Functional control of CRISPR/Cas9 is essential for precise gene *** engineering of guide RNA(gRNA)provides diverse approaches for conditional control of CRISPR/Cas9 function with a variety of chemical reactive ***,previous investigations into chemically engineering gRNA only unidirectionally regulated the CRISPR/Cas9 function via stimuli-induced caging/decaging ***,we propose a combinatory strategy to engineer the dynamics of gRNA in which photocontrolled strand-displacement reactions coupled with sequence designs of gRNA can achieve lightinduced switching-on/off control of CRISPR/Cas9 *** analysis and cellular gene regulation indicate this approach is capable of both activating and deactivating CRISPR/Cas9 activities using light ***,photocontrolled multiplex modulations of gene expression for opposite regulatory effects have also been achieved simultaneously under the same cellular *** work establishes an essential principle for construction of stimuli-induced switching-on/off modulations of gRNA that can greatly enrich the versatility of conditional control for a variety of CRISPR/Cas9-based applications.

Oxide-supported metal single-atom catalysts(SACs)have exhibited excellent catalytic performance for water-gas shift(WGS)***,we report the single-atom catalyst Pt1/FeOx exhibits excellent medium temperature catalytic performance for WGS reactions by the density functional theory(DFT)calculations and experimental *** calculations indicate that H_(2)O molecules are easily dissociated at oxygen vacancies,and the formed*OH and*O are adsorbed on Pt1 single atoms and the adjacent O atoms,*** studying four possible reaction mechanisms,it is found that the optimal WGS reaction pathway is proceeded along the carboxyl mechanism(pathway III),in which the formation of*COOH intermediates can promote the stability of Pt1/FeOx SAC and the easier occurrence of WGS *** energy barrier of the rate-determining step during the entire reaction cycle is only 1.16 eV,showing the high activity for the medium temperature WGS reaction on Pt1/FeOx SAC,which was verified by experimental ***,the calculated turnover frequencies(TOFs)of CO_(2)and H_(2)formation on Pt1/FeOx at 610 K(337℃)can reach up to 1.14×10^(-3)s^(-1)·site^(-1)through carboxyl *** this work,we further expand the application potential of Pt1/FeOx SAC in WGS reaction.

Digitization of Astronomical Photographic Plates of China and Astrometric Measurement of Single-exposure Plates

From the mid-19th century to the end of the 20th century, photographic plates served as the primary detectors for astronomical observations. Astronomical photographic observations in China began in 1901, and over a century, a total of approximately 30,000 astronomical photographic plates were captured. These historical plates play an irreplaceable role in conducting long-term, time-domain astronomical research. To preserve and explore these valuable original astronomical observational data, Shanghai Astronomical Observatory has organized the transportation of plates, taken during nighttime observations from various stations across the country, to the Sheshan Plate Archive for centralized preservation. For the first time, plate information statistics were calculated. On this basis, the plates were cleaned and digitally scanned, and finally digitized images were acquired for 29,314 plates. In this study, using Gaia DR2 as the reference star catalog, astrometric processing was carried out successfully on 15,696 single-exposure plates, including object extraction, stellar identification,and plate model computation. As a result, for long focal length telescopes, such as the 40 cm double-tube refractor telescope, the 1.56 m reflector telescope at Shanghai Astronomical Observatory, and the 1m reflecting telescope at Yunnan Astronomical Observatory, the astrometric accuracy obtained for their plates is approximately 0."1–0."3. The distribution of astrometric accuracy for medium and short focal length telescopes ranges from 0."3 to 1."0. The relevant data of this batch of plates, including digitized images and a stellar catalog of the plates, are archived and released by the National Astronomical Data Center. Users can access and download plate data based on keywords such as station, telescope, observation year, and observed celestial coordinates.

Myeloid-derived Suppressor Cells Activate Liver Natural Killer Cells in a Murine Model in Uveal Melanoma

Objective Elevated myeloid-derived suppressor cells(MDSCs)in many malignancies are associated with the increased risk for metastases and poor ***,a mouse model of intraocular melanoma was established to explore how MDSCs influence liver *** In this study,murine B16LS melanoma cells were transplanted into the posterior compartment(PC)of the eye of C57BL/6 *** from the liver of naive mice and mice bearing melanoma liver metastasis were isolated using isotonic Percoll centrifugation,examined by flow cytometry for their expression of Gr1,CD11b,F4/80,RAE-1,and Mult-1,and further isolated for MDSCs and natural killer(NK)*** effects of MDSCs on NK cells were tested by coculturing and assessing the ability of NK cells to produce interferon-gamma(IFN-γ)by ELISA and NK cell cytotoxicity by 3H-thymidine incorporation *** impact of IFN-γon liver metastases was examined via selectively depleting IFN-γin *** The results showed that mice with liver metastases had increased levels of CD11b+Gr1+F4/80+as well as CD11b+Gr1+F4/80−*** significantly enhanced the generation of IFN-γtogether with the cytotoxicity of the NK ***,these effects were cell-cell *** IFN-γwas not of a toxic nature to the melanoma cells,it profoundly inhibited B16LS cell *** IFN-γin vivo led to increased liver *** All these findings first revealed that MDSCs accumulated in liver metastasis of intraocular melanoma could activate the NK cells to produce an effective anti-tumor immune ***,the MDSCs’performance in different tumor models would need more investigation to boost current immunotherapy modalities.

