Acorn squash(Cucurbita pepo)is an iconic fall vegetable in the United States,known for its unique fruit shape and also prized for its culinary *** is known about the metabolism that underlies the development of fruit quality attributes such as color,sweetness,texture and nutritional qualities in acorn squash,or any other winter squash grown *** provide insight into winter squash fruit and seed development and add to the genomic resources in the Cucurbita genus,RNA sequencing was used to generate an acorn squash fruit and seed transcriptome from the cultivar Sweet ReBA at critical points throughout fruit development.141838600 high-quality paired-end Illumina reads were assembled into 55949 unigenes.85%of unigenes with predicted open reading frames had homology with previously identified genes and over 62%could be functionally *** with the watermelon and cucumber genomes provided confirmation that the unigenes are full-length and comprehensive,covering an average of 90%of the coding sequence of their homologs and 72%of the cucumber and watermelon *** candidate genes associated with carotenoid and carbohydrate metabolism were identified toward a resource for winter squash fruit quality trait *** transcriptome represents a major advance in *** genomics,providing significant new sequence information and revealing the repertoire of genes expressed throughout winter squash fruit and seed *** studies on the genetic basis of fruit quality and future breeding efforts will be enhanced by tools and insights developed from this resource.

elephants are highly social beings with complex individual *** know that elephants have a general interest in death,investigating carcasses,not just limited to kin;however,research does not explore in depth whether individuals change their behavior or personality following traumatic events,such as the death of a *** a captive herd of Asian elephants(elephas maximus)housed at Chester Zoo,UK,we measured social behavior and proximity and personality using the Ten-Item Personality Inventory,and found age-related and relationship-related changes in both behavior and personality following the deaths of herd ***,the herd spent less time socializing and engaging in affiliative behaviors following the death of the adult female when compared to baseline data,yet spent more time engaging in these behaviors after the death of two *** death of the central female had a dramatic impact on her infant calf,resulting in increasingly withdrawn behavior,yet had the opposite effect on her adult daughter,who subsequently established a more integrated role within the *** Stability fell in the motherless calf but rose in an adult female,who had lost her adult daughter,but had a new calf to care *** suggest that the greater impact on the behavior and personality of surviving herd members following the deaths of calves,compared to an adult member,attests to the significance of the unifying role played by calves within an elephant herd.

Digital elevation models(DeMs)are a necessary dataset for modelling the earth’s surface;however,all DeMs contain *** can reduce this error using DeM fusion techniques since numerous DeMs can be available for a ***,the use of a clustering algorithm in DeM fusion has not been previously *** this study a new DeM fusion algorithm based on a clustering approach that works on multiple DeMs to exploit consistency in the estimates as indicators of accuracy and precision is *** fusion approach includes slope and elevation thresholding,k-means clustering of the elevation estimates at each cell location,as well as filtering and smoothing of the fusion *** of the input DeMs,and the products of each step of the fusion algorithm,with a higher accuracy reference DeM enabled a detailed analysis of the effectiveness of the DeM fusion *** main findings of the research were:the k-means clustering of the elevations reduced the precision which also impacted the overall accuracy of the estimates;the number of final cluster members and the standard deviation of elevations before clustering both had a strong relationship to the error in the k-means estimates.

Background High deductible health plans(HDHPs)are associated with the avoidance of both necessary and unnecessary *** hernia repair(UHR)is a procedure that is frequently unnecessarily performed in young children,contrary to best practice *** hypothesized that children with HDHPs,as compared with other types of commercial health plans,are less likely to undergo UHR before 4 years of age but are also more likely to have UHR delayed beyond 5 years of *** Children aged 0-18 years old residing in metropolitan statistical areas(MSAs)who underwent UHR in 2012-2019 were identified in the IBM Marketscan Commercial Claims and encounters Database.A quasi-experimental study design using MSA/year-level HDHP prevalence among children as an instrumental variable was employed to account for selection bias in HDHP ***-stage least squares regression modeling was used to evaluate the association between HDHP coverage and age at *** A total of 8601 children were included(median age 5 years,IQR 3-7).Univariable analysis revealed no differences between the HDHP and non-HDHP groups in the likelihood of UHR being performed before 4 years of age(27.7%vs 28.7%,p=0.37)or after 5 years of age(39.8%vs 38.9%,p=0.52).Geographical region,metropolitan area size,and year were associated with HDHP *** variable analysis demonstrated no association between HDHP coverage and undergoing UHR atears of age(p=0.76)or>5 years of age(p=0.87).Conclusions HDHP coverage is not associated with age at pediatric *** studies should investigate other means by which UHRs in young children can be avoided.

