Background:Exercise training promotes brain plasticity and is associated with protection against cognitive impairment and Alzheimer’s disease(AD).These beneficial effects may be partly mediated by blood-borne factors.Here we used an in vitro model of AD to investigate effects of blood plasma from exercise-trained donors on neuronal viability,and an in vivo rat model of AD to test whether such plasma impacts cognitive function,amyloid pathology,and neurogenesis.methods:mouse hippocampal neuronal cells were exposed to AD-like stress using amyloid-βand treated with plasma collected from human male donors 3 h after a single bout of high-intensity exercise.For in vivo studies,blood was collected from exercise-trained young male Wistar rats(high-intensity intervals 5 days/week for 6 weeks).Transgenic AD rats(mcGill-R-Thyl-APP)were inj ected 5 times/fortnight for 6 weeks at2 months or 5 months of age with either(a)plasma from the exercise-trained rats,(b)plasma from sedentary rats,or(c)saline.Cognitive function,amyloid plaque pathology,and neurogenesis were assessed.The plasma used for the treatment was analyzed for 23 cytokines.Results:Plasma from exercised donors enhanced cell viability by 44.1%(p=0.032)and reduced atrophy by 50.0%(pmyloid-β-treated cells.In vivo exercised plasma treatment did not alter cognitive function or amyloid plaque pathology but did increase hippocampal neurogenesis by~3 fold,regardless of pathological stage,when compared to saline-treated rats.Concentrations of 7 cytokines were significantly reduced in exercised plasma compared to sedentary plasma.Conclusion:Our proof-of-concept study demonstrates that plasma from exercise-trained donors can protect neuronal cells in culture and promote adult hippocampal neurogenesis in the AD rat brain.This effect may be partly due to reduced pro-inflammatory signaling molecules in exercised plasma.

The mesoproterozoic(1.11 Ga)Umkondo large igneous province(LIP)in southern Africa and Antarctica was emplaced inmyr and is dominated by low-Ti tholeiitic doleritic-gabbroic sills.It is of particular interest because it is the least studied LIP in southern Africa with both sublithospheric and lithospheric mantle sources proposed and it coincides with the early assembly of Rodinia,so it has importance in understanding the nature of magmatism and tectonics in and around the Kalahari craton during the mesoproterozoic.In this study,we compiled a large database of existing(~750)and new(~100)major and trace element data for the Umkondo province,as well as 42 new Sr-Nd isotopic measurements,to provide constraints on its magma sources and geochemical evolution.major element compositional variations in the low-Ti tholeiites are explained by low-pressure(1 kbar)three-phase fractional crystallisation(olivine,clinopyroxene and plagioclase)of a parent magma with~10 wt.%mgO in oxidising conditions(QFm+1).Inverse models show that the low-Ti tholeiitic magmas were derived as residual melts after the crystallization of 12%-33%olivine from primary komatiitic-basaltic magmas(up to~20 wt.%mgO)in equilibrium with mantle olivine(Fo90).Low Sm/Yb and TiO2/Yb-Nb/Yb indicate that the primary magmas were derived by 2%-20%shallow(40-50 km)partial melting of spinel lherzolite.High Sm/Yb is restricted to dyke swarms and may imply limited magma production from deeper(up to~70 km)garnet lherzolite-like sources.The low-Ti tholeiites of the Umkondo province are enriched in large ion lithophile elements(Rb-Sr-Cs-K)and depleted in high-field strength elements(Zr-Hf-Nb-Ta),indicating the involvement of crustal material and/or the subcontinental lithospheric mantle.This is supported by covariations in Th/Nb,Nb/Yb,Nb/La and Ce/Sm with generally negativeΔNb.Sr-Nd isotopes lend support to the notion that the Umkondo magmas were derived from depleted and/or enriched sublithospheric mantle sources and subsequently contaminated by enriched lithospheric m

Unequal Distribution of Innovation Efforts for Neglected Tropical Diseases: The Role of Funding Evaluation Criteria

