This study presents the simulation of a MFC with Pseudomonas aeruginosa based on a metabolic flux analysis (MFA) which arises as a linear programming model that served as input for the fuel cell model. The linear model was implemented in Xpress MP? and the coupling model in Comsol Multiphysics?. The in silico model predicted maximum potentials of 0.135 V which were consistent with those obtained in the experimental cell. Afterwards, an optimization platform based on multiobjective optimization approach was implemented aimed to determine genes that increase the average cell power. cysA, cysP and rpoS mutants which were predicted to increase the power of the cell, were experimentally tested with an air cathode fuel cell finding an augment up to 35-fold in the average power density for the rpoS mutant. Power densities were obtained through experimentation in the range of [0.13 - 5] mW/m2. Electron shuttle rise was qualitative corroborated trough cyclic voltammetry tests, which allowed to visualize the augment of the peaks for rpoS mutation.

Human action recognition from skeletal data is an important and active area of research in which the state of the art has not yet achieved near-perfect accuracy on many well- known datasets. In this paper, we introduce the Distribution of Action Movements Descriptor, a novel action descriptor based on the distribution of the directions of the motions of the joints between frames, over the set of all possible mo- tions in the dataset. The descriptor is computed as a normal- ized histogram over a set of representative directions of the joints, which are in turn obtained via clustering. While the descriptor is global in the sense that it represents the overall distribution of movement directions of an action, it is able to partially retain its temporal structure by applying a window- ing scheme. Influence of main dietary chemical constituents on the in vitro gas and methane production in diets for dairy cows

Background： Modification of chemical composition of diets fed to dairy cows might be a good strategy to reduce methane（CH4） production in the rumen. Notable reductions of CH4 production compared to conventional highroughages rations were more frequently observed for very concentrated diets or when fat supplements were used. In these cases, the reduction in the gas emission was mainly a consequence of an overall impairment of rumen function with a reduction of fiber digestibility. These strategies do not always comply with feeding standards used in intensive dairy farms and they are usually not applied owing to the risks of negative health and economic ***, the present study evaluated the effects of seven commercial diets with contents of neutral detergent fiber（NDF）,protein and lipids ranging 325 to 435 g/kg DM, 115 to 194 g/kg DM, and 26 to 61 g/kg DM, respectively, on in vitro degradability, gas（GP）, and CH4 ***： In this experiment, changes in the dietary content of NDF, crude protein（CP） and lipids were always obtained at the expense or in favor of starch. A decreased of the dietary NDF content increased NDF（NDFd） and true DM（TDMd） degradability, and increased CH4 production per g of incubated DM（P 〈 0.001）, but not that per g of TDMd. An increase of the dietary CP level did not change in vitro NDFd and TDMd, decreased GP per g of incubated DM（P 〈 0.001）, but CH4 production per g of TDMd was not affected. Methodological factors affecting gas and methane production during in vitro rumen fermentation evaluated by meta-analysis approach

Effects of some methodological factors on in vitro measures of gas production（GP, mL/g DM）, CH4production（mL/g DM） and proportion（% CH4 on total GP） were investigated by meta-analysis. These factors were considered：pressure in the GP equipment（0 = constant; 1 = increasing）, incubation time（0 = 24; 1 = ≥ 48 h）, time of rumen fluid collection（0 = before feeding; 1 = after feeding of donor animals）, donor species of rumen fluid（0 = sheep; 1 =bovine）, presence of N in the buffer solution（0 = presence; 1 = absence）, and ratio between amount of buffered rumen fluid and feed sample（BRF/FS; 0 = ≤ 130 mL/g DM; 1 = 130–140 mL/g DM; 2 = ≥ 140 mL/g DM）. The NDF content of feed sample incubated（NDF） was considered as a continuous variable. From an initial database of 105 papers, 58 were discarded because one of the above-mentioned factors was not stated. After discarding 17 papers,the final dataset comprised 30 papers（339 observations）. A preliminary mixed model analysis was carried out on experimental data considering the study as random factor. Variables adjusted for study effect were analyzed using a backward stepwise analysis including the above-mentioned variables. The analysis showed that the extension of incubation time and reduction of NDF increased GP and CH4 values. Values of GP and CH4 also increased when rumen fluid was collected after feeding compared to before feeding（＋26.4 and ＋9.0 mL/g DM, for GP and CH4）,from bovine compared to sheep（＋32.8 and ＋5.2 mL/g DM, for GP and CH4）, and when the buffer solution did not contain N（＋24.7 and ＋6.7 mL/g DM for GP and CH4）. The increase of BRF/FS ratio enhanced GP and CH4production（＋7.7 and ＋3.3 mL/g DM per each class of increase, respectively）. In vitro techniques for measuring GP and CH4 production are mostly used as screening methods, thus a full standardization of such techniques is not feasible. Most conventional aerosol neutralizers are based on radioactive sources, which are controlled by strict regulations restricting their handling, transport, and storage. The TSI 3087 soft X-ray （SXR） neutralizer circumvents these legal restrictions. The aim of the present work is to compare the performance of a standalone SXR aerosol neutralizer with that of conventional radioactive aerosol neutralizers based on 85Kr （TSI 3077） and 241Am （Grimm 5522） by performing field tests in a real environmental scenario. The results obtained when the SXR neutralizer was connected to a mobility particle sizer spectrometer （MPS）, different from the device suggested by the manufacturer, were comparable with those obtained with the use of radioactive aerosol neutralizers. In changing the neutralizer, the particle number concentrations, measured with the MPS connected to the SXR neutralizer, almost remained within the 10% uncertainty bounds for the particle size interval 10-300 nm, when diffusion losses inside the SXR tube were considered. Based on our comparisons, the SXR neutralizer can be regarded as a standalone instrument that could solve the problems associated with legal restrictions on radioactive neutralizers and fulfil the need for a portable instrument for different field test purposes.

