Genetic dissection of crown root traits and their relationships with aboveground agronomic traits in maize

The crown root system is the most important root component in maize at both the vegetative and reproductive stages. However, the genetic basis of maize crown root traits(CRT) is still unclear, and the relationship between CRT and aboveground agronomic traits in maize is poorly understood. In this study, an association panel including 531 elite maize inbred lines was planted to phenotype the CRT and aboveground agronomic traits in different field environments. We found that root traits were significantly and positively correlated with most aboveground agronomic traits, including flowering time, plant architecture and grain yield. Using a genome-wide association study(GWAS)coupled with resequencing, a total of 115 associated loci and 22 high-confidence candidate genes were identified for CRT. Approximately one-third of the genetic variation in crown root was co-located with 46 QTLs derived from flowering and plant architecture. Furthermore, 103 (89.6%) of 115 crown root loci were located within known domestication-and/or improvement-selective sweeps, suggesting that crown roots might experience indirect selection in maize during domestication and improvement. Furthermore, the expression of Zm00001d036901, a high-confidence candidate gene, may contribute to the phenotypic variation in maize crown roots, and Zm00001d036901 was selected during the domestication and improvement of maize. This study promotes our understanding of the genetic basis of root architecture and provides resources for genomics-enabled improvements in maize root architecture.

高宽长比移动床膜生物反应器(MBBR)的出水口与进水配水渠距离较近,易产生配水不均问题,造成短流现象,从而影响工艺处理效率。为充分发挥该MBBR的高效处理能力,以青岛市某污水厂宽长比为2.3∶1的MBBR为研究对象,通过采取调整内回流比、封堵部分出水口、调整曝气系统运行工况、降低液位使配水渠形成跌水等措施,在确保系统氧转移效率的同时,缓解了MBBR内短流现象,并通过水质测试和示踪试验进行了验证。在常规运行条件(内回流比为200%、外回流比为100%)下,MBBR的出水氨氮浓度逐渐升高,示踪试验结果证实系统内存在短流现象。当保持外回流比不变、内回流比改为100%和300%时,尽管短流现象有所改善,但综合考虑处理效果和运行能耗等因素,这两种工况均不可取。采用封堵部分出水口和调整曝气系统运行工况的解决方案后,MBBR出水横断面的氧转移效率差距减小,但进水氧转移效率差距增加,因此进一步增加降低液位使配水渠形成跌水措施来保障配水渠进水均匀性。经示踪试验验证,所采用的工艺优化措施解决了进出水配水不均问题,有效缓解了MBBR在常规运行工况下的出水短流现象。

Drought is one of the most important abiotic stresses affecting maize growth and development and therefore resulting in yield loss.Thus it is essential to understand molecular mechanisms of drought stress responses in maize for drought tolerance improvement.The root plays a critical role in plants sensing water deficit.In the present study,two maize inbred lines,H082183,a drought-tolerant line,and Lv28,a drought-sensitive line,were grown in the field and treated with different water conditions(moderate drought,severe drought,and well-watered conditions)during vegetative stage.The transcriptomes of their roots were investigated by RNA sequencing.There were 1428 and 512 drought-responsive genes(DRGs)in Lv28,688 and 3363 DRGs in H082183 under moderate drought and severe drought,respectively.A total of 31 Gene Ontology(GO)terms were significantly over-represented in the two lines,13 of which were enriched only in the DRGs of H082183.Based on results of Kyoto encyclopedia of genes and genomes(KEGG)enrichment analysis,"plant hormone signal transduction"and"starch and sucrose metabolism"were enriched in both of the two lines,while"phenylpropanoid biosynthesis"was only enriched in H082183.Further analysis revealed the different expression patterns of genes related to abscisic acid(ABA)signal pathway,trehalose biosynthesis,reactive oxygen scavenging,and transcription factors might contribute to drought tolerance in maize.Our results contribute to illustrating drought-responsive molecular mechanisms and providing gene resources for maize drought improvement.

Staggered-grid finite-difference(SGFD)schemes have been widely used in acoustic wave modeling for geophysical problems.Many improved methods are proposed to enhance the accuracy of numerical modeling.However,these methods are inevitably limited by the maximum Courant-Friedrichs-Lewy(CFL)numbers,making them unstable when modeling with large time sampling intervals or small grid spacings.To solve this problem,we extend a stable SGFD scheme by controlling SGFD dispersion relations and maximizing the maximum CFL numbers.First,to improve modeling stability,we minimize the error between the FD dispersion relation and the exact relation in the given wave-number region,and make the FD dispersion approach a given function outside the given wave-number area,thus breaking the conventional limits of the maximum CFL number.Second,to obtain high modeling accuracy,we use the SGFD scheme based on the Remez algorithm to compute the FD coefficients.In addition,the hybrid absorbing boundary condition is adopted to suppress boundary reflections and we find a suitable weighting coefficient for the proposed scheme.Theoretical derivation and numerical modeling demonstrate that the proposed scheme can maintain high accuracy in the modeling process and the value of the maximum CFL number of the proposed scheme can exceed that of the conventional SGFD scheme when adopting a small maximum effective wavenumber,indicating that the proposed scheme improves stability during the modeling.