A broad range of organic compounds are known to exist in drinking water sources and serve as precursors of disinfection byproducts（DBPs）.epidemiological findings of an association of increased risk of bladder cancer with the consumption of chlorinated water has resulted in health concerns about *** are thought to be an important category of DBP precursors in ***,little is known about the actual presence of peptides and their DBPs in drinking water because of their high sample complexity and low *** address this challenge and identify peptides and non-chlorinated/chlorinated peptide DBPs from large sets of organic compounds in water,we developed a novel high throughput analysis strategy,which integrated multiple solid phase extraction（SPe）,high performance liquid chromatography（HPLC）separation,and non-target identification using precursor ion exclusion（PIe）high resolution mass spectrometry（MS）.After MS analysis,structures of candidate compounds,particularly peptides,were obtained by searching against the Human Metabolome Database（HMDB）.Using this strategy,we successfully detected 625 peptides（out of 17,205 putative compounds）and 617 peptides（out of 13,297）respectively in source and finished water *** source and finished water samples had 501 peptides and amino acids in *** remaining 116 peptides and amino acids were unique to the finished *** a subset of 30 putative compounds for which standards were available,25 were confirmed using HPLC-MS *** analyzing the peptides identified in source and finished water,we successfully confirmed three disinfection reaction pathways that convert peptides into toxic DBPs.

It is established in the psychological literature that pre-displacement stressors,PTSD symptoms,and psycholo-gical distress are associated among internally displaced ***,existing studies have not demonstrated the mechanism underlying these *** study compared two explanatory models;one with PTSD symptoms severity explaining the indirect association between pre-displacement stressors and psychological dis-tress,and the other with psychological distress explaining the indirect relationship between pre-displacement stressors and PTSD symptoms *** a cross-sectional design,631 women(Mean age=31.18±8.59)were conveniently and purposely selected from the displaced women harboured in two camps in Borno State,Nigeria,due to the Boko-Haram *** were collected utilizing structured questionnaires and subjected to path *** demonstrated that PTSD symptoms severity and psychological distress indirectly explained their respective and independent association with pre-displacement ***,PTSD symptoms had a statis-tically larger standardized indirect effect size,greater indirect effect percentage,and bigger kappa-squared(k2)effect size than psychological *** symptoms and psychological distress may have a reciprocal inﬂuence on each other from pre-displacement ***,PTSD symptoms can better explain the association between pre-displacement stressors and psychological distress than the other way *** outcomes have important implications for the psychological treatment of displaced persons.

DNA methylation has been identified as a powerful and activity-dependent regulator of changes in the brain that may underlie neuroadaptations in response to various types of stimuli,including exposure to drugs of ***,the medial prefrontal cortex(mPFC)projections to the nucleus accumbens(NAc)are critically important for reinstated cocaine-seeking in a rodent model of cocaine *** circuitry undergoes several epigenetic modifications following cocaine exposure,including changes in DNA methylation that are associated with drug-seeking *** have previously shown that methyl supplementation via L-Methionine(MeT)administration attenuates cocaine-seeking behavior and reverses expression and methylation patterns of the immediate early gene c-fos,suggesting that MeT may act by altering the excitability of this circuitry during cocaine *** the current study,male rats were microinjected with an adeno-associated virus overexpressing halorhodopsin in the mPFC,optical fibers were surgically implanted into the NAc,and the rats were given injections of MeT *** underwent acquisition of cocaine self-administration(0.75 mg/kg/infusion,2-h sessions)followed by extinction training in the absence of drug-paired *** reinstatement tests were conducted:cue-induced reinstatement without optogenetic manipulations and cocaine-primed reinstatement with optogenetic inhibition of mPFC-to-NAc *** were no group differences before the cocaine-primed reinstatement session,and all groups showed robust cue-induced *** rats treated with MeT and rats that received mPFC-to-NAc inhibition showed an abolishment of cocaine-primed reinstatement,suggesting that systemic methyl supplementation may act through this critical circuity.
