RNA interference of pax2 inhibits growth of transplanted human endometrial cancer cells in nude mice

The development of human endometrial carcinoma(HEC) is a complex pathologic process involves several oncogenes and tumor suppressor *** full-length paired-box gene 2(pax2),a recently discovered oncogene,promotes cell proliferation and growth and inhibits apoptosis of HEC ***,we examined the effect of pax2 small interfering RNA(siRNA) on the growth of transplanted HEC cells in nude *** expression of Pax2 in 21 cases of normal endometrium and 38 cases of HEC was examined by immohistochemistry(IHC).HEC models were developed by subcutaneously transferring HEC cells into nude mice,followed by treatment with empty lentivirus vector,lentivirus vector-based pax2 siRNA,and phosphate buffered saline,*** weeks later,tumor size was measured,tumor inhibition rate was calculated,and histological analyses were conducted after staining with hematoxylin and *** expression of Pax2 and Bcl-2 was detected by Western blot;proliferating cell nuclear antigen(PCNA) was detected by *** differences were observed in the positive rate of Pax2 between normal endometrium and HEC(14.2% vs.60.5%,Plia stain decreased,tumor growth was inhibited,and PCNA,Pax2,and Bcl-2 expression was reduced in HEC models treated with pax2 *** results indicate that Pax2 expression is related to HEC tumor biology with the increased expression of Pax2 correlated to ***2 siRNA down-regulates Pax2 expression and inhibits tumorigenesis of HEC in nude mice,possibly due to cell apoptosis and the inhibition of tumor proliferation induced by down-regulation of Bcl-2.

Changes of brain neuropeptide Y and its receptors in rats with flurazepam tolerance and dependence

Aim: Anticonvulsant tolerance and dependence are two obstacles that restrict the clinical use of benzodiazepines (BDZ). In order to explore the mechanism of these two adverse reactions, changes of neuropeptide Y (NPY) and its receptors in the hippocampus of rat models, in relation to flurazepam (FZP, a member of BDZ) tolerance and dependence, were investigated. Methods: The mRNA of preproNPY and its receptors (Y1, Y2, and Y5) in the hippocampus were determined by competitive RT-PCR, and the distribution of NPY in the hippocampus was examined by immunohistochemistry. Results: A decrease of preproNPY mRNA in the hippocampus was found in tolerant and dependent rats. The level of preproNPY mRNA in the hippocampus was reversely correlated with the degree of tolerance and dependence, measured by the threshold of pentylenetetrazol-induced seizures. Immunohistochemistry indicated a decrease of NPY-immunoreactive material in neurons of the CA1, CA3, and dentate gyrus regions of both tolerant and dependent rats. The mRNA of NPY receptors Y,and Y5decreased in tolerant rats but did not change in dependent rats. The mRNA of NPY receptor Y2increased in tolerant rats but decreased in dependent rats. Conclusion: A decrease of NPY in the hippocampus might be involved in anticonvulsant tolerance and dependence following long-term treatment with FZP. Y,, Y2, and Y5mRNA were also altered in FZP tolerance and dependence.

Evaluation of the shear stability of aerobic granular sludge from a pilot-scale membrane bioreactor:Establishment of a quantitative method

This work established a quantitative method to access the shear stability of aerobic granular sludge(AGS)and validated its feasibility by using the mature AGS from a pilot-scale(50 tons/day)membrane bioreactor(MBR)for treating real municipal *** results showed that the changing rate( S)of the peak area(S)of granule size distribution(GSD)exhibited an exponential relationship(R^(2)≥0.76)with the shear time(y=a−b·cx),which was a suitable indicative index to reflect the shear stability of different AGS *** limiting granule size(LGS)was defined and proposed to characterize the equilibrium size for AGS after being sheared for a period of time,whose value in terms of Dv50 showed high correlation(R^(2)=0.92)with the parameter *** free Ca^(2+)(28.44-34.21 mg/L)in the influent specifically interacted with polysaccharides(PS)in the granule’s extracellular polymeric substance(EPS)as a nucleation site,thereby inducing the formation of Ca precipitation to enhance its Young’s modulus,while Ca2+primarily interacted with PS in soluble metabolic product(SMP)during the initial granulation ***,the Young’s modulus significantly affected the parameter a related to shear stability(R^(2)=0.99).Since the parameter a was more closely related(R^(2)=1.00)to S than that of the parameter b or c,the excellent correlation(R^(2)=0.99)between the parameter a and the wet density further verified the feasibility of this method.

