Mesenchymal stem cells transduced by PLEGFP-N1 retroviral vector maintain their biological features and differentiation

Background Enhanced green fluorescent protein （EGFP） has been an important reporter gene for gene therapy. Human mesenchymal stem cells （hMSCs） are ideal target cells in cell transplantation and tissue engineering. We investigated their biological characteristics and differentiation mediated by PLEGFP-N1 retroviral transduction.Methods hMSCs were isolated from human bone marrow by density gradient fractionation and adherence to plastic flasks. Individual colonies were selected and cultured in tissue dishes. Packaging cells PT67 were transfected by PLEGFP-N1 retroviral vector , and hMSCs were transduced by viral supernatant infection. Meanwhile, hMSCs-EGFP were identified by immune phenotypes and whether it could differentiate into osteoblasts or adipocytes under conditioned media was investigated.Results The rate of stably transduced hMSCs-EGFP was up to 96% after being screened by G418. hMSCs-EGFP exhibited fibroblast-like morphological features. Flow cytometric analyses showed that hMSCs-EGFP were positive for CD73, CD105, CD166, CD90 and CD44, but negative for CD34 and CD45. In addition, it could functionally be induced into osteocytes or adipocytes under conditioned media. These biological features of hMSCs-EGFP were consistent with those of hMSCs.Conclusions hMSCs transduced by PLEGFP-N1 retroviral vector can be used in vivo securely because they can maintain their biological characteristics and differentiation. It is a simple and reliable way to trace the changes of hMSCs in vivo by EGFP during cell transplantation and gene therapy.

Fabricating low-strain and fast-charging silicon-carbon composite anodes is highly desired but remains a huge challenge for lithium-ion ***,we report a unique silicon-carbon composite fabricated by uniformly dis-persing amorphous Si nanodots(SiNDs)in carbon nanospheres(SiNDs/C)that are welded on the wall of the macroporous carbon framework(MPCF)by vertical graphene(VG),labeled as MPCF@VG@SiNDs/*** high dispersity and amor-phous features of ultrasmall SiNDs(~0.7 nm),the flexible and directed electron/li+transport channels of VG,and the MPCF impart the MPCF@VG@SiNDs/C more lithium storage sites,rapid li+transport path,and unique low-strain property during li+***,the MPCF@VG@SiNDs/C exhibits high cycle stability(1301.4 mAh g^(-1) at 1 A g^(-1) after 1000 cycles without apparent decay)and high rate capacity(910.3 mAh g^(-1),20 A g^(-1))in half cells based on industrial electrode *** assembled pouch full cell delivers a high energy density(1694.0 Wh L^(-1);602.8 Wh kg^(-1))and an excellent fast-charging capability(498.5 Wh kg^(-1),charging for 16.8 min at 3 C).This study opens new possibilities for preparing advanced silicon-carbon com-posite anodes for practical applications.

The vortex generator(VG)and its well-known effect in flow optimization are widely studied and employed across different engineering ***,while the same working principles of VG may be well suited for the applications on surface-cleaning technologies,such promising potential is hardly,if any,explored in the published ***,in the present study,the influence on flow-induced particle resuspension brought by a rectangular vG in a channel flow is investigated with the help of highfidelity computational fluid dynamics *** increases of particle removal forces and resuspension rates are discovered in long,strip-like regions with reduced boundary-layer thickness resulted from the vG-induced vortices,and the enhancement effect is especially significant for configurations with the VG installed at a greater angle of *** is also shown that while the resuspension enhancements on the lower and the upper surfaces of the channel exhibit distinct statistical characteristics,having a VG in the channel improves the overall particle-removing capability of the channel flow by introducing higher surface-averaged removal forces and particle resuspension *** but not least,the increase of resuspension rate is especially significant for the smaller,micron-scale particles which are otherwise hardly disturbed by a VG-less channel flow,and such resuspension-enhancement effectgenerally subsides with increasing particle size.

