Activation of human fibroblast-like synoviocytes by uric acid crystals in rheumatoid arthritis

Hyperuricemia-mediated uric acid crystal formation may cause joint inflammation and provoke the destruction of joints through the activation of inflammasome-mediated innate immune ***,the immunopathological effects and underlying intracellular regulatory mechanisms of uric acid crystal-mediated activation of fibroblast-like synoviocytes(FLs)in rheumatoid arthritis(RA)have not been ***,we investigated the in vitro effects of monosodium urate crystals,alone or in combination with the inflammatory cytokines tumor-necrosis factor(TNF)-a or interleukin(IL)-1b,on the activation of human FLs from RA patients and normal control subjects and the underlying intracellular signaling mechanisms of treatment with these *** urate crystals were able to significantly increase the release of the inflammatory cytokine IL-6,the chemokine CXCL8 and the matrix metalloproteinase(MMP)-1 from both normal and RA-FLs(all P,0.05).Moreover,the additive or synergistic effect on the release of IL-6,CXCL8 and MMP-1 from both normal and RA-FLs was observed following the combined treatment with monosodium urate crystals and TNF-a or *** experiments showed that the release of the measured inflammatory cytokine,chemokine and MMP-1 stimulated by monosodium urate crystals were differentially regulated by the intracellular activation of extracellular signal-regulated kinase and c-Jun N-terminal kinase pathways but not the p38 mitogen-activated protein kinase *** results therefore provide a new insight into the uric acid crystal-activated immunopathological mechanisms mediated by distinct intracellular signal transduction pathways leading to joint inflammation in RA.

Prospective study of Centurion~？ versus Infiniti~？ phacoemulsification systems： surgical and visual outcomes

AIM： To evaluate surgical outcomes（sOs） and visual outcomes（VOs） in cataract surgery comparing the Centurion？ phacoemulsification system（CPs） with the Infiniti？ phacoemulsification system（IPs）.METHODs： Prospective, consecutive study in a singlesite private practice. Totally 412 patients undergoing cataract surgery with either the CPs using the 30-degree balanced？ tip（n=207） or the IPs using the 30-degree kelman？ tip（n=205）. Intraoperative and postoperative outcomes were documented prospectively up to one month follow-up. Nuclear sclerosis（Ns） grade, cumulated dissipated energy（CDE）, preoperative corrected distance visual acuity（CDVA）, and CDVA at one month were recorded. REsULTs： CDE was 13.50% less in the whole CPs compared with the whole IPs subcohort. In eyes with Ns grade III or greater, CDE was 28.87% less with CPs（n=70） compared with IPs（n=44）（P=0.010）. surgical complications were not statistically different between the two subcohorts（P=0.083）, but in the one case of vitreous loss using the CPs, CDVA of 6/4 was achieved at one month. The mean CDVAs（VOs） at one month for Ns grade III and above cataracts were-0.17 log MAR（6/4.5） in the CPs and-0.15 log MAR（6/4.5） in the IPs subcohort respectively（P=0.033）.CONCLUsION： CDE is 28.87% less, and VOs are significantly improved, in denser cataracts in the CPs compared with the IPs. The authors recommend the CPs for cases with denser nuclei.

sCHEDULING TWO GROUPs OF JOBs WITH INCOMPLETE INFORMATION

In real world situations, most scheduling problems occur neither as complete off-line nor as complete on-line models. Most likely, a problem arises as an on-line model with some partial information. In this article, we consider such a model. We study the scheduling problem P(n1,n2), where two groups of jobs are to be scheduled. The first job group is available beforehand. As soon as all jobs in the first group are assigned, the second job group appears. The objective is to minimize the longest job completion time (mak.s.an). We show a lower bound of 3/2 even for very special cases. Best possible algorithms are presented for a number of cases. Furthermore, a heuristic is proposed for the general case. The main contribution of this paper is to discuss the impact of the quantity of available information in designing an on-line algorithm. It is interesting to note that the absence of even a little bit information may significantly affect the performance of an algorithm.

The structural and surface properties of high-quality epitaxial cubic MgxZn1-xO films deposited by pulsed laser deposition (PLD) were studied by X-ray diffracti on and atomic force microscopy respectively. For films of about 500nm thick, sca ns over a 30μm × 30μm area revealed a surface roughness Ra of about 100nm. Th is relatively large surface roughness is primarily attributed to the particulate and outgrowth during the PLD process. A good epitaxial growth on LaAlO3 (LAO ) (100) substrates, however has been obtained for composition x = 0.9, 0.7 and 0 .5 with the heteroepitaxial relationship of (100)■∥(100)LAO (out-of-plane) and (011)■∥(010)LAO (in-plane). These structural qualities suggest that cubic Mgx Zn1-xO alloys films have good potential in a variety of optoelectronic device ap plications.

