Evidence against the participation of a pharmacokinetic interaction in the protective effect of single-dose curcumin against gastrointestinal damage induced by indomethacin in rats

OBJECTIVE： To determine the role of a pharmacokinetic interaction in the protective effect of curcumin against the gastric damage induced by indomethacin administration as such or as its prodrug acemetacin. METHODS： Wistar rats orally received single dose of indomethacin （30 mg/kg） with and without curcumin （30 mg/kg）; gastric injury was evaluated by determining the total damaged area. Additional groups of rats received an oral single dose of indomethacin （30 mg/kg） or its prodrug acemetacin （34.86 mg/kg） in the presence or absence of curcumin （30 mg/kg）. Indomethacin and acemetacin concentrations in plasma from blood draws were determined by high-performance liquid chromatography Plasma concentration-against-time curves were constructed, and bioavailability parameters, maximal concentration （Cmax） and area under the curve to the last sampling time （AUC0_t） were estimated. RESULTS： Concomitant administration of indomethacin and curcumin resulted in a significantly reduced gastric damage compared to indomethacin alone. However, co-administration of curcumin did not produce any significant alteration in the bioavailability parameters of indomethacin and acemetacin after administration of either the active compound or the prodrug. CONCLUSION： Curcumin exhibits a protective effect against indomethacin-induced gastric damage, but does not produce a reduction of the bioavailability of this nonsteroidal anti-inflammatory drug, indomethacin. Data thus suggest that a pharmacokinetic mechanism of action is not involved in curcumin gastroprotection.

Synthesis and Characterization of a Polymeric Material Blended to Bone Forming Elements

A synthetizing material blended with two distinct proteins (collagen and casein) and mineral mixture, was developed in order to evaluate their properties suitable for possible applications in the biomedical such as inducing the regeneration of damaged bone, either due to an accident or illness. Samples were evaluated by 1) Mechanical properties tests under the bending, 2) Scanning electronic microscopy and 3) Infrared spectroscopy were carried out. The results showed that the developed material has breaking strength and structure characteristics associated with the protein used in their composition. This fact suggests that the used protein determines the resistance of the material, in such a way according to the required use, being able to choose appropriate strength and duration either short or long time. The material composition for specific use, in order to find the most suitable mixture for bone replacement, or induce bone recovery, according to the required properties similar to those of damaged living tissue.

Foveal thickness reduction after anti-vascular endothelial growth factor treatment in chronic diabetic macular edema

AIM：To report foveal thickness reduction in eyes with resolution of macular edema and recovery of a foveal depression after one-year of anti-vascular endothelial growth factor（anti-VEGF） therapy for center-involving diabetic macular edema（DME）.METHODS：Foveal thickness was assessed with optical coherence tomography to determine the central subfield foveal thickness（CSFT） and macular volume in 42 eyes with DME（CSFT〉275 μm）. Evaluations also included measurement of best-corrected visual acuity（BCVA）, and were performed at baseline, and upon foveal depression recovery achieved after 12 monthly intravitreal injections of either 1.5 mg/0.06 mL bevacizumab（n=21） or 0.5 mg/0.05 mL ranibizumab（n=21）. Data was compared to 42 eyes of normally sighted, non-diabetic, healthy individuals with similar age, gender and race ***：Mean baseline BCVA was 0.59±0.04 and 0.32± 0.03 log MAR（P〈0.001） after treatment and resolution of DME, with all, but 3 eyes, showing BCVA improvement. Mean CSFT before treatment was 422.0±20.0 μm, and after treatment, decreased to 241.6±4.6 μm（P〈0.001）, which is significantly thinner than CSFT found in control subjects（272.0±3.4 μm; P〈0.001）. Moreover, in 33/42 DM eyes（79%）, CSTF was thinner than the matched control eye. Macular volume showed comparable results, but with lower differences between groups（control：8.5±0.4 mm^3; DME：8.2±1.0 mm^3; P=0.0267）.CONCLUSION：DME eyes show significantly lower foveal thickness than matched controls after DME resolution achieved with one-year anti-VEGF therapy. Further investigation into the reasonsfor this presumable retinal atrophy using fluorescein angiography and functional parameters as well as establishing possible predictors is warranted. This finding should be considered during the treatment of DME.

