The Effects of Graded Abomasal Glucose Infusion on Milk Yield and Composition in Lactating Goats

Four non-pregnant multiparous lactating goats were used in a 4 × 4 latin square experiment in the present study. The four treatments were abomasal infusion of 0, 50, 100, and 150 g d^-1 glucose. The results showed that graded glucose infusion increased milk yield （P 〈 0.01）. The highest milk yield was observed when 50 g d^-1 glucose was infused. An infusion of glucose at 100 or 150 g d^-1 failed to increase milk yield further and milk yield of the 100 g d ^-1 glucose infusion treatment was even significantly lower than that of the 50 g d^-1 treatment （P 〈 0.01）. Though milk fat content was gradually decreased by graded glucose infusions and milk fat yield of the 100 and 150 g d 1 treatments were lower than that of the control （P 〈 0.01）; milk fat yield of the 50 g d i treatment was not depressed and was higher than that of the control （P 〈 0.01）. Graded glucose infusion did not change the contents of milk protein and non-fatty solids. Yields of milk protein （P 〈 0.05）, lactose （P 〈 0.01）, non-fat solids （P 〈 0.01）, and lactose content （P 〈 0.01） of the 50 g d^-1 treatment were higher than those of the control but had no difference among the other three treatments. Contents of those de novo synthesized fatty acids in milk fat remained unchanged or increased and those preformed fatty acids remained unchanged or decreased by graded glucose infusion. Glucose infusion decreased the total yields of C16 ＋ C16：1 and 〉 C16 fatty acids and had no influence on the yield of C4-C14：1 fatty acids （P 〈 0.05）. Results of the present study implied that increased glucose supply had positive effects on the production of milk, milk protein, milk fat, and lactose only when glucose was a limiting factor for milk synthesis. Excessive glucose supply had no further beneficial effects and might depress milk fat synthesis.

The infiltration casting method is widely employed for the preparation of ex-situ composite ***,the production of composite materials using this method must necessitates a comprehensive understanding of the wettability and interface characteristics between the reinforcing phase and the bulk metallic glasses(BMGs).This work optimized the composition of Zr-based BMGs through microalloying methods,resulting in a new set of Zr-based BMGs with excellent glass-forming *** experiments between the Zr-based BMGs melts and W substrates were conducted using the traditional sessile drop method,and the interfaces were characterized utilizing a scanning electron microscope(SEM)equipped with energy dispersive X-ray spectroscopy(EDS).The work demonstrates that the microalloying method substantially enhances the wettability of the Zr-based BMGs ***,the incorporation of Nb element impedes the formation of W-Zr phases,but the introduction of Nb element does not alter the extent of interdiffusion between the constituent elements of the amorphous matrix and W element,indicating that the influence of Nb element on the diffusion of individual elements is minute.

Influence of deformation on precipitation in AZ80 magnesium alloy

Precipitates in the conventionally processed （solution treatment followed by aging） AZ80 alloy are coarse, cellular, and incoherent. They nucleate and grow on the basal planes of the matrix or distribute discontinuously in the alloy. Their unique morphology and undesired distribution make them ineffective for precipitation strengthening. This condition, however, can be modified by applying selected deforma-tion and heat treatment conditions. The effect of deformation and heat treatment on the morphology and distribution of precipitates has been studied. Deformation was introduced by hot extrusion, cold rolling, or equal channel angular pressing （ECAP）. The microstructures were characterized using scanning electron microscopy, transmission electron microscopy, and X-ray diffraction. The results showed that cold de-formation improved precipitation more significantly than hot deformation, and twinning promoted precipitation more effectively than slip. When ECAP was applied, the Bc-route induced more precipitates than the A-route.

