.%.A thermodynamic model was developed based on the ion and molecule coexistence theory,specifically targeting the CaO–SiO_(2)–Al_(2)O_(3)–MgO–TiO_(2)–V_(2)O_(3)–FeO slag *** impact of slag composition on the smelting of vanadium titanomagnetite was assessed,and the thermodynamic model was utilized to identify the optimal high-titanium *** results revealed that increasing the basicity,MgO content,and FeO content within the slag effectively suppressed the reduction of titanium and silicon ***,the calculated activity coefficient of TiO_(2)decreased with higher basicity,MgO,and FeO *** an increase in basicity significantly enhanced the reduction of vanadium oxides,the effects of MgO and FeO contents on vanadium oxide reduction were comparatively less ***,higher basicity and FeO content promoted the formation of calcium titanates,whereas an elevated MgO content favored the formation of magnesium *** smelting results indicated that a lower V_(2)O_(3)content and higher TiO_(2)activity corresponded to a smaller titanium mass fraction in the iron alloy,while the opposite trend was observed for vanadium.
