Identification of epitopes targeted following virus infection or vaccination can guide vaccine design and development of therapeutic interventions targeting functional sites,but can be ***,we employed peptide microarrays to map linear peptide epitopes(LPEs)recognized following SARS-CoV-2 infetion and *** detected by nonhuman primate(NHP)and patient Igms after SARS-CoV-2 infection extensively overlapped,localized to functionally important virus regions,and aligned with reported neutralizing antibody binding *** LPE overlap occurred atfter infection and vaccination,with LPE clusters specifc to each stimulus,where strong and conserved LPEs mapping to sites known or likely to inhibit spike protein ***-specifc LPEs tended to map to sites known or likely to be afected by structural changes induced by the proline substitutions in the mRNA vaccine's S *** LPEs to regions of known functional importance in this manner may acelerate vaccine evaluation and discovery of targets for sile secific therapeutic interventions.

The genetic alterations associated with cell transformation are in large measure expressed in the metabolic phenotype as cancer cells proliferate and change their local environment,and prepare for ***,the fundamental biochemistry of cancer cells is generally the same as in the untransformed cells,but the cancer cells produce a local environment,the TmE,that is hostile to the stromal cells,and compete for *** order to proliferate,cells need sufficient nutrients,either those that cannot be made by the cells themselves,or must be made from simpler ***,in solid tumors,the nutrient supply is often limiting given the potential for rapid proliferation,and the poor quality of the ***,cancer cells may employ a variety of strategies to obtain nutrients for survival,growth and *** much has been learned using established cell lines in standard culture conditions,it is becoming clear from in vivo metabolic studies that this can also be misleading,and which nutrients are used for energy production versus building blocks for synthesis of macromolecules can vary greatly from tumor to tumor,and even within the same *** we review the operation of metabolic networks,and how recent understanding of nutrient supply in the TmE and utilization are being revealed using stable isotope tracers in vivo as well as in vitro.

An Example Using Improved Lefschetz Duality

A theorem of Lambrechts and Stanley is used to find the rational cohomology of the complement of an embedding S^(4n-1)→ S^(2n)× S^m as a module and demonstrate that it is not necessarily determined by the map induced on cohomology by the embedding, nor is it a trivial extension. This demonstrates that the theorem is an improvement on the classical Lefschetz duality.

Current concepts on osteonecrosis of the femoral head

It is estimated that 20000 to 30000 new patients are diagnosed with osteonecrosis annually accounting for approximately 10% of the 250000 total hip arthroplasties done annually in the United States. Thelack of level 1 evidence in the literature makes it difficult to identify optimal treatment protocols to manage patients with pre-collapse avascular necrosis of the femoral head, and early intervention prior to collapse is critical to successful outcomes in joint preserving procedures. There have been a variety of traumatic and atraumatic factors that have been identified as risk factors for osteonecrosis, but the etiology and pathogenesis still remains unclear. Current osteonecrosis diagnosis is dependent upon plain anteroposterior and frog-leg lateral radiographs of the hip, followed by magnetic resonance imaging(mRI). Generally, the first radiographic changes seen by radiograph will be cystic and sclerotic changes in the femoral head. Although the diagnosis may be made by radiograph, plain radiographs are generally insufficient for early diagnosis, therefore mRI is considered the most accurate benchmark. Treatment options include pharmacologic agents such as bisphosphonates and statins, biophysical treatments, as well as joint-preserving and joint-replacing surgeries. the surgical treatment of osteonecrosis of the femoral head can be divided into two major branches: femoral head sparing procedures(FHSP) and femoral head replacement procedures(FHRP). In general, FHSP are indicated at pre-collapse stages with minimal symptoms whereas FHRP are preferred at post-collapse symptomatic stages. It is difficult to know whether any treatment modality changes the natural history of core decompression since the true natural history of core decompression has not been delineated.

The ovary is not an immunologically privileged organ, but a breakdown in tolerogenic mechanisms for ovary-specific antigens has disastrous consequences on fertility in women, and this is replicated in murine models of autoimmune disease. Isolated ovarian autoimmune disease is rare in women, likely due to the severity of the disease and the inability to transmit genetic information conferring the ovarian disease across generations. Nonetheless, autoimmune oophoritis is often observed in association with other autoimmune diseases, particularly autoimmune adrenal disease, and takes a toll on both society and individual health. Studies in mice have revealed at least two mechanisms that protect the ovary from autoimmune attack. These mechanisms include control of autoreactive T cells by thymus-derived regulatory T cells, as well as a role for the autoimmune regulator （AIRE）, a transcriptional regulator that induces expression of tissue-restricted antigens in medullary thymic epithelial cells during development of T cells. Although the latter mechanism is incompletely defined, it is well established that failure of either results in autoimmune-mediated targeting and depletion of ovarian follicles. In this review, we will address the clinical features and consequences of autoimmune-mediated ovarian infertility in women, as well as the possible mechanisms of disease as revealed by animal models.

