Molecular Clone, Expression, and Prediction of Construction and Function to Key Genes of Interleukin Family of Porcine

This research was to clone, express, and analyze the structure and function of major molecules of porcine interleukin family. Genes of porcine interleukin family were cloned by RT-PCR from stimulated porcine PBMC by LPS and PHA, and then expressed in E. coli, and the structure and function of these molecules were predicted by ExPASY. The results showed that genes of IL-4, IL-6, and IL-18 were successfully cloned and expressed. Furthermore, the expression products of recombinant IL-4 and IL-6 both have multiple biological activities. By analyzing these genes with the NCBI/GenBank data, the homologies of the nucleotide acid sequence are 99.25, 99.21, and 100%, respectively, and have great species differences when compared with other animal species. The results of the prediction showed that all these molecules contain several phosphorylation, glycosylation, protein kinase, and signal transduction bonding sites in secondary structure, and all are compact globularity protein in space configuration. These characteristics of structure are the basis for their multiple biological functions. The genes, structure and function of key molecular of porcine interleukin family were successfully cloned, expressed, and analyzed in this paper.

With the advancing of location-detection technologies and the increasing popularity of mobile phones and other location-aware devices,trajectory data is continuously *** large-scale trajectories provide opportunities for various applications,the locations in trajectories pose a threat to individual ***,there has been an interesting debate on the reidentifiability of individuals in the Science *** main finding of Sánchez et *** exactly opposite to that of De Montjoye et al.,which raises the first question:"what is the true situation of the privacy preservation for trajectories in terms of reidentification?''Furthermore,it is known that anonymization typically causes a decline of data utility,and anonymization mechanisms need to consider the trade-off between privacy and *** raises the second question:"what is the true situation of the utility of anonymized trajectories?''To answer these two questions,we conduct a systematic experimental study,using three real-life trajectory datasets,five existing anonymization mechanisms(i.e.,identifier anonymization,grid-based anonymization,dummy trajectories,k-anonymity andε-differential privacy),and two practical applications(i.e.,travel time estimation and window range queries).Our findings reveal the true situation of the privacy preservation for trajectories in terms of reidentification and the true situation of the utility of anonymized trajectories,and essentially close the debate between De Montjoye et *** Sánchez et *** the best of our knowledge,this study is among the first systematic evaluation and analysis of anonymized trajectories on the individual privacy in terms of unicity and on the utility in terms of practical applications.

Crosslinking reactions of high density polyethylene with low peroxide concentrations ranging from 0.1 wt% to 1.0 wt% at temperatures of 170, 180 and 190 ℃ were monitored by rheological measurements. A critical gel forms at the peroxide concentration of 0.2 wt%, where the transition from long chain branching generation to crosslinking network formation could occur. Rheokinetics of crosslinking can be fitted well by Ding-Leonov＇s model. The curing rate k2 at the earlier stage exhibits about 3 times acceleration per 10 ~C with increasing temperature, while the equilibrium modulus G＇ at the fully cured stage is almost independent of temperature. Influences of crosslinking on the subsequent crystallization behaviors were detected by DSC measurements. Above the critical gel concentration, crystallization is largely retarded as evidenced by the lower crystallization temperature Tc and crystallinity Xc due to the network formation. The secondary crystallization valley located at the temperature near 80 ℃ can be observed above the critical concentration, which becomes more evident with the increasing peroxide concentration and curing temperature. This phenomenon provides another evidence of crystallization retardation by the crosslinking network.

Study on the Effects of Cytokine Level in Ducklings with Selenium Poisoning

One-day-old healthy ducklings （n = 100） were randomly allocated into two groups （n = 50, respectively） to investigate the effect of cytokines on ducklings with selenium poisoning. The ducklings in control group （Group Ⅰ） were fed with full rations containing 0.3 mg kg^-1 selenium, and the ducklings in selenium poisoning group （Group Ⅱ ） were fed with seleniumexcessive rations containing 8.0 mg kg^-1 selenium. One mL blood was collected by cardiac puncture at d 15, 30, 60, and 90 （n = 10, respectively）, serum was separated by centrifugation. Contents of cytokines were determined by the radioimmunity method （RIA）. The results showed that the contents of IL-113, IL-213 in Group Ⅱ were higher than that in control group at d 15 （P〉0.05）, but were significantly lower at d 30, 60, and 90 （P〈0.01）; the contents of IL-8β in Group II were significantly lower than that in control group （P〈0.01）; the contents of erythropoietin （EPO） and IGF-2 in Group II were significantly lower in different degree than that in control group at d 60 and 90 （P〈0.01）; the contents of granulocyte- macrophage colony stimulating factor （GM-CSF） in the two groups were lower in different degree and significantly lower at d 60 and 90 （P〈0.01）. It can be concluded that the excessive selenium could result in a decrease in immunity and a series of clinical pathology changes, such as anemia, tissue hemorrhage, and necrosis.

