L-type calcium channel blockers enhance 5-HTP-induced antinociception in mice

AIM: To investigate the involvement of L-type Ca2+ channels in antinociceptive action induced by the 5-HT precursor,5-hydroxytryptophan (5-HTP). METHODS: Female Kunming mice were treated with either 5-HTP (20-80 mg/kg,ip) alone, or the combination of 5-HTP and fluoxetine (2-8 mg/kg, ip), pargyline (15-60 mg/kg, ip), nimodipine (2.5-10 mg/kg, ip), nifedipine (2.5-10 mg/kg, ip), verapamil (2.5-10 mg/kg, ip), CaCl2 (5-20 mmol/L, icv), or EGTA(0.5-3 mmol/L, icv) prior to the hot-plate test (55 C, hind-paw licking latency). In addition, locomotor activity inmice treated with 5-HTP alone was measured using an ambulometer with five activity boxes. RESULTS: Ipinjection of 5-HTP alone had no influence on the spontaneous locomotor activity, whereas dose-dependently in-creased the latency to licking hind-paw in the hot-plate test in mice. The inhibitory effects of 5-HTP on nociceptiveresponse were significantly enhanced by fluoxetine in the mouse hot-plate test. At a sub-effective dose, pargylinecould cause a leftward shift in the dose-response curve of 5-HTP-induced antinociception. Co-administration with5-HTP and nimodipine, nifedipine, or verapamil obviously potentiated the antinociceptive effects elicited by ***, 5-HTP-induced antinociception was antagonized by CaCl2 and enhanced by EGTA injected icv in themouse hot-plate test. CONCLUSION: These findings suggest that systemic administration of 5-HTP may yield theantinociceptive effects, which are related to Ca2+ influx from extracellular fluid through L-type Ca2+ channels.

This study aims to explore the expression of stanniocalcin 2(STC2)gene in breast cancer and its clinical *** patients with breast cancer from Zhongnan Hospital of Wuhan University admitted during March 2014 to October 2014 were enrolled in this *** the tissues used in this experiment included 50 cases of breast cancer tissues and corresponding 50 cases of paracancer normal breast tissues with complete patients'*** real-time quantitative polymerase chain reaction(qPCR)was applied to detect the expression of STC2 gene in 50 cases of breast cancer and paracancer normal breast *** results showed that the expression level of STC2 gene in 50 cases of breast cancer tissues was significantly higher than that in paracancer normal breast tissues(P0.05).In conclusion,the abnormal overexpression of STC2 gene may play a role in the development and progression of breast cancer,and it can be used as an independent metastasis and prognostic factor of breast *** addition,STC2 gene probably promotes the development and metastasis of breast cancer by interacting with estrogen and ER,and it may become a new direction for breast cancer endocrine therapy.

Cell karyotyping in patients with small lymphocytic lymphoma/chronic lymphocytic leukemia （SLL/CLL） is not easy to success, and small genomic lesions （〈5 Mb） are not routinely detected by this method. It is likely that a complete genomic characterization of CLL requires a combination of fluorescence in situ hybridization （FISH）, single nucleotide polymorphism （SNP） array profiling for comprehensive genome-wide analysis of acquired genomic copy number aberrations （aCNAs） and loss-of-heterozygosity （LOH） in dominant clones, and karyotyping for detection of balanced translocations, isochromosomes, and marker chromosomes. SNP array analysis can reveal chromothripsis, a phenomenon by which regions of the cancer genome are shattered and recombined to generate frequent oscillations between the lower and the higher DNA copy number states. This study provided cytogenetic findings in a CLL/SLL patient with v-myc avian myelocytomatosis viral oncogene homolog （C-MYC）-amplification by FISH, in which SNP arrays detected profound genomic upheaval due to chromothripsis that may lead to malignant transformation.

Three new lactones,xylanilyticolides A-C(1-3),were isolated from cultures of the actinomycete Promicromonospora xylanilytica YIM *** structures were elucidated by 1D and 2D NMR spectroscopic data in conjunction with HRESIMS *** 1 exhibited potent cytotoxicities against five human cancer cell lines HL-60,A-549,SMMC-7721,MCF-7 and SW480 with the IC_(50) values of 3.9,15.2,11.2,5.9,and 4.7 pM,respectively.