Geometrical transition properties of vortex cavitation and associated flow-choking characteristics in poppet valves

Poppet valves have become increasingly significant in ensuring precise digital flow rate and pressure control in hydraulic systems,necessitating a more profound understanding of the geometrical properties of cavitation in them,as well as associated flow-choking *** a comparative analysis with experimentally observed cavity images,we found that large eddy simulation(LES)turbulence modeling effectively replicates the geometrical properties of cavitation in these *** analysis demonstrated that cavitation is generated from vortices that result from the interaction between the notch contracta flow and the surrounding fluid *** in the internal or external vena contracta conditions result in fixed or discrete cavities,and the length-to-diameter ratio serves as a measure of the transition between internal and external vena contracta flow *** study establishes a threshold length-to-diameter ratio of approximately 2 for the tested poppet *** specifically,in notch structures with a smaller valve opening,longer sealing length,and smaller throttling angle(corresponding to a larger length-to-diameter ratio),the liquid-to-vapor transfer process is more evident than that in the reverse direction.A long-standing vapor cavity becomes fixed inside the notch,leading to a more pronounced flow-choking *** contrast,for structures with a smaller length-to-diameter ratio,the cavitation process for discrete vapor cavities is more complete,ensuring fluid flow continuity and significantly reducing the occurrence of the flow-choking phenomenon.

Potential role of microRNA-503 in Icariin-mediated prevention of high glucose-induced endoplasmic reticulum stress

The Boundary Element Method for Ordinary State-Based Peridynamics

The peridynamics(PD),as a promising nonlocal continuum mechanics theory,shines in solving discontinuous *** to now,various numerical methods,such as the peridynamic mesh-free particlemethod(PD-MPM),peridynamic finite element method(PD-FEM),and peridynamic boundary element method(PD-BEM),have been ***-BEM,in particular,outperforms other methods by eliminating spurious boundary softening,efficiently handling infinite problems,and ensuring high computational ***,the existing PD-BEM is constructed exclusively for bond-based peridynamics(BBPD)with fixed Poisson’s ratio,limiting its applicability to crack propagation problems and scenarios involving infinite or semi-infinite *** this paper,we address these limitations by introducing the boundary element method(BEM)for ordinary state-based peridynamics(OSPD-BEM).Additionally,we present a crack propagationmodel embeddedwithin the framework ofOSPD-BEM to simulate crack *** validate the effectiveness of OSPD-BEM,we conduct four numerical examples:deformation under uniaxial loading,crack initiation in a double-notched specimen,wedge-splitting test,and threepoint bending *** results demonstrate the accuracy and efficiency of OSPD-BEM,highlighting its capability to successfully eliminate spurious boundary softening phenomena under varying Poisson’s ***,OSPDBEMsignificantly reduces computational time and exhibits greater consistencywith experimental results compared to PD-MPM.

Underwater Pulse Waveform Recognition Based on Hash Aggregate Discriminant Network

Underwater pulse waveform recognition is an important method for underwater object *** existing works focus on the application of traditional pattern recognition methods,which ignore the time-and space-varying characteristics in sound propagation channels and cannot easily extract valuable waveform *** propagation channels in seawater are time-and space-varying convolutional *** the extraction of the waveform features of underwater acoustic signals,the effect of high-accuracy underwater acoustic signal recognition is identified by eliminating the influence of time-and space-varying convolutional channels to the greatest extent *** propose a hash aggregate discriminative network(HADN),which combines hash learning and deep learning to minimize the time-and space-varying effects on convolutional channels and adaptively learns effective underwater waveform features to achieve high-accuracy underwater pulse waveform *** the extraction of the hash features of acoustic signals,a discrete constraint between clusters within a hash feature class is *** constraint can ensure that the influence of convolutional channels on hash features is *** addition,we design a new loss function called aggregate discriminative loss(AD-loss).The use of AD-loss and softmax-loss can increase the discriminativeness of the learned hash *** results show that on pool and ocean datasets,which were collected in pools and oceans,respectively,by using acoustic collectors,the proposed HADN performs better than other comparative models in terms of accuracy and mAP.

The topological pumping has been instrumental in advancing our understanding of topological phase transitions in various physical systems,which can be extended to uncover intriguing higher-order topological phases in the lower-dimensional ***,we propose a theoretical exploration of topological dipole pumping on an acoustic square superlattice by cyclically modulating intracell couplings,which shares the topological nature of an extended three-dimensional system with chiral hinge *** the multipole chiral numbers,we characterize the higher-order topological phases that arise during the *** evolution of topological phase transitions is verified by numerical simulations and shows corner states are transferred across the *** findings can inspire the construction of chiral hinge states in artificial crystals,opening up new possibilities for the design of devices allowing the unidirectional propagation of sound.

Dynamics modeling and optimal control for multi-information diffusion in Social Internet of Things

As an ingenious convergence between the Internet of Things and social networks,the Social Internet of Things(SIoT)can provide effective and intelligent information services and has become one of the main platforms for people to spread and share ***,SIoT is characterized by high openness and autonomy,multiple kinds of information can spread rapidly,freely and cooperatively in SIoT,which makes it challenging to accurately reveal the characteristics of the information diffusion process and effectively control its *** this end,with the aim of exploring multi-information cooperative diffusion processes in SIoT,we first develop a dynamics model for multi-information cooperative diffusion based on the system dynamics theory in this ***,the characteristics and laws of the dynamical evolution process of multi-information cooperative diffusion are theoretically investigated,and the diffusion trend is *** this basis,to further control the multi-information cooperative diffusion process efficiently,we propose two control strategies for information diffusion with control objectives,develop an optimal control system for the multi-information cooperative diffusion process,and propose the corresponding optimal control *** optimal solution distribution of the control strategy satisfying the control system constraints and the control budget constraints is solved using the optimal control ***,extensive simulation experiments based on real dataset from Twitter validate the correctness and effectiveness of the proposed model,strategy and method.
