OBJECTIVE:To investigate if the Liuwei Dihuang pill(LWDHP)can inhibit metastasis to the liver and lungs in mice bearing triple-negative breast cancer(TNBC),and the molecular mechanism underpinning this ***:Ninety-nine TNBC bearing-mice were distributed randomly to five groups:control(Con),paclitaxel(PTX),low-dose LWDHP(LLP,2.3 g·kg^-1·d^-1),middle-dose LWDHP(MLP,4.6 g·kg^-1·d^-1)and high-dose LWDHP(HLP,9.2 g·kg^-1·d^-1).The LWDHP were administered(p.o.)to the agonal *** morphology of BC cells was observed by hematoxylin&eosin *** of axin-2,β-catenin,T cell factor(TCF),cyclin-D1 and vascular endothelial growth factor(VEGF)was detected by western blotting or immunofluorescence.β-catenin/TCF-1 interaction was measured using a co-immunoprecipitation ***:After LWDHP treatment,metastasis of BC cells to the lungs and liver was inhibited,expression of axin-2 was increased,expression of TCF-1,β-catenin,cyclin-D1 and VEGF was decreased,andβ-catenin/TCF-1 interaction was ***:The LWDHP could inhibit metastasis of BC cells to the liver and *** molecular mechanism underlying this action may be regulation of protein expression andβ-catenin/TCF-1 interactions in the Wnt pathway.

老年绝经女性冠心病发生风险的列线图预测模型的构建及验证研究

目的:构建老年绝经女性人群发生冠心病(CHD)的预测模型,并对列线图的预测效率进行验证。方法:回顾性分析2018年3月—2019年2月在安徽省某三级甲等医院心内科就诊并接受冠状动脉(冠脉)造影的639例老年绝经女性的临床资料,根据冠脉造影结果将其分为CHD组(439例)和对照组(200例),同时将研究对象依据7∶3的比例随机分为建模组(447例)和验证组(192例)。在建模组人群中采用多因素回归分析筛选出老年绝经女性人群发生CHD的独立影响因素、构建个性化的列线图模型,并在验证组人群中对模型的预测效率和适用性进行外部验证。结果:建模组人群的多因素logistic回归分析结果表明,每日睡眠时长(OR=0.674,95%CI:0.546~0.833)是老年绝经女性人群发生CHD的独立保护因素(P<0.05),而生育个数(OR=1.798,95%CI:1.391~2.325)、超敏C反应蛋白水平(OR=1.572,95%CI:1.381~1.789)及餐后2h血糖(OR=1.197,95%CI:1.097~1.305)为老年绝经女性人群发生CHD的独立危险因素(P<0.05)。利用上述指标构建列线图预测模型,列线图模型的ROC曲线下面积在建模组及验证中分别为0.815(95%CI:0.773~0.857)、0.823(95%CI:0.755~0.890),建模组及验证组人群中的校准曲线均提示列线图的预测概率与实际发生频率之间具有较好的一致性。临床决策分析曲线提示,当老年绝经女性人群发生CHD的阈值概率处于0.18~0.94时,采用列线图的临床净获益水平最高。结论:老年绝经女性人群发生CHD的列线图预测模型预测能力和适用能力较好,可用于筛查此类人群的CHD发生风险。

Using repeated precise gps campaigns for research of present-day crustal movement and strain in the qinghai-Tibetan Plateau

The problem of crustal movement in the qing-hai-Tibetan Plateau is a hotstopic of the present-day international geodetic world. GPS observations in 1993, 1995 and 1997 are used to survey the three-dimensional displacement of GPS monitoring station in the qinghai-Tibetan Plateau, and to calculate and analyze the present-day crustal movement and strain in the qinghai-Tibetan Plateau. The result of repeated campaign shows that the crustal movement in the qinghai-Tibetan Plateau is still compressing in the north and south directions, extending in the west and east directions, and uplifting in the vertical direction. The relative convergence velocity of the Himalayan block is approximately (19.5±1.7)mm/a, the strain velocity is about (5.5±6.0) mm/a, and the uplifting velocity is some (7.6±5.2) mm/a. The compression velocity of the Tibet block relative to Golmud in the north and south directions is (9.3±4.6) mm/a, the strain velocity of the middle part in the west and east directions is (15.6±6.3) mm/a,

