Unhealthy eating habits with sugar-rich and high-fat foods can bring nutritional imbalance and other chronic diseases. At present, there is an increasing demand in regulating human sub-health through functional foods. Agriophyllum squarrosum(L.) Moq., a pioneer plant native to sand dunes with ecological restoration function, is a traditional food plant of sand regions and Mongolian folk medicine. Numerous studies have confirmed that A. squarrosum seed, also called sand rice, has high nutritional value. However, the long-term health effects of eating sand rice remain unclear. To better explore and evaluate its long-term effects, hereby, ICR mice were fed with sand rice flour at different ratios(control, 60% and 95%) for 100 days. Results show that the growth and blood glucose level curve of these experimental mice were lower and more stable than that of the control. In addition, mice fed pure sand rice ingredient(95%) gained more stable body weight and blood glucose after 28 days, which revealed that sand rice is a food with comprehensive nutritional value and rarely negatively impacts the body growth performance. Our data also demonstrates that, the content of triglyceride(TG), total cholesterol(TC), low-density lipoprotein cholesterol(LDL-C) and high-density lipoprotein cholesterol(HDL-C) provided the lowest level in the blood of mice with a diet of pure sand rice ingredient, 0.48, 4.14, 0.91, 3.03 mmol/L, respectively. Also, T-SOD and GSH-PX activity provided a high level in mice with pure sand rice gradient, which significantly increased by 36%(T-SOD)and 19%(GSH-PX) compared to the control. These results suggest that long term intake of the pure sand rice ingredients is more helpful for stabilizing blood lipid and improve antioxidant capacity and beneficial to people suffering from chronic disease. Growth factors can regulate the proliferation and differentiation ability of cells, keep the internal environment stable and affect the body size. In order to explore the influence of the change of the expression of growth factors on the body size of the Eothenomys miletus in different regions of Hengduan Mountain, the E. miletus in Dali(DL), Jianchuan(JC), Lijiang(LJ), Xianggelila(XGLL) and Deqin(DQ) were collected, and their seven morphological indicators and nine growth factors, including insulin-like growth factor1(IGF1), basic fibroblast growth factor(b FGF), epidermal growth factor(EGF), growth hormone release inhibitory factor(GRIF), platelet-derived growth factor(PDGF), transcription elongation factor1(TCEA1), a disintegrin and metalloprotease17(ADAM7), branched-chain amino acid transaminase1(BCAT1), high mobility group protein1(HMG1)were statistically analyzed. It was found that the body size of E. miletus in five regions of Hengduan Mountain differentiated, the body mass, body length, tail length, forelimb length, hind limb length, IGF, bFGF, EGF, PDGF, TCEA1 content of the E. miletus in DL, JC and LJ are greater than those in XGLL and DQ, and the contents of GRIF, ADAM7, BCAT1 and HMG1 are also different, decrease with the increase of regional latitude. The regional differences in the body size of E. miletus in different regions may be related to the changes in body mass, body length, tail length, forelimb length, hind limb length and the content of growth factors. The increase in the content of growth factors leads to the increase in the body size of E. miletus. It is speculated that growth factors may affect the body size of E. miletus by affecting the bone, epidermis, muscle and proliferation and differentiation of cells.

促丝裂原活化蛋白激酶(mitogen-activated protein kinase,MAPK)级联反应是参与植物生长发育、激素及胁迫的响应信号通路中重要而复杂的信号网络之一。促分裂原激活蛋白激酶(mitogen-activated protein kinase kinase,MAPKK)是其主要成员之一,是该级联反应中位于通路中间、对信号传递起着收集和发散的关键作用的激酶。已有研究表明马铃薯(Solanum tuberosum L.)StMAPKK1基因响应干旱胁迫。本研究采用同源重组的克隆方式构建了StMAPKK1的诱饵载体pGBKT7-StMAPKK1,通过酵母(Saccharomyces cerevisiae)双杂交系统筛选马铃薯c DNA文库,共得到5种StMAPKK1互作蛋白,并利用小规模杂交回转验证对互作真实性进行了验证。通过生物信息学分析鉴定这5种StMAPKK1互作蛋白分别为水解酶(水解O-糖基化合物)、RING-H2亚家族RHE蛋白、氰酸酯酶、ARF GTPase活化因子以及一个含有C2结构域的蛋白。本研究结果为进一步研究马铃薯St MAPKK1所参与信号通路及其生物功能提供理论依据。