Hepatitis B virus(HBV)infection is a serious global public health *** infection may be transmitted through sexual intercourse,parenteral contact or from an infected mother to the baby at birth and,if contracted early in life,may lead to chronic liver disease,including cirrhosis and hepatocellular *** the basis of the HBV carrier rate,the world can be divided in 3 regions of high,medium and low *** major concern is about high endemicity countries,where the most common route of infection remains vertical transmission from mother to *** of all pregnant women and passive immunization with human hepatitis B immunoglobulin are not affordable for many developing *** infection rate can be reduced by modifying behavior,improving individual education,testing all blood donations,assuring asepsis in clinical practice and screening all pregnant ***,availability of a safe and efficacious vaccine and adoption of appropriate immunization strategies are the most effective means to prevent HBV infection and its *** unsolved problem for poorest countries,where the number of people currently infected is high,is the cost of the vaccine.A future challenge is to overcome the social and economic hurdles of maintaining and improving a prevention policy worldwide to reduce the global burden of the disease.

Outlining key inflammation-associated parameters during early phase of an experimental gram-negative sepsis model in rhesus macaques(Macaca mulatta)

The aim of this study was to identify inflammation-associated markers during the early phase of sepsis in rhesus macaque. Four rhesus macaques were given an intravenous dose of 1010 CFU/kg of E. coli. Blood samples were collected before, or 30 minutes, 2, 4, 6 and 8 hours after E. coli infusion. Physiological parameters, bacteremia, endotoxemia, C-reactive protein(CRP), procalcitonin(PCT), and plasma cytokines/chemokines were determined for each animal. Bacteremia was present in all animals from 30 minutes to 3 hours after E. coli infusion whereas endotoxin was detected during the full-time course. CRP and PCT levels remained at detectable levels during the whole experimental window suggesting an ongoing inflammatory process. Signature cytokines and chemokines such as TNF-α, MIP-1α, and MIP-1β peaked about 2 hours after E. coli infusion and decreased thereafter. Plasma IL-6, IL-12 p40, IFN-γ, and IL-1 Ra, as well as I-TAC, MIG, IP-10 and MCP-1, remained at detectable levels after 4 hours of E. coli infusion. This nonhuman primate model could be useful for the assessment of new therapeutics aiming to suppress key inflammatory markers throughout sepsis early phases.

Airborne Nanoparticles Filtration by Means of Cellulose Nanofibril Based Materials

Nanoparticles in air are of particular concern for public health and employee exposure in work-places. Therefore, it is very important to prepare effective filters for their removal. In this work filters were prepared from nanocellulose, i.e. cellulose nanofibrils (CNF). CNF was produced using two methods giving two different qualities of CNF. One quality had negative charges on the fibril surfaces while the other was neutral, and had in addition thinner fibrils compared to the other qualities. Filter samples were produced from water dispersions of CNF, by removal of the water by freeze drying. The performance of the CNF based filters was assessed and compared with filters based on synthetic polymer fibres. The ability to collect NaCl particles with a broad size distribution, ranging from nanometer to micrometer scale, was determined. CNF filters showed quality values comparable with the synthetic polymer based filters. Filters based on both the two CNF qualities had very good filtration efficiency for a given pressure drop across the filter.

Proteins from <i>Punctularia atropurpurascens</i>with Biotechnological Applications

Basidiomycetes are able to biodegrade waste and xenobiotic molecules through the production of extracellular enzymes. For example, white-rot fungi produce lignin-degrading enzymes which are capable of efficiently decolorizing dye solutions. Many mushrooms also produce lectins, a group of proteins which bind specifically to the carbohydrates in glycoconjugates. Several fungal lectins target their specificities towards oligosaccharides present in mammalian glycoproteins, thus constituting excellent ligands for the preparation of affinity adsorbents useful in isolation and characterization of these glycoproteins. In this study we isolated and characterized two different proteins, a lectin and a laccase, present in extracts from Punctularia atropurpurascens. The lectin isolated from the mycelium extract, was immobilized on activated-Sepharose and used to evaluate the interaction with three glycoproteins. The adsorbent was able to efficiently adsorb and elute bovine lactoferrin, constituting a promising tool for the purification of this glycoprotein. In vitro experiments revealed that the lectin also exhibited antimicrobial activity against Aspergillus niger. Laccase activity was detected in the extracellular extract from P. atropurpurascens. This enzyme, in both soluble and immobilized forms, was able to degrade Remazol Brilliant Blue R and Acid Blue 25 dyes. The biological activities found in this fungus demonstrate its potential for various biotechnological applications.
