Multidrug resistance （MDR） is the major impediment to cancer chemotherapy. The expression of lung resistance-related protein （LRP）, a non-ATP-binding cassette （ABC） transporter, is high in tumor cells, resulting in their resistance to a variety of cytotoxic drugs. However, the function of LRP in tumor drug resistance is not yet explicit. Our previous studies had shown that Kinesin KIF4A was overexpressed in cisplatin （DDP）-resistant human lung adenocarcinoma cells （A549/DDP cells） compared with A549 cells. The expression of KIF4A in A549 or A549/DDP cells significantly affects cisplatin resistance but the detailed mechanisms remain unclear. Here, we performed co-immunoprecipitation experiments to show that the tail domain of KIF4A interacted with the N-terminal of LRP. Immunofluorescence images showed that both the ability of binding to LRP and the motility of KIF4A were essential for the dispersed cytoplasm distribution of LRP. Altogether, our results shed light on a potential mechanism in that motor protein KIF4A promotes drug resistance of lung adenocarcinoma cells through transporting LRP-based vaults along microtubules towards the cell membrane. Thus KIF4A might be a cisplatin resistance-associated protein and serves as a potential target for chemotherapeutic drug resistance in lung cancer.

无人机高空航拍图像中车辆像素占比极低,目标可视化信息较少,在目标检测任务中容易漏检和误检。因此,本文提出一种基于改进YOLOX(You Only Look Once X)的无人机高空航拍视角下小尺度车辆精确检测方法。首先,为增强网络对低级特征的提取能力,在原始YOLOX预测头部增加一个160 pixel×160 pixel的浅层特征提取网络;其次,在骨干网络后端嵌入基于归一化的注意力机制模块(Normalization-based Attention Module,NAM),以抑制冗余的非显著特征表达;最后,为了增大小尺度车辆的相对像素比,提升网络捕捉有效特征信息的能力,提出一种基于滑动窗口的图像切分检测方法。试验结果表明,改进YOLOX网络表现出良好的检测效能,检测精度达到了84.58%,优于典型的目标检测网络Faster R-CNN(79.95%)、YOLOv3(83.69%)、YOLOv5(84.31%)及YOLOX(83.10%)。此外,改进YOLOX能够有效解决无人机高空航拍图像中小尺度车辆的漏检和误检问题,且预测框更贴合车辆的实际轮廓;同时,在不同航拍高度的目标检测任务中具有较高的鲁棒性。

The mechanisms of magnetoreception have been proposed as the magnetitebased, the chemical radical-pair and biocompass model, in which magnetite particles, the cryptochrome (Cry) or iron-sulfur cluster assembly 1 (IscA1) may be involved. However, little is known about the association among the molecules. Here we investigated the molecular characterization and the mRNA expression of IscA1 in different developmental stages, tissues and magnetic fields in the migratory brown planthopper (BPH), Nilaparvata lugens. NliscA1 contains an open reading frame of 390 bp, encoding amino acids of 129, with the predicted molecular weight of 14.0 kDa and the isoelectric point of 9.10. Well-conserved Fe-S cluster binding sites were observed in the predicted protein. Phylogenetic analysis demonstrated NliscA1 to be clustered into the insect's IscA1. NliscA1 showed up-regulated mRNA expression during the period of migration. The mRNA expression of NliscA1 could be detected in all the three tissues of head, thorax and abdomen, with the highest expression level in the abdomen. For the macropterous migratory Nilaparvata lugens, mRNA expression of NliscA1 and N. lugens cryptochromel (Nlcry1) were up-regulated under the magnetic fields of 5 Gauss and 10 Gauss in strength (vs. local geomagnetic field), while N. lugens cryptochrome 2 (Nlcry2) remained stable. For the brachyterous non-migratory Nilaparvata lugens, no significant changes were found in mRNA expression of NliscA1, Nlcry1 and Nlcry2 among different magnetic fields. These findings preliminarily reveal that the expression of NliscA1 and Nlcry1 exhibited coordinated responses to the magnetic field. It suggests some potential associations among the putative magneto-sensitive molecules of cryptochrome and iron-sulfur cluster assembly.

Responses of atmospheric carbon dioxide(CO_(2))density to geomagnetic secular variation are investigated using the Whole Atmosphere Community Climate Model-eXtended(WACCM-X).Our ensemble simulations show that CO_(2) volume mixing ratios(VMRs)increase at high latitudes and decrease at mid and low latitudes by several ppmv in response to a 50%weakening of the geomagnetic *** significant changes in CO_(2) are mainly found above~90 km altitude and primarily redetermine the energy budget at~100-110 *** analysis of transformed Eulerian mean(TEM)circulation found that CO_(2) change is caused by enhanced upwelling at high latitudes and downwelling at mid and low latitudes as a result of increased Joule *** further analyzed the atmospheric CO_(2) response to realistic geomagnetic weakening between 1978 and 2013,and found increasing(decreasing)CO_(2) VMRs at high latitudes(mid and low latitudes)*** the first time,our simulation results demonstrate that the impact of geomagnetic variation on atmospheric CO_(2) distribution is noticeable on a time scale of decades.

Curcumin and its derivatives have good electrical and optical properties due to the highly symmetric structure of delocalized π electrons. Apart from that, curcumin and its derivatives can interact with numerous molecular targets, thereby exerting less side effects on human body. The fluorescence emission wavelength and fluorescence intensity of curcumin can be enhanced by modifying its π-conjugated system and β-diketone structure. Some curcumin-based fluorescent probes have been utilized to detect soluble/insoluble amyloid-β protein, intracranial reactive oxygen species, cysteine, cancer cells, etc. Based on the binding characteristics of curcumin-based fluorescent probes with various target molecules, the factors affecting the fluorescence intensity and emission wavelength of the probes are analyzed, in order to obtain a curcumin probe with higher sensitivity and selectivity. Such an approach will be greatly applicable to in vivo fluorescence imaging.

The Clinical Practice Guideline on Acupuncture and Moxibustion: Gastroesophageal Reflux Disease (GERD) (WFAS 007.9-2023), officially released by the World Federation of Acupuncture-Moxibustion Societies (WFAS) on October 9, 2023, represents the inaugural acupuncture and moxibustion clinical practice guideline dedicated to GERD globally. This guideline outlines its purpose, scope, target audience, and relevant environments, along with detailing the acupuncture and moxibustion treatment methodology for GERD, as well as the guideline development process and recommendations. This article specifically emphasizes the recommendations of the guideline, highlighting the crucial importance of both the dissemination and adherence to this guideline to standardize acupuncture and moxibustion treatments for GERD. Such standardization plays a pivotal role in the advancement and widespread utilization of acupuncture and moxibustion in the management of GERD.