TCP基因家族是参与调控植物内源激素的重要转录因子,马铃薯中StTCP15基因受到细胞分裂素、ABA和GA3的诱导表达,并在块茎休眠打破过程中StTCP15基因表达量变化显著。为探究外源6-BA对马铃薯块茎休眠时间、内源激素含量变化及StTCP15基因表达模式的影响,以马铃薯栽培品种‘鄂薯10号’块茎为试验材料,用液相色谱-质谱(LC-MS/MS)和实时荧光定量PCR (RT-qPCR)技术分别测定了10 mg/L6-BA处理下马铃薯块茎内源GA3、ABA、6-BA、IAA的含量及StTCP15基因表达量变化。结果表明外源6-BA处理与对照相比提前1周打破块茎休眠,块茎内源激素含量短时间与对照差异显著,随着时间延长差异减小;在块茎休眠阶段随着贮藏时间的延长处理组与对照组块茎内源激素ABA、GA3和IAA含量升高,6-BA含量下降;休眠打破后ABA和IAA含量降低,GA3和6-BA含量升高;发芽期间4种内源激素含量均升高。RT-qPCR分析结果表明6-BA处理及对照块茎休眠期间StTCP15基因表达量持续升高,休眠解除后表达量下降。StTCP15基因可能参与ABA信号通路,并在马铃薯休眠的解除中发挥作用。

为探索miRNA-mRNA如何参与沙蓬干旱响应和干旱记忆,选择沙蓬干旱敏感生态型AEX为植物材料,开展3轮干旱-复水循环实验后进行转录组和sRNA测序。分别鉴定出341个和7858个响应干旱胁迫的沙蓬miRNA(DEMIs)和mRNA(DEMs)。DEMI-DEM调控网络构建表明,24个沙蓬DEMIs参与调控1423个DEMs,这些靶基因主要参与细胞分化功能。鉴定出两个干旱记忆miRNA(DMMIs)miR156和miR6173,分别调控11个和6个干旱记忆基因(DMGs)。miR156的靶基因包括3个转录因子(TF)AsRWP-RK、AsC3H11和AsWRKY33;miR6173的靶基因包括两个GST(谷胱甘肽S转移酶)。在先前的研究中,AsC3H11和AsWRKY33作为TF分子开关参与沙蓬干旱记忆分级调控网络,因此,沙蓬miR156可作为“miRNA-转录因子-功能基因”网络的候选枢纽。研究解析了沙蓬干旱响应和干旱记忆的分子调控基础,将为荒漠植物中的抗逆遗传资源开发提供理论依据。

Transcriptomic analysis identifies a pan-cancer association of IL27 expression with cancer prognosis and immune microenvironment

Nowadays great progress has been made in targeted therapy and comprehensive therapy for a few cancers. But the prognosis of the vast majority of cancer patients is still weakness. It has been proven that inflammation can affect the development and treatment of cancer. Some previous studies have suggested that inflammation-related interleukins (ILs) play a significant role in the development of some cancers.1,2 ILs act as a considerable role in immunity,3 and inflammation affects the development of cancer. Therefore, the expression profile of IL gene may also be one of the vital markers of early pathological changes, occurrence and progression of cancer.4,5 Currently, studies on the expression of IL27 in cancer are still extremely limited. Our work comprehensively and systematically studied the IL27 gene expression in the generic cancer when compared with the existing IL27 gene related research experiment. We screened out cancer species linked with poor prognosis, and explored the correlation between IL27 gene expression and immune cell infiltration and immune microenvironment in these cancers further, providing an increasing number of the direct basis for subsequent studies. Our findings showed that IL27 can be used to assess prognosis and immune infiltration in patients with colon adenocarcinoma (COAD), glioblastoma multiforme (GBM), head and neck squamous cell carcinoma (HNSC), skin cutaneous melanoma (SKCM) and testicular germ cell tumors (TGCT) as a biomarker.

In this paper, transient stability analysis was focused on Hu-liao HVDC and AC parallel transmission system. The Hu-liao HVDC project was introduced;Simulation method and mathematic models of AC and DC systems were stu-died as well as corresponding regulators and controllers. The dynamic performance and the interaction between AC and DC systems during serious disturbance were researched by detail time-domain simulation. Comparison was also made under different operation schemes. The research will bring important and significant reference for further operation and stability control of Hu-liao HVDC and AC system.

Recently generation of strong magnetic(B)fields has been demonstrated in capacitor coils heated by high power laser pulses[*** et al.,***.3,1170(2013)].This paper will present a direct measurement of B field generated with an open-ended coil target driven by a nanosecond laser pulse using ultrafast proton *** radiographs are analyzed with particle-tracing *** B field at the coil center is inferred to be ~50 T at an irradiance of ~5×10^(14) W·cm^(-2).The B field generation is attributed to the background cold electron flow pointing to the laser focal spot,where a target potential is induced due to the escape of energetic electrons.