Effect of loureirin A against Candida albicans biofilms

Loureirin A is a major active component of Draconis sanguis, a traditional Chinese medicine. This work aimed to investigate the activity of loureirin A against Candida albicans biofilms. 2, 3-Bis-(2-methoxy-4-nitro-5-sulfophenyl)-2 H-tetrazolium-5-carboxanilide(XTT) reduction assay and scanning electron microscopy were used to investigate the anti-biofilm *** inhibitory concentration testing and time-kill curve assay were used to evaluate fungicidal activity. Cell surface hydrophobicity(CSH) assay and hyphal formation experiment were respectively carried out to investigate adhesion and morphological transition,two virulence traits of C. albicans. Real-time RT-PCR was used to investigate gene expression. Galleria mellonella-C. albicans and Caenorhabditis elegans-C. albicans infection models were used to evaluate the in-vivo antifungal effect. Human umbilical vein endothelial cells and C. elegans nematodes were used to evaluate the toxicity of loureirin A. Our data indicated that loureirin A had a significant effect on inhibiting C. albicans biofilms, decreasing CSH, and suppressing hyphal formation. Consistently, loureirin A down-regulated the expression of some adhesion-related genes and hypha/biofilm-related genes. Moreover, loureirin A prolonged the survival of Galleria mellonella and Caenorhabditis elegans in C. albicans infection models and exhibited low toxicity. Collectively,loureirin A inhibits fungal biofilms, and this effect may be associated with the suppression of pathogenic traits, adhesion and hyphal formation.

Crystalline Silica Promotes Rat Fibrocyte Differentiation in Vitro,and Fibrocytes Participate in Silicosis in Vivo

Objective The aim of this study was to investigate the effects of Si O2 on fibrocytes and whether fibrocytes participate in silicosis in vivo. Methods A macrophagocyte（AM）/fibrocyte coculture system was established, and AMs were treated with 100 μg/mL SiO 2. Flow cytometry was used to detect the number of fibrocytes. Real‐time PCR was performed to measure the expression of collagen I, collagen III, and α‐SMA mR na. The levels of collagen I, collagen III, and TGF‐β1 protein were determined by EliSA. Immunohistochemical staining was performed to measure α‐SMA protein expression. A rat silicosis model was induced by intratracheal instillation of SiO 2. Lung histopathological evaluation was conducted using HE and Masson＇s trichrome staining after 1 and 9 weeks. The number of fibrocytes in peripheral blood or lung tissue of rat was detected by flow cytometry. Double‐color immunofluorescence was applied to identify fibrocytes in the lung tissue. Results Peripheral blood monocytes were found to differentiate into fibrocytes in vitro in a time‐dependent manner, and exposure to crystalline silica might potentiate fibrocyte differentiation. In addition, fibrocytes were able to migrate from peripheral blood to the lung tissue, and the number of fibrocytes was increased after SiO 2 exposure. Conclusion Silica exposure potentiates fibrocyte differentiation, and fibrocytes may participate in silicosis in vivo.

Development and characterization of new allohexaploid resistant to web blotch in peanut

Peanut diseases seriously threaten peanut production, creating disease-resistant materials via interspecific hybridization is an effective way to deal with this problem. In this study, the embryo of an interspecific F1 hybrid was obtained by crossing the Silihong(Slh) cultivar with Arachis duranensis(ZW55), a diploid wild species. Seedlings were generated by embryo rescue and tissue culture. A true interspecific hybrid was then confirmed by cytological methods and molecular markers. After treating seedlings with colchicine during in vitro multiplication, the established interspecific F1 hybrid produced seeds which were named as Am1210. With oligonucleotide fluorescence in situ hybridization(Oligo FISH), molecular marker evaluations, morphological and web blotch resistance characterization, we found that: 1) Am1210 was an allohexaploid between Slh and ZW55;2) the traits of spreading lateral branches, single-seeded or double-seeded pods and red seed coats were observed to be dominant compared to the erect type, multiple-seeded pods and brown seed coats;3) the web blotch resistance of Am1210 was significantly improved than that of Slh, indicating the contribution of the web blotch resistance from the wild parent A. duranensis. In addition, 69 dominant and co-dominant molecular markers were developed which could be both used to verify the hybrid in this study and to identify translocation or introgression lines with A. duranensis chromosome fragments in future studies as well.
