Angiogenesis and nerve regeneration induced by local administration of plasmid pBud-coVEGF165-coFGF2 into the intact rat sciatic nerve

Vascular endothelial growth factor(VEGF) and fibroblast growth factor 2(FGF2) are well-known growth factors involved in the regeneration of various tissues and organs, including peripheral nerve system. In the present study, we elucidated the local and systemic effects of plasmid construct рBud-coVEGF165-coFGF2 injected into the epineurium of intact rat sciatic nerve. Results of histological examination of sciatic nerve and multiplex immunoassays of serum showed the absence of immunogenicity and biosafety of plasmid рBud-coVEGF165-coFGF2. Moreover, local administration of plasmid DNA construct resulted in significantly decreased levels of pro-inflammatory cytokines in the peripheral blood, including tumor necrosis factor α(TNFα) and interleukin-12, and significantly increased levels of cytokines and chemokines including Regulated upon Activation, Normal T Cell Expressed and Presumably Secrete(RANTES), epidermal growth factor, interleukin-2, and monocyte chemoattractant protein 1. These changes in the peripheral blood on day 7 after injection of plasmid construct рBud-coVEGF165-coFGF2 show that the plasmid construct has systemic effects and may modulate immune response. At the same time, reverse transcriptionpolymerase chain reaction revealed transient expression of coFGF2, coVEGF165, ratFGF2 and ratVEGFA with direct transport of transcripts from distal part to proximal part of the sciatic nerve. Immunohistochemical staining revealed prolonged presence of VEGFA in sciatic nerve till 14 days post-injection. These findings suggest that local administration of plasmid construct рBud-coVEGF165-coFGF2 at a concentration of 30 ng/μL results in the formation of pro-angiogenic stimuli and, and the plasmid construct, used as a drug for gene therapy, might potentially facilitate regeneration of the sciatic nerve. Phytogeography of the northeastern coast of the Caspian Sea:native flora and recent colonizations

The northeastern coast of the Caspian Sea is one of the youngest territories for plant colonization in Kazakhstan,and the flora of the most recent portion of this coastal area,the New Caspian marine plain,was chosen to study phytogeographical characteristics of plant species,especially as related to alien origin and invasive *** of the recent formation of this flora,I expected that a large proportion of the species may be alien and *** compiling a comprehensive species list from previous research,I identified the taxonomic,life form,ecological,and geographical structures of the *** area belongs to the northern type of deserts with continental climate and low annual *** and life form structures show that the flora is specific to Central Asian *** structure demonstrates a high percentage of halophytes,which resulted from their adaptation to soil salinity and shallow ground waters that occur throughout the study *** of the flora indicate allochtonous properties of the flora （*** derived from areas outside the study area） with a low proportion of native （Caspian） *** species are estimated to comprise 12％-19％ of the *** some species have invasive features,severe arid environmental conditions and high salinity of soils and ground waters decrease invasive capacity of exotic ***,a threat of expansion of potentially invasive plants exists in the New Caspian marine plain due to the fast growth of anthropogenic habitats associated with recent oil developments.

The most important components of electrical vehicles are the battery and the related cooling *** subsystems play a major role in determining the overall electric vehicle *** this study,a novel cooling system with fluid in the battery cell is proposed,by which the energy storage system can be optimized through control of the temperature of the batteries.A sensitivity analysis is conducted considering the maximum temperature,the heat rate,the coolant temperature,and the geometry of the *** numerical simulations show that the parameters for the trapezoidal compartment have an impact on the thermal performance of *** optimal geometry is proposed *** is concluded that for high values of Reynolds number for which the flow becomes turbulent,a decrease in the battery temperature can be obtained thereby avoiding thermal stresses.

Diaspore characteristics and ecological adaptation of Bromus tectorum *** different distribution regions

