Chinese soft-shelled turtles （Trionyx sinens） in culture farms using an artificial warming system in Zhejiang, China, often show typical signs of white-spot disease such as white spots on their bodies, skin lesions, anorexia and eventually death. The sick turtles were mostly 5-80 g in weight. A suspected fungal pathogen was isolated from the sick turtles and verified as Paecilomyces lilacinus by sequence analysis of the internal transcribed spacer （ITS） of its ribosomal DNA （rDNA）. Detailed morphological examinations were also conducted to confirm the white-spot disease.

采用高温固相法成功制备出新型Ca_(5-x)Ga_(6)O_(14):xSm^(3+)应力发光材料。通过X射线衍射、扫描电镜、漫发射光谱、光致激发和发射光谱、荧光衰减曲线、应力发光光谱和热释光光谱等测试详细研究了Ca_(5-x)Ga_(6)O_(14):xSm^(3+)的晶体结构、表面形貌、光致发光和应力发光性能及其发光机理。在404 nm激发下,Ca_(5-x)Ga_(6)O_(14):xSm^(3+)呈现出4个发射峰,分别位于562,599,642,715 nm,对应Sm^(3+)的^(4)G_(5/2)→6Hj(j=5/2,7/2,9/2,11/2)的特征发射。随着Sm^(3+)离子掺杂浓度的增加,发光强度先增强后减弱,在x=0.07时获得最强发射,且衰减时间从1.92 ms缩短至1.30 ms。在滑动摩擦激发下可获得Ca_(5-x)Ga_(6)O_(14):xSm^(3+)的应力发光发射带,且应力发光强度与施加应力满足线性增长,表明该材料在应力传感领域具有潜在应用价值。

利用X射线CT技术研究根系三维构型的进展

【目的】X射线CT扫描具有非破坏性、高分辨率等特点,近年来被广泛应用于研究植物根系三维构型。本文介绍了X射线CT技术研究植物根系的原理和方法,并重点阐述了CT图像提取和定量根系的方法。【主要进展】X射线CT可以穿透土体进而获取土体三维结构,并能通过图像分析准确定量土体内部的细节特征(如根系、孔隙等)。目前根系结构研究中应用的CT主要有医用CT、工业显微CT、同步辐射CT等,分别用于研究不同大小和分辨率要求的样品。应用CT扫描根系样品时,样品的制备和扫描参数的选择非常重要,样品的尺寸、容器的材质、扫描功率和扫描时长等因素都会影响CT图像质量。CT图像中提取根系的方法包括全局阈值法、区域生长法、根系追踪法、深度学习法等,结合图像预处理和后处理能更完整地提取根系。利用ImageJ、Rootrak、RooTh等软件,可以定量根系形态学参数(体积、根长、直径等),还可以分析根系三维分布特征(根系生长角度、根系生长深度等)。【展望】从CT图像中提取根系的算法在实际应用中仍存在很多问题,开发提取根系的先进算法,准确获取根系三维构型并将其定量表征,仍将是CT技术用于根系研究的重要方向。由于CT设备性能的特点,扫描样品尺寸和图像分辨率的矛盾仍然是关键问题,X射线CT硬件设备的创新和发展对其在根系研究中更加广泛和有效的应用具有重要意义。

A low-pathogenicity isolate of listeria monocytogenes from cow’s milk,as screened in mouseand chicken embryonated egg models,was examined for virulence-related phenotypic *** genes (iap,prfA,plcA,hly,mpl,actA,plcB,InlA and InlB) were compared with *** strains 10403S and EGD to elucidate the possible molecular mechanisms of low ***-though *** H4 exhibited similar patterns to strain 10403S in terms of hemolytic activity,invitro growth and invasiveness and even had higher adhesiveness,faster intracellular growth and higherphospholipase activity in vitro,it was substantially less virulent than the strain 10403S in mouse and chickenembryo models (50% lethal dose:10vs.10and 10vs.10,respectively).The genes prfA,plcA andmpl were homologous among *** strains H4,10403S and EGD (>98%).Genes iap,hly,plcB,InlA and InIB of *** 10403S had higher homology to those of strain EGD (>98%) than *** homology of the gene hly between strain 10403S and isolate H4 was 96.9% at the nucleotide level,but 98.7% at the amino acid *** actA gene of isolate H4 had deletions of 105 nucleotides correspondingto 35 amino acid deletions falling Within the proline-rich *** together,this study presents someclues as to reduced virulence to mice and chicken embryos of the isolate H4 probably as a result of deletionmutations of actA.

A one-step dual flow immunochromatographic assay （DICGA）, based on a competitive format, was de- veloped for simultaneous quantification of ochratoxin A （OTA） and zearalenone （ZEN） in corn, wheat, and feed sam- ples. The limit of detection for OTA was 0.32 ng/ml with a detection range of 0.53-12.16 ng/ml, while for ZEN it was 0.58 ng/ml with a detection range of 1.06-39.72 ng/ml. The recovery rates in corn, wheat, and feed samples ranged from 77.3% to 106.3% with the coefficient of variation lower than 15%. Naturally contaminated corn, wheat, and feed samples were analyzed using both DICGA and liquid chromatography-tandem mass spectrometry （LC-MS/MS） and the correlation between the two methods was evaluated using a regression analysis. The DICGA method shows great potential for simple, rapid, sensitive, and cost-effective quantitative detection of OTA and ZEN in food safety control.

