High altitude is an extreme environment that imposes hypoxic pressure on physiological processes,and natives living at high altitudes are more adaptive in certain physiological *** far,epigenetic modifications under extreme changes in hypoxic pressures are relatively less ***,we recruit 32 Tibetan elite alpinists(TEAs),who have successfully mounted Everest(8848 m)at least five *** samples and physiological phenotypes of TEAs and 32 matched non-alpinist Tibetan volunteers(non-TEAs)are collected for ***-wide DNA methylation analysis identifies 23,202 differentially methylated CpGs(P_(adj)0.1)between the two *** differentially methylated CpGs are in hypoxia-related genes such as PPP1R13L,MAP3K7CL,SEPTI-9,and *** addition,Gene ontology enrichment analysis reveals several inflammation-related *** analysis indicates that 12 phenotypes are significantly different between the two *** particular,TEAs exhibit higher blood oxygen saturation levels and lower neutrophil count,platelet count,and heart *** DNA methylation association analysis,we find that two CpGs(cg16687447,cg06947206)upstream of PTEN were associated with platelet *** conclusion,extreme hypoxia exposure leads to epigenetic modifications and phenotypic alterations of TEA,providing us clues for exploring the molecular mechanism underlying changes under extreme hypoxia conditions.

Warming and precipitation are key global change factors driving soil carbon(C)dynamics in terrestrial ***,the effects of warming and altered precipitation on soil microbial diversity and functional genes involved in soil C cycling remain largely *** investigated the effects of warming and increased precipitation on soil C cycling in a temperate desert steppe of Inner Mongolia using metagenomic *** found that warming reduced plant richness,Shannon-Wiener and Simpson *** contrast,increased precipitation signifcantly infuenced Shannon-Wiener and Simpson *** reduced soil microbial species by 5.4%while increased precipitation and warming combined with increased precipitation led to increases in soil microbial species by 23.3%and 2.7%,*** relative abundance of Proteobacteria,which involve C cycling genes,was signifcantly increased by warming and increased *** signifcantly reduced the abundance of GAPDH(Calvin cycle)and celF(cellulose degradation)while it enhanced the abundance of glxR(lignin degradation).Increased precipitation signifcantly enhanced the abundance of pgk(Calvin cycle),coxL(carbon monoxide oxidation),malZ(starch degradation),and mttB(methane production).Moreover,a wide range of correlations among soil properties and C cycling functional genes was detected,suggesting the synergistic and/or antagonistic relationships under scenario of global *** results may suggest that warming is benefcial to soil C storage while increased precipitation negatively affects soil C *** fndings provide a new perspective for understanding the response of microbial communities to warming and increased precipitation in the temperate desert steppe.

Olfactory ensheathing glia promote axonal regeneration in the mammalian central nervous system,including retinal ganglion cell axonal growth through the injured optic ***,it is unknown whether olfactory ensheathing glia also have neuroprotective *** ensheathing glia express brain-derived neurotrophic factor,one of the best neuroprotectants for axotomized retinal ganglion ***,we aimed to investigate the neuroprotective capacity of olfactory ensheating glia after optic nerve *** ensheathing glia cells from an established rat immortalized clonal cell line,TEG3,were intravitreally injected in intact and axotomized retinas in syngeneic and allogeneic mode with or without microglial inhibition or immunosuppressive *** and gene expression analyses were *** bulb-derived primary olfactory ensheathing glia and TEG3 express major histocompatibility complex classⅡ*** and syngenically transplanted TEG3 cells survived in the vitreous for up to 21 days,forming an *** axotomized retinas,only the allogeneic TEG3 transplant rescued retinal ganglion cells at 7 days but not at 21 *** these retinas,microglial anatomical activation was higher than after optic nerve crush *** intact retinas,both transplants activated microglial cells and caused retinal ganglion cell death at 21 days,a loss that was higher after allotransplantation,triggered by pyroptosis and partially rescued by microglial inhibition or ***,neuroprotection of axotomized retinal ganglion cells did not improve with these *** different neuroprotective properties,different toxic effects,and different responses to microglial inhibitory treatments of olfactory ensheathing glia in the retina depending on the type of transplant highlight the importance of thorough preclinical studies to explore these variables.

Synthesis and Characterization of Two [2]Catenanes Based on Phenylene-diacetylene Crown Ethers

Two charged donor-acceptor [2]catenanes were synthesized by the cyclization of the π-acceptor 1,1 ＇-[1, 4-phenylenebis-（methylene）]bis（4,4＇-bipyridinium）bis（hexafluorophosphate）（2） and bis（bromomethyl）benzene with the templates of the π-donor phenylene-diacetylene crown ethers la and lb in the yields of 31%, 35%, respectively. The mass spectra, 1H and 13C NMR spectra confirm that the phenylene-diacetylene crown ethers, with a nonaromatic ~r-system, successfully template the cyclization of CBPQT4＋. The X-ray crystallography analysis shows that the CBPQT4＋ ring encircles around the phenylene motif of the crown ethers, leaving the 1,3-butadiyne fragment out of the cavity of CBPQT4＋ ring. The mechanically interlocked structure of [2]catenanes was stabilized by the cooperative effects of π-stacking and hydrogen bonding interactions.