The invasion of nonnative plants is considered one of the main threats to the structure and function of North American ecosystems. Moreover, they can alter ecosystem processes and reduce biodiversity. In arid and semi-arid region of North America, the species of European annual grass Bromus tectorum L. is an outstanding example of these problems, which not only increase the fire density and change the fire regime, but replace native communities. Therefore, there are amount of researches on B. tectorum, including resource acquisition, water use efficiency and growth. Whereas the relevant research on the morphology of diaspore is scare. Diaspores have a fundamental role in seed germination and seedling establishment. Besides, as an important link between different generations, diaspores have a vital significance on individual reproduction and population extension. Hence, dias- pores under selection for studying have an important implication. This study compares differences in seed mor- phology for Bromus tectorum collected from the United States, Kazakhstan, and Xinjiang of China. The following indices of B. tectorum diaspores were analyzed： size, thickness of covering layers, and micromorphological char- acteristics of the base, middle and transition area of diaspores as well as of the awn. Micromorphology of the lemma and the cross-section of the diaspore were observed by scanning electron microscopy. Results showed that thick- ness of the lemma and the palea of diaspores from B. tectorum-infested grasslands in the United States were re- duced （P〈0.05）, likely because of environmental influences. This reduction facilitated the germination of diaspores and lowered the resistance of B. tectorum to adverse environmental conditions. The length of the awn also in- creased significantly （P〈0.05）, which helped in dispersal and anchoring of diaspores. Venture capital financing officially appeared in the US in1960s and developed there up to the point of becoming a distinct type in private equity financing with its own special features and process steps. Later in1970-1980s this best practice US venture capital model started to spread into emerging economies and other countries. Key two emerging markets in the world today are China and Russia. Showing to be extremely successful in the US, governments of emerging economies made efforts to encourage the formation of the local venture industry. Nowadays the US venture capital model is successfully applied in emerging economies and many other countries. However, when implementing the same model in different countries venture capitalists have to adapt and adjust its strategies, practices and operational activities of venture capital cycle, starting from VC fund organization and up to exit options. One of the reasons for those different VC practices and strategies among countries is distinct institutional environment of each country, which determines certain level of investors’rights protection, regulation, availability of business infrastructure, tax incentives, transparency, etc. The United States, China and Russia have substantially different institutional environments, in terms of regulations, effectiveness of their enforcement, government involvement, etc. Thus, contrasting US venture capital model application in China and Russia, while benchmarking both emerging countries to the US, will allow to better understand how venture capitalists’ respond and adapt to distinct regulatory and social settings. Analysis of historic background and current regulations on VC financing in China and Russia helps to set baseline and understand public policy content, in which venture capital firms have to operate. Identification of genes involved in the formation of soluble dietary fiber in winter rye grain and their expression in cultivars with different viscosities of wholemeal water extract

The grain of rye(Secale cereale L.) used for baking contains a large amount of non-starch polysaccharides,making it an excellent component of functional foods. But rye grain intended for alcohol production and forage use should have a reduced content of these polysaccharides. A comprehensive parameter that can predict the best field of application for winter rye grain is the viscosity of its wholemeal water ***, our understanding of the genetic background underlying this key trait and associated features of rye grain is poor. By analyzing six Russian winter rye cultivars, we identified the most contrasting forms and characterized the peculiarities of their water-soluble carbohydrates capable of influencing the viscosity of water extracts. Then, using phylogenetic and transcriptomic analyses, we identified in the rye genome many genes encoding putative glycosyltransferases and glycosylhydrolases responsible for the synthesis and degradation of arabinoxylans, mixed-linkage glucans, cellulose, and some other polysaccharides. We determined the dynamics of m RNA abundance for these genes at three stages of kernel development. Comparisons of gene expression levels in two contrasting cultivars revealed specific members of multigene families that may serve as promising targets for manipulating non-starch polysaccharide content in rye grain. High-viscosity cultivars were characterized by up-regulation of many glycosyltransferases involved in the biosynthesis of arabinoxylans and other cell-wall polysaccharides,whereas low-viscosity cultivars showed up-regulation of several genes encoding polysaccharidedegrading enzymes.

Development and Validation of Methods for Identification and Quantification of Biological Active Substances in the Fruit of <i>Tetrapleura tetraptera</i>

This study was conducted to identify the biological active substances (BAS) from the fruit of Tetrapleura tetraptera obtained from Ghana. TLC, UV Spectroscopy and differential spectrophotometry methods of analysis were employed in this study. Identified BAS included flavonoids, polysaccharides and amino acids. Following state pharmacopoeia of Ukraine (SPU) requirements to validate the developed techniques were employed in this study, and the following validation properties specificity, linearity, range, precision accuracy (convergence) and robustness was determined. These characteristics did not exceed the eligibility criteria as stated in (SPU) Appendix 2. Thus the technique can be made to the SPU monograph on fruits of Tetrapleura tetraptera.
