Modulation of c-Jun NH<sub>2</sub>-Terminal (JNK) by Cholinergic Autoantibodies from Patients with Sjögren’s Syndrome

Background: We wanted to determine (via an immunopharmacological approach) whether the c-Jun NH2 terminal kinase (JNK) cascade is phosphorylated in the submandibular gland by carbachol and cholinergic autoantibodies (IgG) present in the sera of patients with primary Sj?gren’s syndrome (pSS) by interaction and activation of salivary gland muscarinic acetylcholine receptors (mAChRs). Methods: The JNK, PGE2 and NOS assays were measured in rat sub- mandibular gland with pSS IgG and carbachol alone or in the presence of different blocker agents. Results: pSS IgG- activated M3 mAChRs stimulated JNK phosphorylation whereas the activation of M1 mAChRs by carbachol stimulated JNK phosphorylation involving calcium-activated mechanism. The intracellular pathway leading to pSS IgG-induced biological effects on JNK activity involved activation of protein kinase C (PKC), inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) enzymes. Also, activation of COX-2 and COX-1 by pSS IgG and carbachol-induced PGE2 generation were involved. Conclusion: These results may contribute to better understanding the modulatory role of JNK enzymes by cholinergic autoantibodies from pSS patients acting on mAChR in rat submandibular gland.

Functional Vegetable-Based Sausages for Consumption by Children

The dislike food preferences that children registered regarding vegetables and fish compromise the exposure to those iodine sources and may contribute to cause a nutritional deficiency in this micronutrient, causing serious damage of the brain and the central nervous system. Functional foods are becoming well-established in the market, focused on by an adult consumer more interested in healthy eating habits but without the motivation to optimize natural food preparation or to promote a wide variety of food in their diet. This article presents evidence of a new functional product concept, 100% vegetable, fermented soya based and salted by a red algae, that was approved by 900 parents who chose the sensorial features: distinctive aroma (57.9%), soft texture (38.4%), sausage shape (21%) and attractive color (77.9%). Three different varieties were developed: carrot (50% w/w), sugar-beet (50% w/w) and tomato (40% w/w) mini sausages, with tofu (20% w/w) and 0.8 % (w/w) of seaweed, reflecting trends that the younger consumer demands. The sensorial tests performed, after quality control assays, received very satisfactory acceptance by the potential buyer.

Water-Deficit Response Is Not Affected by Glutathione Deficiency in Arabidopsis thaliana pad2-1 Plants

Changes in glutathione and ascorbate content, hydrogen peroxide and abscisic acid were studied in developing leaves in wild-type (Col-0) and glutathione-deficient mutant pad2-1 lines of Arabidopsis thaliana over a time period of 9 days of drought followed by re-watering. Glutathione deficient mutant (pad2-1) presents mechanisms of acclimation to water stress through the reduction of plant biomass and increase in endogenous concentrations of ascorbate and glutathione. These acclimation responses to stress appeared along with the first symptoms of stress and we suggest here that they are regulated by ABA and H2O2. Normally, wild type plants under conditions of stress require an initial response phase in which a decrease in antioxidants is observed before reaching acclimation through the increase in levels of antioxidants. pad2-1 is more sensitive to stress and reacts to it;however, it did not suffer more oxidative stress than Col-0 plants, even though pad2-1 had higher levels of endogenous H2O2 relative to wild-type. In both water stressed Col-0 and pad2-1 plants increases in ABA were observed, however, more sharply in wild-type stressed plants. Low levels of glutathione together with high levels of H2O2 may regulate endogenous ABA concentrations and could be related to the slow growth rates which were observed during the experiment. The results highlighted the double function of glutathione as an antioxidant and signal molecule and also, the different response patterns of wild-type and pad2-1 when faced with drought stress. The results bring new insights to the responses of pad2-1 under conditions of water stress.

