Extracellular vesicles(EVs)have been identified as promising biomarkers for the noninvasive diagnosis of various ***,challenges in separating EVs from soluble proteins have resulted in variable EV recovery rates and low ***,we report a high-yield(>90%)and rapid(pping(FLOAT).The FLOAT approach utilizes an acoustofluidic droplet centrifuge to rotate and controllably heat liquid *** adding a thermoresponsive polymer flocculant,nanoparticles as small as 20 nm can be rapidly and selectively concentrated at the center of the *** demonstrate the ability of FLOAT to separate urinary EVs from the highly abundant Tamm-Horsfall protein,addressing a significant obstacle in the development of EV-based liquid *** to its high-yield nature,FLOAT reduces biofluid starting volume requirements by a factor of 100(from 20 mL to 200µL),demonstrating its promising potential in point-of-care diagnostics.

Metabolic flexibility has emerged as a critical determinant of CD8+T-cell antitumor activity,yet the mechanisms driving the metabolic flexibility of T cells have not been *** this study,we investigated the influence of the nuclear cap-binding complex(CBC)adaptor protein ARS2 on mature T *** doing so,we discovered a novel signaling axis that endows activated CD8+T cells with flexibility of glucose ***2 upregulation driven by CD28 signaling reinforced splicing factor recruitment to pre-mRNAs and affected approximately one-third of T-cell activation-induced alternative splicing *** these effects,the CD28-ARS2 axis suppressed the expression of the M1 isoform of pyruvate kinase in favor of pKM2,a key determinant of CD8+T-cell glucose utilization,interferon gamma production,and antitumor effector ***,pKM alternative splicing occurred independently of CD28-driven pI3K pathway activation,revealing a novel means by which costimulation reprograms glucose metabolism in CD8+T cells.

The ability to capture the chemical signatures of biomolecules(i.e.,electron-transfer dynamics)in living cells will provide an entirely new perspective on biology and *** can be accomplished using nanoscale optical antennas that can collect,resonate and focus light from outside the cell and emit molecular ***,we describe biologically inspired nanoscale optical antennas that utilize the unique topologies of plant viruses(and thus,are called gold plant viruses)for molecular fingerprint *** electromagnetic calculations for these gold viruses indicate that capsid morphologies permit high amplification of optical scattering energy compared to a smooth *** experimental measurements of various gold viruses based on four different plant viruses,we observe highly enhanced optical cross-sections and the modulation of the resonance wavelength depending on the viral ***,in label-free molecular imaging,we successfully obtain higher sensitivity(by a factor of up to 10^(6))than can be achieved using similar-sized *** virtue of the inherent functionalities of capsids and the plasmonic characteristics of the gold layer,a gold virus-based antenna will enable cellular targeting,imaging and drug delivery.

Growth and patterning of pt Nanowires on Silicon Substrates

This paper describes a simple procedure for growing dense fi lms of pt nanowires directly on silicon substrates by modifying the surface through chemical or physical *** the former,a self-assembled monolayer of(3-mercaptopropyl)-trimethoxysilane(MpTMS)is applied which can strongly bind pt(0)nuclei to the surface through pt S *** attached,the pt(0)nuclei can act as catalytic sites for the growth of pt nanowires along the〈111〉***,relief features are physically created on the surface in order to generate nucleation and binding sites for pt(0)nuclei,due to the higher free energy associated with a rough ***,pt nanowires have been successfully produced in well-defined patterns by scouring grooves on the silicon surface or by photochemically patterning the MpTMS monolayers with a shadow *** have also measured the electrochemical properties of these immobilized or patterned pt *** results provide an effective route to producing dense fi lms of pt nanowires with high surface areas for various electrochemical applications.