Effect of sensor quantity on measurement accuracy of log inner defects by using stress wave

Wood nondestructive testing （NDT） is one of the high efficient methods in utilizing wood. This paper explained the principle of log defect testing by using stress wave, and analyzed the effects of sensor quantity on defect testing results by using stress wave in terms of image fitting degree and error rate. The results showed that for logs with diameter ranging from 20 to 40 cm, at least 12 sensors were needed to meet the requirement which ensure a high testing accuracy of roughly 90% of fitness with 0.1 of error rate. And 10 sensors were recommended to judge the possible locations of defects and 6 sensors were sufficient to decide whether there were defects or not.

Manufacture is facing more furiously competition in the new century. It tends to be globalized. Rapid response and technology innovation have become the key factor to the success of manufacture enterprise. At present, internet-based manufacturing develops rapidly. With the development of engineering material and machining techniques, better cutting properties of metal cutting tools are required. The world is paying more attention to the study of indexable carbide inserts with three-dimensional complex grooves which can affect cutting properties directly and chip-controlling. So the variety and specification of indexable insert develop quickly and the application of insert with three-dimensional complex chip-former becomes wider and wider. Because milling is an interrupted cutting, the process of milling is complicated and the failure of the milling inserts is severe. It is important to choose suitable milling inserts groove for particular machining conditions before actual machining, it will extend tool life and raise productivity. Optimization and choice of inserts with complex groove via internet may serve for manufacture enterprises all over the world. The milling inserts with three-dimensional complex grooves are optimized and chosen via Internet in this paper. The face-milling process is studied, with the mathematics models of cutting force for different shape of cutting edges established, the cutting forces predicted and stress-fields of different insert grooves analyzed by FEM, according to the predictable results and the most suitable insert groove for particular machining conditions are optimized and chosen. The complex groove optimization and choice are based on a client/server model, with the client and server being on different machines across the internet. Communication between the client and server uses the TCP/IP. The result of optimization and choice will be sent to the users via Internet. AIM:To investigate the prognostic value of KRAS mutation,and phosphatase and tensin (PTEN) expression in Chinese metastatic colorectal cancer metastatic colorectal cancer (mCRC) patients treated with ***:Ninety Chinese mCRC patients treated with cetuximab were evaluated for KRAS mutation and PTEN protein expression by DNA sequencing of codons 12 and 13 and immunohistochemistry,*** then selected 61 patients treated with cetuximab,either in combination with chemotherapy,or alone as a second-line or third-line regimen to assess whether KRAS mutation or PTEN protein expression is associated with the response and the survival time of mCRC patients treated with ***:KRAS mutation was found in 30 (33.3%) tumor samples from the 90 patients,and positive PTEN expression was detected in 58 (64.4%) of the 90 *** the 61 patients who were treated with cetuximab as a second-line or third-line regimen,the resistance to cetuximab was found in 22 patients with KRAS mutation and in 39 patients without KRAS mutation,with a response rate of 4.5% and 46.1% respectively (P=0.001),a shorter median progression-free survival (PFS) time of 14 ± 1.3 wk and 32 ± 2.5 wk respectively (P < 0.001),a median overall survival (OS) time of 11 ± 1.2 mo and 19 ± 1.8 mo respectively (P < 0.001),as well as in 24 patients with negative PTEN expression and in 37 patients with positive PTEN expression respectively (P < 0.001),with a responsive rate of 4.2% and 48.6% respectively,a shorter median PFS survival time of 17 ± 2.0 wk and 28 ± 1.9 wk respectively (P=0.07),and a median OS time of 11 ± 1.3 mo and 18 ± 1.9 mo respectively (P=0.004).Combined KRAS mutation and PTEN expression analysis showed that the PFS and OS time of patients with two favorable prognostic factors were longer than those of patients with one favorable prognostic factor or no favorable prognostic factor (P < 0.001).CONCLUSION:KRAS mutation and PTEN protein expression are significantly corr