The fabrication of poly（2,6-pyridinedicarboxylic acid）/MWNTs modified glass electrode（PPDA/MWNTs/GCE） was proposed and used for individual or simultaneous determination of guanine and *** performances of the PPDA/MWNTs/GCE were characterized with cyclic voltammetry（CV）.The modified electrode exhibited enhanced electrocatalytic behavior and good stability for the detection of guanine and *** pulse voltammetry（DPV） was used to determine the concentration of guanine,*** detection limit（S/N = 3） for guanine and adenine was 0.045μmol/L and 0.05μmol/L,*** electrochemical method for the measurement of guanine and adenine in calf thymus DNA was also developed with this modified electrode and the result was satisfactory.

BACKGROUND： The abnormal posture and motor pattern have not stabilized in children with cerebral palsy at early period, thus timely treatment can establish normal postural reflex and motor pattern, and prevent complications of muscle contracture, ankylosis, skeletal deformity, etc. The clinical factors affecting the rehabilitative efficacy of gross motor function in children with cerebral palsy should be observed. OBJECTIVE： To observe the effects of therapeutic occasion, grading of gross motor function and developmental level on the rehabilitative efficacy in children with cerebral palsy. DESIGN： A case-controlled analysis. SETTING： Qilu Children＇s Hospital of Shandong University. PARTICIPANTS： Totally 138 children with cerebral palsy, who were hospitalized for 12 months in the Rehabilitation Center of Qilu Children＇s Hospital, Shandong University, were selected from April 2004 to September 2006, and all the children were diagnosed to be accorded with the standard set by the national seminar on cerebral palsy in 2004. There were 97 males and 41 females, including 55 cases of 0-2 years old, 47 cases of 2-4 years and 36 cases of 4-6 years. Informed contents were obtained from relatives of all the children. METHODS： ① Comprehensive rehabilitation treatment： Vojta method was to induce the children to turn over the body and crawl by stimulating reflective turn over and crawling on belly. Bobath method including trainings of head control, turning over body, keeping sitting position, keeping balance, crawling, keeping standing position, and walking, etc.; The children were massaged by using the maneuvers of push, press, rub, pull, wave, etc. according to the sites and types of palsy. Acupuncture was performed mainly at bilateral motor areas, the needle was retained for 1 hour per time, 6 days continuously every week, and followed by a 1-day interval. ② Prognosis assessment： The gross motor functional grading of the children with cerebral palsy at admission was asses

Background Heme oxygenase-1 （HO-1） can be induced by inflammatory cytokines,oxidation,ischemia,hypoxia,and *** a ＂graft survival protective gene,＂ HO-1 is a hot spot in organ transplantation ***,the role of HO-1 gene expression in the function of human colon adenocarcinoma cell line （Caco-2） cells has not been reported *** The role of HO-1 in the proliferation and migration of Caco-2 cells was analyzed using a stable HO-1 expression *** constructed a recombinant adeno-associated virus plasmid containing the HO-1 gene,heme oxygenase 1 （HMOX1）,which was transfected into Caco-2 intestinal *** identified a number of target genes by global microarray analysis combined with real-time polymerase chain reaction （PCR） and chromatin immunoprecipitation *** Our results showed that significant HO-1 upregulation was demonstrated in the Caco-2 cells after HO-1 *** of HO-1 expression promoted proliferation and invasion in *** CTNND1 gene,a member of the armadillo protein family,was identified as a direct HO-1 target *** Overexpression of HO-1 promotes Caco-2 cell proliferation and migration by targeting the CTNND1 gene.

Increase in permeability of renewable energy sources(RESs)leads to the prominent problem of voltage stability in power system,so it is urgent to have a system strength evaluation method with both accuracy and practicability to control its access scale within a reasonable ***,a hybrid intelligence enhancement method is proposed by combining the advantages of mechanism method and data driven ***,calculation of critical short circuit ratio(CSCR)is set as the direction of intelligent enhancement by taking the multiple renewable energy station short circuit ratio as the quantitative ***,the construction process of CSCR dataset is proposed,and a batch simulation program of samples is developed accordingly,which provides a data basis for subsequent ***,a multi-task learning model based on progressive layered extraction is used to simultaneously predict CSCR of each RESs connection point,which significantly reduces evaluation error caused by weak *** performance and anti-noise performance of the proposed method are verified on the CEPRI-FS-102 bus system,which provides strong technical support for real-time monitoring of system strength.