Porcine circovirus type 2（PCV2）has recently been reported to elicit the unfolded protein response（UPR）via activation of the PERK/e IF2α（RNA-activated protein kinase-like endoplasmic reticulum（ER）kinase/eukaryotic initiation factor 2α）*** study attempted to examine which viral protein might be involved in inducing UPR and whether this cellular event would lead to apoptosis of the cells expressing the viral *** transient expression,we found that both replicase（Rep）and capsid（Cap）proteins of PCV2 could induce ER stress as shown by increased phosphorylation of PERK with subsequent activation of the eI F2α-ATF4（activating transcription factor 4）-CHOP（CCAAT/enhancer-binding protein homologous protein）*** expression,but not Rep,significantly reduced antiapoptotic B-cell lymphoma-2（Bcl-2）and increased caspase-3 cleavage,possibly due to increased expression of *** knockdown of PERK by RNA interference clearly reduced Cap-induced CHOP expression,caspase-3cleavage,and apoptotic cell death possibly by partially rescuing Bcl-2 expression,we propose that there is connection between Cap-induced UPR and apoptosis via the PERK/eI F2α/ATF4/CHOP/Bcl-2 *** study,together with our earlier studies,provides insight into the mechanisms underlying PCV2 pathogenesis.

Porous titanium scaffold with suitable porous architecture exhibits enormous potentials for bone defect ***,insufficient osteointegration and osteoinduction still remain to open as one of the major problems to achieve satisfactory therapeutic *** solve this problem,many studies have been carried out to improve the bioactivity of porous titanium scaff old by surface *** this study,porous Ti6Al4V scaff olds were fabricated using additive manufacturing *** architectures were built up based on a diamond pore structure ***–acid-heat(AH)treatment was applied to create a TiO2 layer on the porous Ti6Al4V scaff old(AH-porous Ti6Al4V).Subsequently,a hydrothermal treatment was employed to enable the formation of HA coating with nanopillar-like morphology on the alkali–acid-heat-treated surface(HT/AH-porous Ti6Al4V).The effects of surface modifications on apatite-forming ability,protein adsorption,cell attachment,cell proliferation and osteogenic gene expression were studied using apatite-forming ability test,protein adsorption assay and in vitro cell culture *** was found that the HT/AH-porous Ti6Al4V exhibited the highest apatite formation ability and best affinity to fibronectin and *** vitro studies indicated that the mesenchymal stem cells(MSCs)cultured on the HT/AH-porous Ti6Al4V presented improved adhesion and differentiation compared with the porous Ti6Al4V and AH-porous Ti6Al4V.

目的探讨镰形棘豆总黄酮对四氯化碳诱发的肝纤维化(HF)大鼠的作用,以及对肝转化生长因子(TGF)-β/Smad信号通路的影响。方法将48只雄性大鼠随机分为正常组(腹腔注射花生油,灌胃给予0.9%NaCl)、模型组(腹腔注射40%CCl4花生油溶液诱导HF模型+灌胃给予0.9%NaCl)、阳性对照组(HF模型+灌胃秋水仙碱0.2 mg·kg^(-1))和低、中、高剂量实验组(HF模型+分别灌胃镰形棘豆总黄酮提取物100、200、400 mg·kg^(-1)),每组8只,连续4周。用苏木精-伊红(HE)、Masson染色方法观察组织病理变化;检测血清肝功能、肝纤维化水平;检测血清炎症因子水平,用荧光定量聚合酶链反应(RT-qPCR)法测定肝中基因的表达水平。结果模型组大鼠肝组织病理损伤严重,大量炎症因子和胶原纤维堆积,而其余给药组均有不同程度缓解。正常组、模型组、阳性对照组和低、中、高剂量实验组的谷丙转氨酶水平分别为(49.28±12.44)、(5885.42±948.37)、(4454.60±489.27)、(4650.47±843.53)、(3761.75±887.30)和(3544.90±1066.75)μg·L^(-1),谷草转氨酶水平分别为(186.90±46.89)、(5936.23±793.81)、(3971.37±780.28)、(4360.30±863.35)、(3943.10±439.47)和(3971.38±631.08)μg·L^(-1),透明质酸水平分别为(45.08±17.16)、(104.32±36.06)、(66.83±20.09)、(70.30±21.07)、(60.00±9.68)和(59.02±10.73)μg·L^(-1),层粘连蛋白水平分别为(23.13±3.89)、(60.85±13.66)、(35.67±9.92)、(39.98±9.39)、(36.55±12.21)和(34.68±24.83)μg·L^(-1),Ⅲ型前胶原水平分别为(24.98±5.34)、(82.58±30.14)、(40.70±16.14)、(51.08±23.21)、(43.60±12.48)、(44.20±11.66)μg·L^(-1),白细胞介素(IL)-1β水平分别为(37.63±1.24)、(46.10±3.23)、(39.22±2.36)、(41.33±0.93)、(40.25±2.04)和(39.18±2.23)pg·mL^(-1),肿瘤坏死因子-α水平分别为(314.58±20.56)、(383.71±16.97)、(349.00±7.93)、(348.88±25.11)、(325.75±27.84)和(335.07±21.33)pg·mL^(-1),TGF-β1 mRNA相对表达水平分别为1.00±0.00、60.99±15.70、9.61±1.59、7.37±1.09、6.41±0.64和6.87±1.09,Smad7 mRNA相对表达水平分别为1.00±0.00、0.34±0.05、0.21±0.03、0.35±0.02、0.38±0.02和0.42±0.03。模型组的上述指标与正常组比较,中、高剂量实验组的上述指标与模型组比较,在统计学上差异均有统计学意义(P<0.05,P<0.01,P<0.001)。结论镰形棘豆总黄酮对CCl4诱导的大鼠肝纤维化具有保护作用,其机制可能与调节TGF-β/Smad通路有关。
