A comparison of perioperative outcomes between extraperitoneal robotic single-port and multiport radical prostatectomy with the da Vinci Si Surgical System

To evaluate outcomes between extraperitoneal robotic single-port radical prostatectomy(epR-spRP)and extraperitoneal robotic multiport radical prostatectomy(epR-mpRP)performed with the da Vinci Si Surgical System,comparison was performed between 30 single-port(SP group)and 26 multiport(MP group)cases.Comparisons included operative time,estimated blood loss(EBL),hospital stay,peritoneal violation,pain scores,scar satisfaction,continence,and erectile function.The median operation time and EBL were not different between the two groups.In the SP group,the median operation time of the first 10 patients was obviously longer than that of the latter 20 patients(P<0.001).The median postoperative hospital stay in the SP group was shorter than that in the MP group(P<0.001).The rate of peritoneal damage in the SP group was less than that in the MP group(P=0.017).The pain score and overall need for pain medications in the SP group were lower than those in the MP group(P<0.001 and P=0.015,respectively).Patients in the SP group were more satisfied with their scars than those in the MP group 3 months postoperatively(P=0.007).At 3 months,the cancer control,recovery of erectile function,and urinary continence rates were similar between the two groups.It is safe and feasible to perform epR-spRP using the da Vinci Si surgical system.Therefore,epR-spRP can be a treatment option for localized prostate cancer.Although epR-spRP still has a learning curve,it has advantages for postoperative pain and self-assessed cosmesis.In the absence of the single-port robotic surgery platform,we can still provide minimally invasive surgery for patients.

Quantitative Determination of Glymphatic Flow Using Spectrophotofluorometry

Following intrathecal injection of fluorescent tracers,ex vivo imaging of brain vibratome slices has been widely used to study the glymphatic system in the rodent brain.Tracer penetration into the brain is usually quantified by image-processing,even though this approach requires much time and manual operation.Here,we illustrate a simple protocol for the quantitative determination of glymphatic activity using spectrophotofluorometry.At specific time-points following intracisternal or intrastriatal injection of fluorescent tracers,certain brain regions and the spinal cord were harvested and tracers were extracted from the tissue.The intensity of tracers was analyzed spectrophotometrically and their concentrations were quantified from standard curves.Using this approach,the regional and dynamic delivery of subarachnoid CSF tracers into the brain parenchyma was assessed,and the clearance of tracers from the brain was also determined.Furthermore,the impairment of glymphatic influx in the brains of old mice was confirmed using our approach.Our method is more accurate and efficient than the imaging approach in terms of the quantitative determination of glymphatic activity,and this will be useful in preclinical studies.

Digitalizing river aquatic ecosystems

Traditional river health assessment relies on limited water quality indices and representative organism activity,but does not comprehensively obtain biotic and abiotic information of the ecosystem.Here,we propose a new approach to evaluate the ecological and health risks of river aquatic ecosystems.First,detailed physicochemical and biological characterization of a river ecosystem can be obtained through pollutant determination(especially emerging pollutants)and DNA/RNA sequencing.Second,supervised machine learning can be applied to perform classification analysis of characterization data and ascertain river ecosystem ecology and health.Our proposed methodology transforms river ecosystem health assessment and can be applied in river management.

Research on intelligent search-and-secure technology in accelerator hazardous areas based on machine vision

Prompt radiation emitted during accelerator operation poses a significant health risk,necessitating a thorough search and securing of hazardous areas prior to initiation.Currently,manual sweep methods are employed.However,the limitations of manual sweeps have become increasingly evident with the implementation of large-scale accelerators.By leveraging advancements in machine vision technology,the automatic identification of stranded personnel in controlled areas through camera imagery presents a viable solution for efficient search and security.Given the criticality of personal safety for stranded individuals,search and security processes must be sufficiently reliable.To ensure comprehensive coverage,180°camera groups were strategically positioned on both sides of the accelerator tunnel to eliminate blind spots within the monitoring range.The YOLOV8 network model was modified to enable the detection of small targets,such as hands and feet,as well as larger targets formed by individuals near the cameras.Furthermore,the system incorporates a pedestrian recognition model that detects human body parts,and an information fusion strategy is used to integrate the detected head,hands,and feet with the identified pedestrians as a cohesive unit.This strategy enhanced the capability of the model to identify pedestrians obstructed by equipment,resulting in a notable improvement in the recall rate.Specifically,recall rates of 0.915 and 0.82were obtained for Datasets 1 and 2,respectively.Although there was a slight decrease in accuracy,it aligned with the intended purpose of the search-and-secure software design.Experimental tests conducted within an accelerator tunnel demonstrated the effectiveness of this approach in achieving reliable recognition outcomes.

