Role of nitric oxide in biological effects of vascular endothelial growth factor

To evaluate the role of nitric oxide in the biological effects of vascular endothelial growth factor(VEGF)and the possible mechanism of VEGF,the in vitro cultured vascular endothelial cells of rabbit aorta were divided into control group,VEGF-treated group and VEGF+N-nitro-L-arginine methyl ester(L-NAME)-treated *** absorbance(A)value of vascular endothelial cells and the levels of prostaglandin(PGI2),endothelin-1(ET-1)and von Willebrand factor(vWF)in the supernatant were observed by water-soluble tetrazo-lium salt assay,radioimmunoassay and enzyme-linked immunosorbent *** A values and PGI2 in VEGF-treated group and VEGF+L-NAME-treated group were higher than those in control group(P<0.05 and P<0.01).The ET-1 and vWF were signiﬁcantly decreased in VEGF-treated group and VEGF+L-NAME-treated group com-pared with the control(P<0.05 and P<0.01).These results indicate that VEGF could promote the proliferation and secretion of PGI2 and inhibit the secretion of ET-1 and vWF in vascular endothelial cells and that L-NAME could inhibit the effect of VEGF *** oxide is an important mediator in the process of stimulating prolifera-tion and regulating secretion of vascular endothelial cells by VEGF.

The effect of the autonomic nerves on the transmural dispersion of ventricular repolarization(TDR)under acute myocardial ischemia in intact canine was *** the monophasic action potential(MAP)recording technique,MAPs of the epicardium(Epi),midmyocardium(Mid)and endocardium(Endo)were recorded simultaneously by specially designed plunge-needle electrodes at the left ventricular free wall under acute myocardial ischemia in 12 open-chest *** 90 and TDR among three myocardial layers as well as the incidence of the early afterdepolarization(EAD)before autonomic nervous stimulation and during autonomic nervous stimulation were *** was found that 10 min after acute myocardial ischemia,TDR was increased from 55±8 ms to 86±15 ms during sympathetic stimulation(P0.05).The EAD was elicited in the Mid of 2 dogs(16%)10 min after acute myocardial ischemia,but the EAD were elicited in the Mid of 7 dogs(58%)during sympathetic stimulation(Pluded that:(1)Sympathetic stimulation can increase the transmural dispersion of repolari-zation and induce early afterdepolarizations in the Mid under acute myocardial ischemia,which provide the opportunity for the ventricular arrhythmia developing;(2)Parasympathetic stimulation has no significant effect on the transmural dispersion of repolarization under myocardial ischemia.

Restrictive Effect of Puerarin on Myocardial Infarct Area in Dogs and Its Possible Mechanism

Summary: To evaluate the protective effect of puerarin on ischemic myocardium in dogs with acute myocardial infarction (AMI) and to reveal its possible mechanism, 10 dogs were randomly divided into puerarin group (group G) and control group (group C). AMI model was established in all dogs. Puerarin or saline was administered over a period of 21 days. Coronary angiography was performed before and after ligation of coronary artery. Eight hemorheological parameters were examined before and 22 days after the operation. The infarct area and vessel density of myocardium were assessed. The infarct area in group G was smaller than that in group C. Angiography 2 h and 22 d after ligation of coronary artery revealed significant augmentation of collateral vessels in group G as compared with control group. The platelet aggregation and the blood viscosity were in- creased during AMI when compared with control phase, and the increased indexes during AMI would be inhibited when puerarin were given. Capillaries and distribution vessel density in is- chemic zone on day 22 showed statistically significant augmentation in group G as compared with control group. Puerarin might improve the opening and formation of coronary collateral circula- tion, and might inhibit the increase of platelet aggregation and the blood viscosity during AMI, and thereby improve microcirculation and restrict myocardial infarct area.

Differential Effects of d, l-Sotalol and d-Sotalol on Isoproterenol-Increased Delayed Rectifier Outward Potassium Current in Guinea Pig Single Ventricular Myocytes

The aim of this study was to compare the effects of d, l-Sotalol and dSotalol on the delayed rectifier K+ outward current in the presence of isoproterenol at different concentrations. Time-dependent delayed rectifier K+ outward currents were measured in isolated guinea pig single myocytes using the whole-cell configuration of the patch-clamp technique. Currents were measured in response to 300 ms depolarizing pulses from a holding potential of -40 mV in three experimental protocols [control, isoproterenol (10^(9)mol/L - 10^(-6) mol/L ), and isoproterenol (10^(-9)mol/L - 10^(-6)mol/L ) plus either d, l-Sotalol (10^(-4) mol/L) or d-Sotalol (10^(-4) mol/L)]. IK tail currents were measured upon repolarization to -40 mV. It was found that Ik was significantly amplified in the presence. of isoproterenol (10^(-9) mol/L- 10^(-6) mol/L) plus d-Sotalol. At 10-8 mol/L isoproterenol, Ik was increased by 92. 7%±17. 1 % (Plus d, l-Sotalol (Pluded that the β-adrenergic blocking property of d, l-Sotalol but not that of dSotalol maintains the delayed rectifier K+ outward current blockade in the presence of isoproterenol in guinea pig myocytes. This might contribute to a superior antiarrhythmic efficacy as compared to d-Sotalol.

