Protective effects of Wuwei Xiaodu Drink against chronic osteomyelitis through Foxp3^(+)CD25^(+)CD4^(+) Treg cells via the IL-2/STAT5 signaling pathway

To explore the effectiveness and safety of a Chinese medicinal decoction Wuwei Xiaodu Drink(WWXDD)in inhibiting chronic osteomyelitis via regulatory T cells signaling.The effective constitutes of WWXDD and osteomyelitis related genes were screened.Target proteins were cross-validated using the Venny database.GO function and KEGG pathway analysis were performed for target proteins,while pharmacological network was constructed.The bone properties were analyzed by HE staining and the concentrations of immune factors were measured by ELISA.The expression of CTLA-4 and Foxp3 mRNA and STAT5,p-STAT5,CTLA-4 and Foxp3 protein were detected using Real-time PCR and Western blot,respectively.FACS was used to analyze the percentages of cells.A total of 117 genes overlapped between 785 target genes of the active compounds of WWXDD and 912 osteomyelitis related genes.Inflammation-related genes,including IL-6,TNFα,IL-1βand IL-2 showed high connection degree in the drug-compound-disease-target network.GO function and KEGG pathway analysis revealed that 117 intersection genes mainly enriched in virus infection related pathways,immune related pathways and chemokine signaling pathway.Furthermore,the development of chronic osteomyelitis was suppressed in model rats after treatment with WWXDD.Meanwhile,the concentrations of IL-2 and CD4^(+)CD25^(+)Foxp3 Treg perr centages together with the levels of p-STAT5,CTLA-4 and Foxp3 were also down-regulated.Furthermore,IL-2 and WWXDD drug-containing serum exhibited opposite effects on regulating IL-2,IL-10,TGF-β1,Foxp3,CTLA4 and STAT5.In addition,a STAT5 phosphorylation inhibitor suppressed the expression of Foxp3 and CTLA-4.WWXDD can treat chronic osteomyelitis through suppressing the main regulating factors of Tregs and interfere its immunodepression.Our results bring a new solution for chronic osteomyelitis.

Fluorophores play a significant role in achieving high-contrast imaging in complex physiological environment.Herein,we present the results of our studies on the development of various fluorophores,such as anti-solvatochromic fluorophores,long wavelength emission two-photon fluorophores,large Stokes shift fluorophores and solid-state fluorophores,to achieve high-contrast bioimaging in physiological environments.in this study,we have primarily focused on the design,optical performance,imaging mechanisms,and the applications of these fluorophores with respect to complex physiological conditions.

Annealing atmosphere-dependent capacitive energy storage

Electrostatic capacitors based on dielectrics with high energy density and efficiency are desired for modern electrical systems owing to their intrinsic fast charging-discharging speed and excellent reliability.The longstanding bottleneck is their relatively small energy density.Herein,we report enhanced energy density and efficiency in the Aurivillius Pb_(2)Bi_(4)Ti_(5)O_(18)films by controlling the post-annealing atmosphere.The results demonstrate that the fabrication atmosphere has significant effects on the film texture and defects.As the increase of the oxygen pressure of annealing atmosphere,the Pb_(2)Bi_(4)Ti_(5)O_(18)films show a preferred growth orientation of(00l)and fewer defects,which leads to a higher polarization and breakdown field for the film annealed in air atmosphere and thus help to achieve an ultrahigh energy density of59.4 J·cm^(-3)and an improved efficiency of 81.2%.Moreover,the film also exhibits excellent cycling reliability and good thermal stability.The Pb_(2)Bi_(4)Ti_(5)O_(18)films show a significant potential application for dielectric capacitors.

A controllable self-localized imaging strategy capable of synchronous in situ tracking of local changes in intracellular bioactive small-molecules

In situ tracking and localization of ubiquitous bioactive small molecules(BSMs)within their native habitats is particularly challenging because of their low-molecular weight and widespread distribution properties.We report the proof of concept of a synchronous in situ imaging strategy,whereby the representative BSM amino-biothiols(ABs)mediate activation of the selflocalizable probe HYPQS,thereby releasing insoluble emissive precipitates to afford holistic distribution information of ABs.Notably,three organelle-targetable ABs inhibitors were innovatively fabricated for directed clearance of ABs in particular organelles,providing a powerful aid for HYPQS to achieve programmed in situ tracking of ABs in different organelles“on demand”.Biological transmission electron microscopy images confirmed that this probe released insoluble emissive precipitates at the reaction sites,which is of primary importance for achieving synchronous in situ tracking of BSMs.Furthermore,the probe HYPQS was successfully applied to monitor the dynamic changes in the endogenous ABs pool during diverse cell events.This strategy opens a promising avenue for investigating the undiscovered functional mechanism of local BSMs in relevant biological processes.