The metabolic benefits associated with long-term physical activity are well appreciated and growing evidence suggests that it involves the gut microbiota.Here we re-evaluated the link between exercise-induced microbial changes and those associated with prediabetes and diabetes.We found that the relative abundances of substantial amounts of diabetes-associated metagenomic species associated negatively with physical fitness in a Chinese athlete students cohort.We additionally showed that those microbial changes correlated more with handgrip strength,a simple but valuable biomarker suggestive of the diabetes states,than maximum oxygen intake,one of the key surrogates for endurance training.Moreover,the causal relationships among exercise,risks for diabetes,and gut microbiota were explored based on mediation analysis.We propose that the protective roles of exercise against type 2 diabetes are mediated,at least partly,by the gut microbiota.

Development of CONTHAC-3D and hydrogen distribution analysis of HPR1000

An in-house code,CONTHAC-3D,was developed to calculate and analyze thermal-hydraulic phenomena in containments during severe accidents.CONTHAC-3D is a three-dimensional computational fluid dynamics code that can be applied to predict gas flow,diffusion,and steam condensation in a containment during a severe hypothetical accident,as well as to obtain an estimate of the local hydrogen concentration in various zones of the containment.CONTHAC-3D was developed using multiple models to simulate the features of the proprietary systems and equipment of HPR1000 and ACP100,such as the passive cooling system,passive autocatalytic recombiners and the passive air cooling system.To validate CONTHAC-3D,a GX6 test was performed at the Battelle Model Containment facility.The hydrogen concentration and temperature monitored by the GX6 test are accurately predicted by CONTHAC-3D.Subsequently,the hydrogen distribution in the HPR1000 containment during a severe accident was studied.The results show that the hydrogen removal rates calculated using CONTHAC-3D for different types of PARs agree well with the theoretical values,with an error of less than 1%.As the accident progresses,the hydrogen concentration in the lower compartment becomes higher than that in the large space,which implies that the lower compartment has a higher hydrogen risk than the dome and large space at a later stage of the accident.The amount of hydrogen removed by the PARs placed on the floor of the compartment is small;therefore,raising the installation height of these recombiners appropriately is recommended.However,we do not recommend installing all autocatalytic recombiners at high positions.The study findings in regard to the hydrogen distribution in the HPR1000 containment indicate that CONTHAC-3D can be applied to the study of hydrogen risk containment.

Connexin 43 remodeling induced by LMNA gene mutation Glu82Lys in familial dilated cardiomyopathy with atrial ventricular block

Background Mutations in the lamin A/C gene （LMNA） may cause familial dilated cardiomyopathy （dilated cardiomyopathy） characterized by early onset atrio-ventricular block （A-V block） before the manifestation of dilated cardiomyopathy and high risk of sudden death due to ventricular arrhythmia, which is very similar to the phenotype of gap junction related heart disease. This study aimed to determine the expression and localization of connexins in neonatal myocytes transfected with wild-type （WT） or mutant LMNA to elucidate how these mutations cause heart diseases. Methods We studied the connexin 43 （Cx43） and connexin 40 （Cx40） expression in cultured neonatal myocytes transfected with wild-type （WT） or mutant LMNA （Glu82Lys （E82K） and Arg644Cys （R644C）） using confocal imaging and Western blotting analysis. Results Cx43 protein expression was reduced by 40% in cells transfected with LMNA E82K than that in cells transfected with WT LMNA cDNA. Confocal imaging showed that the Cx43 located inside the cells by LMNA E82K. By contrast, LMNA E82K mutation had no effect on expression and localization of Cx40. LMNA R644C transfection did not show any significant effects on gap junctions at all. Conclusions Our findings suggest that LMNA E82K significantly reduced the Cx43 expression and altered its localization which may be one of the pathological mechanisms underlying LMNA-related heart disease.

