B-cell lymphoma-2 phosphorylation at Ser70 site-related autophagy mediates puerarin-inhibited the apoptosis of MC3T3-E1 cells during osteoblastogenesis

OBJECTIVE:To explore the relationship between autophagy and apoptosis regulated by puerarin during osteoblastogenesis.METHODS:In this study,the effects of puerarin on the autophagic activity and apoptosis level of osteoblast precursors(MC3T3-E1 cells)was observed.Subsequently,the roles of puerarin on B-cell lymphoma-2(Bcl-2)phosphorylation at different sites in osteoblast precursors were observed.The effect of puerarin on the interaction between Bcl-2 and autophagy regulatory molecule or pro-apoptotic molecule was also investigated using Co-immunoprecipitation assays.In addition,the effect of puerarin on mitochondrial membrane potential of osteoblast precursors was also identified by mitochondrial membrane potential fluorescence probe assays.RESULTS:Our results showed that puerarin can promote the autophagic activity and apoptosis level of MC3T3-E1cells.In addition,puerarin promoted Bcl-2phosphorylation at Ser70 site,and the dissociation of Bcl-2-Beclin1 complex.Moreover,puerarin could enhance the binding of Bcl-2-Bcl-2-Associated X(Bax)complex in MC3T3-E1 cells.Furthermore,puerarin increased the mitochondrial membrane potential of MC3T3-E1 cells.CONCLUSIONS:Therefore,puerarin promotes Beclin1into autophagy flux through Bcl-2 phosphorylation at Ser70,thereby enhancing autophagy of osteoblast precursors,which mediates its anti-apoptotic role during osteoblastogenesis.Furthermore,the dissociation of Bcl-2-Beclin1 complex is conducive to the binding of Bcl-2-Bax complex,which resists the apoptosis of osteoblast precursors via the increased mitochondrial membrane potential.

Boosting Hydrogen Storage Performance of MgH_(2) by Oxygen Vacancy-Rich H-V_(2)O_(5) Nanosheet as an Excited H-Pump

MgH_(2) is a promising high-capacity solid-state hydrogen storage material,while its application is greatly hindered by the high desorption temperature and sluggish kinetics.Herein,intertwined 2D oxygen vacancy-rich V_(2)O_(5) nanosheets(H-V_(2)O_(5))are specifically designed and used as catalysts to improve the hydrogen storage properties of MgH_(2).The as-prepared MgH_(2)-H-V_(2)O_(5) composites exhibit low desorption temperatures(Tonset=185℃)with a hydrogen capacity of 6.54 wt%,fast kinetics(Ea=84.55±1.37 kJ mol^(-1) H_(2) for desorption),and long cycling stability.Impressively,hydrogen absorption can be achieved at a temperature as low as 30℃ with a capacity of 2.38 wt%within 60 min.Moreover,the composites maintain a capacity retention rate of~99%after 100 cycles at 275℃.Experimental studies and theoretical calculations demonstrate that the in-situ formed VH_(2)/V catalysts,unique 2D structure of H-V_(2)O_(5) nanosheets,and abundant oxygen vacancies positively contribute to the improved hydrogen sorption properties.Notably,the existence of oxygen vacancies plays a double role,which could not only directly accelerate the hydrogen ab/de-sorption rate of MgH_(2),but also indirectly affect the activity of the catalytic phase VH_(2)/V,thereby further boosting the hydrogen storage performance of MgH_(2).This work highlights an oxygen vacancy excited“hydrogen pump”effect of VH_(2)/V on the hydrogen sorption of Mg/MgH_(2).The strategy developed here may pave a new way toward the development of oxygen vacancy-rich transition metal oxides catalyzed hydride systems.

An extended numerical model of the first exothermic peak for three dimensional printed cement-based materials

The first exothermic peak of cement-based material occurs a few minutes after mixing,and the properties of three dimensional(3D)printed concrete,such as setting time,are very sensitive to this.Against this background,based on the classical Park cement exothermic model of hydration,we propose and construct a numerical model of the first exothermic peak,taking into account the proportions of C_(3)S,C_(3)A and quicklime in particular.The calculated parameters are calibrated by means of relevant published exothermic test data.It is found that this developed model offers a good simulation of the first exothermic peak of hydration for C_(3)S and C_(3)A proportions from 0 to 100% of cement clinker and reflects the effect of quicklime content at 8%-10%.The unique value of this research is provision of an important computational tool for applications that are sensitive to the first exothermic peak of hydration,such as 3D printing.

