PHANTOM-BASED EDUCATION PROGRAM FOR ACUPUNCTURE MANIPULATION

Epithelial cell adhesion efficacy of a novel peptide identified by panning on a smooth titanium surface

Epithelial attachment via the basal lamina on the tooth surface provides an important structural defence mechanism against bacterial invasion in combating periodontal disease. However, when considering dental implants, strong epithelial attachment does not exist throughout the titanium-soft tissue interface, making soft tissues more susceptible to peri-implant disease. This study introduced a novel synthetic peptide(A10) to enhance epithelial attachment. A10 was identified from a bacterial peptide display library and synthesized. A10 and protease-activated receptor 4-activating peptide(PAR4-AP, positive control) were immobilized on commercially pure titanium. The peptide-treated titanium showed high epithelial cell migration ability during incubation in platelet-rich plasma. We confirmed the development of dense and expanded bl(stained by Ln5) with pericellular junctions(stained by ZO1) on the peptide-treated titanium surface. In an adhesion assay of epithelial cells on A10-treated titanium, PAR4-AP-treated titanium, bovine root and non-treated titanium, A10-treated titanium and PAR4-AP-treated titanium showed significantly stronger adhesion than non-treated titanium. PAR4-AP-treated titanium showed significantly higher inflammatory cytokine release than non-treated titanium. There was no significant difference in inflammatory cytokine release between A10-treated and non-treated titanium. These results indicated that A10 could induce the adhesion and migration of epithelial cells with low inflammatory cytokine release. This novel peptide has a potentially useful application that could improve clinical outcomes with titanium implants and abutments by reducing or preventing peri-implant disease.

Citrus Peel Ethanol Extract Inhibits the Adipogenesis Caused from High Fat-Induced DIO Model

Background: Flavonoids are multi-functional bioactive compounds that have been used as natural compounds against various diseases. Citrus fruit is an important source for bioactive flavonoids with potential anti-obesity benefits. Methods: To determine the anti-obese effects of citrus peel, a 45% high fat diet-induced obesity (DIO) model using C57bl/6 mice was prepared for 10 weeks and then treated orally for 12 weeks with ethanol extracts of citrus peel (300 mg/kg, CP). CP was compared with normal chow diet (C), high fat diet (HF), and the anti-obesity drug orlistat (30 mg/kg, O) as a positive control. HF caused increases in lipid accumulation, body weight gain, and hepatic toxicity compared with the C group. Results: CP treatment reduced body weight gain and decreased epididymal fat, mesenteric fat, and plasma and hepatic TG levels in a similar manner as O treatment. Besides, CP was comparatively more effective than O at increasing high density lipoprotein cholesterol (HDL-c) while reducing hepatic toxicity, which is caused by HF. Fat accumulation in adipose tissue was decreased by CP treatment because of up-regulation of specific lipolysis enzymes such as HSL and AMPK and down-regulation of adipogenesis related genes such as C/EBPα and ACC. The proinflammatory cytokines, TNF-α and IL-6, which are the key factors for regulation of inflammation, were significantly decreased by CP. Conclusion: CP may be a potential natural source for new anti-obesity candidate because of its inhibitory effect on fat synthesis-related inflammation and its positive effect on lipolysis activation.

Garlic（Allium sativum L.） is an important agricultural product,and has variety of dietary and medicinal *** study investigated the protective properties of garlic essential oil（GEO） and its major organosulfur component（diallyl disulfide,DADS） against the development of nonalcoholic fatty liver disease（NAFLD）.C57bl/6J mice were fed a normal diet or high-fat diet（HFD） with/without GEO（25,50,and 100 mg/kg） or DADS（10 and 20mg/kg） for 12 *** significantly increased glucose,insulin,FFA,triglycerides,total cholesterol,AST and ALT serum levels,hepatic triglycerides and total cholesterol levels,and liver steatosis compared to the control *** addition,moderate levels of oxidative stress and inflammation were *** the statistical analysis of metabolomics data,several key metabolites were found to be influenced by HFD *** and DADS dose-dependently exerted antiobesity and antihyperlipidemic effects by reducing HFD-induced body weight gain,adipose tissue weight and serum biochemical *** of 50,100 mg/kg GEO and 20 mg/kg DADS significantly decreased the release of pro-inflammatory cytokines in liver,accompanied by elevated antioxidant capacity via inhibition of cytochrome P450 2E1 expression during NAFLD *** anti-NAFLD effects of GEO and DADS were mediated through down-regulation of sterol regulatory element binding protein-1c,acetyl-CoA carboxylase,fatty acid synthase,and3-hydroxy-3-methylglutaryl-coenzyme A reductase,as well as stimulation of peroxisome proliferator-activated receptor alpha and carnitine *** results demonstrate that GEO and DADS dose-dependently protected obese mice with long-term HFD-induced NAFLD from lipid accumulation,inflammation,and oxidative damage by ameliorating lipid metabolic disorders and oxidative *** dose of 20 mg/kg DADS was equally effective in preventing NAFLD as 50 mg/kg GEO containing the same amount of DADS,which demonstrates that DADS

