A pilot study on Paxlovid therapy for hemodialysis patients with severe acute respiratory syndrome coronavirus 2 infections

We aimed to investigate the safety and efficacy of nirmatrelvir/ritonavir(Paxlovid)therapy for hemodialysis-dependent patients with severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection.Thirteen hemodialysis patients infected with the Omicron variant of SARS-CoV-2 from April 3 to May 30,2022,were recruited.Laboratory parameters and chest CT(computed tomography)imaging were analyzed.The treatment group included six patients who received 150 mg/100 mg of Paxlovid orally once daily for 5 days,whereas the control group included seven patients who received basic treatment.No serious adverse reactions or safety events were recorded.Four control patients progressed to moderate disease,and none in the treatment group showed progression of chest CT findings(P<0.05).Paxlovid therapy tended toward early viral clearance and low viral load on Day 8.Moreover,83.3%of the patients in the treatment group and 57.1%of the patients in the control group turned negative within 22 days.In the Paxlovid treatment group,we found significantly increased levels of lymphocytes(P=0.03)and eosinophils(P=0.02)and decreased levels of D-dimer on Day 8 compared with those on Day 1.Paxlovid therapy showed a potential therapeutic effect with good tolerance in hemodialysis patients.The optimal dose and effectiveness evaluation must be further investigated in a largeer cohort.

Rapid Prediction of Wastewater Index Using CNN Architecture and PLS Series Statistical Methods

Chemical oxygen demand (COD) is an important index to measure the degree of water pollution. In this paper, near-infrared technology is used to obtain 148 wastewater spectra to predict the COD value in wastewater. First, the partial least squares regression (PLS) model was used as the basic model. Monte Carlo cross-validation (MCCV) was used to select 25 samples out of 148 samples that did not conform to conventional statistics. Then, the interval partial least squares (iPLS) regression modeling was carried out on 123 samples, and the spectral bands were divided into 40 subintervals. The optimal subintervals are 20 and 26, and the optimal correlation coefficient of the test set (RT) is 0.58. Further, the waveband is divided into five intervals: 17, 19, 20, 22 and 26. When the number of joint intervals under each interval is three, the optimal RT is 0.71. When the number of joint subintervals is four, the optimal RT is 0.79. Finally, convolutional neural network (CNN) was used for quantitative prediction, and RT was 0.9. The results show that CNN can automatically screen the features inside the data, and the quantitative prediction effect is better than that of iPLS and synergy interval partial least squares model (SiPLS) with joint subinterval three and four, indicating that CNN can be used for quantitative analysis of water pollution degree.

Analysis of the inter-annual variability and southward expansion of red tides in the Zhejiang coastal waters from 1981 to 2018

A time series dataset spanning 39 years(1981-2018) on red tide events in Zhejiang coastal waters was used to study the characteristics of inter-annual spatial and temporal variations. A distinct inter-annual pattern characterized by low frequency, explosive growth and fluctuating decline stages was found over the studied time scale. Most red tide events occurred in parallel to the bathymetric contour, and 95.4% were located to the west of the 50 m isobath. Additionally, the high-incidence area of red tides is expanding southward. In this paper, local sea surface temperature(SST), mariculture area and secondary industry growth rate are introduced and identified as the main factors influencing the nutrient and hydrometeorological conditions. A multivariate nonlinear regression equation based on these factors was constructed, and the goodness of fit coefficient was 0.907. The causes of the annual variation and high-frequency area in the southward expansion were quantitatively analyzed based on the proposed regression model. Finally, the results indicated that 68.7% of the annual occurrence variation of red tide was due to the SST and mariculture area, which are the main impact factors;however,secondary industry growth could compensate for the nutrient deficiency caused by the sharp mariculture area reduction and decreased SST. The background nutrient level, which is elevated by coastal economic development, especially secondary industry, is the main determinant of the southward expansion. Although the trend of the southward expansion of high-frequency areas has not changed, the red tide frequency in coastal cities has decreased by half and remained at a stable level after 2010 due to substantial economic restructuring and environmental protection.