An Integration Strategy to Develop Dual-State Luminophores with Tunable Spectra,Large Stokes Shift,and Activatable Fluorescence for High-Contrast Imaging

Developing dual-state luminophores(DSLs)with strong fluorescence in both the monomer and aggregate states is urgently needed but remains a huge challenge because most current luminophores are either aggregation-induced emission or aggregation-caused quenching molecules.Moreover,limited by the structural conservation of the few existing DSLs,there are not enough response sites that can be used to customize various activatable fluorescent probes for specific molecular imaging.Herein,we engineered a general integration strategy for the fabrication of such DSLs with excellent photophysical properties.The DSLs,with their tunable spectra,a large Stokes shift(>170 nm),and achievable near-infrared(NIR)emission,show great potential for high-contrast imaging.Importantly,DSLs can be used as a universal platform for probe customization due to their activatable fluorescence through protection-deprotection of the phenolic hydroxyl group.Based on this,an NIR fluorescent probe DSL-Gal was developed for sensing of β-galactosidase in solutions,senescent cells,and liver metastases with high contrast,further confirming the superiority and universal feasibility of DSLs in probe design.The integration strategy may provide a novel approach for the generation of other DSLs and have great potential applications in bioimaging.

Quantifying the carbon source of pedogenic calcite veins in weathered limestone: implications for the terrestrial carbon cycle

The continent is the second largest carbon sink on Earth’s surface.With the diversification of vascular land plants in the late Paleozoic,terrestrial organic carbon burial is represented by massive coal formation,while the development of soil profiles would account for both organic and inorganic carbon burial.As compared with soil organic carbon,inorganic carbon burial,collectively known as the soil carbonate,would have a greater impact on the long-term carbon cycle.Soil carbonate would have multiple carbon sources,including dissolution of host calcareous rocks,dissolved inorganic carbon from freshwater,and oxidation of organic matter,but the host calcareous rock dissolution would not cause atmospheric CO2drawdown.Thus,to evaluate the potential effect of soil carbonate formation on the atmospheric p CO2level,different carbon sources of soil carbonate should be quantitatively differentiated.In this study,we analyzed the carbon and magnesium isotopes of pedogenic calcite veins developed in a heavily weathered outcrop,consisting of limestone of the early Paleogene Guanzhuang Group in North China.Based on the C and Mg isotope data,we developed a numerical model to quantify the carbon source of calcite veins.The modeling results indicate that4–37 wt%of carbon in these calcite veins was derived from atmospheric CO2.The low contribution from atmospheric CO2might be attributed to the host limestone that might have diluted the atmospheric CO2sink.Nevertheless,taking this value into consideration,it is estimated that soil carbonate formation would lower 1 ppm atmospheric CO2within 2000 years,i.e.,soil carbonate alone would sequester all atmospheric CO2within 1 million years.Finally,our study suggests the C–Mg isotope system might be a better tool in quantifying the carbon source of soil carbonate.

Multiple sclerosis(MS)is a chronic and incurable autoimmune disease of the central nervous system(CNS)that results in neuro-inflammation and demyelination.Current therapies for MS alleviate symptoms to some extent but fail to realize myelin regeneration.Hence,it is crucial to identify effective targets to promote remyelination.As a widely expressed transcriptional regulator,peroxisome proliferatoractivated receptor-coactivator-1α(PGC-1α)expresses in different cells with high energy consumption,including skeletal muscle cells,brown fat cells,neurons,astrocytes,and oligodendrocyte precursor cell(OPC)[1].Targeting PGC-1αmay be beneficial for CNS development and various neurodegenerative diseases.

