Pharmacological inhibition of cannabinoid receptor 1 stimulates gastric release of nesfatin-1 via the mTOR pathway

AIM To determine whether Nucb2/nesfatin1 production is regulated by the cannabinoid system through the intracellular m TOR pathway in the *** Sprague Dawley rats were treated with vehicle, rimonabant, rapamycin or rapamycin+rimonabant. Gastric tissue obtained from the animals was used for biochemical assays: Nucb2 m RNA measurement by real time PCR, gastric Nucb2/nesfatin protein content by western blot, and gastric explants to obtain gastric secretomes. Nucb2/nesfatin levels were measured in gastric secretomes and plasma using enzyme-linked immunosorbent assay. RESULTS The inhibition of cannabinoid receptor 1(CB1) by the peripheral injection of an inverse agonist, namely rimonabant, decreases food intake and increases the gastric secretion and circulating levels of Nucb2/nesfatin-1. In addition, rimonabant treatment activates m TOR pathway in the stomach as showed by the increase in pm TOR/m TOR expression in gastric tissue obtained from rimonabant treated animals. These effects were confirmed by the use of a CB1 antagonist, AM281. When the intracellular pathway m TOR/S6 k was inactivated by chronic treatment with rapamycin, rimonabant treatment was no longer able to stimulate the gastric secretion of Nucb2/*** The peripheral cannabinoid system regulates food intake through a mechanism that implies gastric production and release of Nucb2/Nesfatin-1, which is mediated by the m TOR/S6 k pathway.

Silicon(Si)photonics has recently emerged as a key enabling technology in many application fields thanks to the mature Si process technology,the large silicon wafer size,and promising Si optical *** monolithic integration by direct epitaxy of III-V lasers and Si photonic devices on the same Si substrate has been considered for decades as the main obstacle to the realization of dense photonics *** considerable progress in the last decade,only discrete III-V lasers grown on bare Si wafers have been reported,whatever the wavelength and laser *** we demonstrate the first semiconductor laser grown on a patterned Si photonics platform with light coupled into a waveguide.A mid-IR GaSb-based diode laser was directly grown on a pre-patterned Si photonics wafer equipped with SiN waveguides clad by SiO_(2).Growth and device fabrication challenges,arising from the template architecture,were overcome to demonstrate more than 10 mw outpower of emitted light in continuous wave operation at room *** addition,around 10%of the light was coupled into the SiN waveguides,in good agreement with theoretical calculations for this butt-coupling *** work lft an important building block and it paves the way for future low-cost,large-scale,fully integrated photonic chips.

Background In beef cattle, more than 50% of the energy input to produce a unit of beef is consumed by the female that produced the calf. Development of genomic tools to identify females with high genetic merit for reproductive function could increase the profitability and sustainability of beef *** Genome-wide association studies(GWAS) were performed using a single-step genomic best linear unbiased prediction approach on pregnancy outcome traits from a population of Angus–Brahman crossbred heifers. furthermore, a validation GWAS was performed using data from another farm. Heifers were genotyped with the Bovine GGP f250 array that contains 221,077 SNPs. In the discovery population, heifers were bred in winter breeding seasons involving a single round of timed artificial insemination(AI) followed by natural mating for 3 months. Two phenotypes were analyzed: pregnancy outcome to first-service AI(PAI;n = 1,481) and pregnancy status at the end of the breeding season(PEBS;n = 1,725). The heritability was estimated as 0.149 and 0.122 for PAI and PEBS, respectively. In the PAI model, one quantitative trait locus(QTL), located between 52.3 and 52.5 Mb on BTA7, explained about 3% of the genetic variation, in a region containing a cluster of γ-protocadherin genes and SLC25A2. Other QTLs explaining between 0.5% and 1% of the genetic variation were found on BTA12 and 25. In the PEBS model, a large QTL on BTA7 was synonymous with the QTL for PAI, with minor QTLs located on BTA5, 9, 10, 11, 19, and 20. The validation population for pregnancy status at the end of the breeding season were Angus–Brahman crossbred heifers bred by natural mating. The assessment of detrital zircon age records is a key method in basin analysis,but it is prone to several biases that may compromise accurate sedimentary provenance *** to ultrahigh temperature(HT-UHT)metamorphism(especially if T>850℃)is herein presented as a natural cause of bias in provenance studies based on U-Pb detrital zircon ages,since zircon from rocks submitted to these extreme and often prolonged conditions frequently yield protracted,apparently concordant,geochronological *** age spreading can result from disturbance of the primary U-Pb zircon system,likewise from(re)crystallization processes during multiple and/or prolonged metamorphic *** this contribution,available geochronological data on Archean,Neoproterozoic and Palaeozoic HT-UHT metamorphic rocks,acquired by different techniques(SIMS and LA-ICP-MS)and showing distinct compositions,are reassessed to demonstrate HT-UHT metamorphism may result in modes and age distributions of unclear geological *** a consequence,it may induce misinterpretations on UPb detrital zircon provenance analyses,particularly in sedimentary rocks metamorphosed under such extreme temperature *** evaluate the presence of HT-UHT metamorphism-related bias in the detrital zircon record,we suggest a workflow for data acquisition and interpretation,combining a multi-proxy approach with:(i)in situ U-Pb dating coupled with Hf analyses to retrieve the isotopic composition of the sources,and(ii)the integration of a petrochronological investigation to typify fingerprints of the HT-UHT metamorphic *** proposed workflow is validated in the investigation of one theoretical and one natural example allowing a better characterization of the sedimentary sources,maximum depositional ages,and the tectonic setting of the *** workflow allows to the appraisal of biases imposed by HT-UHT metamorphism and resulting disturbances in the U-Pb detrital zircon record,particularly for sedimentary

