This study aimed to describe a case of a patient with the spontaneous dissolution of zona pellucida(ZP)during cleavage embryo development after intracytoplasmic sperm injection(ICSI).This patient experienced early abortion due to fetal chromosome 8 abnormality and received ICSI at 31 years of ***-lapse monitoring of embryo cleavage and blastocyst culture was performed,and we found that the ZP of embryos 1,2,4,and 5 dissolved gradually during the cleavage stage of embryo *** blastocysts that developed from 2 pronuclear zygotes were analyzed using next-generation sequencing,and one frozen-thawed blastocyst was transferred,resulting in intrauterine *** results of amniotic fluid puncture showed normal fetal *** our knowledge,this is the first report of ZP dissolution during the cleavage-stage embryo;however,the exact mechanism remains *** morphological changes inferred that the granular matter in the periocular space was related to the dissolution of ZP through time-lapse observation.

The concentration of organochlorine pesticides（OCPs） in tissues and organs of silver carp（Hypophthalmichthys mofitrix） from Guanting Reservoir were investigated to evaluate the pollution potential and distribution of OCPs. A total of 16 OCPs were measured and the concentrations were in the range of 1.61-69.01 ng/g wet weight（ww） for total OCPs, 0.16- 0.75 ng/g ww for HCB. 0.75 -26.80 ng/g ww for SHCH（sum of α-,β-, γ- and δ-HCH） and 0.68-35.94 ng/g ww for SDDT（ sum of p, p＇-DDE, p, p＇-DDD, o, p＇-DDT and p, p＇- DDT）. The mean concentrations of total OCPs, HCB, ∑HCH, and ∑DDT were 18.04, 0.96, 7.14 and 9.28 ng/g ww, respectively. Among the organochlorine pesticides, β-HCH and p, p＇-DDE were the most dominant compounds in tissue and organ with the average concentrations of 4.42 and 8.14 ng/g, respectively. The results obtained in this study show that the levels of 16 OCP residues found in silver carps are low and pose no threat to human health and wildlife fed upon them on the basis of existing related quality guidelines. However, recent input of lindane and DDT might still exist in the area investigated and further investigation should be carried on.

Based on the crystal structure of AHAS/sulfonylurea complex, we obtained 296 compounds with low binding energy towards AHAS via virtual screening. One series of them have been synthesized. Preliminary bioassay indicated that some compounds displayed good herbicidal activity on rape and bamygrass and inhibited AHAS to some extent. This study indicated the rationality of our molecular design based on the crystal structure of target enzyme.

Grain number per-panicle is one of the most important components for rice yield. Spikelets on the primary and secondary branches determine the grain number per-panicle in rice. In this study, we identified a natural mutant, gnp4, lack of lateral spikelet on the secondary branches in the field condition. In addition, the Gnp4 and Lax1-1 double mutant showed dramatically reduced secondary branches and spikelets in panicle at reproductive stage, and tillers at vegetative stage. By map-based cloning approach, and using four F2 segregating populations, the Gnp4 gene was finally mapped to a 10.7-kb region on the long arm of chromosome 4 in rice. In this region, only one gene was predicted, and genomic DNA sequencing of the 10.7-kb region showed no nucleotide differences between the mutant and wild type. Interestingly, we found that the methylation level of several cytosines in the promoter CpG islands region of the predicted gene in gnp4 were different from the wild type. Thus, we propose that the DNA methylation changes at these sites may induce to decrease expression level of Gnp4, consequently, resulting in phenotypic variation.

Polycyclic aromatic hydrocarbon in urban soil from Beijing, China

Polycyclic aromatic hydrocarbons （EPA-PAHs） in the urban surface soils from Beijing were determined using gas chromatography and mass spectrometry （GC-MS）. It is significantly complementary for understanding the PAHs pollution in soil of integrated Beijing city on the basis of the information known in the outskirts. The total concentration of 16 EPA-PAH was from 0.467 to 5.470 μg/g and was described by the contour map. Compound profiles presented that the 4-, 5- and 6-ring PAHs were major compositions. The correlation analysis showed that PAHs have the similar source in the most sampling sites and BaP might be considered as the indicator of PAHs. Characteristic ratios of anthracene （An）/（An＋ phenanthrene （Phe））, fluoranthene （Flu）/（Flu/ pyrene （Pyr）） and benzo[a]pyrene （BaP）/laenzo[g,h,i]perylene （BghiP） indicated that the PAHs pollutants probably mainly originated from the coal combustion and it was not negligible from vehicular emission. The level of PAHs in our study area was compared with other studies.

目的探讨慢性阻塞性肺疾病急性加重期(acute exacerhation of chronic obstructive pulmonary disease,AECOPD)患者治疗前后Th17/Treg免疫平衡、呼出气一氧化氮(fraction of exhaled nitric oxide,FeNO)水平变化及对预后的影响。方法选取AECOPD患者126例,采用肺功能检测仪测定第1秒用力呼气量(forced expiratory volume in one second,FEV1)、第1秒用力呼气量占预计值百分比(FEV1%)、用力肺活量(forced vital capacity,FVC)、用力肺活量占预计值百分比(FVC%)、第1秒用力呼气量占用力肺活量的百分比(FEV1/FVC)。并使用一氧化氮测定仪检测FeNO水平,采用全身炎性反应综合征(systemic inflammaton response syndrome,SIRS)评分进行评分,采用流式细胞仪检测血清辅助性T细胞17(T help 17cells,Th17)、调节性T细胞(T Regulatory cells,Treg)水平,比较治疗前后患者肺功能指标、FeNO水平、SIRS评分、Th17/Treg水平等指标变化情况。结果治疗3个月后,AECOPD患者FEV1%、FEV1/FVC、Th17、Th17/Treg水平显著高于治疗前,FeNO、血清Treg水平、SIRS评分显著低于治疗前(P<0.05)。126例AECOPD患者治疗3个月后存活94例(74.60%);死亡32例(25.40%)。存活组FEV1%、FEV1/FVC、Treg水平显著高于死亡组,FeNO、Th17、Th17/Treg及SIRS评分显著低于死亡组(P<0.05)。多因素Logistic回归分析结果显示,治疗前FEV1%、FEV1/FVC、Treg是AECOPD患者预后的危险因素(P<0.05);FeNO、Th17、Th17/Treg、SIRS评分是AECOPD患者预后的保护因素(P<0.05)。结论AECOPD患者治疗后肺功能提高,Th17/Treg、FeNO降低,肺功能、Th17/Treg免疫平衡、FeNO水平对AECOPD患者疗效评估及预后判断具有重要意义。