Analysis of structural dynamic characteristics of a high speed light special catamaran

In this paper,structural dynamic characteristics of a high-speed light special catamaran-wave piercing catamaran are analyzed using the FEA software MSC-NASTRAN. The dynamic reduction method is introduced to eliminate the local vibration modes in order to obtain the whole ship＇s mode shapes. In the post-processor, a lot of accessorial methods are adopted to eliminate the local vibrations, so that the whole ship＇s mode shapes can be identified. The modal analysis indicates that the dynamic reduction method fits for mode shapes identifying. In the end,the test results of a catamaran named Frederick G. Greed are used for reference to validate the obtained results. The comparison process shows that the results are credible. A special mode shape, which is quite different with that of conventional monohull ship, is also pointed out. The obtained results provide a valuable reference for the coming computation of catamaran＇s vihration characteristics.

It is investigated the properties of a diode-pumped Nd: Sr5 （PO4）3 F laser that is passively Q- switched by a thin, single crystal GaAs wafer. The short high-peak-power pulses of the laser have been obtained. The pulse energies, pulse widths and pulse repetition rates for different conditions have been measured and the experimental results show that GaAs is an excellent passive Q- switch of the diode-pumped Nd: Sr5（PO4）3F lasers.

Preparation of silver nanoparticles through the reduction of straw-extracted lignin and its antibacterial hydrogel

Silver nanoparticles(Ag NPs)have attracted attention in the field of biomaterials due to their excellent antibacterial ***,the reducing and stabilizing agents used for the chemical reduction of Ag NPs are usually toxic and may cause water *** this work,Ag NPs(31.2 nm in diameter)were prepared using the extract of straw,an agricultural waste,as the reducing and stabilizing *** analysis revealed that the straw extract contained lignin,the structure of which possesses phenolic hydroxyl and methoxy groups that facilitate the reduction of silver salts into Ag *** surfaces of Ag NPs were negatively charged due to the encapsulation of a thin layer of lignin molecules that prevented their *** the prepared Ag NPs were added to the precursor solution of acrylamide,free radical polymerization was triggered without the need for extra heating or light irradiation,resulting in the rapid formation of an Ag NP-polyacrylamide composite *** inhibition zone test proved that the composite hydrogel possessed excellent antibacterial ability due to the presence of Ag *** prepared hydrogel may have potential applications in the fabrication of biomedical materials,such as antibacterial dressings.

Numerical Simulation of Radiation Transport to Improve Microalgae Cultivation in an Air-lift Photobioreactor

A comprehensive numerical model is developed to simulate the growth of microalgae under light/dark cycling *** purpose of this study is to predict the growth rate of Chlorella vulgaris cultivated in photobioreactors(PBRs)in order to improve the light conditions for microalgae and enhance the photosynthetic *** fluid dynamics(CFD)is used to simulate its internal hydrodynamic *** Lagrangian method is employed to track the movement of microalgae *** radiative transfer equation(RTE)is used to obtain light intensity *** combination of light radiation field and microalgae cell motions is used to construct the light history and they are integrated into the model of the photosynthetic units(PSU)to calculate the microalgae growth *** numerical results demonstrate that enhanced light/dark cycling frequency with ordered mixing can promote efficient microalgae *** effect of the vortex flow field generated by the baffles in an air-lift PBR is analyzed for increasing microalgae growth *** using the 1:1 baffle spacing,the biomass production of microalgae is increased by 41.8% compared to the original PBR.

Selenate oxyanion-intercalated NiFeOOH for stable water oxidation via lattice oxygen oxidation mechanism

Transition metal-based compounds can serve as pre-catalysts to obtain genuine oxygen evolution reaction（OER） electrocatalysts in the form of oxyhydroxides through electrochemical ***,the role and existence form of leached oxygen anions are still ***,we selected iron selenite-wrapped hydrated nickel molybdate（denoted as NiMoO/FeSeO） as a pre-catalyst to study the oxyanion *** is surprising to find that SeO2-exists in the catalyst in the form of intercalation,which is different from previous studies that suggest that anions are doped with residual elements after electrochemical activation,or adsorbed on the catalyst *** experiment and theoretical calculations show that the existence of SeO42-intercalation effectively adjusts the electronic structure of NiFeOOH,promotes intramolecular electron transfer and O-O release,and thus lowers the reaction energy *** expected,the synthesized NiFeOOH-SeO only needs 202 and 285 mV to attain 100 and 1000 mA cm-2in 1 M ***,the anion exchange membrane water electrolyzer（AEMWE） consisting of NiFeOOHSeO anode and Pt/C cathode can reach 1 A cm-2at 1.70 V and no significant attenuation within 300 *** findings provide insights into the mechanism,by which the intercalated oxyanions enhance the OER performance of NiFeOOH,thereby facilitating large-scale hydrogen production through AEMWE.