Hearing Disability in Colombia Self-Perception and Associated Factors 2002-2008

Introduction: Hearing disability is a condition that affects normal ear function, as much in adulthood as in the first years of life. According to the 2005 Census, 6.3% of the Colombian population has some type of disability, of which 17.4% have hearing limitations, including those with hearing devices. Elucidating the conditions of this population and identifying the factors related to hearing disability will permit the management of strategies from different sectors to mitigate the consequences associated with this limitation. Objective: To estimate the self-perception of hearing disability in Colombia and to determine the factors associated with this limitation. Materials and Methods: The present study is cross-sectional, based on the analysis of secondary information obtained from the Registry for Localization and Characterization of Persons with Disability (RLCPD) during the 2002-2008 period. Socio-demographic and healthcare variables were analyzed. Results: 13.6% (102,648/750,377) of the population reported hearing limitations even with the use of special hearing devices. 43.52% (44,041) of people over 3 years of age could neither read nor write. 29.39% (30,145) of people who reported this limitation are not affiliated to any sort of health insurance system. Factors found to be associated with hearing limitations were: socioeconomic stratus (OR: 1.33;CI 95% 1.25;1.42), illiteracy (OR: 1.44;CI 95% 1.42;1.46) and lack of affiliation to a health insurance system (OR: 1.03;CI 95% 1.01;1.04). Conclusion: People registered with hearing disability live under vulnerable conditions;among them, most/the majority pertained to a segment of the population with low economic resources and had difficulty obtaining/accessing work, education and healthcare services.

Over-Production of P60 Family Proteins, Glycolytic and Stress Response Proteins Characterizes the Autolytic Profile of <i>Listeria monocytogenes</i>

Listeria monocytogenes is a foodborne pathogen capable of surviving under challenging conditions both outside and inside the host. During the transition from exponential to stationary phase it experiences a series of environmental changes that require an appropriate response to maintain cell viability. In this study the autolytic behaviour of a L. monocytogenes strain was investigated by two-dimensional electrophoresis. The study was done at the permissive autolysis temperature, 30℃ and at 20℃, an autolysis non-permissive temperature. An autolytic strain proteome was also compared to a non-autolytic strain at the permissive autolysis temperature. The autolytic strain proteome at 30℃ in comparison to 20℃ evidenced increased synthesis of the P60 autolysin, glycolytic enzymes and proteins related with environmental stress responses. The over-production of P45 autolysin, was observed when the autolytic strain proteome was compared with the non-autolytic strain. The proteomes at the non-permissive temperature and the proteome of the non-autolytic strain were characterized by a diminished synthesis of several stress related proteins. The lack of autolysis seems to be associated to the over-production of proteins linked to fatty acid and amino acid synthesis, transcription regulation and cell morphogenesis as evidenced by the proteome at the non-permissive temperature and the non-autolytic strain. Autolysis proteome evidenced the over-production of P60 autolysins, glycolysis and stress proteins whereas the proteome obtained in conditions of absence of autolysis reveal a completely different group of proteins. Possible targets to activate listerial autolysis were identified.

Sustainability Assessment of Wastewater Systems: An Environmental and Economic Approach

To address current challenges regarding sustainable development of wastewater treatment and provide scientific support in decision procedures towards sustainable solutions, new approaches, frameworks and methodologies about different possible solutions and their potential sustainability implications are needed. One way to facilitate sustainability assessment of wastewater is Life Cycle Assessment (LCA) methodology;however, it fails to map the full scope of wastewater impacts. This paper presents a framework to evaluate the performance of Wastewater Treatment Facilities (WWTF) taking into consideration various factors for insuring environmental sustainability. A total of nine indicators, seven environmental and two economic related to four wastewater treatment facilities, were assessed. Apart from evaluating the sustainability, this study also discussed the link of life cycle approach and social aspects of wastewater. The results show that for the environmental dimension using LCA provides information on different types of environmental activities and different impact categories. LCA can thus be used to quantify and compare the multiple types of impacts caused by one type of use or emission, as well as the various resource uses or emissions that contribute to one type of impacts. For the economic dimension, there is still a need for consistent and robust indicators and methods. The empirical results suggest that the environmental sustainability framework can be used in the first phase of the decision procedure that leads to a strategic choice for sustainable resource recovery from wastewater in developing countries. This motives researchers and decision-makers to consider the whole picture, and not just individual aspects, when considering different futures scenarios.