Diatomic Fe-Fe catalyst enhances the ability to degrade organic contaminants by nonradical peroxymonosulfate activation system

Atomically dispersed catalysts have been widely studied due to their high catalytic activity and atom utilization.Single-atom catalysts have achieved breakthrough progress in the degradation of emerging organic contaminants(EOCs)by activating peroxymonosulfate(PMS).However,the construction of atomically dispersed catalysts with diatomic/multiatomic metal active sites by activating PMS to degrade pollutants is still seldom reported,despite the unique merits of atom-pair in synergistic electronic modulation and breaking stubborn restriction of scaling relations on catalytic activity.We have synthesized Fe1-N-C,Fe_(2)-N-C,and Fe_(3)-N-C catalysts with monoatomic iron,diatomic iron,and triatomic iron active center,respectively.The results show that the catalytic degradation activity of Fe_(2)-N-C is twice that of Fe1-N-C and Fe_(3)-N-C due to its unique Fe_(2)N6 coordination structure,which fulfilled the complete degradation of rhodamine B(RhB),bisphenol A(BPA),and 2,4-dichlorophenol(2,4-DP)within 2 min.Electron paramagnetic resonance(EPR)and radical quenching experiments confirmed that the reaction was a nonradical reaction on the catalyst surface.And singlet oxygen and Fe(IV)are the key active species.

HPV infection is the primary cause of cervical intraepithelial neoplasia(CIN),with persistent high-risk HPV infection being the leading factor in the development of cervical cancer.In addition to the application of large loop excision of the transformation zone(LLETZ)for the treatment of cervical pre-cancerous lesions,various ablative techniques,including thermal ablation,cryotherapy,carbon dioxide(CO_(2))laser therapy,and focused ultrasound therapy also play significant roles in the management of cervical intraepithelial neoplasia.This review presents a comprehensive overview of the pathophysiology of cervical HPV infection and discusses the ablative methods commonly used in clinical practice,along with their indication and contradiction,especially in women with high-grade squamous intraepithelial lesions.The aim is to identify safe and effective treatment strategies for treating cervical intraepithelial neoplasia,thereby avoiding under-or over-treatment,which may reduce the incidenceandprogr ssion of cervical cancer through timely diagnosis and treatment.

Synthesis of Organic-Inorganic Hybrid Aluminum Hypophosphite Microspheres Flame Retardant and Its Flame Retardant Research on Thermoplastic Polyurethane

Aluminum hypophosphite microspheres(AHP) were synthesized by hydrothermal method using NaH2PO2·H2O and AlCl3·6H2O as raw materials, and then the AHP microspheres were polymerized by surface polymerization of micro-nanospheres with cyclic cross-linked poly(cyclotriphosphazene-co-4,4'-sulfonyldiphenol)(PZS). A new organic-inorganic poly(phosphonitrile)-modified aluminum hypophosphite microspheres(PZS-AHP) were synthesized by encapsulation and applied to flame retardant thermoplastic polyurethane(TPU). The microstructure and chemical composition of the PZS-AHP microsphere were characterized by scanning electron microscopy, transmission electron microscopy, Fourier transform infrared spectroscopy and X-ray spectroscopy. The thermal stability of PZS-AHP microsphere was explored with thermogravimetric analysis. Thermogravimetric data indicate that the PZS-AHP microspheres have excellent thermal stability. The thermal and flame-retarding properties of the TPU composites were evaluated by thermogravimetric(TG), limited oxygen index tests(LOI), and cone calorimeter test(CCT). The TPU composite achieved vertical burning(UL-94) V-0 grade and LOI value reached 29.2% when 10 wt% PZS-AHP was incorporated. Compared with those of pure TPU, the peak heat release rate(pHRR) and total heat release(THR) of TPU/10%PZS-AHP decreased by 82.2% and 42.5%, respectively. The results of CCT indicated that PZS-AHP microsphere could improve the flame retardancy of TPU composites.

Critical evaluation of the glucose oxidase-like activity of gold nanoparticles stabilized by different polymers

Polymer stabilizers are widely used to synthesize gold nanoparticles(Au NPs)to prevent their aggregation and improve their stability.Although stabilizers are known to greatly influence both the structure and size of Au NPs,limited efforts explore their effects on the activity of Au NPs for biocatalysis.Herein,different polymers are used as stabilizers to synthesize Au NPs.For the glucose oxidase-like activity,we find that without the spatial barrier from stabilizers,naked Au NPs show significantly high catalytic activity as well as the worst stability.Among the polymers,polyacrylic acid-stabilized Au NPs exhibit the highest activity,whose Vmax(0.74μM·s^(−1))is higher than that of the natural glucose oxidase(0.37μM·s^(−1))due to the smallest particle size(<2 nm)and the weak spatial resistance of polyacrylic acid.These variable catalytic results derive from the comprehensive effects including size,steric hindrance,and electronic effect.However,further selectivity and activity tests have exposed shortcomings.They possess universal activities for aldose oxidation,whereas cannot retain activities in typical physiological environments.Our findings highlight the role of polymer stabilizers in imposing effects on the glucose oxidase-like activity of Au NPs and provide a basis for further Au NPs engineering and applications.