Background: International research and innovation efforts for neglected tropical diseases have increased in recent decades due to disparities in overall health research funding in relation to global burden of disease. However, within the field of neglected tropical diseases some seem far more neglected than others. In this research the aim is to investigate the distribution of resources and efforts, as well as the mechanisms that underpin funding allocation for neglected tropical diseases. methodology: A systematic literature review was conducted to establish a comprehensive overview of known indicators for innovation efforts related to a wide range of neglected tropical diseases. Articles were selected based on a subjective evaluation of their relevance, the presence of original data, and the breadth of their scope. This was followed by thirteen in-depth open-ended interviews with representatives of private, public and philanthropic funding organizations, concerning evaluation criteria for funding research proposals. Results: The findings reveal a large difference in the extent to which the individual diseases are neglected with notable differences between absolute and relative efforts. Criteria used in the evaluation of research proposals relate to potential impact, the probability of success and strategic fit. Private organizations prioritize strategic fit and economic impact;philanthropic organizations prioritize short-term societal impact;and public generally prioritize the probability of success by accounting for follow-up funding and involvement of industry. Funding decisions of different types of organizations are highly interrelated. Conclusions: This study shows that the evaluation of funding proposals introduces and retains unequal funding distribution, reinforcing the relative neglect of diseases. Societal impact is the primary rationale for funding but application of it as a funding criterion is associated with significant challenges. 目的钙摄入水平低以及体内维生素D不足是导致亚洲女性罹患骨质疏松的危险因素。此项研究的目的是评估高钙维生素D强化牛奶(HCm)对健康的中国绝经后妇女的体内维生素D状态和骨吸收标志物进行干预的影响。方法本试验对象为55岁以上,绝经至少5年的女性。共有124名女性进行了总体健康状况和骨密度检查筛选。63名女性被随机分为对照组和试验组,试验组在12w内每日饮用两次高钙/维生素D强化奶(含900 mg钙、96 mg镁、2.4 mg锌和6.4μg维生素D)。2组分别在试验开始、试验2 w、8 w和12 w时测量血矿物质、25(OH)维生素D3和Ⅰ型胶原C端肽(CTX)水平。结果对照组女性平均年龄为63±4.6岁,高钙强化奶(HCm)组女性平均年龄为62±3.8岁。所测得的每日钙摄入量的基线值为HCm组260～482 mg,对照组为252～692 mg。12 w后HCm组的平均身体质量指数(BmI)显著降低(Pm组的血25(OH)维生素D3水平显著提高(33.13～39.49 nmol/L),而对照组则维持不变(29.27～28.21 nmol/L)。HCm组的血CTX水平在0 w和2 w之间下降了25%,并保持不变直至第12w。从第2w开始HCm组和对照组相比血CTX水平就出现了显著差异(Pm),在2至4w内能显著提高中国绝经妇女体内维生素D水平,并减少骨吸收破坏。如果长期持续地饮用含高钙和维生素D的强化奶(HCm),能减少骨吸收和骨量丢失的风险,从而减少患骨质疏松和由此引起骨折的风险。

Cellular metabolism is the fundamental process by which cells maintain growth and self-renewal.It produces energy,furnishes raw materials,and intermedi-ates for biomolecule synthesis,and modulates enzyme activity to sustain normal cellular functions.Cellular metabolism is the foundation of cellular life processes and plays a regulatory role in various biological functions,including pro-grammed cell death.Ferroptosis is a recently discovered form of iron-dependent programmed cell death.The inhibition of ferroptosis plays a crucial role in tumorigenesis and tumor progression.However,the role of cellular metabolism,particularly glucose and amino acid metabolism,in cancer ferroptosis is not well understood.Here,we reviewed glucose,lipid,amino acid,iron and sele-nium metabolism involvement in cancer cell ferroptosis to elucidate the impact of different metabolic pathways on this process.Additionally,we provided a detailed overview of agents used to induce cancer ferroptosis.We explained that the metabolism of tumor cells plays a crucial role in maintaining intracellu-lar redox homeostasis and that disrupting the normal metabolic processes in these cells renders them more susceptible to iron-induced cell death,resulting in enhanced tumor cell killing.The combination of ferroptosis inducers and cel-lular metabolism inhibitors may be a novel approach to future cancer therapy and an important strategy to advance the development of treatments.

