Skeletal stem.and progenitor cells(SSPCs) perform.bone m.intenance and repair. With age, they produce fewer osteoblasts and m.re adipocytes leading to a loss of skeletal integrity. The m.lecular m.chanism. that underlie this detrim.ntal transform.tion are largely unknown. Single-cell RNA sequencing revealed that Notch signaling becom.s elevated in SSPCs during aging. To exam.ne the role of increased Notch activity, we deleted Nicastrin, an essential Notch pathway com.onent, in SSPCs in vivo. m.ddle-aged conditional knockout m.ce displayed elevated SSPC osteo-lineage gene expression, increased trabecular bone m.ss, reduced bone m.rrow adiposity, and enhanced bone repair. Thus, Notch regulates SSPC cell fate decisions, and m.derating Notch signaling am.liorates the skeletal aging phenotype, increasing bone m.ss even beyond that of young m.ce. Finally, we identified the transcription factor Ebf3 as a downstream.m.diator of Notch signaling in SSPCs that is dysregulated with aging, highlighting it as a prom.sing therapeutic target to rejuvenate the aged skeleton.

A Novel Approach to m.asure the Total Antioxidant Power of Wines through Near Infrared Spectroscopy and Its Relevance in Hum.n Nutrition

Wine is the product of the alcoholic ferm.ntation of the grape (Vitis vinifera). As such, it is a hydroalcoholic solution with a variable content of nutritional and functional (polyphenols) m.lecules, the latter involved in its antioxidant potential. The organoleptic variables of wine, together with its potential positive effects on health of a light-m.derate intake, have always been topics of great interest within the cultures. In the label of wine, alcohol is the only declared variable. On the other hand, there is no inform.tion about the content of “positive” m.lecules, such as those associated with the antioxidant power. This value could be very im.ortant to classify the wines, helping oenologist and nutritionist in qualifying them.as a com.onent of m.diterranean diet. m.reover, one of the m.st critical aspects in evaluating the role of wine in hum.n health through epidem.ological prospective studies is the quality of the products used and their antioxidant potential. This research aim.d to optim.ze and validate an analytical approach based on a portable device (SCIO?), using NIRS (near-infrared reflectance spectroscopy). It allows the m.asure of TAP (total antioxidant power) of wine through the glass. Research findings are prom.sing. To the best of our knowledge, this is the first tim. that an easy-to-use and cheap hand-held scanner is validated to m.asure the TAP of a beverage.

种子休眠妨碍了向日葵种的研究及其在改良栽培向日葵（Helianthus annuus L.）的育种程序中的应用。测验了数种种子处理方法,测定其在打破许多野生向日葵种的强烈的种子休眠系统上的效率。第一个处理（称为完整处理）包括种子的机械多次划破、于100毫克/升赤霉酸液中浸泡1小时及去除种壳。其他处理是该程序的简化和三个应用赤霉酸和2-氯乙基-次磷酸的完整种子处理。结果表明:去除种壳和种皮的处理最为有效,使许多种（包括极难萌发的一年生沙漠种）的萌发率提高到90%以上,将多次划破的种子置于100毫克/升赤霉酸液中浸泡1小时能促进某些种的萌发。将完整种子置于100毫克/升赤霉酸液中浸泡1小时或于500毫克/升2-氯乙基-次磷酸液中浸泡1小时均不能促进萌发。完整种子置于100毫克/升或100毫克/升赤霉酸液中培养10天可促进萌发,但形成非正常苗,虽然完整处理复杂,但其萌发率高,且材料间变异最小,因而推荐完整处理作为最佳的方法。许多作物育种家都关注把野生向日葵种作为改良栽培向日葵（Helianthus annuus L.）的基因资源,但强烈的种子休眠系统妨碍了这些物种的利用。在所有的野生种中均存在不同强度的种子休眠系统,但在一年生沙漠种*** Heiser、*** Blake和***亚种fephrods（***）Heiser（Heiser等,1969）尤为强烈。这些旱生种作为耐旱基因资源具有特别重要的价值。据恶,某些多年生种的休眠也非常强烈（Heiser,1916）。为确保野生向日葵种子萌发良好,已应用了数种方法。Heiser等（1969）报道一般最有效的方法是将种子播种于盆内,每移出室外3周至1个月,使其置于不同温度（包括冻融）之下。这种方法可促进萌发,但极少超过50%,且对一年生旱生种无效。对旱生种也应用了其他方法,包括湿润和干燥交替、多次换水冲洗和浸泡种子,但萌发率极少达到10%。据研究（Kum.r和Sastry,1974、1975,zimmerman,1977,Fick,1978,Harada,1982）,乙烯萌发混合物,2-氯乙基-次磷酸可促进刚收获的栽培向日葵种子的萌发,赤霉酸（GA3）和苯甲基腺嘌呤也有同样的效果（Kum.r和Sastry 1974,1975）,去除种壳也可促进萌发（Kum.r和Sastry 1974,1975,Harada,1982）。本研究的目的是比较野生向日葵种种子的几种萌发方法,从而确定一种简单又有效的程序。

Breast cancer is the num.er one cause of can- cer deaths am.ng Hispanic wom.n in the United States, and in m.xico, it recently becam. the prim.ry cause of cancer deaths. This m.lign- nancy represents a poorly understood and un- derstudied disease in Hispanic wom.n. The ELLA Binational Breast Cancer Study was es- tablished in 2006 as a m.lti-center study to as- sess patterns of breast tum.r m.rkers, clinical characteristics, and their risk factors in wom.n of m.xican descent. We describe the design and im.lem.ntation of the ELLA Study and provide a risk factor com.arison between wom.n in the U.S. and those in m.xico based on a sam.le of 765 patients (364 in the U.S. and 401 in m.xico). Com.ared to wom.n in m.xico, U.S. wom.n had significantly (p m.an live births) and breastfeeding rates (57.5% vs. 80.5%), higher use of oral contraceptives (60.7% vs. 50.1%) and horm.ne replacem.nt therapy (23.3% vs. 7.6%), and higher fam.ly history of breast cancer (15.7% vs. 9.0%). Re- sults show that differences in breast cancer risk factor patterns exist between m.xico and U.S. wom.n. We provide lessons learned from.the conduct of our study. Binational studies are an im.ortant step in understanding disease pat- terns and etiology for wom.n in both countries.