This study investigated the in vitro fermentation profiles of human milk oligosaccharides(HmOs) by different dominating infant microbiota(***-, ***-and Bacteroides-dominated inocula). We sought to evaluate fecal microbiota composition of healthy infant donors in Guangzhou(including breast-feeding and formula-feeding). Further, we were expected to study in vitro fermentation outcomes of six commercial HmOs [i.e., 2 -fucosyllactose(2 -FL), 3-fucosyllactose(3-FL), 3 -sialyllactose(3 -SL), 6 -sialyllactose(6 -SL), lacto-Ntetraose(LNT) and lacto-N-neotetraose(LNnT)], fructo-oligosaccharide(FOS) and galacto-oligosaccharides(GOS) by using individual infant fecal inocula. methods Nineteen healthy donors were selected at Guangzhou, China. Nine infants were qualified and enrolled in the experiment. Each 50 mg of carbohydrate sample was equivalently weighted into anaerobic tubes and fermented with fecal slurry at 37 °C water bath with gentle shaking for various time periods. Total gas production of each tube was measured by inserting the needle through rubber stopper with a graduated syringe. The concentration of short chain fatty acids(SCFAs) was determined by gas chromatography, and the concentration of organic acids(OAs) was determined by high performance liquid chromatography. DNA extraction from the fermented products was performed using the OmEGA Soil DNA Kit(m5635-02)(Omega Bio-Tek, Norcross, GA, USA), following the manufacturer s instruction. The nearly full-length bacterial 16 S rRNA from each purified DNA sample was performed to investigate microbiota composition. Results Fecal microbiota composition and microbiota classification It was noteworthy that three distinct clusters of infant gut microbiota were shown among nineteen infants. Three distinct groups were characterized by the bacteria abundance, denoted as B. longum-, B. breve-and Bacteroides-dominated inocula. Using e^(+)e^(−)annihilation data corresponding to an integrated luminosity of 2.93 fb^(−1)taken at the center-of-mass energy√s=3.773 GeV with the BESIII detector,a joint amplitude analysis is performed on the decays D^(0)→π^(+)π^(−)π^(+)π^(−)and D^(0)→π^(+)π^(−)π^(0)π^(0)(non-η).The fit fractions of individual components are obtained,and large interferences among the dominant components of the decays D^(0)→a_(1)(1260)π,D^(0)→π(1300)π,D^(0)→ρ(770)ρ(770),and D^(0)→2(ππ)_(S)are observed in both *** the obtained amplitude model,the CP-even fractions of D^(0)→π^(+)π^(−)π^(+)π^(−)and D^(0)→π^(+)π^(−)π^(0)π^(0)(non-η)are determined to be(75.2±1.1_(stat).±1.5_(syst.))%and(68.9±1.5_(stat).±2.4_(syst.))%,*** branching fractions of D^(0)→π^(+)π^(−)π^(+)π^(−)and D^(0)→π^(+)π^(−)π^(0)π^(0)(non-η)are measured to be(0.688±0.010_(stat.)±0.010_(syst.))%and(0.951±0.025_(stat.)±0.021_(syst.))%,*** amplitude analysis provides an important model for the binning strategy in measuring the strong phase parameters of D^(0)→4πwhen used to determine the CKm angleγ(ϕ_(3))via the B^(−)→DK^(−)decay.

The study investigates school travel to and from schools as perceived by parents in Abu Dhabi, United Arab Emirates. Far fewer children are cycling or walking to school, and more and more are being driven to school by car. Traffic conditions, environmental factors, distance to school, road’s infrastructures, walking or biking with groups, schools efforts to educate children about active travel, and preferences of parents to accompany children to school were significantly perceived as important. The absence of active travel culture in the community was seen as a significant factor. Parent’s gender, child’s gender, child’s grade level, number of cars owned by the family, nationality and number of children were significantly associated with parents’ decisions to allow active travel to school. Results show that Abu Dhabi must establish aggressive strategic plans to promote active-commuting to school as the main mode of transportation. Current conditions in Abu Dhabi City are not conducive to active travel to school.