Improved prediction of clay soil expansion using machine learning algorithms and meta-heuristic dichotomous ensemble classifiers

soil swelling-related disaster is considered as one of the most devastating geo-hazards in modern ***,proper determination of a soil’s ability to expand is very vital for achieving a secure and safe ground for ***,this study has provided a novel and intelligent approach that enables an improved estimation of swelling by using kernelised machines(Bayesian linear regression(BLR)&bayes point machine(BPM)support vector machine(sVM)and deep-support vector machine(D-sVM));(multiple linear regressor(REG),logistic regressor(LR)and artificial neural network(ANN)),tree-based algorithms such as decision forest(RDF)&boosted trees(BDT).Also,and for the first time,meta-heuristic classifiers incorporating the techniques of voting(VE)and stacking(sE)were *** independent scenarios of explanatory features’combination that influence soil behaviour in swelling were *** results indicated BLR as possessing the highest amount of deviation from the predictor variable(the actual swell-strain).REG and BLR performed slightly better than ANN while the meta-heuristic learners(VE and sE)produced the best overall performance(greatest R2 value of 0.94 and RMsE of 0.06%exhibited by VE).CEC,plasticity index and moisture content were the features considered to have the highest level of *** binary classifiers(sVM,D-sVM and BPM)gave better accuracy(average accuracy and recall rate of 0.93 and 0.60)compared to ANN,LR and ***-driven diagnostic test indicated that the meta-heuristic models’best performance occurred when ML training was conducted using k-fold validation ***,it is recommended that the concepts developed herein be deployed during the preliminary phases of a geotechnical or geological site characterisation by using the best performing meta-heuristic models via their background coding resource.

Nanoscale plasma-activated aerosol generation for in situ surface pathogen disinfection

Plasma treatment constitutes an efficient method for chemical-free disinfection.A spray-based system for dispensing plasma-activated aerosols onto surfaces would facilitate disinfection of complex and/or hidden surfaces inaccessible to direct line-of-sight(for example,UV)*** complexity and size of current plasma generators(for example,plasma jet and cometary plasma systems)-which prohibit portable operation,together with the short plasma lifetimes,necessitate a miniaturized in situ technique in which a source can be simultaneously activated and administered on-demand onto ***,we demonstrate this possibility by combining two nanoscale technologies for plasma and aerosol generation into an integrated device that is sufficiently small and *** is generated on a carpet of zinc oxide nanorods comprising a nanoneedle ensemble,which when raised to a high electric potential,constitutes a massive point charge array with near-singular electric fields to effect atmospheric *** plasma is then used to activate water transported through an underlying capillary wick,that is subsequently aerosolized under MHz-order surface acoustic *** show that the system,besides being amenable to miniaturization and hence integration into a chipscale device,leads to a considerable improvement in plasma-activation over its macroscale cometary discharge predecessor,with up to 20%and 127%higher hydrogen peroxide and nitrite ion concentrations that are respectively generated in the plasma-activated ***,in turn,leads to a 67%reduction in the disinfection time to achieve 95%bacterial load reduction,therefore demonstrating the potential of the technology as an efficient portable platform for on-demand field-use surface disinfection.

The pollen tube clear zone: Clues to the mechanism of polarized growth

Pollen tubes usually exhibit a prominent region at their apex called the ＂clear zone＂ because it lacks light refracting amyloplasts. A robust, long clear zone often associates with fast growing pollen tubes, and thus serves as an indicator of pollen tube health. Nevertheless we do not understand how it arises or how it is maintained. Here we review the structure of the clear zone, and attempt to explain the factors that contribute to its formation. While amyloplasts and vacuolar elements are excluded from the clear zone, virtually all other organelles are present including secretory vesicles, mitochondria, Golgi dictyosomes, and the endoplas-mic reticulum （ER）. secretory vesicles aggregate into an inverted cone appressed against the apical plasma membrane. ER elements move nearly to the extreme apex, whereas mitochondria and Golgi dictyosomes move less far forward. The cortical actin fringe assumes a central position in the control of clear zone formation and maintenance, given its role in generating cytoplasmic streaming. Other likely factors include the tip-focused calcium gradient, the apical pH gradient, the influx of water, and a host of signaling factors （small G-proteins）. We think that the clear zone is an emergent property that depends on the interaction of several factors crucial for polarized growth.

Calcium at the Cell Wall-Cytoplast Interface

Attention is given to the role of Ca^2＋ at the interface between the cell wall and the cytoplast, especially as seen in pollen tubes. While the cytoplasm directs the synthesis and deposition of the wall, it is less well appreciated that the wall exerts considerable self control and influences activities of the cytoplasm. Ca^2＋ participates as a crucial factor in this two way communication. In the cytoplasm, a [Ca^2＋] above 0.1 μM, regulates myriad processes, including secretion of cell wall components. In the cell wall Ca^2＋ , at 10μM to 10 mM, binds negative charges on pectins and imparts structural rigidity to the wall. The plasma membrane occupies a pivotal position between these two compartments, where selective channels regulate influx of Ca^2＋ , and specific carriers pump the ion back into the wall. In addition we draw attention to different factors, which either respond to the wall or are present in the wall, and usually generate elevated[Ca^2＋ ] in the cytoplasm. These factors include：（i） stretch activated channels; （ii） calmodulin; （iii） annexins; （iv） wall associated kinases; （v） oligogalacturonides; and （vi） extracellular adenosine 5＇-triphosphate. Together they provide evidence for a rich and multifaceted system of communication between the cytoplast and cell wall, with Ca^2＋ as a carrier of information.