Changes in physical activity,sedentary behavior,and self-reported cognitive function in cancer survivors before and during the COVID-19 pandemic:A cross-sectional study

Background Physical activity(PA)and reductions in sedentary behavior(SED)may mitigate cancer-related cognitive impairment.The purpose of this study was to examine(a)the associations between changes in PA,SED,and cognitive function in cancer survivors before and during the coronavirus disease 2019(COVID-19)pandemic;and(b)clinical subgroups that moderate this association.methods A cross-sectional survey was administered online to adult cancer survivors globally between July and November of 2020.This was a secondary analysis of a cross-sectional survey examining changes in self-reported PA and quality of life in cancer survivors before and during the COVID-19 pandemic.Self-reported Questionnaires assessed moderate-to-vigorous PA(mVPA)using the modified Godin Leisure Time Exercise Questionnaire,cognitive function using the Functional Assessment of Cancer Therapy-Cognitive Function(FACT-Cog)scale,and SED using the Domain-specific Sitting Time questionnaire.Cancer survivors were classified into no change in behavior,desirable change(i.e.,increase mVPA to meet PA guidelines or decrease SED by≥60 min/day),and undesirable change(i.e.,decrease mVPA tomin/week or increase SED by≥60 min/day).Analysis of covariance examined differences in FACT-Cog scores across the activity change categories.Planned contrasts compared differences in FACT-Cog scores between cancer survivors with(a)no meaningful change vs.any change,and(b)a desirable change vs.an undesirable change.Results There were no significant differences in FACT-Cog scores across activity-change categories in the full sample of cancer survivors(n=371;age=48.6±15.3 years(mean±SD)).However,cancer survivors who were diagnosed≥5 years ago(t(160)=–2.15,p=0.03)or who received treatment≥5 years ago(t(102)=–2.23,p=0.03)and who had a desirable change in activity reported better perceived cognitive abilities than those who had an undesirable change.Conclusion PA promotion efforts should consider reducing SED in addition to maintaining mVPA in long-term cancer survivors to miti

采用一锅法将生物柴油副产物粗甘油(CG)转化为生物基多元醇(CG-polyol),并以糠醛渣(FR)为增强填料,共混发泡制备出一种FR增强生物基聚氨酯(PU/FR)泡沫复合材料。通过对PU/FR泡沫的结构形貌、热稳定性、发泡时间、密度和压缩强度进行表征,探究了糠醛渣粒径(0.25 mm、 0.09 mm样品分别标记为FR60和FR180)和添加量对PU/FR泡沫性能的影响。结果表明:通过热转化法合成的CG-polyol酸值为1.9 mg/g、羟值为406 mg/g、黏度为1 092 mPa·s,该多元醇适合用于制备PU泡沫。FR的加入延长了发泡时间,最大上升时间和不粘时间分别由未添加时的29和31 s提高到37和39 s,泡孔结构更加完整,泡孔尺寸减少,破碎现象明显减少。FR添加量≤5%时,可有效提高泡沫的密度和压缩强度;当添加量相同时,FR180填料对泡沫的性能提升更显著;当FR180添加量为5%时制备的PU/FR180-5泡沫复合材料的压缩强度达到最大为0.153 3 mPa,相比未添加FR的泡沫提高了28.1%,此时密度为0.051 0 g/cm~3,导热系数为0.032 8 W/(m·K),复合材料的综合性能较好。同时FR的添加可适当提高PU泡沫材料的热稳定性,相比未添加FR的泡沫的最大热解速率温度(T_(max))453℃,PU/FR180-5泡沫的T_(max)达到463℃。