目的比较鞘内注射孤啡肽在Swiss albino和C57 bl两种小鼠中所引发的痛觉超敏的特点。方法以随机、双盲、画笔轻触法,观察鞘内注射孤啡肽50 min内两种小鼠痛觉超敏反应。结果在Swiss albino小鼠中,孤啡肽引起的痛觉超敏持续50 min,在5 fmol～50 pmol的剂量范围内,没有观察到量效关系。在C57 bl小鼠中,孤啡肽引起的痛觉超敏持续30 min,在10fmol～1nmol的剂量范围内,呈现出钟形的量效曲线。结论鞘内注射孤啡肽的作用有小鼠种系间的区别。

Hepatocellular carcinoma （HCC） is a lethal liver malignancy worldwide. In this study, we reported that protein phosphatase magnesium-dependent 16 （PPM1D） was highly expressed in the majority of HCC cases （approximately 59%） and significantly associated with high serum a-fetoprotein （AFP） level （P = 0.044）. Kaplan- Meier and Cox regression data indicated that PPM1D overexpression was an independent predictor of HCC- specific overall survival （HR, 2.799; 95% CI, 1.346-5.818, P = 0.006）. Overexpressing PPM1D promoted cell viability and invasion, whereas RNA interference-mediated knockdown of PPM1D inhibited proliferation, invasion, and migration of cultured HCC cells. In addition, PPM1D suppression by small interfering RNA decreased the tumorigenicity of HCC cells in vivo. Overall, results suggest that PPM1D is a potential prognostic marker and therapeutic target for HCC.

The role of the vascularized bone marrow component as a continuous source of donor-derived hematopoietic stem cells that facilitate tolerance induction of vascularized composite allografts is not completely *** this study,vascularized composite tissue allograft transplantation outcomes between recipients receiving either conventional bone marrow transplantation(CBMT)or vascularized bone marrow(VBM)transplantation from Balb/c(H2d)to C57bl/6(H2b)mice were *** high-or low-dose CBMT(1.5×10^(8)or 3×10^(7)bone marrow cells,respectively)was *** addition,recipients were treated with costimulation blockade(1 mg anti-CD154 and 0.5 mg CTLA4Ig on postoperative days 0 and 2,respectively)and short-term rapamycin(3 mg/kg/day for the first posttransplant week and then every other day for another 3 weeks).Similar to high-dose conventional bone marrow transplantation,5/6 animals in the vascularized bone marrow group demonstrated long-term allograft survival(>120 days).In contrast,significantly shorter median survival was noted in the low-dose CBMT group(~64 days).Consistently high chimerism levels were observed in the VBM transplantation ***,low levels of circulating CD4^(+)and CD8^(+)T cells and a higher ratio of Treg to Teff cells were maintained in VBM transplantation and high-dose CBMT recipients(>30 days)but not in low-dose VBM transplant ***-specific hyporesponsiveness was shown in tolerant recipients in *** of the vascularized bone marrow component after secondary donor-specific skin transplantation did not affect either primary allograft or secondary skin graft survival.

Effects of feeding a diet containing Gymnema sylvestre extract: attenuating progression of obesity in C57bl/6J mice

Objective: To investigate the effect of Gymnema sylvestre extract(GS) on initial anti-obesity, liver injury, and glucose homeostasis induced by a high-fat diet(HFD). Methods: The dry powder of GS was extracted with methanol, and gymnemic acid was identified by high performance liquid chromatography as deacyl gymnemic acid. Male C57bl/6J mice that fed on either a normal diet, normal diet containing 1 g/kg GS(CON+GS) HFD, or HFD containing 1.0 g/kg GS(HFD+GS) for 4 weeks were used to test the initial anti-obesity effect of GS. Body weight gain and food intake, and serum levels about lipid and liver injury markers were measured. Histopathology of adipose tissue and liver stained with hematoxylin and eosin(H&E) and oil-red O were analyzed. After 4 weeks of GS extract feeding, intraperitoneal glucose tolerance test(IPGTT) was performed. Results: The methanol extracts of GS exerted significant anti-obesity effects in HFD+GS group. They decreased body weight gain, a lower food and energy efficiency ratio, and showed lower serum levels of total cholesterol(TC), triglyceride(TG), low-density lipoprotein(LDL)-cholesterol, very-low density lipoprotein(VLDL)-cholesterol and leptin compared with the HFD group. The decreases of abdominal as well as epididymal fat weight and adipocyte hypertrophy, lipid droplets in liver, and serum levels of aspartate aminotransferase(AST) and alanine transaminase(ALT) were also observed. The CON+GS group showed an effect of glucose homeostasis compared to the CON group. Conclusions: This study shows that GS provide the possibility as a key role in an initial anti-obesity effects feeding with a HFD.