Statistical description of depth-dependent turbulent velocity measured in Taihu Lake, China

Quantitative description of turbulence using simple physical/mathematical models remains a challenge in classical physics and hydrologic dynamics. This study monitored the turbulence velocity field at the surface and bottom of Taihu Lake, in China, a large shallow lake with a heterogeneous complex system, and conducted a statistical analysis of the data for the local turbulent structure. Results show that the measured turbulent flows with finite Reynolds numbers exhibit properties of non-Gaussian distribution. Compared with the normal distribution, the Levy distribution with meaningful parameters can better characterize the tailing behavior of the measured turbulence. Exit-distance statistics and multiscaling extended self-similarity(ESS) were used to interpret turbulence dynamics with different scale structures. Results show that the probability density function of the reverse structure distance and the multiscaling ESS can effectively capture the turbulent flow dynamics varying with water depth. These results provide an approach for quantitatively analyzing multiscale turbulence in large natural lakes.

Enhanced electrochemical performance of Si/C electrode through surface modification using SrF_(2) particle

The silicon-based material exhibits a high theoretical specific capacity and is one of the best anode for the next generation of advanced lithium-ion batteries(LIBs).However,it is difficult for the silicon-based anode to form a stable solid-state interphase(SEI)during Li alloy/de-alloy process due to the large volume change(up to 300%)between silicon and Li4.4Si,which seriously limits the cycle life of the LIBs.Herein,we use strontium fluoride(SrF_(2))particle to coat the silicon-carbon(Si/C)electrode(SrF_(2)@Si/C)to help forming a stable and high mechanical strength SEI by spontaneously embedding the SrF_(2) particle into SEI.Meanwhile the formed SEI can inhibit the volume expansion of the silicon-carbon anode during the cycle.The electrochemical test results show that the cycle performance and the ionic conductivity of the SrF_(2)@Si/C anode has been significantly improved.The X-ray photoelectron spectroscopy(XPS)analysis reveals that there are fewer electrolyte decomposition products formed on the surface of the SrF_(2)@Si/C anode.This study provides a facile approach to overcome the problems of Si/C electrode during the electrochemical cycling,which will be beneficial to the industrial application of silicon-based anode materials.

Highly efficient electroreduction of CO_(2) by defect single-atomic Ni-N_(3) sites anchored on ordered micro-macroporous carbons

Microporous supports typically fail to fully expose active sites for electrolytes and CO_(2) molecules,and this usually results in low current density for the electrocatalytic CO_(2) reduction reaction(CO_(2)RR).To overcome the biggest obstacle and facilitate commercial applications,defective single-atomic Ni-N_(3) sites anchored to ordered micro-macroporous N-doped carbon(Ni-N/OMC)have been prepared by the pyrolysis of the Ni-ZIF-8@PS(ZIF=zeolitic imidazolate framework)and are intended to provide enhanced CO_(2)RR with a current density at an industrial level.This Ni-ZIF-8@PS is constructed of nickel-based ZIF-8 embedded in the three-dimensional(3D)highly ordered polystyrene spheres(PS).The 3D ordered micro-macroporous architecture of Ni-N/OMC could facilitate the mass transfer of substrates to the accessible defective single-atomic Ni-N_(3) sites through micropores(0.6 nm)and macropores(~200 nm)interconnected by 50 nm channels.In a flow cell,Ni-N/OMC exhibits almost 100.0%CO Faraday efficiency(FECO)between−0.2 and−1.1 V vs.RHE and an industrial level CO partial current density of 208 mA cm^(−2).It has a turnover frequency of 1.5×10^(5) h^(−1) at−1.1 V vs.RHE in 1 M KOH electrolyte,which exceeds that of most reported nickel-based electrocatalysts.This excellent CO_(2)RR performance for Ni-N/OMC makes it a state-of-the-art electrocatalyst for CO_(2)RR.Theoretical calculations show that the defective Ni-N_(3) site can lower the energy of*COOH formation compared with that of the Ni-N4 site,thereby accelerating CO_(2)RR.Ni-N/OMC can also be utilized as a cathodic catalyst in Zn-CO_(2) battery,exhibiting high CO selectivity in the discharge process and excellent stability.This work paves a pathway to rational design of highly efficient electrocatalysts with 3D hierarchically ordered micro-macroporous architecture for CO_(2)RR towards industrial production and commercial applications.