Triclosan(TCS)has been manufactured as an antibacterial compound for half a century.Currently,it is widely used in various personal care products;however,its potential adverse effects raise a lot of attention.Here,we create a long-term oral administration mouse model and identify the corresponding hepatotoxicity of TCS.We discover that daily intragastric administration of 10 mg/kg TCS to mice for 12 weeks results in severe hepatic fibrosis.Further study displays that hepatic iron increased 18%,23%and 29%upon oral TCS treatment for4,8 and 12 weeks,respectively.Accompanied by hepatic iron variation,splenic and duodenal iron are increased,which indicates systemic iron disorder.Not only excessive iron accumulated in the liver,abnormal hepatic malondialdehyde,prostaglandin synthase 2 and glutathione peroxidase 4 are pointed to ferroptosis.Additional study uncovers that hepcidin expression increases 7%,10%,4%in serum and 2.4-,4.8-,and 2.3-fold on transcriptional levels upon TCS exposure for 4,8 and 12 weeks,individually.Taken together,the mice in the TCS-treated group show disordered systemic iron homeostasis via the upregulated hepatic hepcidin-ferroportin axis.Meanwhile,both hepatic iron overload(systemic level)and hepatocyte ferroptosis(cellular level)are accused of TCS-induced liver fibrosis.Ferriprox■,an iron scavenger,significantly ameliorates TCS-induced liver fibrosis.In summary,this study confirms the impact of TCS on liver fibrosis;a critical signal pathway is also displayed.The significance of the current study is to prompt us to reevaluate the“pros and cons”of TCS applications.

Transcriptomic profiling of sorghum leaves and roots responsive to drought stress at the seedling stage

Drought stress affects the growth and productivity of crop plants including sorghum.To study the molecular basis of drought tolerance in sorghum,we conducted the transcriptomic profiling of sorghum leaves and roots under drought stress using RNA-Seq method.A total of 510,559,and 3 687 differentially expressed genes(DEGs)in leaves,3 368,5 093,and 4 635 DEGs in roots responding to mild drought,severe drought,and re-watering treatments were identified,respectively.Among them,190 common DEGs in leaves and 1 644 common DEGs in roots were responsive to mild drought,severe drought,and re-watering environment.Gene Ontology(GO)enrichment analysis revealed that the GO categories related to drought tolerance include terms related to response to stimulus especially response to water deprivation,abscisic acid stimulus,and reactive oxygen species.The major transcription factor genes responsive to drought stress include heat stress transcription factor(HSF),ethylene-responsive transcription factor(ERF),Petunia NAM,Arabidopsis ATAF1/2 and CUC2(NAC),WRKY transcription factor(WRKY),homeodomain leucine zipper transcription factor(HD-ZIP),basic helix-loop-helix transcription factor(bHLH),and V-myb myeloblastosis viral oncogene homolog transcription facotr(MYB).Functional protein genes for heat shock protein(HSPs),late-embryogenesis-abundant protein(LEAs),chaperones,aquaporins,and expansins might play important roles in sorghum drought tolerance.Moreover,the genomic regions enriched with HSP,expansin,and aquaporin genes responsive to drought stress could be used as powerful targets for improvement of drought tolerance in sorghum and other cereals.Overall,our results provide a genome-wide analysis of DEGs in sorghum leaves and roots under mild drought,severe drought,and re-watering environments.This study contributes to a better understanding of the molecular basis of drought tolerance of sorghum and can be useful for crop improvement.

Manipulating strain mode and degree that can be applied to epitaxial complex oxide thin films have been a cornerstone of strain engineering.In recent years,lift-off and transfer technology of the epitaxial oxide thin films have been developed that enabled the integration of heterostructures without the limitation of material types and crystal orientations.Moreover,twisted integration would provide a more interesting strategy in artificial magnetoelectric heterostructures.A specific twist angle between the ferroelectric and ferromagnetic oxide layers corresponds to the distinct strain regulation modes in the magnetoelectric coupling process,which could provide some insight in to the physical phenomena.In this work,the La_(0.67)Sr_(0.33)MnO_(3)(001)/0.7Pb(Mg_(1/3)Nb_(2/3))O_(3)-0.3PbTiO_(3)(011)(LSMO/PMN-PT)heterostructures with 45.and 0.twist angles were assembled via water-etching and transfer process.The transferred LSMO films exhibit a fourfold magnetic anisotropy with easy axis along LSMO.A coexistence of uniaxial and fourfold magnetic anisotropy with LSMO[110]easy axis is observed for the 45°Sample by applying a 7.2 kV cm^(−1)electrical field,significantly different from a uniaxial anisotropy with LSMO[100]easy axis for the 0°Sample.The fitting of the ferromagnetic resonance field reveals that the strain coupling generated by the 45°twist angle causes different lattice distortion of LSMO,thereby enhancing both the fourfold and uniaxial anisotropy.This work confirms the twisting degrees of freedom for magnetoelectric coupling and opens opportunities for fabricating artificial magnetoelectric heterostructures.

In the past 5 years,there has been significant research interest in the intrinsic magnetic topological insulator family compounds MnBi2+2nTe4+3n（where n=0,1,2…）.In particular,exfoliated thin films of MnBi2Te4have led to numerous experimental breakthroughs,such as the quantum anomalous Hall effect,axion insulator phase and high-Chern number quantum Hall effect without Landau levels.However,despite extensive efforts,the energy gap of the topological surface states due to exchange magnetic coupling,which is a key feature of the characteristic band structure of the system,remains experimentally elusive.The electronic structure measured by using angle-resolved photoemission （ARPES） shows significant deviation from ab initio prediction and scanning tunneling spectroscopy measurements,making it challenging to understand the transport results based on the electronic structure.This paper reviews the measurements of the band structure of MnBi2+2nTe4+3nmagnetic topological insulators using ARPES,focusing on the evolution of their electronic structures with temperature,surface and bulk doping and film thickness.The aim of the review is to construct a unified picture of the electronic structure of MnBi2+2nTe4+3ncompounds and explore possible control of their topological properties.