Human metapneumovirus(HMPV) infection is one of the leading causes of hospitalization in young children with acute respiratory illness. In this study, we prospectively collected respiratory tract samples from children who were hospitalized with acute lower respiratory tract infection in six hospitals in China from 2017 to 2019. HMPV was detected in 145 out of 2733 samples(5.3%) from the hospitalized children. The majority of HMPV-positive children were under the age of two(67.6%), with a median age of one year. HMPV can independently cause acute lower respiratory tract infection in young children, while all patients showed mild clinical symptoms. Of all the co-infected patients, HMPV was most commonly detected with enterovirus(EV) or rhinovirus(RhV)(38.0%),followed by respiratory syncytial virus(RSV)(32.0%). The highest detection rate occurred from March to May in both northern and southern China. Out of 145 HMPV positive samples, 48 were successfully typed, of which 36strains were subgrouped into subtypes A2c(75%), eight strains were included in subtype B1(16.7%), and four strains were included in subtype B2(8.3%). Moreover, 16 A2c strains contained 111-nucleotide duplications in the G gene. Twenty-seven complete HMPV genomes were successfully obtained, and 25(92.6%) strains belonged to subtype A2c, whereas one strain was included in subgroup B1 and another was included in subgroup B2. A total of 277 mutations were observed in the complete genomes of 25 A2c strains. All results presented here improve our understanding of clinical characteristics and molecular epidemiology of HMPV infection in children.

ELAID:detecting integer-Overflow-to-Buffer-Overflow vulnerabilities by light-weight and accurate static analysis

The Integer-Overflow-to-Buffer-Overflow(IO2BO)vulnerability has been widely exploited by attackers to cause severe damages to computer systems.Automatically identifying this kind of vulnerability is critical for software security.Despite many works have been done to mitigate integer overflow,existing tools either report large number of false positives or introduce unacceptable time consumption.To address this problem,in this article we present a static analysis framework.It first constructs an inter-procedural call graph and utilizes taint analysis to accurately identify potential IO2BO vulnerabilities.Then it uses a light-weight method to further filter out false positives.Specifically,it generates constraints representing the conditions under which a potential IO2BO vulnerability can be triggered,and feeds the constraints to SMT solver to decide their satisfiability.We have implemented a prototype system ELAID based on LLVM,and evaluated it on 228 programs of the NIST’s SAMATE Juliet test suite and 14 known IO2BO vulnerabilities in real world.The experiment results show that our system can effectively and efficiently detect all known IO2BO vulnerabilities.

Uric acid promotes neuronal differentiation of human placenta-derived mesenchymal stem cells in a time- and concentration-dependent manner

Uric acid is an important, naturally occurring serum antioxidant. The present study investigates the use of uric acid for promoting proliferation and neuronal differentiation of mesenchymal stem cells derived from human placenta tissue. Human placenta-derived mesenchymal stem cells were pre-induced in the presence of either 0, 0.2, 0.4 or 0.8 mM uric acid in combination with 1 mM β-mercaptoethanol for 24 hours, followed by exposure to identical uric acid concentrations and 5 mM β-mercaptoethanol for 6 and 10 hours. Cells developed a neuronal-like morphology, with formation of interconnected process extensions, typical of neural cells. Immunocytochemistry and immunofluorescence staining showed neuron specific enolase positive cells were present in each group except the control group. A greater number of neuron specific enolase positive cells were observed in 0.8 mM uric acid in combination with 5 mM β-mercaptoethanol at 10 hours. After 24 hours of induction, Nissl bodies were detected in the cytoplasm of all differentiated cell groups except the control group and Nissl body numbers were greatest in human placenta-derived mesenchymal stem cells grown in the presence of 0.8 mM uric acid and 5 mM β-mercaptoethanol. These results suggest uric acid accelerates differentiation of human placenta-derived mesenchymal stem cells into neuronal-like cells in a time-and concentration-dependent manner.