Genesis of ultra-deep dolostone and controlling factors of largescale reservoir:A case study of the Sinian Dengying Formation and the Cambrian Longwangmiao Formation in the Sichuan Basin

This paper investigates the origin of ultra-deep dolostone and the factors influencing large-scale dolostone reservoirs,focusing on the Sinian Dengying Formation and the Cambrian Longwangmiao Formation in the Sichuan Basin.The study involves petrology,microscale X-ray diffraction,trace element analysis,and C-O-Sr-Mg isotope experiments to provide a detailed analysis.The research findings indicate that the Dengying and Longwangmiao formations comprise six types of matrix dolostone and four types of cement.The Dengying Formation,which developed under a sedimentary background of a restricted platform,contains special microbial and microcrystalline dolostones.The dolomite grains are small(line,and calcite dolostones with coarser and more ordered crystals exhibit similar δ^(13)C and^(87)Sr/^(86)Sr values to microbial and microcrystalline dolostone.During the sedimentary period of the Dengying Formation,ancient marine conditions were favorable for microbial survival.Microorganisms induced the direct precipitation of primary dolomite in seawater,forming microbial and microcrystalline dolostones during the seawater diagenesis period.During the subsequent diagenesis period,dolostones underwent the effects of dissolution-recrystallization,structures,and hydrothermal fluids.This resulted in the formation of dolostone with coarser crystals,a higher degree of order,and various types of cement.The Longwangmiao Formation was developed in an interplatform beach characterized by special particle dolostone.The particle dolostone has a large grain size(>30μm),high order degree(Min=0.7),small unit cell parameters,high Na content(Max=432 ppm),and low Fe and Mn content.The δ^(2

In this work,a new method to solve the Reynolds equation including mass-conserving cavitation by using the physics informed neural networks(PINNs)is proposed.The complementarity relationship between the pressure and the void fraction is used.There are several difficulties in problem solving,and the solutions are provided.Firstly,the difficulty for considering the pressure inequality constraint by PINNs is solved by transferring it into one equality constraint without introducing error.While the void fraction inequality constraint is considered by using the hard constraint with the max-min function.Secondly,to avoid the fluctuation of the boundary value problems,the hard constraint method is also utilized to apply the boundary pressure values and the corresponding functions are provided.Lastly,for avoiding the trivial solution the limitation for the mean value of the void fraction is applied.The results are validated against existing data,and both the incompressible and compressible lubricant are considered.Good agreement can be found for both the domain and domain boundaries.

BACKGROUND Ulcer colitis(UC)is a chronic,nonspecific,and noninfectious inflammatory bowel disease.Recently,Toll-like receptors(TLRs)have been found to be closely associated with clinical inflammatory diseases.Achieving complete remission in patients with intermittent periods of activity followed by dormancy is challenging.Moreover,no study has explored the mechanism by which Kuicolong-yu enema decoction retains traditional Chinese medicine enemas to attenuate the inflammatory response in UC.AIM To explore the mechanism by which Kuicolong-yu enema decoction retains traditional Chinese medicine enemas to attenuate the inflammatory response in UC.METHODS This prospective clinical study included patients who met the exclusion criteria in 2020 and 2021.The patients with UC were divided into two groups(control and experimental).The peripheral blood of the experimental and control groups were collected under aseptic conditions.The expression of TLR4 protein,NF-κB,IL-6,and IL-17 was detected in the peripheral blood of patients in the experimental group and control group before and 1 month after taking the drug.linear co rrelation analysis was used to analyze the relationship between the expression level of TLR4 protein and the expression levels of downstream signal NF-κB and inflammatory factors IL-6 and IL-17,and P<0.05 was considered statistically significant.RESULTS There were no significant differences in the patient characteristics between the control and experimental groups.The results showed that the expression levels of TLR4 and NF-κB in the experimental group were significantly lower than those in the control group(P<0.05).The levels of IL-6 and IL-17 in the experimental group were significantly lower than those in the control group(P<0.05).The TLR4 protein expression in the experimental group was positively correlated with the expression level of downstream signal NF-κB and was positively correlated with the levels of downstream inflammatory cytokines IL-6 and IL-17(r=0.823,P<0.05).CONCLUSION Kuicolong-yu enema

Efficacy and safety of Chinese herbal medicine as adjunctive therapy in sepsis patients with bloodstream infection:a propensity-matched analysis

OBJECTIVE:To investigate the efficacy and safety of Chinese herbal medicine in treating sepsis patients with bloodstream infection.METHODS:A 6-year retrospective study was carried out at a university hospital in China.Adult sepsis patients with bloodstream infection were included.The primary outcome was 28-day mortality after admission.Propensity score method was used to adjust for possible confounding.28-day mortality was estimated by KaplanMeier analysis and compared using the log-rank test.Cox regression analysis was carried out to identify factors impacting in-hospital mortality outcomes.RESULTS:Following the application of the propensity score method,a total of 176 patients were included.The all-cause 28-day mortality in the control group and Chinese herbal medicine group was 21.6%and 14.8%,respectively.Kaplan-Meier survival analysis showed that Chinese herbal medicine was associated with a lower hazard ratio(HR)in all-cause 28-day death compared with the control group[HR=0.44,95%CI(0.22,0.90),Plications were similar between the two groups(P>0.05).Blood-activating and stasis-eliminating herb administration was associated with reduced inhospital mortality among sepsis patients with bloodstream infection[HR=0.54,95%CI(0.34,0.94),Pliminating herb,might have certain efficacy and safety in treating sepsis patients with bloodstream infection.Clinicians should prescribe blood-activating and stasis-eliminating herb in treating these two coalescent critical diseases as long as no contraindications exist.However,further studies are needed to validate our results.