To investigate the adjuvant potential of porcine IL-4 and IFN-γ in mice and pigs, the genes of porcine IL-4 and IFN-γ were cloned and the recombinant mammalian expression plasmids were constructed for in vivo expression of the cytokines. Adjuvant effects of recombinant expression plasmids of IL-4 and IFN-γ （pcDNA-IL-4, pcDNA-IFN-γ） co-administrated with Cysticercus cellulosae crude antigen or TSOL18 recombinant protein antigen have been carried out in mice and pigs, respectively. We have demonstrated that recombinant plasmids of the cytokines as an adjuvant could induce stronger immune response in mice and pigs. With the C. cellulosae parasite antigen, porcine pcDNA-IL-4 induced higher specific antibody of immunized mice than pcDNA-IFN-γ. But pcDNA-IFN-γ is significantly stronger than that of no adjuvant or empty plasmids with the antigen control group. For the TSOL18 recombinant protein antigen vaccine, pcDNA-IL-4 still had a stronger ability to enhance specific antibody in swine than pcDNA-IFN-γ （P 〈 0.01）, but the immune protective rate was lower in challenged pigs （only 68.7%）. Although pcDNA-IFN-γ showed lower specific antibody, the protection rate was very high （91%） than other group （P 〈 0.01）. This study indicated that the recombinant expression plasmids of porcine IL-4 and IFN-γ display stronger adjuvant effects to C. cellulosae vaccine, further research should be carried out for understanding of the interaction mechanism.

Synthesis and Photocatalytic Activity of One-dimensional ZnO-Zn2SnO4 Mixed Oxide Nanowires

Mixed oxide photocatalysts, ZnO-Zn2SnO4 （ZnO-ZTO） nanowires with different sizes were prepared by a simple thermal evaporation method. The ZnO-ZTO nanowires were characterized with a scanning electron microscope, X-ray diffraction, high-resolution transmission electron microscopy, energy-dispersive spectrom- eter, and X-ray photoelectron spectra. The photocatalytic activity of the ZnO-ZTO mixed nanowires were studied by observing the photodegradation behaviors of methyl orange aqueous solution. The results suggest that the ZnO-ZTO mixed oxide nanowires have a higher photocatalytic activity than pure ZnO and Zn2SnO4 nanowires. The photocatalyst concentration in the solution distinctly affects the degradation rate, and our results show that higher photodegradation efficiency can be achieved with a smaller amount of ZnO-ZTO nanowire catalyst, as compared to the pure ZnO and ZTO nanowires. Moreover, the photocatalytic activity can also be enhanced by reducing the average diameter of the nanowires. The activity of pure ZnO and ZTO nanowires are also enhanced by physically mixing them. These results can be explained by the synergism between the two semiconductors.

Superoxide dismutase 2(SOD2)-mediated gene therapy has significant protective effects against kanamycin-induced hearing loss and hair cell loss in the inner ear,but the underlying mechanisms are still ***,an in vivo aging model of mitochondrial DNA(mtDNA)4834 deletion mutation was established using D-galactose,and the effects of noise or kanamycin on inner ear injury was *** subjected to mtDNA4834 mutation via D-galactose administration showed hearing loss characterized by the disruption of inner ear structure(abnormal cell morphology,hair cell lysis,and the absence of the organ of Corti),increased SOD2 promoter methylation,and an increase in the degree of *** to noise or kanamycin further contributed to the effects of ***2 overexpression induced by viral injection accordingly counteracted the effects of noise and kanamycin and ameliorated the symptoms of hearing loss,suggesting the critical involvement of SOD2 in preventing deafness and hearing-related *** PI3K and MAPK signaling pathways were also regulated by noise/kanamycin exposure and/or SOD2 overexpression,indicating that they may be involved in the therapeutic effect of SOD2 against age-related hearing loss.

Saposhnikovia divaricata is a valuable Chinese medicinal herb; the transformation from vegetative growth to reproductive growth may lead to the decrease of its pharmacological activities. Therefore, the study of bolting and flowering for Saposhnikovia divaricata is warranted. The present study aimed to reveal differentially expressed genes(DEGs) and regularity of expression during the bolting and flowering process, and the results of this study might provide a theoretical foundation for the suppression of early bolting for future research and practical application. Three sample groups, early flowering, flower bud differentiation, and late flowering(groups A, B, and C, respectively) were selected. Transcriptomic analysis identified 67, 010 annotated unigenes, among which 50, 165 were differentially expressed including 16, 108 in A vs B, and 17, 459 in B vs C, respectively. Gene ontology(GO) and Kyoto Encyclopedia of Genes and Genomes(KEGG) pathway functional classification analysis were performed on these differentially expressed genes, and five important pathways were significantly impacted(P ≤ 0.01): plant circadian rhythm, other glycan degradation, oxidative phosphorylation, plant hormone signal transduction, and starch and sucrose metabolism. Plant hormone signal transduction might play an important role in the bolting and flowering process. The differentially expressed indole-3-acetic acid(IAA) gene showed significant down-regulation during bolting and flowering, while the transport inhibitor response 1(TIR1) gene showed no significant change during the bolting process. The expression of flowering related genes FLC, LYF, and AP1 also showed a greater difference at different development stages. In conclusion, we speculate that the decrease in auxin concentration is not caused by the degrading effect of TIR1 but by an alternative mechanism.
