Vascular endothelial cells and oxidation reduction system play an important role in the pathogenesis of atherosclerosis(AS).If these conditions are disordered,it will inevitably lead to plaque formation and even rupture.Astragaloside IV(AsIV)and salvianolic acid B(Sal B)are the main active ingredients of Astragalus membranaceus and Salvia miltiorrhiza,respectively,and found to ameliorate vascular endothelial dysfunction and protect against oxidative stress in recent studies.However,it is still unknown if the combination of AsIV and Sal B(AsIV+Sal B)can inhibit the development of plaque through amplifying the protective effect of vascular endothelial cells and anti-oxidative stress effect.To clarify the role of AsIV+Sal B in AS,we observed the efficacy of each group(Control,Model,AsIV,Sal B,and AsIV+Sal B)by biomolecular assays,such as observing the pathological morphology of the aorta by oil red O staining,evaluating the level of oxidative stress and endothelial cells in the serum by the Elisa test,and analyzing the changes of all small molecule metabolites in liver tissue by UPLC-QTOF-MS.Results showed that AsIV,Sal B and AsIV+Sal B decreased the deposition of lipid in the arterial wall,so as to exert the effect of anti-oxidant stress and vascular endothelial protection,where the inhibitory effect of AsIV+Sal B was the most obvious.Metabonomics analysis showed that Sal B regulated the metabolic pathways of arginine and proline.AsIV regulated glycerol metabolism and saturated fatty acid biosynthesis metabolism.AsIV+Sal B is mainly related to the regulation of the citrate cycle(TCA cycle),alanine,aspartic acid,and glutamate metabolism,cysteine,and methionine metabolism.Succinic acid and methionine are synergistic metabolites that exert an enhancing effect when AsIV and Sal B were used in combination.In conclusion,we demonstrated that AsIV acompanied with Sal B can be successfully used for anti-oxidative stress and vascular endothelial protection of AS,and succinic acid and methionine are the synergistic metabolite

少数民族特色村寨是少数民族历史、地域、文化等形成和演变的缩影,是中华民族文化的有效载体,集中体现了中华文明的多元化和多样性。保护与发展少数民族特色村寨是保护中华文化多样性的重要举措,也是新时期民族工作的重要组成部分。湖南作为多民族省份,保护与发展少数民族特色村寨取得了一定成绩,积累了丰富的经验,同时也面临诸多困境。从非遗传承的视角来看,新时期要从民族特色村寨的现实需求出发,贯彻落实乡村振兴发展策略,把握发展的方向,既要重视智慧发展,也要重视文化传承,利用特色村寨的民族特色,建设美丽乡村,建立新型农村社会管理体制,提高特色村寨的建设水平。

为了解长春地区犬猫源金黄色葡萄球菌(Staphylococcu saureus,S.aureus)和伪中间型葡萄球菌(Staphylococcus pseudintermedius,SP)耐药基因和毒力基因携带情况,利用高通量测序技术对17株S.aureus与38株SP所携带的耐药基因与毒力基因进行分析,根据Kirby-Bauer纸片扩散法的结果分析耐药基因型与表型的关系,同时进行了MLST分型。结果表明S.aureus和SP对常规抗菌药物表现中介或耐药,但对呋喃妥因敏感率为100%;β-内酰胺类耐药基因bla_(Z)检出率较高,分别为100%和57.9%;各有1株同时携带bla_(Z)和bla_(NDM);S.aureus中检出2株携带磷霉素类耐药基因fosA;SP中检出21株携带多药耐药基因cfr;各检出3株携带多药外排耐药基因mdfA;共检出4株MRSA和11株MRSP;优势ST型分别为ST25和ST1430;在大多数分离菌中携带有γ-溶血素hlgB、黏附素clf、fnb、杀白细胞素LukF-PV、LukS-PV等毒力基因。分离株耐药性严重。研究结果可为兽医临床中治疗凝固酶阳性葡萄球菌感染用药提供数据支持。

The syndrome of dampness stagnancy due to spleen deficiency(DSSD)is relatively common globally.Although the pathogenesis of DSSD remains unclear,evidence has suggested that the gut microbiota might play a significant role.Radix Astragali,used as both medicine and food,exerts the effects of tonifying spleen and qi.Astragalus polysaccharide(APS)comprises a macromolecule substance extracted from the dried root of Radix Astragali,which has many pharmacological functions.However,whether APS mitigates the immune disorders underlying the DSSD syndrome via regulating gut microbiota and the relevant mechanism remains unknown.Here,we used DSSD rats induced by high-fat and low-protein(HFLP)diet plus exhaustive swimming,and found that APS of moderate molecular weight increased the body weight gain and immune organ indexes,decreased the levels of interleukin-1β(IL-1β),IL-6,and endotoxin,and suppressed the Toll-like receptor 4/nuclear factor-κB(TLR4/NF-κB)pathway.Moreover,a total of 27 critical genera were significantly enriched according to the linear discriminant analysis effect size(LEfSe).APS increased the diversity of the gut microbiota and changed its composition,such as reducing the relative abundance of Pseudoflavonifractor and Paraprevotella,and increasing that of Parasutterella,Parabacteroides,Clostridium XIVb,Oscillibacter,Butyricicoccus,and Dorea.APS also elevated the contents of short-chain fatty acids(SCFAs).Furthermore,the correlation analysis indicated that 12 critical bacteria were related to the body weight gain and immune organ indexes.In general,our study demonstrated that APS ameliorated the immune disorders in DSSD rats via modulating their gut microbiota,especially for some bacteria involving immune and inflammatory response and SCFA production,as well as the TLR4/NF-κB pathway.This study provides an insight into the function of APS as a unique potential prebiotic through exerting systemic activities in treating DSSD.