A comprehensive comparison of different regression techniques and nature-inspired optimization algorithms to predict carbonation depth of recycled aggregate concrete

The utilization of recycled aggregates(RA)for concrete production has the potential to offer substantial environmental and economic advantages.However,RA concrete is plagued with considerable durability concerns,particularly carbonation.To advance the application of RA concrete,the establishment of a reliable model for predicting the carbonation is needed.On the one hand,concrete carbonation is a long and slow process and thus consumes a lot of time and energy to monitor.On the other hand,carbonation is influenced by many factors and is hard to predict.Regarding this,this paper proposes the use of machine learning techniques to establish accurate prediction models for the carbonation depth(CD)of RA concrete.Three types of regression techniques and meta-heuristic algorithms were employed to provide more alternative predictive tools.It was found that the best prediction performance was obtained from extreme gradient boosting-multi-universe optimizer(XGB-MVO)with R^(2) value of 0.9949 and 0.9398 for training and testing sets,respectively.XGB-MVO was used for evaluating physical laws of carbonation and it was found that the developed XGB-MVO model could provide reasonable predictions when new data were investigated.It also showed better generalization capabilities when compared with different models in the literature.Overall,this paper emphasizes the need for sustainable solutions in the construction industry to reduce its environmental impact and contribute to sustainable and low-carbon economies.

Observation and verification of surface electrical explosion driven by radial-distributed pulsed current in laboratory lightning strike test

The laboratory lightning test is essential for assessing the effectiveness of lightning strike protection(LSP).Particularly,direct lightning strike damage can be performed with pulsed current injection into the specimen.This paper focuses on the dynamic process of arc plasma and shock wave behaviour in the vicinity of the‘strike’point.A rod-plate discharge load is built for testing aluminium and coated plate under 40-kA-level pulsed current.The visualisation of the luminous discharge plasma and its flow field via high-speed photography(from different angles)is meticulously designed and implemented,synchronised with electro-physical diagnostics.The results indicate some new mechanisms for lightning strike damage,apart from the impulse heat loading from the thermal arc.The transient current injection through the arc root concentrates on a thin skin layer(skin-depth effect),with the radial-attenuated current density,driving asynchronously electrical explosions on the plate surface.The inhomogeneous Joule heating of the plate leads to outwardly propagating phase transition and shock wave along the conductive surface.In addition,the electro-thermal instability is observed and regarded as the seed of irregular erosion region.Spectroscopic information reveals two different plasma states of main discharge arc channel and adjacent surface electrical explosion.The correspondence of the physical mechanism of electrical explosion and optical radiation is established.Microscopic images for different regions depict erosion characteristics and summarise influencing factors,further confirming the mechanism above.The research clarifies the role of skin-depth effect in transaction arc erosion for electrode,complements the electrical explosion theory with unevenly distributed current and helps optimise strategies of LSP.

A Deep Mantle Source for the Late Neoarchean Metamorphosed Basalts in Eastern Hebei,North China Craton:Insights from Whole-Rock Geochemistry and Sm-Nd Isotopes,and Zircon U-Pb-Hf Isotopes

The basalts within the greenstone belt worldwide serve as an ideal target to decipher the nature of Archean mantle sources and further to extend the understanding of the early stages of Earth's evolution.To provide important insights into the issues,we carried out a detailed investigation of whole-rock geochemistry and Sm-Nd isotopes,and zircon U-Pb-Hf isotopes for the Late Neoarchean metamorphosed basalts in eastern Hebei,North China Craton.U-Pb isotopic dating using the LA-ICPMS on zircons reveals that the basalts in eastern Hebei erupted at ca.2.48-2.51 Ga and subsequently experienced multiple regional metamorphic events at 2477 and 1798 Ma,respectively.The metamorphosed basalts are featured by low SiO_(2),MgO,K_(2)O+Na_(2)O,and high Fe O contents,endowed with the subalkaline and high-Fe tholeiitic affinities.The radiogenic initial Nd and Hf isotope values and correlations among V,Ni and Cr contents strongly imply that the basalts experienced significant clinopyroxene and olivine fractionation and minor crustal contamination during magma evolution.They are also characterized by the relatively low total REE contents and exhibit significant depletions to moderate enrichments in the LREE contents,indicating the derivation from a deep mantle source in an Archean proto-mantle plume setting.
