Achi.vable supercooli.g for the formati.n of methane hydrate from water emulsi.ns was studi.d i. seven di.ferent crude oi.s and i. decane. The experi.ents were performed under constant rate cooli.g from + 20 to-15 °C and a pressure of methane of 12 MPa. i. was demonstrated that the shapes and posi.i.ns of the resulti.g survi.al curves depend on the densi.y, vi.cosi.y and di.persi.e power of oi. samples used i. the experi.ents, as well as on the degree of oi. oxi.ati.n. i. addi.i.n, results of the experi.ents on i.e freezi.g under the same emulsi.ns are presented. The results obtai.ed i. the work allowed us to di.cuss the possi.i.i.y and features of pri.ary and secondary nucleati.n of the hydrate and i.e i. the systems under consi.erati.n.

Studi.s on natural vari.ti.n are an i.portant tool to unravel the geneti. basi. of quanti.ati.e trai.s i. plants. Despi.e the si.ni.i.ant roles of phytohormones i. plant development, i.cludi.g root archi.ecture, hardly any studi.s have been done to i.vesti.ate natural vari.ti.n i. endogenous hormone levels i. plants. Therefore, i. the present study a range of hormones were quanti.i.d i. root extracts of thi.teen Arabi.opsi. thali.na accessi.ns usi.g a ultra performance li.ui. chromatography tri.le quadrupole mass spectrometer. Root system archi.ecture of the set of accessi.ns was quanti.i.d, usi.g a new parameter （mature root uni.） for complex root systems, and correlated wi.h the phytohormone data. Si.ni.i.ant vari.ti.ns i. phytohormone levels among the accessi.ns were detected, but were remarkably small, namely less than three-fold di.ference between extremes. For cytoki.i.s, relati.ely larger vari.ti.ns were found for ri.osi.es and glucosi.es, as compared to the free bases. For root phenotypi.g, length-related trai.s--lateral root length and total root length--showed larger vari.ti.ns than lateral root number-related ones. For root archi.ecture, antagoni.ti. i.teracti.ns between hormones, for example, i.dole-3-aceti. aci. to trans-zeati. were detected i. correlati.n analysi.. These fi.di.gs provi.e conclusi.e evi.ence for the presence of natural vari.ti.n i. phytohormone levels i. Arabi.opsi. roots, suggesti.g that quanti.ati.e geneti. analyses are feasi.le.

Phytohormones, auxi.s i. parti.ular, play an i.portant role i. plant development and producti.i.y. Earli.r data showed posi.i.e i.pact of exogenous auxi. on potato (Solanum tuberosum L.) tuberi.ati.n. The ai. of thi. study was to generate potato plants wi.h i.creased auxi. level predomi.antly i. tubers. To thi. end, a pBi.B33-tms1 vector was constructed harbori.g the Agrobacteri.m auxi. bi.synthesi. gene tms1 fused to tuber-speci.i. promoter of the class i.patati. gene (B33-promoter) of potato. Among numerous i.dependently generated B33:tms1 li.es, those wi.hout vi.i.le di.ferences from control were selected for detai.ed studi.s. i. the majori.y of transgeni. li.es, tms1 gene transcri.ti.n was detected, mostly i. tubers rather than i. shoots. i.doleaceti. aci. (i.A) content i. tubers and the auxi. tuber-to-shoot rati. were i.creased i. tms1-expressi.g transformants. The organ-speci.i. i.crease i. auxi. synthesi. i. B33:tms1-transformants accelerated and i.tensi.i.d the process of tuber formati.n, reduced the dose of carbohydrate supply requi.ed for i. vi.ro tuberi.ati.n, and decreased the photoperi.di. dependence of tuber i.i.i.ti.n. Overall, a posi.i.e correlati.n was observed between tms1 expressi.n, i.A content i. tubers, and sti.ulati.n of tuber formati.n. The revealed properti.s of B33:tms1 transformants i.ply an i.portant role for auxi. i. potato tuberi.ati.n and offer prospects to magni.y potato producti.i.y by a moderate organ-speci.i. enhancement of auxi. content.

Forest soi. profi.es of two dunes wi.hi. the European belt of i.land dunes were analysed i. the laboratory. We carri.d out respi.ometri. measurements of carbon di.xi.e producti.n and oxygen consumpti.n for every hori.on of the studi.d soi.s whi.e si.ultaneously quanti.yi.g the organi. matter and humi.i.y. Oxygen consumpti.n and carbon di.xi.e excreti.n decreased exponenti.lly wi.h depth. The oxygen consumpti.n decrease was less rapi. than the decrease i. carbon di.xi.e producti.n. We found a stati.ti.al si.ni.i.ant li.ear dependence between oxygen consumpti.n and carbon di.xi.e excreti.n, and organi. matter content and soi. water capaci.y. Respi.ati.n processes i. the profi.es were di.i.ed i.to two strata;oxygen respi.ati.n domi.ated i. the fi.st and fermentati.n processes i. the second. We esti.ated total respi.ati.n i. the studi.d profi.es for an area of 1 m2 down to around 1 m depth. We concluded that when assessi.g the soi.’s role i. carbon cycli.g i. an ecosystem, i. i. necessary to consi.er both the respi.atory and fermentati.n strata, as both produce large quanti.i.s of carbon di.xi.e. The mai. factor determi.i.g carbon di.xi.e producti.n i.tensi.y i. organi. matter content;thus the di.tri.uti.n of organi. matter i. the soi. profi.e determi.es carbon cycli.g i.tensi.y.

The recombi.ant exopolyphosphatase PPX1 wi.h a speci.i. acti.i.y of ~300 U/mg protei. was puri.i.d from the strai. of Saccharomyces cerevi.i.e wi.h the i.acti.ated PPN1 gene transformed by the expressi.n vector carryi.g the yeast PPX1 gene. The recombi.ant PPX1 was si.i.ar to the PPX1 of wi.d strai.s i. i.s substrate speci.i.i.y and requi.ement for cati.ns. PPX1 had the hi.h substrate speci.i.i.y to polyphosphates. The preparati.n was sui.able for polyphosphate assay i. the presence of orthophosphate and nucleosi.e phosphates not hydrolyzed by PPX1. The yi.ld of the enzyme preparati.n was 250 assays per 1 g of the bi.mass. The recombi.ant PPX1 was successfully used i. polyphosphate assay i. di.ferent yeast speci.s and some foodstuffs.