BACKGROUND Treatment of postoperative anastomotic stenosis for colorectal cancer is often challenging,especially for patients who do not respond well to endoscopy.In cases where patients have undergone an enterostomy,the stenosis can be easily resolved through magnetic compression.However,common magnetic compre-ssion techniques cannot be performed on those without enterostomy.We design-ed a novel Y–Z deformable magnetic ring(Y–Z DMR)and successfully applied it to a patient with a stenosis rectal anastomosis and without enterostomy after rectal cancer surgery.CASE SUMMARY We here report the case of a 57-year-old woman who had undergone a laparo-scopic radical rectum resection(Dixon)for rectal cancer.However,she started facing difficulty in defecation 6 months after surgery.Her colonoscopy indicated stenosis of the rectal anastomosis.Endoscopic balloon dilation was performed six times on her.However,the stenosis still showed a trend of gradual aggravation.Because the patient did not undergo an enterostomy,the conventional endoscopic magnetic compression technique could not be performed.Hence,we imple-mented a Y–Z DMR implemented through the anus under single channel.The magnetic ring fell off nine days after the operation and the rectal stenosis was relieved.The patient was followed up for six months and reported good defeca-tion.CONCLUSION The Y–Z DMR deformable magnetic ring is an excellent treatment strategy for patients with rectal stenosis and without enterostomy.

To the Editor:Varicocele(VC)is a vascular disease and considered as the main cause of male infertility.^([1])The incidence of VC in the common male population was 4.4–22.6%,of which the incidence of primary infertility was 35–40%,while the incidence of secondary infertility was as high as 80%.^([2])The exact pathophysiological mechanism of male infertility caused by VC and regulative molecules are still unclear.Clear and definite molecular markers of VC disease are helpful for early prevention and timely treatment.This study aimed to screen candidate regulative molecules playing a role in male infertility caused by VC.

Objective This study aimed to determine the current epidemiological status of PLWHA aged≥50 years in China from 2018 to 2021.It also aimed to recommend targeted interventions for the prevention and treatment of HIV/AIDS in elderly patients.Methods Data on newly reported cases of PLWHA,aged≥50 years in China from 2018 to 2021,were collected using the CRIMS.Trend tests and spatial analyses were also conducted.Results Between 2018 and 2021,237,724 HIV/AIDS cases were reported among patients aged≥50 years in China.The main transmission route was heterosexual transmission(91.24%).Commercial heterosexual transmission(CHC)was the primary mode of transmission among males,while non-marital non-CHC([NMNCHC];60.59%)was the prevalent route in women.The proportion of patients with CHC decreased over time(Z=67.716,P<0.01),while that of patients with NMNCHC increased(Z=153.05,P<0.01).The sex ratio varied among the different modes of infection,and it peaked at 17.65 for CHC.The spatial analysis indicated spatial clustering,and the high-high clustering areas were mainly distributed in the southwestern and central-southern provinces.Conclusion In China,PLWHA,aged≥50 years,were predominantly infected through heterosexual transmission.The primary modes of infection were CHC and NMNCHC.There were variations in the sex ratio among different age groups,infected through various sexual behaviors.HIV/AIDS cases exhibited spatial clustering.Based on these results,the expansion of HIV testing,treatment,and integrated behavioral interventions in high-risk populations is recommended to enhance disease detection in key regions.

Plant height influences plant architecture,lodging resistance,and yield performance.It is modulated by gibberellic acid(GA)metabolism and signaling.DELLA proteins,acting as central repressors of GA signaling,integrate various environmental and hormonal signals to regulate plant growth and development in Arabidopsis.We examined the role of two DELLA proteins,GmRGAa and GmRGAb,in soybean plant height control.Knockout of these proteins led to longer internodes and increased plant height,primarily by increasing cell elongation.GmRGAs functioned under different light conditions,including red,blue,and far-red light,to repress plant height.Interaction studies revealed that GmRGAs interacted with the blue light receptor GmCRY1b.Consistent with this,GmCRY1b partially regulated plant height via GmRGAs.Additionally,DELLA proteins were found to stabilize the protein GmSTF1/2,a key positive regulator of photomorphogenesis.This stabilization led to increased transcription of GmGA2ox-7b and subsequent reduction in plant height.This study enhances our understanding of DELLA-mediated plant height control,offering Gmrgaab mutants for soybean structure and yield optimization.

The pathogenesis of hypertrophic cardiomyopathy(HCM)is very complicated,particularly regarding the role of circular RNA(circRNA).This research pays special attention to the relationships of the circRNA-mediated network,including RNA-RNA relationships and RNA-RNA binding protein(RNA-RBP)relationships.We use the parameter framework technology proposed in this paper to screen differentially expressed circRNA,messenger RNA(mRNA),and microRNA(miRNA)from the expression profile of samples related to HCM.And 31 pairs of circRNA and mRNA relationship pairs were extracted,combined with the miRNA targeting database;145 miRNA-mRNA relationship pairs were extracted;268 circRNA-mRNA-miRNA triads were established through the common mRNA in the 2 types of relationship pairs.Thus,268 circRNA-miRNA regulatory relationships were deduced and 30 circRNARBP relationship pairs were analyzed at the protein level.On this basis,a circRNA-mediated regulatory network corresponding to the two levels of RNA-RNA and RNA-RBP was established.And then the roles of circRNA in HCM were analyzed through circRNA-mRNA,circRNA-miRNA,and circRNA-RBP,and the possible role in disease development mas inferred.