Experimental Study on the Effect of Ligustrazine in the Prevention of Intimal Proliferation of Deendothelial Artery

To evaluate the possibility of employing ligustrazine in the prevention of restenosis, the effects of ligustrazine on the intimal thickening of air injured carotid artery of rats were investigated, and the effects of ligustrazine on the proliferation of rabbit aortic median smooth muscle cells (SMCs) cultivated in vitro were examined. Artery injury model of 18 rats of about 3 months old was established by Fishman air dry method. Fourteen days after operation, the maximal artery intimal and medial thickness of the control and ligustrazine group was measured on the image analysis system. Using cell counting and thymidine ( 3H TdR) up take method, we also examined the effects of ligustrazine on the proliferation of aortic median SMC from 4 rabbits. Ligustrazine was found to inhibit the proliferation and 3H TdR up take of SMC in a dose dependent manner in vitro ( P <0.05 or P <0.01 vs control). It also inhibited the intimal thickening of rat arteries after deendothelialization. The maximal intimal thickness of ligustrazine group was much thinner than that of the control (35.9±3.8 μm vs 80.2±23.4 μm, P <0.01). It was showed that ligustrazine could be used for prevention of restenosis in clinical practice.

The mechanism of vascular endothelial growth factor (VEGF) on the prevention of restenosis after angioplasty was investigated. The cultured vascular endothelial cells (VEC) were incubated with the conditioned medium (CM) from vascular smooth muscle cells (VSMC) infected with recombinant adenoviruses containing the hVEGF 165 gene. To observe the effects of VEGF on proliferation and NO, ET, 6-keto-PGF1α secretion of VEC, WST-1 method, Griess method and radioimmunoassay were used respectively. The PDGF-B mRNA transcription in VECs was detected by RT-PCR. It was showed that NO, 6-keto-PGF1α and OD value were markedly increased in a dose-dependent manner in the VEGF-treated groups as compared with those in the control group, while ET and PDGF-B mRNA were significantly decreased in the VEGF-treated groups (P<0.05 or P<0.01). Adenovirus vector mediated hVEGF 165 gene could promote the proliferation of VECs and improve NO, PGI 2 secretion, inhibit ET secretion and PDGF-B mRNA transcription in the VECs. The above results offered further theoretical evidence for VEGF on the prevention of restenosis after angioplasty.

目的研究动物在体中层心肌细胞的电生理特性与体表心电图U波形成的关系.方法利用单相动作电位记录技术,同步记录16只兔子左心室心外膜下、中层、心内膜下心肌的单相动作电位,以下同剂量的Sotalol (1、 1.5、 2.0 mg/kg)作为工具药进行研究 .结果应用Sotalol前心电图上无U波,应用Sotalol 1.5 mg/kg后出现U波,随着Sotalol外膜相比有显著差异,心外膜下心肌的复极时程与体表心电图T波的顶点相一致,而中层心肌的复极时程与U波的终点相一致.结论 U波的产生可能是由于中层心肌复极延迟所致.

Effect of Salvia Miltiorrhiza on Coronary Collateral Circulation in Dogs with Experimental Acute Myocardial Infarction

Summary: To evaluate the effect of Salvia Miltiorrhiza on coronary collateral circulation in dogs with experimental acute myocardial infarction, 12 dogs were randomly divided into Salvia Miltiorrhiza group (group D) and control group (group C). Acute myocardial infarction model was established in all dogs, Salvia Miltiorrhiza or saline was administered to the 2 groups respectively for 21 days. Coronary angiography was performed before and after ligation of coronary artery. The capillaries and distribution vessel density in tissue sections of myocardia were assessed. Angiography 2 h and 22 days after ligation of coronary artery revealed significant augmentation of collateral vessels, and capillaries and distribution vessel density in ischemic zone on day 22 showed statistically significant augmentation in group D as compared with that of controls ( P luded that Salvia Miltiorrhiza might improve the opening and formation of coronary collateral circulation of ischemic myocardia in dogs and thereby protecting myocardia from ischemia.

Cytochrome P450 (CYP)-dependent metabolites of arachidonic acid, epoxyeicosatrienoic acids (EETs), have been suggested to be an endothelium-derived hyperpolarizing factor (EDHF) . However, the interaction or relation between EDHF and endothelial nitric oxide synthase (eNOS) is still to be elucidated. In the present study, the regulation of eNOS by endogenous EDHF is examined. The cytochrome P450 epoxygenase BM3F87V is cloned into the mammalian expression vector pCB6. Cultured bovine aortic endothelial cells (BAECs) less than 4 passages are used and transfected with BM3F87V. The effects of endogenous EETs result from BM3F87V transfection on eNOS are assessed in the endothelial cells by Western blot and Northern blot, and eNOS activity is also measured by the conversion of L-arginine to L-citrulline. Compared to transfection with the empty pCB6 vector, transfection of BAECs with BM3F87V significantly elevates the levels of eNOS protein expression, which is markedly inhibited by treatment with CYP inhibito