BACKGROUND The estimation of left ventricular ejection fraction(LVEF)by 2D echocardiography(2D-ECHO)is the most used tool to assess LV systolic function(LVSF).Global longitudinal strain(GLS)has recently been suggested as a superior method for several *** study explored the association and prevalence of LV systolic dysfunction(LVSD)by using these methods in patients with end-stage renal disease(ESRD)and severe hyperparathyroidism(SHPTH);both associated with cardiovascular events(CEs).AIM To evaluate the myocardial function in patients with ESRD and SHPTH by using the GLS and LVEF measured through conventional *** In 62 patients with ESRD and SHPTH,asymptomatic,and without a history of CEs,LVSF was evaluated by 2D-ECHO,obtaining the EF,by the Simpson biplane method,and GLS by speckle *** The total patients with ESRD had a preserved LVEF(>50%)but abnormal GLS(<13.55%).Additionally,multivariate analysis showed an independent association of GLS and serum parathyroid hormone(PTH),LV mass index,and ***,PTH was independently associated with lateral e'wave and tricuspid regurgitation *** In patients with SHPTH linked to ESRD,the use of GLS by 2D-ECHO is a more sensitive tool than LVEF for detecting LVSD.

Dengue is a global health problem without current specific treatment nor safe vaccines *** severe dengue is related to pre-existing non-neutralizing dengue virus(DENV)antibodies,the role of T cells in protection or pathology is *** cutaneous DENV infection in immunocompetent mice we previously showed the generation of PNA+germinal centers(GCs),now we assessed the activation and proliferation of B and T cells in draining lymph nodes(DLNs).We found a drastic remodelling of DLN compartments from 7 to 14 days post-infection(dpi)with greatly enlarged B cell follicles,occupying almost half of the DLN area compared to*24%in na?ve *** clusters of proliferating(Ki-67+)cells inside B follicles were found 14 dpi,representing*33%of B cells in DLNs but only*2%in noninfected *** GCs,we noticed an important recruitment of tingle body macrophages removing apoptotic *** contrast,the percentage of paracortex area and total T cells decreased by 14–16 dpi,compared to *** randomly distributed Ki-67+T cells were found,similar to non-infected ***69 expression by CD4+and CD8+T cells was minor,while it was remarkable in B cells,representing 1764.7%of change from basal levels 3 *** apparent lack of T cell responses cannot be attributed to apoptosis since no significant differences were observed compared to noninfected *** study shows massive B cell activation and proliferation in DLNs upon DENV *** contrast,we found very poor,almost absent CD4+and CD8+T cell responses.

Influence of Inorganic Salts during the Photooxidative Process of Nitrobenzene

Into the photooxidation process, several factors such as pH, time of irradiation, dose of UV light, lamp power, contaminant concentration, turbidity of the solution and the presence of salts can interfere with the photodegradation of pollutants. This research aims to evaluate the influence of salts： NaCl, MgCl2, CaCI2, BaC12, CuCl2, Na2SO4, MgSO4, MnSO4, FeSO4, CuSO4, Na3PO4, K2CrO4 and K2Cr207 in concentrations of 0.0005 M, 0.005 M and 0.05 M during photodegradation of aqueous solution of 59.5 mg/L of nitrobenzene at a pH of 2.5. It was observed that the presence of salts such as CuCI2, CuSO4, FeSO4, K2CrO4 and K2Cr207 interfere negatively in the system UV/H202 applied for degradation of nitrobenzene; possibly by oxidation of Fe2＋ to Fe3＋ and Cu＋ to Cu2＋ in the Cr case, due to the difficulty of transforming the Cr6＋ to Cr3＋ or because these solutions have color and act as radiation absorbing filter.