Abstract Objective The incidence of hypertension in Tibet ranks highest among all Chinese provinces. This may be due to genetic changes caused by Tibet＇s unique natural environment and agrarian lifestyle, prompting us to investigated the relationship between gene polymorphisms and hypertension. Methods Blood samples were collected from 229 hypertensive participants and 372 healthy （control） participants from five Tibetan counties. Seventeen single nucleotide polymorphisms were investigated for their connection to hypertension. Results The C allele at rs2070744 of the NOS3 gene was shown to be significantly associated with hypertension （P=0.0443; OR=1.636）. Additionally, the T allele of rs4961 of the ADD gene was correlated with hypertension in women （P=0.03124; OR=1.584）. Conclusion In this study we found that the NOS3 and ADD genes were related to a high incidence of hypertension among Tibetans. N053 gene plays a role in regulating vascular tone and blood vessel diameter, which may be altered by the low-oxygen environment of Tibet. ADD is involved in water and salt metabolism, which is consistent with the high-salt diet of Tibetans. The correlations elucidated by our study were different from those of other ethnic groups, indicating that these findings may be specific to the Tibetan people.

AIM:To investigate the incidence of gastroesophageal reflux disease(GERD) and its related risk factors in Uygur and Han Chinese adult in Urumqi,***:A population-based cross-sectional survey was undertaken in a total of 972 Uygur(684 male and 288 female) aged from 24 to 61 and 1023 Han Chinese(752 male and 271 female) aged from 23 to 63 *** participants were recruited from the residents who visited hospital for health examination from November 2011 to May *** participant signed an informed consent and completed a GERD questionnaire(GerdQ) and a lifestyle-food frequency questionnaire *** whose Gerd Q score was ≥ 8 and met one of the following requirements would be enrolled into this research:(1) being diagnosed with erosive esophagitis(EE) or Barrett's esophagus(BE) by endoscopy;(2) negative manifestation under endoscopy(non-erosive reflux disease,NERD) with abnormal acid reflux revealed by 24-h esophageal pH monitoring;and(3) suffering from typical heartburn and regurgitation with positive result of proton pump inhibitor ***:According to Gerd Q scoring criteria,340 cases of Uygur and 286 cases of Han Chinese were defined as *** incidence in Uygur was significantly higher than in Han Chinese(35% vs 28%,χ2 = 11.09,P < 0.005),Gerd Q score in Uygur was higher than in Han Chinese(7.85 ± 3.1 vs 7.15 ± 2.9,P < 0.005),and Gerd Q total score in Uygur male was higher than in female(8.15 ± 2.8 vs 6.85 ± 2.5,P < 0.005).According to normalized methods,304(31%) cases of Uygur were diagnosed with GERD,including 89 cases of EE,185 cases of NERD and 30 cases of BE;256(25%) cases of Han Chinese were diagnosed with GERD,including 90 cases of EE,140 cases of NERD and 26 cases of *** incidence in Uygur was significantly higher than in Han Chinese(31% vs 25%,χ2 = 9.34,P < 0.005) while the incidences were higher in males of both groups than in females(26% vs 5% in Uygur,χ2 = 35.95,P < 0.005,and 19.8% vs 5.2% in Han,χ2 = 5.48,P < 0.02

The role of the Indonesian Throughflow （ITF） in the influence of the Indian Ocean Dipole （IOD） on ENSO is investigated using version 2 of the Parallel Ocean Program （POP2） ocean general circulation model. We demonstrate the results through sensitivity experiments on both positive and negative IOD events from observations and coupled general circulation model simulations. By shutting down the atmospheric bridge while maintaining the tropical oceanic channel, the IOD forcing is shown to influence the ENSO event in the following year, and the role of the ITF is emphasized. During positive IOD events, negative sea surface height anomalies （SSHAs） occur in the eastern Indian Ocean, indicating the existence of upwelling. These upwelling anomalies pass through the Indonesian seas and enter the western tropical Pacific, resulting in cold anomalies there. These cold temperature anomalies further propagate to the eastern equatorial Pacific, and ultimately induce a la Nifia- like mode in the following year. In contrast, during negative IOD events, positive SSHAs are established in the eastern Indian Ocean, leading to downwelling anomalies that can also propagate into the subsurface of the western Pacific Ocean and travel further eastward. These downwelling anomalies induce negative ITF transport anomalies, and an E1 Nifio-like mode in the tropical eastern Pacific Ocean that persists into the following year. The effects of negative and positive IOD events on ENSO via the ITF are symmetric. Finally, we also estimate the contribution of IOD forcing in explaining the Pacific variability associated with ENSO via ITE