海上丝绸之路是东方外交史领域的一个重要研究课题。东海丝路是海上丝绸之路的一个重要组成部分,而中日丝绸贸易则是东海丝路中的核心贸易。文章从中日丝绸贸易尤其是日本丝织业发展轨迹的视角切入,回答了曾给东北亚各国带来巨大贸易红利并持续千余年的东海丝路,为何在明清之际由盛转衰,以至湮没于历史的长河中。亦即,通过对“中国丝织技术和制品的单项东输与日本丝织业的不断进步”“明朝的‘海禁’政策与日本丝织业的奋起直追”“中日‘丝银’贸易的展开与日本蚕丝业的快速赶超”三个具体问题的探讨,大跨度、纵向考察中日丝绸贸易的得失,尤其是日本丝织业从无到有、由弱变强的成功经验,并从中总结出五点启示性的结论。

Gentisate-1,2-dioxygenase（GDO） is one type of nonheme iron enzyme which belongs to the cupin *** catalyzes a number of oxidative C-C bond cleavage reactions in the biodegradation of toxic and xenobiotic compounds.A biomimetic iron（II） complex,[（hydrotris（3,5-diphenylpyrazole-1-yl）borate）FeII（hydrotris（3,5-diphenylpyrazole-1-yl）borate）],which represents a functional model of GDO was investigated by Density Function Theory（DFT）.The iron（II） 1,4-dihydroxy-2-naphthoate complex combined with dioxygen to generate the corresponding iron（III） species,and then another dioxygen will be added to form an iron（III） superoxide *** superoxide can attack the C1 carbon of the carboxysemiquinonato group to form an alkylperoxide *** p-hydroxy group on the aromatic ring passes by the O-O cleavage of the alkylperoxo intermediate to form an anhydride intermediate through ***,the metal-coordinated hydroxide participates in the hydrolysis of the anhydride species to afford the ring cleavage product.

A theoretical study for the hydrolysis mechanisms several selected substrates catalyzed by α/β-hydroylase superfamily were investigated by means of ONIOM method with a two-layered model. Three hydrolysis pathways were examined and the most favored one was determined, which result was consistent with the experimental data. The intermediate was less stable and the serine catalytic triad played an important key role. The interactions between the carboxyl groups of the substrates and NH 2 groups of residues and other substrates were discussed in detail.

Enzymes that cleave glycosidic bonds are widely investigated in carbohydrate ***,biophysical,and genetic studies suggest that these enzymes cleave glycosidic bonds by a largely dissociative mechanism generating an oxocarbenium ion *** have examined the fully mechanism of glycosidic cleavage with density functional theory by B3LYP method at 6-31g（d,p）basis *** results indicate that this reaction experiences a hemiaminal intermediate,which could lose a molecule of water directing to an imine ***,the cyclization of hemiaminal intermediate will leads to the target products after elimination of two molecules of water.

High throughput single-cell RNA-seq has been successfully implemented to dissect the cellular and molecular features underlying ***,an elaborate and comprehensive transcriptome reference of the whole blood system is ***,we profiled the transcriptomes of 7551 human blood cells representing 32 immunophenotypic cell types,including hematopoietic stem cells,progenitors and mature blood cells derived from 21 healthy *** high sequencing depth and coverage,we constructed a single-cell transcriptional atlas of blood cells(ABC) on the basis of both protein-coding genes and long noncoding RNAs(lncRNAs),and showed a high consistence between ***,putative lncRNAs and transcription factors regulating hematopoietic cell differentiation were *** common transcription factor regulatory networks were activated in neutrophils and monocytes,lymphoid cells dramatically changed their regulatory networks during ***,we showed a subset of nucleated erythrocytes actively expressing immune signals,suggesting the existence of erythroid precursors with immune ***,a web portal offering transcriptome browsing and blood cell type prediction has been ***,our work provides a transcriptional map of human blood cells at single-cell resolution,thereby offering a comprehensive reference for the exploration of physiological and pathological hematopoiesis.