Symptomatic changes in postmenopause with different methods of hormonal therapy

Objective: The diversity of opinions on the adverse effects of medications used to treat postmenopausal symptoms has prompted the use of various routes and mechanisms of action that need to be explored because bioavailability of the medications can vary. In order to select the appropriate route of administration for hormonal therapy (HT), it is necessary to determine baseline therapeutic efficacy. Design: We designed a prospective, randomized study consisting of four groups of postmenopausal wo-men: group 1 received oral conjugated estrogens, group 2 received a synthethic steroid, group 3 received estradiol nasally in spray form, and group 4 used transdermal estradiol in the form of patches. Criteria used to evaluate effectiveness was the Greene scale, which evaluate six components. These criteria were applied to each patient before hormonal intervention and then each month for 6 months. Luteinizing hormone (LH), follicle stimulating horone (FSH) and estradiol concentration were determined by chemiluminescence. Student’s t-test was used for intra-group comparisons before and after treatment. Results: There was a significant decrease in the vasomotor and sexual component (p < 0.05) with the use of four HT types. For depression, a difference was observed with synthetic steroids and oral estrogens. Upon analyzing the somatic component there was a decrease in symptoms with nasal and transdermal routes. Psychological changes were observed with the use of oral synthethic steroids and transdermal patches. Anxiety component demonstrated differences with nasal spray and oral estrogens, although all HT forms in this component showed a pattern of irregular changes. Conclusions: Changes in the response could be due each route of administration and medication used. Absorption variability may exist, which has repercussions in the control of symptoms and should be taken into consideration when selecting the appropriate route of administration for patients beginning HT.

Effect of Microwave Power on the Zn2SnO4 Synthesis and Its Use for Photodegradation of Phenol

Spinel structure Zn2SnO4 was successfully synthesized by microwave-assisted hydrothermal process. The effects of the microwave power on the formation and physical properties of the Zn2SnO4 particles are discussed. The products were characterized by X-ray diffraction, atomic force microscopy, infrared spectroscopy, and N2 adsorption. The results indicated that the microwave power had important influence on the formation of the spinel phase. The results also revealed that the physical properties of Zn2SnO4 particles did not change with the increase of the microwave power above 600 W, with 20 min of reaction time. Furthermore, the photocatalytic activity of the Zn2SnO4 particles for the phenol degradation under sunlight was also investigated.

Health Risk Assessment in Children by Arsenic and Mercury Pollution of Groundwater in a Mining Area in Sonora, Mexico

A highly acidic spill from one of the most important copper mines in the Americas (Buenavista del Cobre) occurred in Sonora, Mexico in August, 2014. 40,000 m3 of metals-enriched sulphate solution escaped into the Sonora River representing the worst ecological disaster in Mexican history. The spill affected the economy, water accessibility and the health of residents near the river. Despite the economic importance of mining in this area, no environmental assessment was undertaken. There is no information about the health impact of this event on the local population. The study assesses the health risk among children exposed to arsenic and mercury via groundwater in five communities located in a mining area into the Sonora River Basin, Mexico. Local drinking water from the localities was sampled one year after the spill and it was analyzed for arsenic and mercury by Atomic Absorption Spectrometry coupled with Hydride Generation (AAS/HG). Further, some of the traditional local foods were sampled and analyzed for the same elements. Results indicate that levels of arsenic in drinking water did not exceed the Mexican Norm (25 μg As/L) or the EPA Guideline (10 μg As/L). The concentrations of mercury exceeded the WHO and Mexican Legislation value (1.0 μg Hg/L). The Hazard Quotient (HQ) values for arsenic were >1 in 42% of children exposed and for mercury in 67% of the children. Foods concentrations contained in the ranges of 9.2 to 62.0 μg As/Kg and 0.28 to 42 μg Hg/Kg for arsenic and mercury respectively. These values are below the Codex Alimentarius limits. Children affected by mining activities are at risk of developing chronic diseases associated with low arsenic and mercury exposure via groundwater consumption, without consideration of the contribution from these metals by other important exposures routes.

Acute fatty liver of pregnancy associated with severe acute pancreatitis:A case report

Acute fatty liver of pregnancy is a rare disease that affects women in the third trimester of pregnancy. Although infrequent, the disease can cause maternal mortality. The diagnosis is not always clear until the pregnancy is terminated, and significant complications, such as acute pancreatitis, can occur. Pancreatic involvement typically only occurs in severe cases after the development of hepatic and renal impairment. To date, little knowledge is available regarding how the disease causes pancreatitis. Treatment involves supportive measures and pregnancy interruption. In this report, we describe a case of a previously healthy 26-year-old woman at a gestational age of 27 wk and 6 d who was admitted with severe abdominal pain and vomiting. This case illustrates the clinical and laboratory overlap between acute fatty liver of pregnancy and pancreatitis, highlighting the difficulties in differentiating each disease. Furthermore, the hypothesis for this overlapping is presented, and the therapeutic options are discussed.