Nucleic acid amplification and quantification via polymerase chain reaction(pCR)is one of the most sensitive and powerful tools for clinical laboratories,precision medicine,personalized medicine,agricultural science,forensic science and environmental *** multiplex pCR,characterized by low power consumption,compact size and simple operation,is ideal for timely diagnosis at the point-of-care(pOC).Although several fast/ultrafast pCR methods have been proposed,the use of a simple and robust pCR thermal cycler remains challenging for pOC ***,we present an ultrafast photonic pCR method using plasmonic photothermal light-to-heat conversion via photon–electron–phonon *** demonstrate an efficient photonic heat converter using a thin gold(Au)film due to its plasmon-assisted high optical absorption(approximately 65%at 450 nm,the peak wavelength of heat source light-emitting diodes(LEDs)).The plasmon-excited Au film is capable of rapidly heating the surrounding solution to over 150℃ within 3 *** this method,ultrafast thermal cycling(30 cycles;heating and cooling rate of 12.7960.93℃ s^(-1) and 6.660.29℃ s^(-1),respectively)from 55℃(temperature of annealing)to 95℃(temperature of denaturation)is accomplished within 5 *** photonic pCR thermal cycles,we demonstrate here successful nucleic acid(λ-DNA)*** simple,robust and low cost approach to ultrafast pCR using an efficient photonic-based heating procedure could be generally integrated into a variety of devices or procedures,including on-chip thermal lysis and heating for isothermal amplifications.

Green hydrogen is anticipated to play a major role in the decarbonization of the mobility *** chemical storage in CO_(2)-neutral synthetic liquid fuels is advantageous in terms of safety and reliability compared to other hydrogen storage developments,and thus represents a complementary building block to developments in electric and hydrogen mobility for the low-carbon transition in the mobility *** development is especially relevant for transport sectors which will have no alternatives to liquid fuels in the foreseeable *** this paper,three alternative technological routes for the chemical storage of hydrogen in CO_(2)-neutral synthetic liquid fuels are identified and comparatively evaluated in terms of feedstock potential,product potential,demand for renewable electricity and associated costs,efficiency as well as expected market *** all three routes exhibited similar levels of overall efficiencies,electricity-based liquid fuels in Germany are currently limited by the high cost and limited supply of renewable *** contrast,liquid fuels generated from biogenic waste have a constant supply of biogenic feedstock and are largely independent from the supply and cost of renewable electricity.

Effect of aging treatment on irradiation-induced segregation of high Mn-Cr steel

To investigate the effect of aging treatment on irradiation-induced segregation of high Mn-Cr steel, specimens for electron-beam irradiation were prepared from the high Mn-Cr austenitic steel which was solution treated at 1 373 K for 1 h and aging treated at 573 K for 1 000 h, respectively. The electron-beam irradiation was performed at 573 K up to doses of 5.4 dpa in a 1 250 kV HVEM and irradiation-induced segregation analyses were carried out by an EDX in a 200 kV FE-TEM. The results show that void formation is not observed in both solution treated and aging treated ones. The amount of Cr segregation at the grain boundary decreases in the aged one; however, that of Mn is not changed in solution treated one.

To detect anaerobic bacteria Clostridium sp . and Bacteroides fragilis in intrahepatic stones by molecular genetic method Methods DNA was extracted from 59 stone samples and subjected to polymerase chain reaction (pCR) amplification targeting the 16S rRNA gene of Clostridium sp . and the glutamine synthetase gene of Bacteroides fragilis Single-strand conformational polymorphism (SSCp) analysis was performed to identify the Clostridium sp Results 16S rRNA gene sequences for Clostridium sp. were identified in 49 stones (83%, 49/59) The two most common groups were detected in 19 (41%) and 17 (37%) of the 46 samples using SSpC analysis, and 25/59 (42%) stones were tested positive for Bacteroides fragilis Conclusions Anaerobes such as Clostridium sp and Bacteroides fragilis present in intrahepatic stones and may play a role in stone formation pCR is a useful technique to detect fastidious pathogens, which are difficult to culture SSCp of pCR products is a rapid method in differentiating bacterial species

Hepatic tight junctions:From viral entry to cancer metastasis

The tight junction (TJ) is a critical cellular component for maintenance of tissue integrity, cellular interactions and cell-cell communications, and physiologically functions as the "great wall" against external agents and the surrounding hostile environment. During the host-pathogen evolution, viruses somehow found the key to unlock the gate for their entry into cells and to exploit and exhaust the host cells. In the liver, an array of TJ molecules is localized along the bile canaliculi forming the blood-biliary barrier, where they play pivotal roles in paracellular permeability, bile secretion, and cell polarity. In pathology, certain hepatic TJ molecules mediate virus entry causing hepatitis infection; deregulation and functional abnormality of the TJ have also been implicated in triggering liver cancer development and metastasis. All these findings shed new insights on the understanding of hepatic TJs in the development of liver disease and provide new clues for potential intervention.