Knockout of C6orf120 in Rats Alleviates Concanavalin A-induced Autoimmune Hepatitis by Regulating Macrophage Polarization

Objective The effect of the functionally unknown gene C6orf120 on autoimmune hepatitis was investigated on C6orf120 knockout rats(C6orf120^(-/-))and THP-1 cells.Method Six–eight-week-old C6orf120^(-/-)and wild-type(WT)SD rats were injected with Con A(16 mg/kg),and euthanized after 24 h.The sera,livers,and spleens were collected.THP-1 cells and the recombinant protein(rC6ORF120)were used to explore the mechanism in vitro.The frequency of M1 and M2 macrophages was analyzed using flow cytometry.Western blotting and PCR were used to detect macrophage polarization-associated factors.Results C6orf120 knockout attenuated Con A-induced autoimmune hepatitis.Flow cytometry indicated that the proportion of CD68^(+)CD86^(+)M1 macrophages from the liver and spleen in the C6orf120^(-/-)rats decreased.C6orf120 knockout induced downregulation of CD86 protein and the mRNA levels of related inflammatory factors TNF-α,IL-1β,and IL-6 in the liver.C6orf120 knockout did not affect the polarization of THP-1 cells.However,rC6ORF120 promoted the THP-1 cells toward CD68^(+)CD80^(+)M1 macrophages and inhibited the CD68^(+)CD206^(+)M2 phenotype.Conclusion C6orf120 knockout alleviates Con A-induced autoimmune hepatitis by inhibiting macrophage polarization toward M1 macrophages and reducing the expression of related inflammatory factors in C6orf120^(-/-)rats.

UPLC-Q-TOF/MS-Based Serum Metabolomics Reveals Potential Anti-tumor Mechanism of Banxia Xiexin Decoction in Colorectal Cancer Mice

Objective: To clarify the potential mechanism of Banxia Xiexin Decoction(BXD) on colorectal cancer(CRC) from the perspective of metabolomics. Methods: Forty male C57BL/6 mice were randomly divided into normal control(NC), azoxymethane/dextran sulfate sodium(AOM/DSS) model, low-dose BXD(L-BXD), high-dose BXD(H-BXD) and mesalamine(MS) groups according to a random number table, 8 mice in each group. Colorectal cancer model was induced by AOM/DSS. BXD was administered daily at doses of 3.915(L-BXD) and 15.66 g/kg(H-BXD) by gavage for consecutive 21 days, and 100 mg/kg MS was used as positive control. Following the entire modeling cycle, colon length of mice was measured and quantity of colorectal tumors were counted. The spleen and thymus index were determined by calculating the spleen/thymus weight to body weight. Inflammatory cytokine and changes of serum metabolites were analyzed by enzyme-linked immunosorbent assay kits and ultra performance liquid chromatography-quadrupole/time-of-flight mass spectrometry(UPLC-Q/TOF-MS), respectively. Results: Notably, BXD supplementation protected against weight loss, mitigated tumor formation, and diminished histologic damage in mice treated with AOM/DSS(P<0.05 or P<0.01). Moreover, BXD suppressed expression of serum inflammatory enzymes, and improved the spleen and thymus index(P<0.05). Compared with the normal group, 102 kinds of differential metabolites were screened in the AOM/DSS group, including 48 potential biomarkers, involving 18 main metabolic pathways. Totally 18 potential biomarkers related to CRC were identified, and the anti-CRC mechanism of BXD was closely related to D-glutamine and D-glutamate metabolism, phenylalanine, tyrosine and tryptophan biosynthesis, arginine biosynthesis, nitrogen metabolism and so on. Conclusion: BXD exerts partial protective effects on AOM/DSS-induced CRC by reducing inflammation, protecting organism immunity ability, and regulating amino acid metabolism.

Ground state of Rydberg-dressed Bose gas confined in a toroidal trap

The experimental realization of Rydberg dressing technology in ultracold atomic systems provides another superior platform for studying novel states of matter and macroscopic quantum phenomena.In this work,based on the mean-field theory,we have investigated the ground-state phases of a two-component Bose–Einstein condensate with Rydberg interaction and confined in a toroidal trap.The effects of the Rydberg interaction and external potential,especially the Rydberg blockade radius,on the ground-state structure of such a system have been investigated in full parameter space.Our results show that the Rydberg blockade radius,which can be regarded as another controllable parameter,can be used to obtain a variety of ground-state phases.More interestingly,it is found that for weak Rydberg interactions,the Rydberg blockade radius breaks the spontaneous rotational symmetry of the system,leading to the formation of a discrete unit cell structure.For strongly interacting cases,it can be used to realize different orders of discrete rotational symmetry breaking.