Lichens are symbiotic organisms that comprise a fungus and a photosynthetic partner wich are recognized as a good indicator of climate ***,our understanding of how aridity affects the diversity of saxicolous lichens in drylands is still *** evaluate the relationship between saxicolous lichen diversity and aridity in a central México dryland,a geographical transect was established of 100 km to build an aridity gradient in the semiarid zone of the State of Querétaro,Mexico,comprising ten sampling sites with a 10 km *** richness,abundance and diversity of soil lichen species were recorded using two sampling methods:the quadrat-intercept and the line-intercept method,to compare their performance in assessing soil lichen diversity in *** number of species and Shannon diversity of saxicolous lichens were higher at intermediate values of the aridity index(AI=0.10-0.34).Quadrat intercept and point intercept methods gave quite similar results,which means that the selected method does not influence the results in a significant *** study confirms the role of saxicolous lichens as climate change indicators and reveals the importance of the sampling method selection in the evaluation of different parameters of soil lichen diversity in drylands.

Unique biogenesis and kinetics of hornwort Rubiscos revealed by synthetic biology systems

Hornworts are the only land plants that employ a pyrenoid to optimize Rubisco’s CO_(2) fixation,yet hornwort Rubisco remains poorly *** we assembled the hornwort Anthoceros agrestis Rubisco(AaRubisco)using the Arabidopsis thaliana SynBio expression system and observed the formation of stalled intermediates,prompting us to develop a new SynBio system with *** cognate *** successfully assembled AaRubisco and Rubisco from three other hornwort *** *** Rubisco,AaRubisco assembly is not dependent on RbcX or *** characterization reveals that hornwort Rubiscos exhibit a range of catalytic rates(3–10 s−1),but with similar affinity(∼30μM)and specificity(∼70)for CO_(2).These results suggest that hornwort Rubiscos do not comply with the long-held canonical catalytic trade-off observed in other land plants,providing experimental support that Rubisco kinetics may be phylogenetically ***,we observed a 50%increase in AaRubisco catalytic rates when RbcX was removed from our SynBio system,without any reduction in *** biology,biochemistry,and proteomic analysis suggest that subtle differences in Rubisco large-subunit interactions,when RbcX is absent during biogenesis,increases the accessibility of active sites and catalytic turnover ***,this study uncovered a previously unknown Rubisco kinetic parameter space and provides a SynBio chassis to expand the survey of other Rubisco *** discoveries will contribute to developing new approaches for engineering Rubisco with superior kinetics.

IBD5 polymorphisms in inflammatory bowel disease: Association with response to infliximab

AIM: Inflammatory bowel diseases (IBD) are multifactorial pathologies of unknown etiology. One susceptibility locus, IBD5, has been mapped to chromosome 5q31. We analyzed our Spanish cohorts of Crohn's disease (CD) and ulcerative colitis (DC) patients to determine whether this locus is associated with IBD, and to ascertain the main clinical phenotype influenced by this risk factor. The kind of interaction, either genetic heterogeneity or epistasis, between this IBD5 susceptibility region and the NOD2/CARD15 gene mutations was studied as well. finally, we assessed whether this locus can predict response to infliximab therapy. METHODS: A case control study was performed with 274 CD and 211 UC patients recruited from a single center and 511 healthy ethnically matched controls. Two polymorphisms were genotyped in the IBD5 locus and three in the CARD15/NOD2 gene. RESULTS: Our results evidence association only with CD especially with the fistulizing phenotype and in the absence of NOD2/CARD15 variants (mutant allele frequency in patients vs controls: OR = 2.03, 95% CI = 1.35-3.06, Pfrequency of the IBD5 homozygous mutant genotype significantly increased in CD patients lacking response to infliximab (RR = 3.88, 95% CI = 1.18-12.0, Pfound within the worse prognosis group. CONCLUSION: The IBD5 variants may enhance an individual carrier's risk for CD, mainly in the absence of the NOD2/CARD15 mutations and in fistulizing patients. The data presented suggest the potential role of the 5q31 polymorphisms as markers of response to infliximab.

Pro-Inflammatory Substances and Cognition in the Dallas Heart Study

Cognitive decline in late adulthood might be partially mediated by subclinical generalized vascular disease. If so, atherogenic factors such as pro-inflammatory cytokines might be mid-life targets for prevention or treatment. Dallas Heart Study subjects (n = 997;mean age = 42.94 ± 10.2 yrs) underwent blood assays of pro-inflammatory biomarkers associated with atherosclerosis and 8 years later completed a cognitive outcome measure, the Montreal Cognitive Assessment (MoCA). Markers included C-reactive protein (CRP), Interleukin-18 (IL-18), Lipoprotein-associated phospholipase (LP-PLA2), and Monocyte Chemoattractant Protein (MCP-1), with Apolipoprotein E4 (ApoE4) as a potential modifier. We found weak evidence for LP-PLA2 and CRP as predictors of cognitive scores. No relationship was found between elevated MCP-1, IL-18 and cognition. Presence of the ApoE4 allele did not impact the relationship between biomarkers and cognitive function. Levels of atherogenesis-related pro-inflammatory blood biomarkers did not predict cognitive function in middle-aged adults after an interval of 8 years.
